1.Effect of Electroacupuncture on Expression of Ghrelin and mRNA Expression of Its Receptor in Functional Dyspesia Rats.

OBJECTIVETo explore the effect of electroacupuncture (EA) on the expression of Ghrelin and mRNA expression of its receptor in functional dyspepsia (FD) rats.
METHODSTotally 80 rats were divided into the normal group, the model group, the drug therapy group, and the EA group according to random digit table, 20 in each group. FD model was duplicated by clipping tail modeling. Drug containing cisapride [2 mL/100 g, 0.09 g/(kg x d)] was administered to rats in the drug therapy group from the 3rd day after successful modeling, once per day. EA at Zusanli (ST36) (0.3-0.5 cun) and Taichong (LR3) (0.1-0.2 cun) was performed in the EA group. The twirling of needle was performed to the subsidence of needle, and then the needle was connected to HANS-200A Acupoint Nerve Stimulating Device using disperse-dense wave at 2 Hz, 2 mA, 30 min each time, once per day. Six days consisted of one therapeutic course, two courses in total with an interval of one day. The intestinal propulsive rate of ink was observed. Ghrelin protein expression in gastric tissue was detected by Western blot. mRNA expression of growth hormone secretagogue receptor (GHS-R) in stomach, hypothalamus, and hippocampus was detected using Real-time PCR respectively.
RESULTSCompared with the normal group, the intestinal propulsive rate of ink, Ghrelin protein expression in gastric tissue, mRNA expression of GHS-R in stomach, hypothalamus, and hippocampus decreased in the model group (P < 0.05, P < 0.01). Compared with the model group, the intestinal propulsive rate of ink, Ghrelin protein expression in gastric tissue, mRNA expression of GHS-R in stomach, hypothalamus, and hippocampus increased in the EA group (P < 0.01); mRNA expression of GHS-R in stomach, hypothalamus, and hippocampus increased in the drug therapy group (P < 0.01). Compared with the drug therapy group, Ghrelin protein expression in gastric tissue, mRNA expression of GHS-R in hypothalamus increased in the EA group (P < 0.05, P < 0.01).
CONCLUSIONEA could regulate Ghrelin content and GHS-R mRNA expression of FD rat hypothalamus, hippocampus, and gastric tissue, and promote the intestinal propulsive rate of ink.

2.The relationship of serum placental growth factor and carotid atherosclerosis in patients with type 2 diabetes mellitus

Objective To explore the relationship between serum placental growth factor (PLGF) and carotid atherosclerosis (CAS) in patients with type 2 diabetes mellitus (T2DM ). Methods Serum PLGF were determined by ELISA in 53 patients with T2DM ,who were divided into T2DM without carotid plague (T2DM group ,n= 27) and T2DM with carotid plague (T2DM + CAS group ,n= 26). Fasting plasma glucose (FPG) ,glycosylated hemoglobin A1c (HbA1c) and some other metabolic variables were also measured. The results were compared to those of 27 healthy controls (NC group). Results Serum PLGF levels were significantly higher in T2DM group and T2DM + CAS group than in NC group (P<0.01). T2DM+CAS group showed significantly higher serum levels of PLGF compared to that of T 2DM group(P<0.01).Correlation analysis showed that serum PLGF was positively correlated with TC ,LDL‐C , hsC‐RP and IL‐6 in T2DM patients (P<0.01). Similarly ,multiple regression analysis showed that LDL‐C and IL‐6 were the independent factors of serum PLGF level (P<0.05). Furthermore ,Serum PLGF was positively associated with carotid plague. Conclusion PLGF is positively related with chronic inflammation factors in T2DM patients. Serum PLGF may play an important role in the occurrence and development of carotid atherosclerosis and other macroangiopathy in T 2DM.
3.The study on genes trans-regulated by XTP4 using suppression subtractive hybridization technique

