1.The association between polymorphism of HLA-DRB1 allele and systemic sclerosis and renal damage in Han Chinese of Henan Province

Objective To investigate the association between polymorphism of HLA-DRB1 allele and systemic sclerosis (SSc) and scleroderma-associated renal damage in Han Chinese of Henan Province.Methods Eighty-one SSc patients and 90 normal controls were recruited in this study,among them 59 patients had renal damage (SRD).The genotyping was carried out by nest PCR-SBT and gene clone.Comparisons between groups were performed with x2 test or exact probabilities.Multivariable Logistic regression was used to evaluate the association between prevalence of SSc or SRD and the possible relevant alleles.Results Thirty-three HLA-DRB1 alleles were discovered from the specimens,including 27 in SSc specimens,and 22 in SRD.Among them,the allele frequencies of HLA-DRB1 * 040501 (8.64%), * 110101 (11.11%), * 150201(8.02%) were higher in SSc patients than those of the controls (1.67%,4.44%,2.22% respectively).After adjusted for other factors,HLA-DRBl * 040501 (P=0.010,OR =5.839,95%CI:1.518-22.460)、* 110101(P=0.019,OR=3.060,95%CI:1.204-7.772)、* 150201(P=0.010,OR=4.780,95%CI:1.444-15.821 )were identified as independent risk factors for SSc.And the allele frequencies of HLA-DRB1*040501 (9.32%),* 150201 (7.63%) were higher in SRD patients than those of the controls (1.67%,2.22% respectively).After adjusted for other factors,HLA-DRB1 * 040501 (P=0.008,OR=6.363,95%CI:1.614-25.087) and * 150201 (P=0.030,OR =4.043,95 %CI:1.147-14.243 ) were identified as independent risk factors for SRD.Conclusion Our data suggest that HLA-DRB1 * 040501,* 110101,* 150201 may be susceptible genes for SSc and the HLA-DRB1 * 040501,* 150201 may be susceptible genes for SRD in Han Chinese of Henan Province.
2.The effects of rosuvastatin on the liver and kidney function and muscle pathology in rats

ObjectiveTo study the liver and kidney function and muscle effects of rosuvastatin in rats.MethodsIn this study,twenty healthy male rats (9-month-old) were randomly divided into two groups:normal control group and rosuvastatin treatment group.The first treated group was given conventional feed; The second treated group was given conventional feed and oral rosuvastatin [5mg/( kg · d)] of 12 weeks.The level of liver and kidney function and creatine kinase,histological changes of liver and muscle were examined before and after the treatment.ResultsOur findings demonstrate that the level of liver and kidney function and creatine kinase was no significant difference among the two experimental groups,also no difference before and after treatment( P >0.05).At the same time,liver and muscle showed no abnormal pathology.Conclusion These findings collectively indicate that rosuvastatin shows no significant side effects in the liver and kidney function and muscle pathology compared with control group.
3.Chemical constituents from traditional Chinese medicine Siegesbeckia pubescens.

Column chromatography on silica gel was used to study the chemical constituents of traditional Chinese medicine Siegesbeckia pubescens. The chemical structures of the separated compounds were elucidated by spectroscopic data analyses. As a result, eighteen compounds were obtained and identified as 3, 4'-dimethoxy quercetin(1), 3, 3', 4'-trimethoxy quercetin(2), 3, 3'-dimethoxy quercetin(3), 7, 3', 4'-trimethoxy luteolin(4), ursolic acid(5), 2β,19α-dihydroxyursolic acid(6), 2β-hydroxyursolic acid (7), stigmasterol-7-one(8), 5α, 8α-epidioxy-24(R)-methyl-cholesta-6, 22-diene-3β-ol(9), β-sitosterol(10), 2, 6-di(3-hydroxy-4-methoxyphenyl)-3, 7-dioxacyclo [3. 3. 0] octane (11), aurantiamide acetate (12), 3-(m-hydroxyl-p-methoxy)-N-(2'-p-hydroxyl-phenethyl)-2E-acrylamide(13), p-hydroxy benzaldehyde (14), m-hydroxy-p-methoxy benzaldehyde (15), 3, 4, 5-trimethoxybenzoic acid(16), monoethyl malonate(17), and p-hydroxylcinnamic acid(18). Among them, compounds 1-9, 11-18 were isolated from this plant for the first time.

