3.Biological characterization of C57 mouse bone marrow mesenchymal stem cells using a whole bone marrow adherent culture technique

BACKGROUND:Under mitomycin C treatment, feeder cells appear to have restricted proliferation, but they are stil able to secret different cytokines. Non-mesenchymal stem cells from the bone marrow and secreted factors in plasma maintain the micro-environment suitable for the growth of mesenchymal stem cells that can improve the yield of mesenchymal stem cells.
OBJECTIVE:To study the biological characteristics of C57 mouse bone marrow mesenchymal stem cells isolated using a whole bone marrow adherent culture technique.
METHODS:Using the whole bone marrow adherent culture technique, purified and amplified C57 mouse bone marrow mesenchymal stem cells were harvested. cellproliferation kinetics, immune cellsurface markers, multiple differentiation potential and cellcycle were detected.
RESULTS AND CONCLUSION:Using the whole bone marrow culture, mouse bone marrow mesenchymal stem cells were harvested and capable of adhering to the plastic culture vessel. The obtained cells expressed CD45, CD105 and Sca-1, but were negative for CD34, CD33 and C-kit. The doubling time was (57.11±1.5) hours. The cells could be induced to differentiate into osteoblasts, adipocytes and chondrocytes. The cellcycle analysis showed that 64%of cells were in G 0-G 1 phase. These indicates that C57 mouse bone marrow mesenchymal stem cells isolated using a whole bone marrow adherent culture technique have biological characteristics of mesenchymal stem cells.
5.Comparison of effects of different pterygium surgeries

AIM: To compare the clinical outcomes of pterygium excision followed by translocation of adjacent conjunctival autograft with stem cells or transplantation of conjunctival autograft with stem cells to treat and prevent the recurrence of pterygium.METHODS: Fifty patients (60 eyes) were divided into 2groups, and pterygium excision followed by translocation of adjacent conjunctival autograft with stem cells (group A) and pterygium excision followed by transplantation of conjunctiva autograft harvested from counter-side limbus with stem cells (group B) were performed respectively.The surgical time of each case was recorded.RESULTS: All the 50 cases (60 eyes) of pterygium were recruited. The epithelization of cornea and conjunctiva in all cases finished in 2-5d, During the follow-up of 6-12mo, pterygium reoccurred in 2 cases (1 in each group), with no difference in recurrence rate. The mean surgical time was 25± 8min for group A,43± 11min for group B, the difference being statistically significant (P<0.05, t-test).CONCLUSION: Both methods, pterygium excision followed by translocation of adjacent conjunctival autograft with stem cells or transplantation of conjunctival autograft with stem cells are effective on treating and reducing the recurrence of pterygium. Compared with the latter, pterygium excision followed by translocation of adjacent conjunctival autograft with stem cells is simpler and easier to perform, and is preferred by the patients too.
6.Analysis on Repeated Offline of Files in Database in No.1 Military Medical Project

Objective To explore the cause of repeated offline of files in the database of No.1 Military Project and find the method to solve the problem. Methods The abnormal offline of files in database is not by chance,and its document in oracle is not very clear. Using the knowledge from internet,the fault was repeatedly simulated on test machine and the source of the fault was found. Results Solving plans and countermeasures were put forward. Conclusion The database of the No.1 Military Medical Project is the core of military hospital work,so its importance is self-evident. The technical support of No. 1 Military Project is strengthened,the right maintenance direction of the database is pointed out as well as the right solution to deal with the problem.
7.Energy calibration and electrical system design of HK.ESWL-008 child extracorporeal shock wave lithotripter

Objective To design an electrical system of extracorporeal shock wave lithotripter for children and adults uses.Methods The shock wave kernel parameters of child energy section of Dornier Delta Ⅱ lithotripter were tested such as peak sound pressure,pressure pulse width and pressure pulse rise time,and then energy calibration was executed for HK.ESWL008 child extracorporeal shock wave lithotripter to make the kernel parameters of the two lithotripters the same or similar to each other.An electrical system was designed with PLC system as the core control unit,involving in wave source 3D motion,large and small C-arms motion,water circulation system,B-type ultrasound positioning system,high-frequency X-ray imaging system,high-voltage condenser charging and discharging system,console and bedside-box-controlled display system as well as electromagnetic compatibility.Results Within A,B,C,1 and 2 energy sections for child lithotripsy the maximum and average differences between the kernel parameters of the two lithotripters were 7.1% and 3.8% respectively.There was no difference between the clinical experience by the two lithotripters,and HK.ESWL-008 child extracorporeal shock wave lithotripter passed EMC and safety detections by Guangdong Medical Devices Quality Surveillance and Test Institute of CFDA.Conclusion HK.ESWL-008 child extracorporeal shock wave lithotripter can be used for children lithotripsy clinically.
8.Protective effects of adenosine on cultured rat hippocampal neurons after oxygen-glucose deprivation

AIM:To investigate the protective effects of adenosine on cultured rat hippocampal neurons after oxygen-glucose deprivation. METHODS:The control and adenosine-treated hippocampal neurons cultured for 12 d were exposed to oxygen-glucose deprivation environment for 0.5-4 h and then cultured with original medium in normoxia for 24 h . The soma area,survival rate, effluxes of lactate dehydrogenase (LDH)and apoptosis of neurons were observed. RESULTS:The soma area, effluxes of lactate dehydrogenase from neurons and apoptosis were increased while survival rate of neurons was decreased after oxygen-glucose deprivation compared with those pre-oxygen-glucose deprivation. Compared with the control, after oxygen-glucose deprivation the soma area, effluxes of lactate dehydrogenase from neurons and apoptosis were decreased, however, the survival rate of neurons was increased in the adenosine group.CONCLUSION: Oxygen-glucose deprivation can lead to the severe damage of cultured hippocampal neurons, and adenosine can reduce neuronal injury induced by oxygen-glucose deprivation.
9. Simultaneous Determination of Curcumin and Germacrone in Curcuma wenyujin by Ultrasound-Assisted Ionic Liquid-Reversed Phase Liquid Chromatography

