2.Systematic Review of Jinkui Shenqi Pills on Hypertension

Objective To evaluate the efficacy and safety of Jinkui Shenqi Pills on hypertension through Meta-analysis and systematic review. Methods Articles about intervention of modified Jinkui Shenqi Pills in hypertensive clinical randomized controlled trials were retrieved from CNKI, Wanfang database, VIP, Cochrane Library, PubMed, and Embase. They were all searched from inception to June 2016. Cochrane system evaluation manual bias risk assessment method was used to evaluate the quality of the articles, and RevMan5.3 software was used for Meta-analysis. Results Nine articles involving 776 cases were included in the study. Meta-analysis showed that the total effective rate of modified Jinkui Shenqi Pills was better than that of conventional Western medicine treatment [OR=2.67, 95%CI (1.73, 4.11), P<0.000 01], and combination therapy was superior to conventional Western medicine alone [OR=3.27, 95%CI (1.99, 5.39), P<0.000 01]. However, the efficacy of modified Jinkui Shenqi Pills alone was equivalent to the conventional Western medicine alone [OR=1.16, 95%CI (0.43, 3.09), P=0.77). Conclusion Modified Jinkui Shenqi Pills can improve the clinical symptoms of the patients with hypertension, and combination treatment can improve the antihypertensive effect. However, the quality of the included research literature is too low, yet more large-scale, multi-center, high-quality clinical randomized controlled trials are needed to further verify.
3.Neuroprotective effect of fucoidan against 6-OHDA-induced damage and its mechanism on MN9D cell line

Objective To study the neuroprotective effect of fucoidan against 6-OHDA-induced damage on MN9D cell line and preliminarily explore its mechanism.Methods The MN9D cells were treated with different concentrations of 6-OHDA(12.5,25,50,100 and 200 μmol/L)for 24 h and with 50 μmol/L 6-OHDA for different time courses(6,12,24 and 48 h)as well.Based on the above experiment,the best concentration(50 μmol/L)of 6-OHDA and time course(24 h)were selected and the damaged model of MN9D cell line was induced accordingly.The MN9D cells were pretreated with 0.01,0.1 and 1.0 mg/mL fucoidan for 1 h,and then co-treated with 50 μmol/L 6-OHDA for 24 h.The viability of MN9D cells was detected by MTT assay;the LDH leakage was detected by bioassay;the level of intracellular xidative stress was observed by 2',7'-dichlorodihydrofluorescein diacetate assay.Results MTT metabolic value decreased following the increase of 6-OHDA concentration or the extension of processing time.Treatment with 50 μmol/L 6-OHDA for 24 h induced a significant decrease of MTT metabolic rate and an increase of LDH leakage rate in the MN9D cells.Pretreatment with 0.1 and 1.0 mg/mL fucoidan inhibited the death of 6-OHDA-induced cells,increased the MTT metabolic value and decreased the LDH leakage.Cell morphological changes were consistent with the biochemical experiments.Treatment with 50 μmol/L 6-OHDA for 8 h could significantly increased the generation of reactive oxygen species induced by 6-OHDA,while pretreatment with 1.0 mg/mL fucoidan for 1 h showed antagonism on that.Conclusion Fucoidan can protect MN9D cells from the death induced by 6-OHDA and the underlying mechanism may be involved in the antioxidant capacity of fucoidan.
4.Changes of bilirubin, SOD, IL-1, IL-6 and WBC count before and after Guasha treatment in rats

Objective To study experimentally the mechanism of Guasha therapy in curing diseases. Methods All Wistar rats (n=40) were randomly divided into the normal group, group with 0.5 hour after Guasha (0.5 h group), group with 6 hours after Guasha (6 h group) and group with 12 hours after Guasha (12 h group). The arterial blood samples were collected and given blood routine examination. The levels of bilirubin, superoxide dismutase (SOD), interlukin- 1 (IL-1) and IL-6 were detected respectively . Results In the 6 h group the WBC count decreased significantly and the level of the total bilirubin increased significantly (P<0.05) compared with those in the normal group. The level of SOD decreased significantly in the 12 h group compared with that in the normal group (P<0.05).[WTHZ] Conclusion The Guasha therapy can inhibit the inflammatory reaction of WBC and raise the level of the total bilirubin , which may be its one of the mechanisms for abating of fever and curing diseases.
5.Application of high quality of nursing service in psychiatric ward