4.The study on the correlation between serum mannose-binding lectin and renal injury in rheumatoid arthritis

Objective To detect the serum level of mannose binding lectin (MBL) in patients with renal injury induced by rheumatoid arthritis (RA), and to investigate the role of MBL in the pathogenesis of renal injury in RA. Methods ELISA was used to measure the serum MBL level of 19 RA patients with renal injury, 49 RA patients without renal injury and 40 healthy individuals. The clinical features and laboratory markers were compared and analyzed by chi-square test, two independent samples t-test and Spearman's correlation analysis. Results Compared with RA patients without renal injury, the number of tender and swollen joints [(15±9) vs (9±11)], duration of morning stiffness [(2.9±1.3) vs (2.3±1.6) h] and extraarticular manifestations (42.1% vs 16.3%) in RA patients with renal injury were significantly higher (P<0.05or P<0.01). There was no significant difference between the two groups in RA disease duration and jointdeformity(P>0.05). In patients with renal injury, the level of platelet count [(376±155)×109/L vs (304±121)×109/L], CIC[(4.3±3.0) vs (2.9±3.3) g/L], ESR[(79±46) vs (53±31) mm/1 h], RF[(77±42) vs (52±49)U/ml], CRP[(32±28)vs (23±18)mg/L], IgG[(11.7±2.6)vs (8.4±2.4)g/L], C3[(1.18±0.53)vs (0.94±0.21) g/L] were higher than those in RA patients without renal injury (P<0.01 or P<0.05); the level of Alb [(26±13) vs (30±9) g/L] was lower than that in RA patients without renal injury (P<0.05). The level of serum MBL in the two groups of RA patients was significantly lower than that in the healthy group [(3.1±0.5)mg/L](P<0.01), and the level of serum MBL in RA patients with renal injury [(1.7±1.2) mg/L] was higher than that in RA patients without renal injury [(1.4±1.3) mg/L](P<0.05). The level of serum MBL in RA patients with renal injury showed a positive correlation with IgG, C3, CRP and 24 h urine protein level (r=0.6, 0.6, 0.47, 0.57; P<0.05). Conclusion Renal injury in RA patients is immune complex dependent. The serum level of MBL is higher in patients with renal injury, therefore, high-concentration MBL may be one of a potential causes of renal injury in RA patients.
5.The impact of negative life events on risky mentation in college students: mediating role of personality

Objective:To explore the mediation effect of personality between negative life events and risky mentation of university students.Methods:A cross sectional investigation was conducted among 8 379 freshmen with the adolescent self-rating life events check list (ASLEC), the prodromal questionnaire(PQ-16) and the Eysenck personality questionnaire (EPQ). The data were analyzed by SPSS 23.0 and AMOS 24.0.Results:The total score of negative life events scale((31.16±0.58) vs (15.19±0.15)), the scores of neuroticism((58.20±0.36) vs (41.59±0.13)) and psychoticism((53.07±0.29) vs (47.71±0.08)) in the risk psychological state group were significantly higher ( t=26.611, 42.270, 17.286, all P<0.01), and the score of introversion-extroversion factor was significantly lower((49.83±0.42) vs (55.88±0.13), t=-13.634, P<0.01) than those in the risk-free psychological state group. There was a positive correlation between the scores of risk psychological state and negative life events( r=0.290, 0.334, both P<0.01), and the scores of risk psychological state and negative life events were positively correlated with the scores of personality neuroticism and psychoticism ( r=0.139-0.469, all P<0.01) in both risk psychological state and risk-free psychological state group.The risk psychological state score of college students was negatively correlated with the inside and outside personality score( r=-0.070, P<0.01), and the score of negative life events was not correlated with introversion-extroversion personality score in the risk psychological state group, while the score of risk psychological state, negative life events and introversion-extroversion personality score were negatively correlated in the risk-free psychological state group ( r=-0.177, -0.080, P<0.01). The personality of college students played a complete mediating role between negative life events and risk psychological state in the risk psychological state group, while the personality of college students in the risk-free psychological state group played a partial mediating role between negative life events and risk psychological state, accounted for 71.43% of the total effect. Conclusion:Negative life events not only directly lead to the risky mentation of college students, but also affect the risky mentation of college students by the mediation effect of introverted and extroverted tendency and unstable emotion.

