3.The expression of BMP-2 and BMP-6 in rat facial nerve motorneurons after facial nerve injury

?Objective: To study the effects of endogenous bone morphogenic protein 2(BMP 2) and 6(BMP 6) in rat facial nerve after injury. Methods: Facial nerve trunk was cut and then anastomosed in 30 adult male Sprague Dawley rats,another 6 rats without operation were used as the controls. Every 6 rats were killed 6 ,72 h,1, 2 and 4 weeks respectively after operation and nerve specimens were immunohistochemically examined for BMP 2 and BMP 6 expression. Result:In the control nerve the gray level of BMP 2 and BMP 6 were 210.89?8.21 and 232.03?9.25 respectively. 6, 72 h,1, 2 and 4 weeks after operation the gray level of BMP 2 were 242.83?11.01,240.67?7.91,234.46?5.28,232.12?7.27 and 220.71 ?10.19;that of BMP 6 210.75?5.19,204.08?10.85,198,91?8.58,186.37?4.11 and 184.62? 8.45 ,respectively. Conclusion: BMP 2 may play a role in the early irritable actions when facial nerve was injured. However, BMP 6 might have functions in the regeneration of facial nerve during later period after injury.
4.Probability of premature mortality due to four chronic diseases in Jiangmen City from 2012 to 2021

Objective:
To investigate the mortality of malignant tumors, diabetes, cardio-cerebrovascular diseases and chronic respiratory diseases and trends in probability of premature mortality due to these four chronic diseases in Jiangmen City, Guangdong Province from 2012 to 2021, so as to provide the evidence for perfecting the chronic disease control strategy.
Methods:
The mortality of malignant tumors, diabetes, cardio-cerebrovascular diseases and chronic respiratory diseases among household registered residents in Jiangmen City from 2012 to 2021 were collected from the Guangdong Provincial Population Death Information Registration Management System, and the crude mortality, standardized mortality by the population of the Fifth National Population Census in China in 2000 and probability of premature mortality were calculated. The trends in mortality and probability of premature mortality were analyzed using average annual percent change (AAPC), and whether achieving the targets for the probability of premature mortality due to four chronic diseases in 2025 and 2030 were evaluated.
Results:
A total of 226 012 deaths occurred due to four chronic diseases in Jiangmen City from 2012 to 2021, and the overall crude mortality and standardized mortality rates were 569.22/105 and 283.29/105, with a reduction in the probability of premature mortality from 15.04% to 12.05% (AAPC=-2.403%, Z=-7.603, P<0.001). The probability of premature mortality due to four chronic diseases decreased from 19.21% to 16.38% in males, and from 10.42% to 7.58% in females (AAPC=-1.893% and -3.085%, Z=-5.262 and -9.811, both P<0.001). The probability of premature mortality due to diabetes appeared a tendency towards a rise (AAPC=2.317%, Z=2.548, P=0.034), and the probability of premature mortality due to cardio-cerebrovascular diseases showed a tendency towards a decline (AAPC=-4.826%, Z=-13.590, P<0.001), while no significant changing trend was seen in the probability of premature mortality due to malignant tumors or chronic respiratory diseases (AAPC=-0.751% and -2.461%, Z=-1.532 and -1.730, P=0.125 and 0.122). The predicted probability of premature mortality due to four chronic diseases was 10.92% in 2025 and 9.66% in 2030 in Jiangmen City, which were both lower than the target (11.21% and 9.81%).
Conclusions
The probability of premature mortality due to four chronic diseases appeared a tendency towards a decline in Jiangmen City from 2012 to 2021, which can reach the target in 2025 and 2030. Males should be given a high priority for interventions of chronic diseases, and diabetes control should be reinforced.
5.Variable pulsed Nd:YAG laser and intense pulsed light for the treatment of port wine stain in animal models

ObjectiveTo study the therapeutic effect of variable pulsed Nd∶YAG laser and intense pulsed light (IPL) on port wine stain(PWS) in an animal model.MethodsThe combs of 38 Leghorn chickens served as the animal model of PWS,which were randomly classified into 4 groups:blank control group receiving no irradiation,Nd:YAG group receiving Nd∶YAG laser irradiation for 2 sessions,IPL group receiving IPL irradiation for 2 sessions,and combination group receiving 1 session of Nd∶YAG laser irradiation followed by 1 session of IPL irradiation 45 days later.One week after the final irradiation,the rnorphology of combs was observed,and tissue specimens were resected from the combs for the analysis of histological changes and blood vessel density.The data were assessed by using one-way ANOVA.ResultsAfter 2 sessions of irradiation,the color of comb skin was lightened,and light microscopy showed the thickening of vascular layers in the dermis,blockage of blood vessels and decrease in the number of blood vessels.The count of blood vessels per high power field(× 400) was significantly lower in the Nd∶YAG group(17.92 ± 3.63),IPL group(8.08 ± 1.56)and combination group(7.08 ± 1.31) than in the control group(37.08 ± 3.97,all P < 0.01 ),but was similar between the combination group and IPL group (P > 0.01),and higher in the Nd∶YAG group than in the combination group(P < 0.01 ).ConclusionsVariable pulsed Nd∶YAG laser and IPL,alone or in combination,can be used in the treatment of PWS,however,the effect of IPL alone or in combination with Nd:YAG seems superior to that of Nd∶YAG laser alone.
6.Feasibility of real-time triplane strain rate imaging for quantitative assessment of left atrial function in normal subjects

