2.The Value of MR Cisternography in Epidermoids of the Cerebellopontine Angle

Objective To evaluate the usefulness of MR cisternography in epidermoids of the cerebellopontine angle (C-P angle).Methods The findings of MR cisternography of epidermoids of the C-P angle in 19 cases confirmed by surgery were analysed retrospectirey.The signal intensity of tumors,the effect on nerves and vessels by tumor were evaluated.Results MR cisternography depicted tumors as slight-medium hypointense (n=19) to CSF. 84% of patients had involvement of the trigeminal nerves (n=16); 42% of patients had involvement of the facial nerve and the acoustic nerve (n=8); 26% of patients had involvement of the internal carotid artery (n=5); 21% of patients had involvement of the vertebral-base artery (n=4). Conclusion MR cisternography contributes to show the anatomic details of the C-P angle and clearly depicte the relationship between tumors and nerves as well as vessels. Thus it can provide useful informations for the presurgical planning and approach of C-P angle epidermoids.
3. Simultaneous determination of nine compounds in Bergenia purpurascens by HPLC-MS

OBJECTIVE: To develop a method for the determination of arbutin, gallic acid, bergenin, protocatechuic acid, chlorogenic acid, catechin, epicatechin, epicatechin gallate, and ferulic acid in Bergenia purpurascens by HPLC-MS/MS. METHODS: The samples were separated on a Diamonsil C18 column (4.6 mm × 150 mm, 5 μm) by gradient elution using methanol and 0.2‰(V/V) formic acid aqueous solution as the mobile phase at a flow rate of 0.8 mL · min-1. The column temperature was room temperature. Multiple-reaction monitoring (MRM) scanning was employed for quantification with switching electrospray ion source polarity in negative mode. RESULTS: Complete separation was achieved within 10 min for the nine compounds (arbutin, gallic acid, bergenin, protocatechuic acid, chlorogenic acid, catechin, epicatechin, epicatechin gallate, ferulic acid): The nine regression equations showed linear relationships between peak areas and contents of each compound. The average recoveries (n=9) of the compounds listed above were 100.2%, 100.0%, 99.8%, 99.9%, 97.9%, 101.2%, 99.8%, 101.7%, 102.4% and RSD were 0.80%, 3.06%, 2.72%, 3.28%, 3.58%, 1.44%, 0.95%, 0.96%, 0.48%, respectively. CONCLUSION: The method is simple, accurate and has good reproducibility, thus can be used for the determination of the nine compounds in Bergenia purpurascens.
4.Influence of Human Chorionic Gonadotropin on Testicular Germ Cell Apoptosis of Bilateral Cryptorchidism in Rats

Objective To investigate the influence of human chorionic gonadotropin on testicular germ cell apoptosis of bilateral cryptorchidism in rats.Methods Immature male rats (22 day-old Sprague Dawley) were subjected to bilateral cryptorchidism and injected with 20 U human chorionic gonadotropin from the 22th day to the 34th day every other day. Sham operation rats were as control group. At age 35 days and 60 days, rats were sacrificed for detection germ cell apoptosis by terminal- deoxynucleotidyl transferase mediated d-UTP nick end labeling (TUNEL). Results There were significant difference of apoptosis index (AI) between bilateral cryptorchidism and sham operation groups(P0.05).Conclusion AI of cryptorchidism increases and HCG addes the number of apoptotic germ cells.
5.Value of difusion weighted MR imaging in intracranial epidermoids

Objective To evaluate the usefulness and specificity of diffusion weighted imaging (DWI) in the diagnosis of intracranial epidermoid Methods Fifteen patients with surgically proved intracranial epidermoids were evaluated with T 1 , T 2 weighted imaging, proton density weighted imaging and fast fluid attenuated inversion recovery (FLAIR) sequences followed by echo planar DWI Results DWI depicted all tumors ( n =15) as strong hyperintensity relative to CSF and brain tissues DWI was also sensitive in assessing small residual foci ( n =4) and recurrence of tumor ( n =3) Conclusion DWI is highly sensitive and specific in demonstrating epidermoids, and also in postoperatively assessing small residual foci and recurrence of tumors
6.Compatibility evaluation of Chinese medicines injections based on isothermal titration calorimetry.