Objective To explore the influence of XTP4 on genomic expression profile of hepatocyte through screening and cloning of genes trans-regulated by XTP4-expressing plasmid. Methods The expressive vector pcDNA3.1(-)-XTP4 was constructed by routine molecular biological methods, and suppression subtractive hybridization (SSH) method was employed to detect the mRNA differentially expressed by the HepG2 cells transfected with pcDNA3.1(-)-XTP4 and pcDNA3.1(-), respectively, using lipofectamine. The twice enriched PCR products were subcloned into T/A vectors to set up the subtractive library. Amplification of the library was carried out in E. coli strain JM109. The screened cDNA were sequenced and analyzed in GenBank with Blast search after PCR. Results The amplified subtractive library containd 21 positive clones. Colony PCR analysis showed that there were 16 clones containing 200-1000bp inserts. Sequence analysis was performed and 9 kinds of encoding sequences were achieved. These genes trans-regulated by XTP4 protein involved in hepatic fibrogenesis, tumorgenesis, mitochondrial function, and cell growth regulation. Conclusions The findings obtained by SSH provide significant data for a preliminary understanding of the biological function of a new identified gene-XTP4. These results will pave the way for the study of the molecular mechanism of the transactivating effects of HBxAg and the development of new therapy for chronic hepatitis B.

Objective To explore the therapy measurement of iron deficiency anemia(IDA)associated helicobacter pylori(HP)infection among children.Methods Eight hundred and ninty nine cases entrusted children aged from 2 to 7 years old were measured hemoglobin(Hb),mean corpuscular volume(MCV)mean corpuscular hemoglobin(MCH),mean corpuscular hemoglobin concentration(MCHC)and serum ferri,serum fettin(SF),HP-IgG antibodies.Divided the HP-IgG positive cases into two groups for treatment,The treatment group adopted omeprazole,amoxicillin and metronidazole to cure them for two weeks,then oral ferrous sulfate sugar liquid.The contrast group purely take the ferrous sulfate sugar liquid.After eight weeks re-examined the Hb and SF.Results Totally 279 cases were diagnosed as Hp-positive,620 as HP-negative and 99 children diagnosed as IDA,among the 99 cases IDA,58 cases with HP infection(20.8%) and 41 cases with non-HP infection(6.7%).That means the number of IDA cases with HP infection was higher than those with non-HP infection(?2=39.46 P
6.Application of BIS monitoring and Ramsay score in the prevention of unplanned tracheal extubation in ICU patients

Objective To investigate the application value of BIS monitoring and Ramsay score in the prevention of unplanned tracheal extubation in ICU patients.Methods 93 patients were enrolled in this study,they were divided into the experimental group(47 cases)and the control group(46 cases) using random number method.They received sedation regimens with BIS monitoring and Ramsay score or Ramsay score respectively.Occurrence rate of unplanned extubation was compared between the two groups.Results The occurrence rate of unplanned extubation was significantly lower in the experimental group than that of the control group.Conclusions BIS monitoring and Ramsay score is a suitable ways for the management of sedation of intubated patients.
7.Effect of phacoemulsification on corneal endothelium of cataract with lupus nephritis

To evaluate the influence of phacoemulsification on corneal endothelium of senile cataract with lupus nephritis (LN).
●METHODS:This clinical trial involved 40 cataract patients with lupus nephritis (40 eyes), and 50 cases (50 eyes) without lupus nephritis. All of them underwent phacoemulsification+lOL implantation. The parameters of corneal endothelial cell including central corneal endothelium cell density ( CED ), average area of endothelial cell ( AVE) and coefficient of variation ( CV) were recorded by corneal endothelial microscope pre -operation and at one month after operation. The data were analyzed by SPSS 13. 0 statistical software.
● RESULTS: Both LN group and control group, the morphology of coneal endothelial was statistical significant differences between pre - operation and 1mo postoperation. The CED was lower, AVE and CV were higher ( P < 0. 05, respectively). A significant decrease in CED was seen in the LN group than did in the control group (P < 0. 05). Compared to control group,the post -operative AVE and CV in LN group was significantly increased (P<0. 05).
● CONCLUSlON: The corneal endothelial cell in lupus nephritis patients is more fragile. Safe and reliable operation should be selected for these patients.
8.Double incision trabeculectomy combined with phacoemulsification and intraocular lens implantation for acute angle closure glaucoma with cataract