Objective To evaluate the feasibility of real-time triplane strain rate imaging for the quantitative assessment of left atrial function in normal subjects. Methods Totally 40 healthy volunteers were enrolled. Left atrial expansion index (LAEI), left atrial passive emptying fraction (LAPEF) and left atrial active emptying fraction (LAAEF) were measured using real-time triplane volume analysis. Mean systolic peak strain rate (MSRs), mean early diastolic peak strain rate (MSRe) and mean late diastolic peak strain rate (MSRa) were measured using real-time triplane strain rate imaging. The results were compared respectively. Results MSRs correlated positively with LAEI (r=0.61, P<0.01). MSRe correlated inversely with LAPEF (r=-0.67, P<0.01), so did MSRa with LAAEF (r=-0.78, P<0.01). Conclusion Real-time triplane strain rate imaging is feasible for the quantification of left atrial function in normal subjects.
7.Effect of carboxymethyl chitosan on ATP-sensitive K~+ currents in single ventricular myocyte of rat

Objective To investigate the effect of carboxymethyl chitosan on ATP-sensitive K+ currents(IKATP) in single ventricular myocyte of rat.Methods Single ventricular myocyte of rat was isolated with enzymatic dissociation technique.Whole-cell patch clamp technique was used.The stimulating protocol was:holding potential-40 mV,command potential-100 mV to +50 mV,step potential + 10 mV,duration 200 ms,stimulating interval 6 s.Results Carboxymethyl chitosan inhibited the ATP-sensitive K+ channel(KATP channel) in a dose-dependent manner.When holding potential was-60 mV and stimulating potential was + 50 mV,carboxymethyl chitosan(0.1%,0.2%,0.3%) decreased the amplitude values of IKATP to 66.1%?9.9%,50.3%?13.3%and 39.8%?9.5%(n=7)of control,respectively.The changes of IKATP under other command potentials were consistent with this.Conclusion Carboxymethyl chitosan inhibits KATP channel in single ventricular myocyte of rat in a dose-dependent manner,and may be one of the mechanisms of antiarrhythmia.
8.CLONING OF FULL LENGH cDNA AND EXPRESSION OF RAT SMALL GTP-BINDING PROTEIN TC10 IN FACIAL NUCLEUS INDUCED BY INJURY

Objective To explore the expression of small GTP-binding protein TC10 during nerve injury and construct eukaryotic expression vector carrying rat TC10 cDNA. Methods Total RNA was extracted from rat facial nucleus, and the change of rat TC10 full-length cDNA was investigated by RT-PCR. PCR product was inserted into eukaryotic expression vector pcDNA3 to construct the pcDNA 3-TC10. The plasmid was identified by restriction enzyme analysis and sequencing analysis. Results After injury, the expression of TC10 rapidly increased, Restriction enzyme assay and sequencing assay showed that the construct we got was rat TC10 full-length cDNA construct.

10.Study of Immobilization of Yeast Alcohol Dehydrogenase Cross-linked Magnetic Chitosan Microspheres

Magnetic chitosan microspheres(M-CS) were prepared and used for yeast alcohol dehydrogenase(YADH) immobilization.The optimum technology and the properties of immobilized YADH were studied.The optimal immobilization conditions for YADH were:20ml of 0.25mg/ml of YADH in phosphate buffer(0.05mol/L,pH 7.0) reacted with 50mg of magnetic M-CS at 4℃ for 2h.The microspheres were characterized by transmission electron microscopy,the results showed that M-CS were regular sphere with an average size of 30nm and had magnetic response characteristic.The M-CS suspended in H2O solution was easily precipitated and separated by magnetic field.Mechanical strength and crosslinking degree of M-CS were influenced by the ratio of carrier and immobilized YADH,ion concentration in phosphate buffer and pH of the solution.The immobilization was slightly influenced by the reaction temperature.The immobilization would improve its thermal,basic resistant and acid resistant stability.After the immobilized enzyme was kept between 35℃ to 75℃ for one hour,it still had 70 % of initial enzyme activity,when it was kept pH between 5.0 to 7.4 for one hour,it still had 80% of initial enzyme activity.The optimal reaction temperature of the immobilized enzyme was 40℃ compared to 30℃ of the free YADH,the optimal reaction pH of the immobilized enzyme was 6.8 as same as one of the free enzyme.Storaged at the temperature of 4℃ for 30 days without any protection by reagent,the free enzyme only kept 26% of the original activity but the immobilized enzyme still retained 60% of the activity.The immobilized enzyme maintained 70% of the activity after circular use 5 times.The Km value of immobilized YADH for Pyruvate was 2.58 mmol/L compared to 3.31 mmol/L of the free YADH,it would reduce its appetency for the substrate.
