Objective Pathologically, sometimes it is rather difficult to make differential diagnosis among hepatocellular, cholangiocellular and mixed type of hepatic tumors. The aim of this study was to analyze the immunohistochemical features of mixed type of hepatocarcinoma and to propose some useful diagnostic criteria. Methods Fifteen cases of the hepatic tumor were examined by routine histological and immunohistochemical methods with S P technique. Results The mixed type of hepatic tumor accounted for 3.39% of all cases of primary hepatocarcinoma in the same period. Histologically, all the mixed type of hepatic tumors revealed some characteristics of hepatocellular and cholangiocellular carcinomas, one of them was collision tumor. Immunohistochemically, 10 cases of the tumor were positive for ? fetal protein(AFP) in the hepatocellular carcinoma area, while all 15 tumors were negative for AFP in all cholangiocellular carcinoma areas. Cytokeratin(CK8) was positive in the hepatocellular carcinoma areas of all mixed tumor, whereas positive in cholangiocellular carcinoma areas of 13 cases; CK19 was positive in cholangiocellular carcinoma areas of all the tumors, while positive in hepatocellular carcinoma areas of 13 cases. Bcl 2 gene product was positive in 66.7% of this tumor, in which it was mainly expressed in cholangiocellular tumor area and in hepatocellular areas of five cases. AB/PAS stain for mucin was positive in cholangiocellular tumor regions of all the tumors. Conclusions Diagnosis of mixed hepatocarcinoma should be made mainly based on combined analysis of the data from pathological features, immunohistochemical phenotypes and histochemical stain for mucinous material.

Objective To observe the influences of 1,25-(OH)2D3 and salvia miltiorrhiza on hematopoiesis reconstruction and T cell subsets after allogeneic bone marrow transplantation in rats. Methods Donor rats were divided randomly into four groups, namely,Group A: aGVHD group;Group B: 1,25-(OH)2D3 group;Group C: salvia miltiorrhiza group;and Group D: two-drug combination group. We observed the survival time, peripheral blood cells, clinical manifestations of aGVHD and the changes of rat T cell subsets. Results Compared with those in aGVHD group,in every drug intervention group, the time for illness attack of aGVHD was delayed,aGVHD scoring was reduced and the average survival time was obviously prolonged(P

Objective To investigate the effect of 1,25-(OH)2D3 and salvia miltiorrhiza on T cell subsets and cytokines in rats after allogeneic bone marrow transplantation. Methods Female Wistar rats were used as recipients and male SD rats were used as donors. All Wistar rats were divided into aGVHD group and intervention groups at random, and the intervention groups were further divided into 1,25-(OH)2D3 group, salvia miltiorrhiza group and combination group. The changes of T cell subsets (CD4+ and CD8+) and cytokine (IL-2,IFN-?,IL-4 and IL-10) in every group were detected and measured. Results When the presentation of aGVHD was relatively conspicuous,CD4+ and CD8+ were increased, and the increase of CD4+ was predominant.Compared with the difference prior to transplantation, the difference was statistically significant (P

Objective To extract envelope of heart sounds exactly, for the purpose of the further analysis of its characteristics. Methods The way that envelope extraction of heart sounds based on key-points was given. The points of local peak and valley were calculated firstly, and then heart sound envelope was gotten by the interpolation of these points. Results Compared with the envelope extracted by Hilbert-transform and mathematical morphology, respectively, the outline of heart sounds was extracted more accurately, and its time-domain characters were acquired by this method. Conclusion The envelope of heart sound is extracted correctly by this method, which is useful for the further analysis.

Object To assess the genetic diversity among ten natural populations in Ophiopogon japonicus (L f ) Ker Gawl and O. bodinieri Levl (Liliaceae) in Sichuan Province Methods Forty random decamer primers were screened for Random Amplified Polymophic DNA (RAPD) fragments Results Based on cluster analysis of 515 DNA bands amplified by 11 primers, a DNA molecular dendrogram was established, and the relationship of the populations and seeds between O japonicus and O bodinieri was set up Conclusion 1 Distinct genetic differences and extensive genetic diversity are presented among the samples The genetic differences are related to morphological differences, but not to geographic regions; 2 Quite genetic differences are presented between O japonicus and O bodinieri, it suggests that they have farther relationships

Objective To explore the effects of forepaw sensorimotor deprivation in early life on hippocampus-dependent spatial reference learning and memory in rats. Methods Newborn SD rats were randomly assigned to experiment group (deprivation of forepaw sensorimotor function, n=53) and control group(n=55). Rats of postnatal day 13 (PN13) in experiment group were seleeted, and models of forepaw sensorimotor deprivation were established by microsurgical technique. Open field tests and Morris water maze tests were performed during the time periods of PN25(PN21-31), PN35 (PN31-39), PN45(PN41-50) and PN60(PN56-64) to evaluate the locomotor activity and spatial reference learning and memory, respectively. Results In open field tests, there was no significant difference in parameters of locomotor activity and exploratory behavior between the two groups (P>0.05). In Morris water maze tests, eontrol group performed significantly better than experiment group during training sessions and probe tests on PN25 and PN35 (P<0.05). While on PN45, although there was no significant difference between the two groups during training sessions, control group performed significantly better than experiment group during probe tests (P<0.05). Conclusion The deprivation of forepaw sensorimotor in early life has no signifieant effect on the locomotor activity and exploratory behavior of rats, but can impair the spatial reference learning and memory.

Objective To observe transient receptor potential melastatin 7-like (TRPMTL) expression changes post myocardial infarction (MI) in mouse cardiac fibroblast (CF).Methods TRPM7 expression and Ca2+ influx in CF from MI and control mice were quantified by mRNA RT-PCR and whole cell patch clamp technique. Results (1) TRPM7 expression was significantly upregulated post MI and Ca2+ influx of CF were significantly increased post MI [ (7.4 ± 0.7 ) pA/pF vs.( 16.2 ± 1.7) pA/pF, P < 0.01 ] and Ca2+ influx of CF increased 3-fold under lower pH condition; (2) These effects could be blocked by knock-out TRPM7 gene with SiRNA.conclusion TRPM7L upregulation post MI and under lower pH condition are responsible for increased Ca2+ influx in CF.

AIMTo determine 3 kinds of biologically active components (9 compounds) in Gardenia, namely geniposidic acid, gentiobioside, gardenoside, genipin geniposide, chlorogenic acid, crocin 1, crocin 2, crocin 3 and crocetin simultaneously and quantitatively.
METHODSAn HPLC method detected by 3 different UV-vis waves--240 nm for geniposidic acid, geniposide, gardenoside, genipin gentiobioside, 330 nm for chlorogenic acid, 440 nm for crocin 1, crocin 2, crocin 3 and crocetin respectively, has been developed: Phenomenex Luna C18 column(250 mm x 4.6 mm ID, 5 microns) with linear gradient elution using methanol-acetotrile (9:1) and 0.3% aqueous formic acid.
RESULTSThis method successfully determined 9 compounds in Gardenia (from 5 different areas) simultaneously and quantitatively.
CONCLUSIONThis is a more reasonable and credible quality control method for the Chinese traditional medicine.
