1.Pharmacokinetics of gastrodin from Tiangou Jiangya capsule in rats.

The paper aims to study the pharmacokinetic parameters of gastrodin in rats effected by compound compatibilitiy and different doses of Tiangou Jiangya capsule. The extracts from Gastrodiae Rhizoma( equivalent to gastrodin 16.82 mg x kg(-1) and Tiangou jiangya capsule (equivalent to gastrodin 8.410, 16.82, 33.64 mg x kg(-1)) were oral administrated to rats respectively. The plasma were taken at various time points and treated with acetonitrile to measure the contents of gastrodin by HPLC method. The mean plasma concentration-time data were analyzed by 3P97 pharmacokinetic software and the pharmacokinetic parameters between groups were treated by SPSS 16.0. The results showed that gastrodin in rat was fitted to one-compartment model, Cmax and AUC of Tiangou Jiangya capsule were in direct proportion to oral administration, and t1/2Ka had nothing to do with doses, which indicated that gastrodin was fitted first-order rate transfter process in vivo. Morever, comparison with the Gastrodiae Rhizoma extract, isodose gastrodin in Tiangou Jiangya capsule showed a significant decrease for Cmax, Ke and increase for t1/2Ke, V/Fc, this indicated that compound compatibility can delay the absorbtion of gastrodin, prolong the resident time and promote the distribution in vivo, but its bioavailability is not significantly effected.

2.Observation of the effect of comprehensive rehabilitation on patients with stroke in different periods

ObjectiveTo observe the effect of comprehensive rehabilitation treatment on clinical nerve function deficiency (CNFD) and activities of daily living (ADL) of the patients with stroke. Methods45 stroke sufferers from different period were divided into three groups (1st group: in one month; 2nd group: one to six months; 3rd group: over six months). Before and after the comprehensive rehabilitation treatment was given, the CNFD, ADL and clinical effect were assessed.ResultsAfter comprehensive rehabilitation treatment, the CNFD of 1st group, and the ADL score of 1st group and 2nd group were improved significantly, and the 1st group's was better than the others. The total effective rate also increased in 1st and 2nd group. Conclusions Early rehabilitation treatment can improve the patient's neural motion function and the activities of daily living.
3.Protective effect of tetrahydroxystilbene glucoside on hippocampal neurons damage induced by glutamate in rats

ObjectiveTo observe the protective effect of tetrahydroxystilbene glucoside (TSG) on rats' hippocampal neuronal damage induced by glutamate (Glu) in the culture.MethodsHippocampus was isolated from newborn SD rats and dispersedly cultured in the medium for 9 days. Neurons were incubated with TSG (5—100μmol/L) for 24h, the cells were washed twice with Lock's solution without Mg2+,then Glu 500 μmol/L was added. Thirty min later, the reaction was terminated by washing the monolayer cells twice with the Lock's solution and then cultures were kept at 37℃ for 24h. Cell viability was measured by MTT method and cell membrane damage was determined by LDH leakage; with Fluo-3/AM as an intracellular calcium indicator and added into the bathing medium, fluorescent intensity of intracellular free calcium were observed through laser scanning confocal microscopy (LSCM).ResultsAfter the treatment with 5—100μmol/L TSG for 24h, the decrease of cell viability and the increase of LDH leakage caused by Glu was obviously resisted dose dependently. TSG inhibited increase of Ca2+ in cytoplasm, compared with model group.ConclusionTSG can significantly promote the cell viability and reduce the cell membrane damage in Glu treating hippocampal neurons. The neuroprotective activities of TGS is mediated by inhibiting Ca2+ overload in cytoplasm.
4.Osteoporosis in Graves′ disease and the characteristics of bone turnover markers

Objective To evaluate osteoporosis and the bone turnover markers in patients with Graves′ disease(GD) and the association of bone turnover markers including serum osteocalcin ( OCN ),I procollagen Nterminal propeptide( PINP),serum alkaline phosphatase ( ALP),and β- I collagen carboxyl-terminal peptide ( β-CTX)with thyroid hormone levels.Methods A total of 163 patients with Graves′ disease were enrolled.Serum thyroid hormone,OCN,PINP,and β-CTX levels were measured by the method of ethchemiluminescence.Serum ALP,calcium,phosphorus,and urinary calcium/creatinine ratio were measured by automatic biochemical system.Bone mineral density ( BMD ) of lumbar spines 1-4 ( L 1-4 ) and the proximal left femur was detected by dual energy xray absorptiometry.Results GD cases enrolled were composed of 43 men (26.4% )and 120 women ( 73.6% ),with an average age of ( 47 ± 1 ) years.There were 126 cases ( 77.3% ) with normal BMD,and 37 cases ( 22.7% ) with osteoporosis.OCN,ALP,PINP,β-CTX,and free T3 levels were significantly higher in the group of osteoporosis than those in the group with normal BMD( P<0.05 or P<0.01 ) after adjusting for sex,age,and BMI.The levels of OCN,P1NP,and β-CTX were positively correlated with serum FT3 level ( all P<0.01 ) and were also progressively increased according to the quartile groups of FT3 levels ( P<0.01 ) after sex,age,and BMI were adjusted.Conclusion Patients with Graves′ disease had a high incidence of osteoporosis with characristic high rate of bone turnover,which is related to the raised thyroid hormone levels.

Methods By using the nuclear fast red-crystal violet staining method and situ TUNEL technology, the quantitative changes of apoptosis in neural epithelium in gold hamster were analyzed. 10.Role of Ultrasound Contrast Time-intensity Curves in Hepatic Tumor Characterization and the Evaluation of Radiofrequency Ablation Effect

Purpose To explore the value of ultrasound contrast time-intensity curves in hepatic tumor qualitative diagnosis and evaluation of tumor ablation effect. Materials and Methods Forty-two patients who were clinically diagnosed as hepatic cancer or cirrhosis underwent contrast enhanced ultrasound for the tumor lesions within the liver, dynamic analysis of the lesions were executed with software SonoLiver CAP, the nature of the tumors were determined and the results were compared with those of the enhanced MRI. Ultrasound guided biopsy was followed with radiofrequency ablation treatment, two-dimensional and contrast enhanced ultrasound were executed two weeks after the ablation, and the results were compared with enhanced MRI findings during corresponding period. Results 52 tumor nodules were found with two-dimensional and color Doppler ultrasound, 65 (33 hyperplastic nodules, 32 malignant lesions) with CEUS, 65 (21 hyperplastic nodules, 44 malignant lesions) with ultrasound contrast time-intensity curves, and 61 (20 hyperplastic nodules, 41malignant lesions) with contrast enhanced MRI; pathological examination revealed that 26 of the lesions were benign and 39 were malignant;there was statistically significant difference (χ2=4.561, P<0.05) between the results of ultrasound contrast time-intensity curves and simple contrast enhanced ultrasound for the assessment of the nature of tumor;no statistically significant difference (χ2=0.003, P>0.05) was found between ultrasound contrast time-intensity curves and enhanced MRI for the assessment of the nature of tumor;there was statistically significant difference (χ2=5.298, P<0.05) between ultrasound contrast time-intensity curves and simple contrast enhanced ultrasound for the assessment of whether the tumor was malignant or benign, when taking biopsy results as the reference standard. Conclusion With ultrasound contrast time-intensity curves, dynamical, intuitive, and quantitative evaluation can be obtained for the perfusion differences between the lesion and the surrounding liver parenchyma, which provides more accurate information for qualitative diagnosis of hepatic tumors thus contains great application value in the evaluation of the effects of radiofrequency ablation for hepatic tumors.
