Iodine is the essential element of the human body. Both of iodine deficiency and iodine excess can produce different biological effects on organisms in various degree . It not only can damage the thyroid gland but also influence the nervous system and oxidize system, even damage hereditary material and immune system. The studies of the different biological effects of iodine deficiency and iodine excess on the cells and organisms can play an important role in unveiling the mechanisms of iodine deficiency disease and iodine excess disease. It also can offer the solution for preventing and curing the relevant disease of iodine.
2.Influence of aluminum hydroxide adjuvant on a murine model of allergic rhinitis.

OBJECTIVE:
To investigate the influence of aluminum hydroxide adjuvant on a murine model of allergic rhinitis (AR) and to confirm an appropriate method of establishing a mouse model of AR.
METHOD:
Establishing two types of BALB/c mice models of AR, one was identified as Local group which was characterized through intranasal sensitization and challenge using ovalbumin (OVA), and the other Systemic group which was made by intraperitoneal sensitization with OVA plus aluminum hydroxide and intranasal challenge through OVA. Then the numbers of sneezing and nasal rubbing were counted after the last challenge and the eosinophils in the nasal mucosa of mice models were observed and counted though Luna stain. Furthermore, morphological hyperplasia was examined in intraepithelial goblet cells and submucosal glands with HE stain. In addition, interlukin (IL) -4, IL-5, OVA specific IgE (sIgE) and interferon (IFN)-gamma in nasal lavage fluid (NLF) and serum of mice were examined u sing enzyme linked immunosorbent assay (ELISA).
RESULT:
The counts of sneezing and nasal rubbing in local group were more than those in systemic group and eosinophilia in the nasal mucosa of former group was greater than that in the latter one. Morphological hyperplasia was stronger in intraepithelial goblet cells and submucosal glands in local group compared with that in systemic group. Furthermore, the contents of IL-4, IL-5 and sIgE increased in the NLF and serum of mice of local group compared to those of systemic one. However, the production of IFN-gamma of mice in local group decreased when compared with that in Systemic group.
CONCLUSION
OVA plus aluminum hydroxide adjuvant may promote Th1 type immune response as well as Th2 response. OVA intranasal sensitization and challenge locally is an appropriate way in the establishment of AR mice models.

3.Abnormity rates of blood lipids and uric acid and their relationship analysis in health examination population in Yangzhou city

Objective To investigate the abnormity of blood lipids and uric acid ,and to analyze their relationship in health exami-nation population over 18 years in Yangzhou .Methods Totally 28 202 individuals receiving the health examination from October 2011 to August 2012 in the physical examination center of this hospital were enrolled in our study .Total cholesterol(TC) ,triglycer-ide(TG) ,low density lipoprotein cholesterol(LDL-C) ,high density lipoprotein cholesterol(HDL-C) and blood uric acid(UA) were detected ,and the relationship between age and sex with the abnormity of blood lipids and uric acid and the relationship between UA levels and blood lipids were analyzed .Results Among the detection population ,the detection rates of abnormal TG ,abnormal TC , abnormal HDL-C ,abnormal LDL-C and hyperuricemia were 31 .5% ,32 .1% ,15 .9% ,18 .4% and 17 .3% respectively .Compared with females ,males had the higher rates of abnormal blood lipids and UA (P<0 .05) .The rate of abnormal blood lipids was in-creased with age increase before 50 years old ,the > 50 -70 years old group had the highest abnormity rate of blood lipids ,then which was gradually decreased after 70 years old .The morbidity rate of hyperuricemia was increased with age .Compared with nor-mal UA group ,the hyperuricemia group had the higher rate of abnormal blood lipids (P<0 .01) .Conclusion The health examina-tion population over 18 years have the higher abnormity rate of blood lipids and uric acid ,which is significantly correlated with age and sex ;the increase of uric acid has certain correlation with abnormal blood lipids metabolism .Both blood lipids and uric acid are the risk factors of cardiocerebralvascular diseases ,rational dietary structure and periodic physical examination have the important significance to prevent the occurrence of these diseases .
4.Clinical characteristics of septic arthritis in patients with systemic lupus erythematosus: an analysis of 12 cases

