By using gene SOEing PCR, Arg (CGC) in the expression fragment of chitinase gene (chi58) from Chaetomium cupreum was mutated synonymously to Arg (AGA), which is a bias code of Pichia pas-toris yeast. The expression plasmid pPIC9K-chi58A was constructed and transformed into GS115 strain through electroporation. The chitinase activity reached 101.71 U/mL?3.33 U/mL after induced with 120 h, which was the 3 fold of the original strain (31.83 U/mL?4.85 U/mL). Additionally, recombinant yeast showed good genetic stability after 10 cycle. SDS-PAGE analysis suggested that the weight of protein was 58 kD.

Small and medium-sized enterprises (SMEs) are important components in Chinese medicine industry. However, the lack of big brand is becoming an urgent problem which is critical to the survival of SMEs. This article discusses the concept and traits of Chinese medicine of big brand, from clinical, scientific and market value three aspects. Guided by market value, highlighting clinical value, aiming at the scientific value improvement of big brand cultivation, we put forward the key points in cultivation, aiming at obtaining branded Chinese medicine with widely recognized efficacy, good quality control system and mechanism well explained and meanwhile which can bring innovation improvement to theory of Chinese medicine. According to the characters of SMEs, we hold a view that to build multidisciplinary research union could be considered as basic path, and then, from top-level design, skill upgrading and application three stages to probe the implementation strategy.

Major changes and updates of Clinical and Laboratory Standards Institute (CLSI) document M100-S22 for performance standards for antimicrobial susceptibility testing were introduced in the article,which includes ( 1 ) general changes; (2) changes of drugs recommended for testing and reporting;( 3 ) changes of interpretive criteria ( breakpoints ) and comments ; ( 4 ) changes of quality control and others; and (5) changes of appendixes and glossaries.

Mitogen activated protein kinase(MAPK)is one of the four biggest signal transduction systems which contain four subtribes named p38,ERK5/BMK1,ERK and JNK/SAPK respectively.Previous studies have shown that MAPK pathway is involved in growth,cell differentiation,perishing,the synchronization of cell function and so on.ERK/MAPK,one of the members of MAPK family,and Sma-and MAD-related(Smad)play important roles in the proliferation of fibroblast,modulating inflammation mediator production as well as transferring factors activity in the process of pulmonary fibrosis(PF).This review focuses on relationship between Smads and ERK in the pathogenesis and progression of PF.

Objective To screen and clone the target genes transactivated by nonstructural protein 5A (NS5A) of hepatitis C virus (HCV). Methods The mRNA was isolated from HepG2 cells transfected with pcDNA3.1(-)-NS5A and pcDNA3.1(-) empty vector, respectively. Suppression subtractive hybridization (SSH) method was employed to analyze the differentially expressed DNA sequence between the two groups. The obtained sequences were searched for homologous DNA sequence from GenBank. The new gene with no homology with known genes in this database was confirmed, and electric polymerase chain reaction was conducted for cloning the full-length DNA of the new gene and in conjunction with Kozak rule and the terminus of polyadenyl signal sequence. The reverse transcription PCR (RT-PCR) was used to amplify the new gene from mRNA of HepG2 cell as the template. The coding sequence of the new gene was deduced according to the nucleotide sequence. Results A new gene with unknown function was named as NS5ATP3. The nucleotide sequence of the NS5ATP3 gene and its corresponding amino acid have been determined, which contained 1 572nt and 524aa. The sequence of the NS5ATP3 gene was deposited into GenBank, with the accession number AF529364. Conclusions NS5ATP3 gene transactivated by HCV NS5A protein was cloned and identified successfully by combining molecular biological technology and bioinformatics technique. These results will pave the way for the study of the molecular mechanism of the transactivating effects of HCV NA5A protein and the development of new therapy for chronic hepatitis C.