Objective To explore the effect of high quality of nursing service on increasing the life quality of patient, confidence, self-esteem and patient satisfaction in psychiatric ward .Methods Forty-eight patients in hospital in the department of psychiatry were chosen , were carried out the high quality of nursing service, that is the concept of “take the patient as the center” by way of holistic nursing, and were fully implemented the responsibility nursing in basic nursing , disease observation, treatment nursing, rehabilitation training of drug and health education .The life quality and satisfaction of patients were evaluated by quality of life scale, self esteem scale and satisfaction questionnaire before and after the implementation of high quality of nursing service activities.Results The score of psychological and social subscale in the quality of life scale was (27.33 ±22.33) three months after the implementation of high quality of nursing service activities , and was lower than (30.21 ±23.80) before the implementation, and the difference was statistically significant (t =2.054, P<0.05).The score of the power and energy subscale was (33.54 ±18.12) three months after the implementation, and lower than (35.86 ±18.03) before the implementation, and the difference was statistically significant (t=2.033, P<0.05).The score of self-esteem scale was (25.85 ±11.27) three months after the implementation, and higher than ( 23.94 ±10.88 ) before the implementation, and the difference was statistically significant (t=2.236, P<0.05).The score of high quality of service satisfaction questionnaire was (13.79 ±3.41) three months after the implementation , and higher than ( 11.79 ±4.64 ) before the implementation, and the difference was statistically significant (t =2.582, P <0.05).Conclusions The development of high quality nursing service activities can improve the patients ’ quality of life in the department of psychiatry, enhance their self-esteem and confidence to overcome the disease , and increase patient satisfaction.
6.Improvement of cardiac function by recombinant adenovirus Ad-hBNP in rats with chronic heart failure

Objective To evaluate the therapeutic effect of hBNP on rats with chronic heart failure (CHF). Methods Thirty CHF rats defined by echocardiography at 12 weeks post abdominal aortic constriction were randomly divided into Ad-hBNP group (2. 5 × 1010 VP/ml NS Ad-hBNP 1 ml/week ×4,n = 14), Ad-Track group ( n = 8 ), placebo group ( NS, n = 8 ), 10 sham-operated rats served as control group. After 4 weeks treatment, cardiac function was evaluated by echocardiography and hemodynamic measurements. Heart weight (HW) and HW/body weight (BW) ratio were determined. Results IVS,LVPW, LVEDD and LVESD were significantly reduced in the Ad-hBNP group [(2. 34 ±0. 29)mm, (2. 28 ± 0. 18)mm, (6. 50 ±0. 42)mm, (3.54 ±0. 59) mm] than those in the Ad-Track group[(2. 71 ±0. 35) mm,(3.02 ±0.85)mm, (7.71 ±0.83)mm, (4.72 ±0.80)mm] and in the NS group [(2.78 ±0.23)mm,(2. 83 ± 0. 53 ) mm, (7. 34 ± 0. 97 ) mm, (4. 55 ± 0. 77 ) mm, all P < 0. 05]. The LVEF and LVFS of the Ad-hBNP group [(79. 27 ±7.01 )%, (43.38 ±6. 73)%] were significantly higher than in the Ad-Track group[(70.85±4.81)%, (35.72 ±3.68)%] and in the NS group[(69.67 ±6.90)%, (34.91 ±5.10)%, all P <0. 01]. HR[(417.48 ±32. 57) beats/min, (446. 85 ±61.49) beats/min, P <0. 05;(440. 83 ±32. 18) beats/min , P <0. 05], LVEDP[( - 4. 24 ±4. 00) mm Hg( 1 mm Hg =0. 133 kPa);(21.99 ±6. 80) mm Hg, P <0. 01; ( 18.00 ± 12. 25)mm Hg, P<0. 01] were significantly decreased and while LVSP[(131.79 ±15.76) mm Hg; (112.99 ±32.35) mm Hg, P<0.05; (117.13 ±15.26)mmHg], +dP/dtmax[(5037.20 ±430.41) mm Hg/s; (4217.40 ± 1354. 15)mm Hg/s, P <0.05;(4310. 50 ± 1293.97 ) mm Hg/s, P < 0. 05] and - dP/dtmax [( - 4382. 00 ± 1304. 79 ) mm Hg/s;(-3725.00±791.34) mm Hg/s, P < 0.05; ( - 3890.00 ± 1043.73) mm Hg/s, P < 0.05] were significantly increased in Ad-hBNP group than in Ad-Track group and NS group ( all P < 0. 05 ). HW and HW/BW were also decreased in Ad-hBNP group than in the Ad-Track group and the NS group. Conclusion Exogenous hBNP improved the cardiac function and attenuated remodeling in CHF rats.
7.Validation study of chronic kidney disease epidemiology collaboration predictive equations based on serum creatinine and cystatin C in Chinese chronic kidney disease patients

