Objective To explore distribution and genetypes of plasmid-mediated quionlone resistance genes qnrA.qnrB and qnrS in Enterobacteriaceae isolates iu Renmin Hospital of Wuhan University.Methods The qnrA,qnrB and qnrS genes in Enterobaeteriaceae isolates including nonrepetitive 129 isolates of E.coli.13 isolates of E.cloacae and 37 isolates of k pneunoniae were detected by PCR.Antibiotic suseeptibility testing for 15 antibiotics were also performed by K.B in virto.MICs of ciprofloxacin were determined by agar dilution methed.Plasmid conjugatable test wag applied to examine whether qnrA,qnrB and qnrS genes were located in conjugatable plasmid.For qnr-positive strains,integrase I and SHV-1,TEM-1,CTX-M,OXA-I,OXA-Ⅱ,OXA-Ⅲ,DHA and EBC β-lactamases genes were examined.Results 25 qnr-positive isolates were detected among 179 Enterobaeteriaceae isolates,including 6 qnrA-positive isolates,9 qnrB-positive isolates and 10 qnrS-positive isolates,which were presented in 3.35%.5.02%,and 5.59%respectively.All positive isolates were susceptible to imipenem but resistance to some other drugs.2 qnrA-positive isolates and 4 qnrB-positive isolates of them were susceptible to Quinolones.The plasmid wag eonjugatable in 2 qnrA-positive isolates and 4 isolates carrrying qnrB and qnrS.23 qnrA-positive isolates harbored type 1 Integrase,but one isolate with both qnrB and qnrS did not carry Type 1 Integrage.14 isolates of them were TEM-1 producing strains.6 isolates were OXA-Ⅲ-producing strains.and 7 isolates of them were EBC-producing strains.Conciusions In HuBei province,a low prevalence of qnrA,qnrB and qnrS wag determined in Enterebacteriaceae isolates.Muhidrng-resistance was found in qnr-pesitive isolates and qnr genes were detected in quinolone susceptible strains.Extendedspectrum β-lactamages could be presented in qnr-positive isolates too.

BACKGROUND: Different processes of thinking may lead to the same results, which represent one of the forms of the complexity of the human brain.OBJECTIVE: To analyze the similarity in the results of thinking and difference in the thinking process by means of questionnaires.DESIGN: A comparative analysis of the answers in the final term examination with the difference in answers analyzed by x2 test with contingency table.SETTING: Department of Physiology, Medical College of Henan University of Science and Technology.PARTICIPANTS: This study was conducted between June and November,2004, involving totally 300 volunteered second-year medical students (150male and 150 female, aged 20-22 years) of the medical school of Henan Science and Technology University who took their final examination in physiology.METHODS: The first "question for thinking" in the final examination was taken as an example, and the papers with full score for this question (completely correct group, n=42) and zero score (totally wrong group, n=49)were chosen, from which one paper was randomly selected from each group. The first 10 words in the answer to the question was used as the control and compared with the answers of the other papers word by word.Each same word was given a score of 1 and otherwise a score of 0, and the number of the same words and different words were counted for comparison. Meanwhile the total words of the answer were also counted and compared. The difference in the answers was analyzed with x2 test with contingency table.MAIN OUTCOME MEASURES: The difference in the first 10 words between completely correct group and totally wrong group.RESULTS: Totally 42 students in the complete correct group and 49 in totally wrong group were included in the final result analysis. The number of the total words of the answer was different between the students in spite of the same scores. The first 10 words of the chosen paper in completely correct group was significantly different from those in the other 41 papers (x2=270.978, P ＜ 0.01); there was also significant difference in the first 10words between the chosen paper and the other 41 papers in totally wrong group (x2=285.153, P ＜ 0.01).CONCLUSION: Longitudinal thinking as well as lateral thinking processes are different between persons.