7.Clinical Study of Three Trembling Needles for Early Parkinson's Disease

Objective To observe the clinical efficacy of needling the three trembling points plus rehabilitation and taking levodopaand benserazide hydrochlo-ride.Method Ninety-three patients with early Parkinson's disease were randomized into group A of 32 cases, group B of 30 cases, and group C of 31 cases. Group A was intervened by needling the three trembling points plus rehabilitation and taking levodopaand benserazide hydrochlo-ride, group B by rehabilitation and taking levodopaand benserazide hydrochlo-ride, while group C by taking levodopaand benserazide hydrochlo-ride. UPDRSⅢ scores and Berg balance scale (BBS) scores were observed.Result After intervention, UPDRSⅢ scores and BBS scores were significantly changed in all groups (P<0.05). UPDRSⅢ scores of group A were markedly different form that of group B and C (P<0.05). BBS scores of group A and B were markedly different form that of group C (P<0.05).Conclusion Needling the three trembling points plus rehabilitation and taking levodopaand benserazide hydrochlo-ride can improve motor function and equilibrium function in the early Parkinson's disease patients.

An offline two-dimensional system combining a rat cardiac mascle cell membrane chromatography time-of-flight mass spectrometry (CMC-TOF/MS) with a high performance liquid chromatography time-of-flight mass spectrometry (HPLC-TOF/MS) was established for investigating the parent components and metabolites in rat urine samples after administration of the roots of Aconitum carmichaeli. On the basis of the analysis of the first dimension, retention components of the urine sample were collected into 30 fractions (one fraction per minute). Then offline analysis of the second dimension was carried out. 34 compounds including 24 parent alkaloids and 10 potential metabolites were identified from the dosed rat urine, and then binding affinities of different compounds on cell membranes were compared and influences of some functional groups on activity were estimated with the semi-quantification and curve fitting method. As a result, binding affinities decreased along with the process of deacylation, debenzoylation and demethylation, which may be related to the alleviation of toxicity in the procedure of herb processing or metabolism. Moreover, some minor components in rat urine (Songorine, 14-benzoylneoline, Deoxyaconitine, etc. ) exerted relatively strong affinity on cell membranes are worth exploring. The results delivered by the system suggest that the CMC can be applied to in vivo study.
10.Analysis of points selection pattern in acupuncture treatment of sleep apnea syndrome based on data mining

Objective: To explore the points selection pattern of acupuncture for sleep apnea syndromes by data mining technique. Methods: Clinical literature about acupuncture therapy for sleep apnea syndromes was derived from China National Knowledge Infrastructure (CNKI), Wanfang Academic Journal Full-text Database (Wanfang), Chongqing VIP Database (CQVIP), PubMed and Science Direct between the time that databases were created and March 25th,2017. Relevant excel database was established and descriptive studies and association rules were analyzed. Results: The most frequently used point was Lianquan (CV 23) and the most frequently used meridian was the Stomach Meridian. The analysis of association rules showed that the clinical choice of acupuncture points was highly correlated, among which the combination of the highest degree of confidence and the highest degree of support was Shenmen (HT 7) and Sishencong (EX-HN 1); Lieque (LU 7), lianquan (CV 23) and Zhaohai (KI 6). Conclusion: Acupuncture treatment of sleep apnea syndromes has specific selection rules of points, providing certain references for clinical and scientific research.