Objective To validate the four chronic kidney disease epidemiology collaboration (CKD-EPI) predictive equations based on serum creatinine (SCr) and cystatin C (Cys C) in Chinese CKD patients,and try to develop the GFR predictive equations for Chinese CKD patients.Methods254 CKD patients were randomly selected from four Grade ⅢA hospitals in different regions in China from September 2007 to December 2010.Clearance of dual plasma sampling 99mTc-DTPA was used to measure glomerular filtration rate (rGFR) in 254 CKD patients.The serum concentration of Cr and Cys C were measured.CKD-EPI SCr equation,Cys C equation,Cys C equation adjusted for age,sex and race,SCr/Cys C combinated equation adjusted for age,sex and race were used to estimate GRF ( labeled as eGFR1,eGFR2,eGFR3 and eGFR4,respectively).The correlation,bias and precision of eGFRs were compared with rGFR by Wilcoxon signed rank test,intraclass correlation coefficient (ICC) and Spearman correlation analysis.The deviation degree between rGFR and different eGFRs was compared via Bland-Altman graph.The accuracy within 15%,25%,30% ( P15,P25,P30) and the staging correctness of eGFR against CKD at different stages was calculated.ResultsThe rGFR in 254 CKD participants was [ 48.07 (26.19 -92.97 )] ml · min -1·(1.73 m2) -1.The Spearman correlatiou coefficients (CC) of eGFR and rGFR varied within the range of 0.873 - 0.896 ( P =0.000 ).The intra-class CC ( ICC ) varied within the range of 0.920 - 0.942.The correlation of eGFR4 was the best.The absolute deviations of 4 eGFRs and deviation precision were eGFR4 <eGFR3 < eGFR2 < eGFR1.The 95% confidence intervals for the regression line of 4 eGFRs shown by Bland-Altman graphs were 92.5,87.3,83.0 and 76.1 ml · min-1 · ( 1.73 m2 ) -1,respectively,with the best result of eGFR4.For P30,the correctness of 4 eGFRs were eGFR4 > eGFR3 > eGFR2 > eGFR1,but no significant difference was found by Chi square test (x2 =6.448,P =0.092).The overall correctness rate in 4 eGFRs against CKD stages were 48.4% -57.5%,with the highest consistency of eGFR4,but their staging correctnessratewerenotideal(Kappa values were 0.405,0.348,0.366 and 0.463,respectively).Conclusions Compared with CKD-EPI SCr equation,no advantage was found in CKD-EPI Cys C equation.The Cys C equation adjusted by age and sex shows a little advantages over CKD-EPI Cys C equation in bias,precision,correlation and accuracy.The CKD-EPI SCr/Cys C combinated equation adjusted by age,sex and race has advantage over other three equations not only in bias,precision,correlation and accuracy,but also in staging correctness.However,the validation of this equation is still not fairly ideal for Chinese CKD patients.Based on these findings,it is essential for the Chinese CKD patients to develop SCr/Cys C combined predictive equation which adjusted by age,sex or other factors.(Chin J Lab Med,2012,35:798-804)
8.Inhibitory effect of ginsenoside-Rg3 on lung metastasis of mouse melanoma transfected with ribonuclease inhibitor.