BACKGROUND: Previous research has demonstrated that bone marrow mesenchymal stem calls (BMSCs) differentiate into varying mesoderm-derived mesenchymal cells, such as myocardial cells.OBJECTIVE: To investigate the feasibility of construction of engineered myocardial tissues with polylactic-co-glycolic acid (PLGA) olymer and cardiomyocyte-like cells derived from BMSCs in vitro.METHODS: BMSCs were isolated from bone marrow of SD rats by density gradient cantrifugation. The third passage cells were induced with the culture medium including 5-aza for 4 weeks. After successful induction, the cells were trypsinized and suspended, then the cell suspension was added to the polylactic-co-glycolic acid square slowly and cultured in the incubator for 14 days. The morphological changes were observed before and after induction under phase contrast microscope. The cardiomyocyte-like cells were identified by immunofluorescanca staining. The naked eyes were used to observe the morphology of engineered myocardial tissues. The ultrastructures of the engineered myocardial tissues were viewed with a transmission electron microscope.RESULTS AND CONCLUSION: BMSCs of primary culture formed cell colonies at 14 days. The passaged cells were larger than those of primary culture. After induction by 5-aza, the cells presented long spindle and aligned in parallel. The expression of specific proteins of cardiac troponin Ⅰ (cTnl) in induced BMSCs was positive. Transmission electron microscopy showed that the engineered myocardial tissues had myofilaments, Z line-like substances. With PLGA and cardiomyocyte-like cells derived from BMSCs, the engineered myocardial tissues which resemble native cardiac tissues can be successfully constructed in vitro.

Objective　To investigate the effect of turning scleral flap of self-body over drainage in trabeculectomy.Methods　The surgery was performed on one eye of a rabbit while　the other eye of the same rabbit was conducted with the routine trabeculectomy as the control group. The filtering bleb, intraocular pressure and pathology were observed. Results　The intraocular pressure in the experimental eye was much lower than that in the control group 3～12 weeks after the operations (P＜0.05).As it was within 2 weeks after the surgeries, but there was no significant filtering difference (P＞0.05).The rate of eyes with functional bleb in the experimental group was significantly higher than that in the control group at the postoperative 2～7 weeks .Conclusion　This study suggests the turning of self-body over drainage surgery can help to maintain filtering function for a long time and to prevent complication after filtering surgery.

Objective To investigate the effects of arginine combined with leptin in treatment of rats with a-cute pancreatitis. Methods Sixty Wister rats were randomly divided into sham operation group (group A), acute pancreatitis group (group B),arginine group (group C),leptin group (group D),arginine plus leptin group (group E),with 12 rats in each group. Acute panereatitis model was developed by retrograde injection of 5% sodium tauro-cholate into cholangiopancreatography in rats. After 6 hours, medicine were given by intraperitoneal injection, ab-dominal aortic blood and pancreatic specimens were taken for measurement of tumor necrosis factor-α(TNF-α), ser-um amylase,nuclear factor-κB (NF-κB) activity of pancreatic tissue, and pathological damage score of pancreatic tissue. Results TNF-α,Amylase,NF-κB activity, pathologic scores were(1.206±0.446)μg/L, (1617.2±18.8) U/L,0.174±0.013,0.6±0. 2 in group A;(3.201±0.321) μg/L, (4643.8±303.4) U/L,0.397±0. 025,7.8±1.3 in group B;(1.845±0.404) μg/L,(3370.9±173.2)U/L,0.255±0.026,3.9±1.1 in group C;(1.996±0.374) μg/L, (3693.7±208.0)U/L,0.274±0.062,4.2±1.2 in group D;(1.440±0.287)μg/L, (2501.0±771.4)U/L,0.211±0.027,2.3±1.0 in group E. Compared with group A, the indicators were significantly in-creased in group B (P < 0.05). Compared with group B, these indicators were reduced in group C and D (P <0.05). There was no significant difference between group C and group D. These indicators in group E were signifi-cantly lower than those in group C and D (P < 0.05). NF-κB in pancreatic tissue were positively correlated with other indicators(P <0.01). Conclusions In treatment of acme pancreatitis in rats, arginine combined with leptin has idenditfied effect, and the effect of combination therapy is better than that of separate medicine.