The paper is to report the establishment of a method for quickly evaluating compatibility of Chinese medicines injections. Isothermal titration calorimetry (ITC) was used to evaluate compatibility of Yiqifumai (YQFM) and vitamin C injection (Vc)/5% glucose injection (5% GS). The diversification of Gibbs free energy (deltaG), enthalpy (deltaH) and entropy (deltaS) were used to decide reaction types of colliquefaction procedures of different injections. The reactive profiles were used to determine signs and quantity of heat. And high performance liquid chromatography (HPLC) was used as a supportive method for ITC. Then, feasible binding sites were analyzed based on the information of spatial structures of major compositions. During the colliquefaction procedure of YQFM and Vc, [deltaH] > T[deltaS], so, the reaction is enthalpy-driving. And the reactive profile showed that a big deal of heat was given out during the procedure. Obviously, chemical reactions happened and the major compositions changed. On the other side, the reaction of YQFM mixed with 5% GS was entropy-driving, because [deltaH] < T[deltaS]. The reactive profile showed there was a little heat gave out. So, non-chemical reactions happened, and the major compositions did not change. The conformity existed between the results of ITC and HPLC. ITC could be used to evaluate the compatibility of Chinese medicines injections because of the advantages of ITC, such as real time, fast, sensitive and having more parameters.
7.Indication of contrast-enhanced ultrasound in evaluating of portal vein patency

Objective To evaluate the usefulness of baseline ultrasound(BUS) and contrast-enhanced ultrasound(CEUS) in diagnosis of portal vein(PV) patency and to explore the indication of CEUS. Methods The diagnostic capability of BUS and CEUS were assessed in 64 cases according to definition of PV on BUS,which was divided into high, moderate and low definition. Results Thirty-five patients with portal vein thrombosis(PVT) and 29 patients with normal portal vein were confirmed. The diagnostic accuracies by using BUS and CEUS were 93.1% and 96.6% with high definition of PV,56.3% and 87.5% with moderate definition,and 68.4% and 89. 5% with low definition, respectively. Conclusions In the case of high definition of PV,the validity of diagnosing of PV patency with BUS may be similar to that with CEUS,thus introduction of CEUS would probably not be necessary. Otherwise CEUS is recommended.
8.Construction of OTX1 Lentiviral Vector and Overexpression Research

OTX1 gene is one of the pivotal transcriptional factors involved in the neurogenesis.In order to overexpress the OTX1 gene in distinct cell types and find out its contribution to the proliferation and differentiation of stem cells in vitro,OTX1 cDNA was subcloned into lentiviral vectors.The resulting constructions pDUETOTX1,pDUETGFPOTX1 and pDUETGFP were packaged in 293 cells producing viral particles to transduce 293T cells,SY5Y cells,mouse embryonic stem cells and E15 neural stem cells.It was proved that the transferred OTX1 gene was located in the nuclei of the transduced cells in stead of plasma.Lentivirus is an ideal vector delivering gene to different cells.The overexpression of OTX1 in transduced 293T cells were validated by Western blot and immunofluorescence.

Endogenous ouabain (EO) is a recently found hormone that may be secreted from adrenal cortex. As an endogenous mammalian analogue of cardiac glycosides and an inhibitor of the sodium pump, it regulates the body fluid balance, urine sodium extraction and vasoconstrictive tone, and thus plays an important role in the pathogenesis of hypertension and some other cardiovascular disorders. This articke reviews its biological features, receptor and antibody, detection, effects on diseases and relationship with endothelial cells.

10.Quality control of shuanghuanglian freeze-dried powder for injection based on its HPLC-ELSD fingerprints and biological profiles.

This paper is aimed to provide the methods of quality control and bioassay of traditional Chinese medicine injections including bioassay method. Shuanghuanglian freeze-dried powder for injection (SFPI) was chosen as study object. HPLC-ELSD fingerprints of SFPI had been established and the samples were differentiated by similarity calculation. Meanwhile, biological profiles of SFPI on Escherichia coli had been established by microcalorimetry. The similarity values were calculated using the correlation coefficient, based on quantitative thermo-kinetic parameters (T2m, Tj, I%). The results indicated that HPLC-ELSD fingerprints, which showed content changes of chemical components, could not monitor minimal variation of different samples, especially that of biological pollutants, while biological profiles could sensitively detect antibiotic activity alterations of the samples, which were kept under specific conditions. In conclusion, characterized by two-dimension, microcalorimetry could supply thermograms as biological profiles characterized to describe the bioactivity of drugs. This study could clearly demonstrate that the correlative detection was proposed as an efficient strategy for quality control of SFPI, based on HPLC-ELSD fingerprints and biological profiles, which could detect quality fluctuation of samples early and quickly and predict the potential adverse drug events (ADE) for ensuring clinical safety.