Objective To analyze the clinical characteristics of systemic lupus erythematosus (SLE) patients with septic arthritis.Methods Twelve SLE patients with septic arthritis admitted to Peking Union Medical College Hospital from January 1990 to December 2012 were retrospectively analyzed.Results The median duration from onset of SLE to septic arthritis was 8.5 months (ranged from 1 to 120 months).All patients had received higher doses of steroid therapy (equal to prednisolone 1 mg ·kg-1 ·d-1) and immunosuppressant,and 5 of them had received methylprednisolone pulse therapy.The infectious manifestations included joint pain (all cases),swelling (5 cases),reduced joint function (8 cases) and fever (10 cases).All patients were oligo-arthritis.The hip and knee joints were most commonly involved.Three patients were infected by Salmonella,4 patients were infected by S.aureus,and 3 patients were infected by tuber culosis.Conclusion When SLE patients with long term steroid and immunosuppressant therapy developedacute joint pain and swelling,septic arthritis should be considered.Synovial fluid Gram stain,culture,blood culture and arthroscopy should be performed promptly.Appropriate antibiotics should be administered,and early treatment can improve the outcomes.
5.Functional analysis of Leptospira interrogans sphingomyelinase hemolysin genes and their transcriptional level alterations in the infected host cells

Objective To determine the hemolytic activity of products of sphingomyelinase hemolysin encoding genes of Leptospira interrogaas, and the transcriptional level alterations in the infected host cells. Methods By using genomic DNAs of pathogenic L. interrogans serovar serogroup Icterohaemorrhagiae serovar Lai strain Lai and serogroup Pomona serovar Pomona strain Luo, and non-pathogenic L. biflexa serogroup Sama-ranga serovar Patoc strain Patoc Ⅰ as templates, PCRs were performed to amplify entire sph1-sph4 genes. The amplified products were sequenced after T-A cloning. Prokaryotic expression systems of sph1-sph4 genes were re-spectively constructed, and the expressions of target recombinant proteins rSph1-rSph4 were examined by SDS-PAGE. Ni-NTA affinity chromatographic column was used to extract the expressed rSph1-rSph4. Hemolytic ac-tivities of rSph1-rSph4 on sheep blood agar plate were identified. Transcription alterations of sphl-sph4 genes in L. interrogans strain Lai after infected J774A. 1 cells were measured by real-time fluorescence quantitative RT-PCR. Results From genomic DNAs of both L. interrogans strain Lai and Luo, but not from that of L. biflexa strain Patoc Ⅰ , the target fragments of sph1-sph4 genes could be amplified. All the cloned sph1-sph4 genes had 100% nucleotide sequence identities compared to the corresponding reported sequences. The constructed pro-karyotic expression systems were able to efficiently express the target recombinant proteins rSph1-rSph4, respec-tively. All the rSph1-rSph4 had hemolytic activities, and among the four products rSph2 displayed the strongest hemolytic activity. After L. interrogaas strain Lai infecting J774A. 1 cells, the transcriptional levels of sph1-sph4 genes were remarkably up-regulated, especially for mRNA levels of sph2 and sph4 genes. Conclusion sph1- sph4 genes exist only in pathogenic L. interrogans species, and their products have hemolytic activity. The up-regulation of sph1-sph4 gene transcriptional levels in L. interrogans strain Lai after infected cells implies that the sphingomyelinase hemolysins may play important roles in the process of L. interrogans infection in hosts.
6.Protective Effect of Garlicin on Irradiation Injury in Mice

Objective To study the protective effect of garlicin on radiation injury in mice. Methods Sixty mice were randomly divided into five groups:model control group, solvent control group and garlicin treatment groups[0.17, 0.33, 1.00 g/(kg?d)]. After treated by gavage for 20 consecutive days, the mice were exposed to 60Co-? irradiation for one time and the effect of the irradiation on the amounts of WBC in peripheral blood, the ratio of micronucleus cells in marrow, the content of DNA in marrow cells and the superoxide dismutase (SOD) activity were measured. Results Compared with the exposure group, garlicin increased the amounts of WBC, decreased the micronucleus frequency of marrow cells, increased the content of DNA in marrow cells and increased the SOD activity in mice. Conclusion Garlicin shows a protective effect on irradiation injury in mice.
7.Quantification of posterior capsular opacification after cataract surgery

AIM: To evaluate posterior capsular opacification (PCO) with hydrophobic acrylic intraocular lens (IOL, Sensar AR40e) and silicone IOLs after cataract surgery, to use a software program developed to provide an objective assessment of the amount of PCO in the digital images of the posterior capsule to quantify PCO. METHODS: Ninety-eight eyes underwent standardized phacoemulsification and "in the bag" IOL placement, were randomized to receive a three piece lens of hydrophobic acrylic or silicone, but lens materials were different in one case. In year 1 and 2, digitized retro-illumination images were taken from the posterior capsule. Images were analyzed by POCO software program, removing the Purkinje light reflexes, contrast enhancement, filtering to enhance low-density PCO. RESULTS: The percentage of PCO were 0.32±0.13 of hydrophobic acrylic IOLs in year 1, compared with 0.39±0.17 of silicone (P=0.37). In year 2, the percentage of PCO were 0.42±0.20 with hydrophobic acrylic IOLs and 0.34±0.18 with silicone IOLs (P=0.50). Of those patients with PCO in year 1 and 2, severity grades were 0.50±0.30 and 0.82±0.58 of hydrophobic acrylic cases, compared with 0.63±0.35 and 0.55±0.35 of patients with silicone IOLs (P=0.52,P=0.69) with no statistical significance.CONCLUSION: The POCO system is capable of producing an objective and repeatable measure of PCO that is relevant to assessing techniques of PCO prevention.
8.Treatment of allergic rhinitis with normal saline nasal irrigation at different temperature.