OBJECTIVETo investigate the effect of ginsenoside-Rg3 on lung metastasis of ribonuclease inhibitor (RI) gene-transfected mouse B16 melanoma.
METHODSC57BL/6 mice were iv injected with parental or RI-transfected B16 melanoma cells. Lung metastasis was assessed by the number of surface tumor nodules. Mice were divided into 6 groups. Group I, II and III of mice were given parental, mock-transfected and RI-transfected B16 melanoma cells, respectively while in group IV, V and VI, Rg3 (1.5 mg/kg, iv q.o.d. x 10) was given to mice bearing parental, mock-transfected and RI-transfected B16 melanoma, respectively. Micovessel density (MVD) of the lung metastatic tumor was assessed by immunohistochemical staining of factor VIII-R expression.
RESULTSThe number of tumor nodules was significantly decreased in mice injected with RI-transfected B16 melanoma (Gp III, compared to Gp I and II). Rg3 treatment per se could also decrease the number of lung tumor nodules but to a lesser extent (Gp IV and V compared to Gp III). However, Rg3 synergized with RI transfection resulting in most significant inhibition of lung metastasis (Gp VI). Mice in Gp I and II died within 26 days of the experiment, whereas all the mice in Gp VI were alive during the observation period of one and one half month. MVD was significantly decreased in the lung tumor nodules in mice injected with RI-transfected B16 melanoma. It was further decreased when additional Rg3 was given (Gp VI).
CONCLUSIONTransfection of ribonuclease inhibitor gene significantly reduces the metastatic potential of B16 melanoma. Ginsenoside-Rg3 has a synergistic effect.

9.Fucoidan: advances in the study of its anti-inflammatory and anti-oxidative effects.

Fucoidan is a natural polysaccharide extracted from brown seaweeds, with a wide variety of biological features, especially the anti-inflammatory and anti-oxidative effects. Studies indicated that the anti-inflammatory effect of fucoidan related to its capacity to interact with the selectin or scavenger receptor on the cell membrane. Fucoidan can also inhibit the synthesis and release of reactive oxygen species (ROS), as well as promote its clearance, showing the anti-oxidative activity.

10.XBP-1 interacts with estrogen receptor alpha (ERalpha).

Estrogen receptor alpha (ERalpha) has been a primary target of treatment as well as a prognostic indicator for breast cancer. The level of human X-box binding protein 1 (XBP-1) mRNA was related with that of ERalpha in breast tumors and was over-expressed in some breast tumors. These previous studies suggested that XBP-1 may interact with ERalpha. XBP-1 has two isoforms, XBP-1S and XBP-1U, as the result of unique splicing. GST pull-down assay showed that both XBP-1S and XBP-1U bound to ERalpha in vitro. The binding of XBP-1S to ERalpha was stronger than that of XBP-1U to ERalpha. Co-immunoprecipitation revealed that the binding was in a ligand-independent manner. XBP-1S and XBP-1U interacted with the region of ERalpha that contains a DNA-binding domain. The ERalpha-interacting regions on XBP-1S and XBP-1U have been mapped to two regions, the N-terminal basic region leucine zipper domain (bzip) and the C-terminal activation domain. These findings suggest that XBP-1S and XBP-1U may participate in ERalpha signaling pathway through the mediation of ERalpha.