Objective To analyze the distribution and antimicrobial susceptibility of the pathogenic isolates during 2009-2014 for rational use of clinical antibiotics.Methods The clinical data were reviewed and analyzed retrospectively to examine the distribution and resistance profile of the pathogenic isolates.Strain identification and antimicrobial susceptibility test were conducted by BioMerieux VITEK-2 automatic microbiology analyzer and the associated identification cards.All results were interpreted according to CLSI standards.Results A total of 6 393 strains of pathogenic bacteria were isolated during the six-year period.The top three pathogens were Escherichia coli (33.6 ％),Enterococcus faecalis (19.4 ％),and Streptococcus agalactiae (13.7 ％).In addition,8,1,and 10 strains ofListeria spp.were isolated in 2009,2010,2014,respectively.A total of 40 strains of anaerobic bacteria had been isolated since 2012.The isolates were mainly from genital tract (53.7 ％) and uterine cavity (21.3 ％),followed by blood stream (8.2 ％),including venous blood samples (6.1％) and umbilical cord blood samples (2.2 ％).There were 165 (2.6 ％) cases of surgical site infections.Escherichia,Enterococcus,and Streptococcus were the top three bacteria isolated from both uterine cavity and venous blood samples.The top three bacteria isolated from surgical sites were Escherichia,Enterococcus,and Staphylococcus.The bacteria isolated from umbilical cord blood samples were mostly Streptococcus,Escherichia,and Enterococcus.Anaerobic bacteria were mainly isolated from blood samples,which accounted for 82.5 ％ of all anaerobic bacteria.Listeria monocytogenes isolates were mainly from venous blood samples or uterine cavity.Overall,the antimicrobial susceptibility profile of the pathogenic isolates was relatively stable during the period from 2009 to 2014.Most antibiotics showed good activity against these isolates.Conclusions The pathogens isolated from this gynecology and obstetrics hospital during 2009-2014 period are mainly Escherichia coli,Enterococcusfaecalis,and Streptococcus agalactiae.Most isolates were susceptible to the commonly used antimicrobial agents.In addition to a few MRSA strains,ESBLs-producing Escherichia coli isolates were the main multidrug resistant organisms.

In order to study the therapeutic time window of dimethylaminoethyl ginkgolide B mesylate(XQ-1H) in the permanent focal ischemia of rat,we used the rat model of the permanent middle cerebral artery occlusion (pMCAO).Doses of 15.6,7.8 and 3.9 mg/kg of XQ-1 H were intravenously administered at 0.5,1,2,3 h after MCAO,respectively.Neurological scores,infarct sizes,water contents and pathological changes in each interval were determined at 72 h after MCAO.It was observed that XQ-1 H administered at 0.5 and 1 h after MCAO significantly reduced the cerebral infarct size and edema,and produced significant reductions in the neurological deficits.The protective effect of XQ-1H on the neuron cells was proved by pathological observations.In addition,the contents of MDA,lactate,and the activities of SOD were measured.Reduction in the contents of MDA and lactate and enhancement in the activities of SOD were attributed to the pretreatment of XQ-1H at 0.5 and 1 h.Our results showed that the therapeutic time window of XQ-1H extended for up to 1 h after MCAO.

Objecfive To investigate the association between the exon 2,3,4 of MHC class Ⅰ chain-related gene-B(MICB)and ulcerative colitis(UC)in Chinese Han.Methods Using polymerase chain reaction single-stranded conformation polymorphism,allele frequency of MICB exon 2,3 and 4 in 105 patients with UC and 213 age and sex matched healthy controls were genotyped.All of the studied individuals were Chinese Han.Results Allele frequency of MICB 0106 was increased in patients with UC as compared with normal controls(19.0%vs 8.9%,P=0.000,Pc＜0.001,OR=2.402,95%CI:1.488-3.879).The frequency of MICB 0106 was increased significantly in patients with extensive colitis (24.4%vs 8.9%,P=0.000,Pc＜0.001,OR=3.294,95%CI:1.800-6.027),moderate and severe disease(24.1%vs 8.9%,P=0.000.Pc＜0.001,OR=3.294 95%CI:1.893-5.576)and in those with extra intestinal manifestations(20.5%vs 8.9%,P=0.002,Pc=0.012,OR=2.626,95%CI:1.418-4.861).Furthermore,MICB 0106 allele was higher in frequency in the male patients with UC (22.1%vs 8.0%,P=0.001,Pc=0.006,OR=3.276,95%CI:1.737-6.178)and the patients more than 40 years old(28.8%vs 8.3%,P=0.000,Pc＜0.001,OR=4.500,95%CI:2.381-8.504)as compared with healthy controls.Conclusion MICB 0106 allele is positively associated with UC,especially with extensive colitis,moderate and severe disease,presence of extra intestinal manifestations,male gender and age of more than 40 years in Chinese Han in Hubei province.
