1.Quantitative evaluation of the hydroxyl radical level after global brain ischemia by intracranial microdialysis under stereotaxis

Objective To compare the strength of oxidative stress from different concentrations of oxygen administered by the accumulation of hydroxyl radicals during early reperfusion after global brain ischemia.Methods Sixteen adult male Mongolian gerbils with microdialysis probes implanted in the hippocampal CA1 were divided randomly (random number)into two groups (n =8 in each).All gerbils of both groups were subjected to 10 min bilateral carotid artery occlusion (BCAO).Then the following intervention:(a) immediate 30% O2 (near normoxia,NO group ) and (b ) immediate 100% O2 (hyperoxia,HO group).The accumulation of hydroxyl radicals (·OH)in hippocampus during reperfusion was estimated by measuring 2,3-dihydroxybenzoic acid (DHBA ) and 2,5-DHBA in microdialysis perfusate.Results Immediately after the onset of reperfusion,two groups showed markedly elevated DHBA,which returned to baseline gradually.Compared with the NO group,the HO group showed significantly higher peak DHBA and slower recovery.Conclusions Hydroxyl radical accumulation was more sensitive to inhalation of high concentration O2 during early reperfusion of global cerebral ischemia.

4.Effects of propofol on expression of death-associated protein ktnase mRNA in a rat model of traumatic brain injury

Objective To investigate the effects of propofol on death-associated protein kinase (DAPK) mRNA expression in a rat model of traumatic brain injury (TBI). MethodsSixty male Wistar rats with a mean age of 3-4 months and mean body weight of 250-300 g were randomly divided into 6 groups ( n = 10 each) : Ⅰ normal control;Ⅱ sham operation; Ⅲ TBI; Ⅳ normal saline (NS) +TBI;Ⅴ fat emulsinn+ TBI and Ⅵ propefol + TBI. TBI was produced according to Feeney et al. In group Ⅲ-Ⅵ the animals were killed and their brains were removed at 24 h after brain contusion. NS 2 ml·kg-1 · h-1 , 10% fat emulsion 2 ml · kg-1 · h-1 and propofol 2 ml·kg-1·h-1 were infused iv for 4 h after lead trauma in group Ⅳ, Ⅴ and Ⅵ respectively. DAPK mRNA expression in the brain tissue was determined by RT-PCR and neuronal apeptosis was assessed by TUNEL. Results The expression of DAPK mRNA and neuronal apoptosis were significantly increased in the TBI group as compared with normal control and sham operation groups. Propefol infusion significantly attenuated neuronal apoptosis and reduced DAPK mRNA expression as compared with TBI, NS and FE groups. ConclusionPropofol can protect the brain from traumatic injury by suppression of the expression of DAPK mRNA.

Objective To introduce an interpersonal relationship regulating technique in the static state of Qigong to enrich modern clinical psychotherapies. Methods Based on previous therapeutic experience, we introduce one specific technique of the Systematic Psychotherapy of TCM (SPT): Interpersonal relationship regulating technique in the static state of Qigong. Results Interpersonal relationship regulating technique in the static state of Qigong is a specific and practical technique of SPT, which has been gradually standardized and been mature with more than two decades of innovation and accumulation. Conclusion Interpersonal relationship regulating technique in the static state of Qigong has great clinical value.

8.Clinical analysis of 82 patients with myasthenic crisis

Objective To evaluate the precipitating factor,mortality,complications of myasthenia gravis crisis and risk factors of prolonged intubation.Methods Totally 82 case records of patients who had been adimitted for having 87 episodes of myasthenia crisis over a period of 15 years from January 1985 to February 2000 and their age, gender, precipitating factors, complications of intubation, and prolonged intubation factors were retrospectively reviewed. Results In 75% of myasthenic crisis patients having precipitating factors, infection was the most often served as the precipitating factors, and then the wound, fatigue, manopause and delivery came the next, 24% of precipitating factors were related to medication. Nine were identified with independent predictors of prolonged intubation. Among the 82 myasthenia crisis patients, 12 died, with a mortality of 14%.Conclusion The prevention and treatment for intubation complications may be the best way for further shortening intubation time and reducing the mortality.
9.Expression of AMCase mRNA in nasal mucosa of allergic rhinitis in SD rats.

OBJECTIVE:
To examine AMCase mRNA expression levels in nasal mucosa of allergic rhinitis SD rats.
METHOD:
Thirty SD rats were chosen and randomly divided into the experimental group and the control group. 20 in experimental group and 10 in the control group. AR model rats were established through repeated intraperitoneal shot of ovalbumin (OVA) for 2 weeks and consequently confirmed by local challenge with OVA for 1 week. The control group was treated by the same method with Physiological saline water instead of OVA. After the last excitation allergic rhinitis was diagnosed according to the accumulation score about nasal symptom. The septal mucosa of all rats were used to diagnose pathologically by HE dyeing. AMCase mRNA in nasal mucosa, obtained from the bilateral nasal mucosa in two groups, were used to do reverse transcriptive polymerase chain reaction. Real-time polymerase chain reaction was used to examine AMCase mRNA expression levels.
RESULT:
(1) The results showed definitely that there were positive expression of AMCase mRNA in normal nasal mucosa. This expression increase significantly during nasal allergy (P < 0.05). (2) The increased expression of AMCase mRNA in allergic rhinitis are related with the nasal symptoms score (r = 0.411, P < 0.05).
CONCLUSION
Increased expression of AMCase mRNA in nasal mucosa in allergic rhinitis model might play roles in the pathogenesis of AR, and Restrain the enzyme activity could become new treatment targets of allergic rhinitis.

10.Clinical and pathological features of Miyoshi myopathy with dysferlin protein deficient

Objective To investigate the clinical and pathological features of Miyoshi myopathy(MM) with dysferlin protein deficient. Methods The clinical and pathological data of the 3 patients with MM were analysed. Results 3 patients were onset at youngster.The clinical manifestation were myastheria and myoatrophy in distal of lower limbs.1 case combined myalgia and tumefaction in lower limbs at early stage of onset;1 case showed myathenia in proximal of lower limbs.The level of serum creatine phosphokinase (CK) was significantly ligher in the 3 cases (7543 IU/L, 5657 IU/L, 8721 IU/L respectively). The level of serum lactic dehydrogenase (LDH) was significantly higher in the 2 cases (456 IU/L ,636 IU/L respectively).The result of muscle pathology was showed myogenic damage in all the cases. The expression of dysferlin protain in membrane of muscle cells was completely deficient, although the expression of dystrophin was normal. Inflammatory cells infiltration was found in 1 case's muscle tissue. Conclusions The clinical characters of MM patient are onset at youngster,myasthenia and myoatrophy in lower limbs.The deficit of dysferlin protain can be found by pathology.