OBJECTIVETo investigate the influence of normal saline nasal irrigation with different temperature on allergic rhinitis (AR) and to confirm the appropriate temperature of nasal irrigation.
METHODSSixty patients with AR were randomly divided into 3 groups (according to random number table) and received 15 °C, 25 °C or 40 °C normal saline nasal irrigation respectively, and 20 healthy controls received no treatment. Visual analogue scale ( VAS) was used to assess the symptoms among the control group and the various intervention groups and the enzyme linked immunosorbent assay (ELISA) was applied to detect the contents of histamine, leukotriene C4 (LTC4), house dust mite D1 specific IGE (sIGE) and eosinophil cationic protein (ECP) among the control group and the various intervention groups. GraphPad Prism 5 software was used to analyze the data.
RESULTSThere was no statistical difference between 15 °C normal saline nasal irrigation group and 25 °C normal saline nasal irrigation group. Pre-intervention and post intervention whether is VAS scores of rhinorrhea, sneezing, nasal obstruction and nasal pruritis or in contents of histamine, LTC4, sIgE and ECP showed no difference (all P > 0.05). However, the symptoms of (sneezing VAS score: 3.765 ± 0.291; nasal obstruction VAS score: 3.529 ± 0.365). The difference was significant (t = 4.678, P < 0.001; t = 3.901, P < 0.01). The contents of histamine [ (21.78 ± 0.62) ng/ml] and LTC4 (17.43 ± 0.67 ng/ml) were also decreased after intervention of 40 °C normal saline nasal irrigation [ histamine content: (18.82 ± 0.52) ng/ml; LTC4 content: (13.14 ± 0.59) ng/ml ]. The difference was significant (t = 3.632, P < 0.01; t = 4.79, P < 0.001). Nonetheless, there were no statistical differences in VAS scores of rhinorrhea (t = 2.02, P = 0.051) and nasal pruritis (t = 1.984, P =0.056) and in contents of sIgE (t = 1.891, P = 0.066) and ECP (t = 2.021, P = 0.05). There were statistical differences between the 40 °C group and the 25 °C group normal saline nasal irrigation group in symptoms of sneezing (t = 2.060, P = 0.048) and nasal obstruction (t = 2.1053, P = 0.048). and inflammatory factors of histamine (t = 2.078, P = 0.045) and LTC4 (t = 2.149, P = 0.038). In symptoms of rhinorrhea and nasal pruritis and local production of sIgE and ECP, there was no statistical difference (all P > 0.05).
CONCLUSION40 °C normal saline nasal irrigation can improve the symptoms of sneezing and nasal obstruction and can alleviate the local levels of inflammatory factors like histamine and LTC4 in AR patients, and is the appropriate irrigation temperature.

9.The expression of TXNDC5 in synovial membranes, synovial fluid and blood of patients with rheumatoid arthritis

Objective To investigate the expression of thiredoxin domain containing 5 (TXNDC5) in the synovial tissues and blood samples of various arthritic conditions and autoimmune diseases to further confirm the previous findings, investigate the relations between the expression level of TXNDC5 and clinical parameters of RA. Methods The expression of TXNDC5 in the synovium was quantitatively analyzed by immunohistochemistry, real-time quantitative PCR and Western blotting. The levels of TXNDC5 in blood and synovial fluid was determined using sandwich ELISA in patients with RA, osteoarthritis (OA), systemic lupus erythematosus (SLE), ankylosing spondylitis (AS) and normal controls. One-way ANOVA, LSD test and Spearmen' s correlation were used for statistical analysis. Results Immunohistochemistry indicated that TXNDC5 expression was significantly higher in the synovial tissues of RA (100%, 40±9) than in those of OA and AS(200%,4±4). Real time PCR and western blotting confirmed the above findings (P<0.01). Sandwich-ELISA indicated significantly elevated level of TXNDC5 in the blood and synovial fluid of patients with RA (A=1.31±0.37), but not in those of OA, SLE, and AS, the healthy controls (P<0.01). The level of TXNDC5 in the blood of RA patients (A=0.8185±0.299) was positively correlated with the level of anti-CCP (r=0.350, P =0.027). Conclusion The results suggest that the pronounced increase of TXNDC5 expression may stimulate synovial pannus formation in the hypoxic environment of RA.