[Objective]To evaluate the possibility and clinical value of total en bloc spondylectomy for the treatment of spinal tumors. [Method]Four cases of primary tumor of the thoracolumbar spine were treated with total en bloc spondylectomy TES through a single posterior approach performed in three cases(T10 solitary plasmacytoma,T8 hemangioma,and T12 hemangioma),and one case through a single stage anterior and posterior combined approach(L4 neuroblastoma with paraspinal mass).Improvements in osteotomy apparatus and surgical technique of TES were investigated.[Result]All patients attained significant clinical improvement after surgery without any severe complications.Patients were observed for 13 to 22 months.There was no local recurrence and distal metastasis by the last follow-up.[Conclusion]Total en bloc spondylectomy includes resection of the involved vertebra in two major blocs,rather than a piecemeal pattern.It is one of the most effective therapies for spinal tumors.Howerer,some components of TES remain to be further improved.

Diminished apoptosis,due to overexpression of antiapoptotic proteins, is involved in tumorigenesis and chemoresistance. Antisense oligodeoxynucleotide (ASODN) has been used to inhibit the expression of antiapoptotic proteins(f.g bcl 2, bcl x L , survivin),aimed to induce apoptosis or enhance chemosensitivity.The combination of ASODN with conventional treatment is possibly a reasonable approach in furture cancer therapy. Objective To evaluate gastric bypass on relieving type 2 diabetes mellitus. Methods From September 2009 to September 2010,Thirty two cases of type 2 diabetes mellitus patients underwent gastric bypass operation in Xijing Hospital.Preoperative FBG,2hPG,HbAlc,weight,blood pressure and fatty liver were compared with that on 1 month,6 months,12 months after operation. Result There was no significant postoperative complications.FBG on 1 month,6 months,12 months postop and preoperative level was respectively 7.8 ± 2.2,7.7 ± 2.2,7.2 ± 1.8 and 11.1 ± 2.7 ( mmol/L),P ＜ 0.05.2hPG was 10.2 ± 2.6,10.5±2.8,10.5 ±3.1 and 14.0 ±3.5 (mmoL/L),P＜0.05.HbAlc was 7.6％ ±1.4％,7.5％ ± 1.7％,7.1％ ±1.9％ and9.0％±2.3％,P＜0.05.FBG[(6.9±1.5) mmol/L],2hPG[(10.0±3.2) mmol/L] and HbAlc[ (6.9％ ± 1.9％ ) ] on 12 months after operation were lower than preoperative[ FBG ( 10.7 ± 2.9 ) mmol/L,2hPG ( 14.3 ± 4.1 ) mmol/L,HbAlc ( 8.8％ ± 2.0％ ) ] in patients with BMI ＜25 kg/m2 (P ＜ 0.05 ).In patients with preoperative BMI ≥ 25 kg/m2,FBG ( 7.5 ± 2.3 ) mmol/L,2hPG ( 11.3 ±2.9) mmol/L HbAlc (7.3％ ± 1.9％ ),12 mos pest were lower than that of preoperative levels of [FBG(11.7 ±2.3) mmol/L,2hPG(13.5 ±2.4) mmol/L,HbAlc(9.2％ ±2.7％)] (P ＜0.05).Postoperative blood pressure became normal in 5 out of 6 patients complicating preoperative high blood pressure as evaluated on 12 months after operation. Fatty liver ameliorated in 8 out of 17 patients.Conclusions Gastric bypass significantly improves glycometabolism and comorbidity in type 2 diabetes patients.

Diabetic retinopathy is one of the most common and serious microvascular complications of diabetes, and it is a very complex pathological process, displaying multi-factor, multi-stage characteristic in the outcome. Recent studies have shown that the pathogenesis of diabetic retinopathy is closely related to oxidative stress, and antioxidant therapy can effectively improve the condition. Heme oxygenase-1(HO-1)is a widely existing antioxidant defense enzyme, which can reduce the injury caused by oxidative stress and plays an important physiological role. Studies have indicated that in retina HO-1 can be highly induced by hyperglycemia. Moreover, the expression level of HO-1 by man-made changes can accelerate or postpone the diseases progression. That means HO-1 for the diagnosis and treatment of diabetic retinopathy has satisfactory applied foreground. In this article we would like to review the role of HO-1 in diabetic retinopathy.