Abstract?AIM: To observe the effect of acute angle closure glaucoma with cataract patients treated with glaucoma trabeculectomy combined two incision phacoemulsification and intraocular lens ( IOL ) implantation ( two incision triple surgery) .?METHODS: Patients admitted in our hospital during Jan.2013 to Jan.2016.The acute angle closure glaucoma with cataract patients in 40 cases ( 58 eyes ) were randomly divided into two groups:the observation group of 20 cases ( 29 eyes ) with two incision triple surgery treatment, and the control group of 20 cases ( 29 eyes ) with single incision triple after treatment.Visual acuity, intraocular pressure, bleb, corneal endothelial cell density, the area at 1mo before and after surgery, and postoperative complications of two groups were analyzed.?RESULTS: Postoperative visual acuity of two groups were significantly improved, but there was no significant difference between groups ( P >0.05 ); postoperative intraocular pressure, bleb formation rate of the observation group were 14.41 ±1.38mmHg, 90%, and the control group 14.40 ±1.40mmHg, 86% without statistical significance ( P>0.05 ), corneal endothelial cell density and area of observation group after 1mo were 1696.6±300. 8/mm2 , 540.8±71.6μm2 , and control group 1410.6±288.5/mm2 and 594.3 ±72.8μm2 with significant differences ( P<0.05).The incidence rate of compli-cation was 17%in the observation group, and 21%in control group (P>0.05).?CONCLUSION: For patients with acute angle closure glaucoma and cataract given two incision triple surgery and single incision triple surgery treatment can get good outcomes, but the effect of double incision on corneal endothelial cell injury is less.
9.The Biologic Characteristics of a Strain of Cellulosimicrobium cellulans and Its Utilization of Several Kinds of Benzoic Compounds

A strain of Cellulosimicrobium cellulans Ha8 was studied on its morphological, biological characteristics and its utilization of several kinds of benzoic compounds, the results showed this strain was Gram-positive, the long rod-shaped cells were changed into short rod-shape gradually. pH value from pH 6.0 to pH 9.0 and the temperature from 20 ℃ to 40 ℃ were good for its growth. It could not only hydrolyze protein and starch, use cellulose and pectin, decomposite chitin, liquify gelatin and fix nitrogen, but also use phenol, xylene, benzoic, cinnamic acids and diphenlamine as the sole carbon resource for its growth. It could tolerate 0 mmol/L~30 mmol/L, 0 mmol/L~8 mmol/L, 0 mmol/L~30 mmol/L, 0 mmol/L~15 mmol/L and 0 mmol/L ~ 40 mmol/L of benzoic acids, phenol, xylene, cinnamic acids and diphenlamine seperately, but could not use 2,4-dinitrophenol, o-Nitrophenol, 2-Methoxyphenol, aminobenzenesulfonic acid, catechol and o-Phenanthroline as its sole carbon resource.
10.The study on levofloxacin resistance and gyrA gene mutation of Helicobacter pylori in Jinhua Zhejiang province

Objective To analyze levofloxacin resistance in Helicobacter pylori (Hp) and the sequence difference of gyrA gene in levofloxacin resistance and sensitive Hp strains. To explore the function of gyrA gene mutation in the development of levofloxacin resistance Hp strain.Methods From July 2007 to December 2008 in Department of Gastroenterology, Jinhua People hospital of Zhejiang Province, the gastric mucosa from gastroscopy biopsy of chronic gastritis and peptic ulcer patients were cultured in Hp selective medium under microaerobic condition at 37 degrees for three to five days. The Hp strains were isolated and identified by oxidase test, catalase test,urease test and UreA gene detecting. The levofloxacin susceptibility was determined by E-test and then resistance and sensitive strains were screened. The genomic DNA of Hp strains was isolated. The gyrA gene was amplified by PCR and the sequences were analyzed. Results 38 clinical isolated Hp strains were passed the levofloxacin susceptibility E-test, among those the minimum inhibitory concentration (MIC) of 12 strains was over 1.0 μg/ml, the percentage of resistant strain was 31.58%, while the sensitive stains was 68.42%. The gyrA gene sequence result indicated 10 resistant strain with 261, 271 and 272 10 site mutation, 2 strains with C261A mutation, one strain with C261G mutation, two strains with G271A mutation, 2 strains with A272G mutation, 2 strains with C261A,G271T andA272G mutation, 1 strain with C261G and A272G mutation. However, no mutation sites were found in 26 sensitive strains. Conclusion The rate of levofloxacir-resistance in isolated clinical Hp strain was high. The drug-resistance was associated with 261,271 and 272. site mutation of gyrA gene.