OBJECTIVE: To establish a method for the simultaneous determination of curcumin and germacrone in Curcuma wenyujin by ultrasound-assisted ionic liquid-reversed phase liquid chromatography. METHODS: An Agilent ZORBAX Eclipse Plus C18 column (4. 6 mm × 250 mm, 5 μm) was used. Gradient elution was conducted with the mobile phase consisting of acetonitrile and 0.1% phosphoric acid at the flow rate of 1.0 mL·min-1. The detection wavelength was set at 265 nm, the flow rate was set at 1.0 mL·min-1, and the column temperature was maintained at 30 ℃. RESULTS: The result showed that curcumin and germacrone had the highest extraction yield when the [BMIM]PF6 methanol solution concentration of 0.6 mol·L-1 was used as the extraction solvent and the solid-liquid ratio was 1:20. Under the optimized condition, good linearity of curcumin was shown for curcumin in the range of 0.041 2-2.06 μg (r=0.999 9)and the average recovery was 98.31%. The good linearity of germacrone was shown for germacrone in the range of 0.010 04-0.502 μg (r=0.999 8)and the average recovery was 97.62%. CONCLUSION: This method is convenient, environmentally friendlygreen and highly reproducible. It is beneficial important to study the extraction method of the active gradients of the traditional Chinese medicine Curcuma wenyujin active gradient and its quality control of Curcuma wenyujin.
10.Effect of ulinastatin combined with early enteral nutrition on severe acute pancreatitis and its effect on nuclear factor-κB and Toll-like receptor 4

Objective:To study the effect of ulinastatin combined with early enteral nutrition on severe acute pancreatitis and its effect on nuclear factor-κB(NF-κB) and Toll-like receptor 4 (TLR4).Methods:Ninety severe acute pancreatitis patients who were treated in Central Hospital of Lijin County from January 2016 to January 2020 were selected and were divided into U+EEN group (ulinastatin combined with early enteral nutrition therapy) and EEN group (early enteral nutrition therapy) by random number table method, with 45 patients in each group. Curative effect, complications, nutritional indicators, immunoglobulins and inflammatory factors were detected and compared with analysis of variance. Western blot was used to detect the expression of NF-κB and TLR4 in pancreatic tissue in two groups.Results:The hospitalization time, ICU admission time, intestinal ventilation time, hospitalization costs and organ failure rate, pancreatic cysts, diabetes, chronic pancreatitis, incidence of sepsis in U + EEN group were lower than those in EEN group: (2.1 ± 0.4) months vs. (2.4 ± 0.6) months, (16.9 ± 2.1) d vs. (21.7 ± 2.8) d, (23.7 ± 3.8) d vs. (27.4 ± 4.1) d, (11.4 ± 1.5) thousand Yuan vs. (14.1 ± 2.1) thousand Yuan and 8.9%(4/45) vs. 20.0%(9/45), 13.3%(6/45) vs. 28.9%(13/45), 11.1%(5/45) vs. 24.4%(11/45), 8.9%(4/45) vs. 26.7%(12/45), 6.7%(3/45) vs. 22.2%(10/45), and the differences were statistically significant ( P<0.05). The levels of prealbumin (PA), albumin (ALB) and total protein (TP) after treatment in U + EEN group were higher than those in EEN group: (107.4 ± 6.5) mg/L vs. (102.8 ± 4.7) mg/L, (46.1 ± 3.5) g/L vs. (43.4 ± 2.8) g/L, (55.9 ± 3.4) g/L vs. (53.7 ± 3.1) g/L, and the differences were statistically significant ( P<0.05). The levels of IgG, IgA, IgM after treatment in U+EEN group were higher than those in EEN group: (10.5 ± 1.6) g/L vs. (9.5 ± 1.3) g/L, (8.9 ± 1.4) mg/L vs. (8.3 ± 1.2) mg/L, (60.5 ± 3.6) mg/L vs. (55.9 ± 3.4) mg/L, the levels of tumor necrosis factor (TNF)-α, interleukin (IL)-6 and IL-8 in U+EEN group were lower than those in EEN group: (25.1 ± 2.9) mg/L vs. (30.6 ± 4.1) mg/L, (20.1 ± 1.9) mg/L vs. (24.6 ± 1.5) mg/L, (17.8 ± 1.9) mg/L vs. (20.1 ± 2.3) mg/L, and the differences were statistically significant ( P<0.05). The expressions of NF-κB and TLR4 protein in pancreatic tissue of patients in U + EEN group were significantly lower than those in EEN group (0.3 ± 0.2 vs. 0.5 ± 0.2, 0.2 ± 0.1 vs. 0.5 ± 0.1, P<0.05). Conclusions:Ulinastatin combined with early enteral nutrition can significantly improve the nutritional status and immune function and improve the prognosis of patients with severe acute pancreatitis, which may be related to ulinastatin′s reduction effect of NF-κB and TLR4′s expressions.
