1.The effects of transfection of a truncated BMP-II receptor on the prolife ration of Tca8113 cells

Objective: To study the effects of BMPs signals on the proliferation of tongue cancer Tca8113 cells. Methods: Th e cDNA of truncated BMP-II receptor was transfected into Tca8113 cells by usin g FuGENE6 transfection kit, the transfected cells were named Tca8113ZR. The pro liferation and DNA synthesis of Tca8113 and Tca8113ZR cells were investigated b y MTT assay,FCM and BrdU analysis. Results: In MTT assay the A value of Tca8113 and Tca8113ZR cells was 0.47?0.01 and 0.35?0.01 (P0.05).Conclus ion: BMPs might be involved in the development of squamous cell carc inoma of tongue.
2.Association of HLA-DRB1,DQA1,DQB1 in Patients with Bullous Pemphigoid

Objective To investigate the association between HLA classⅡgenes and bullous pem-phigoid(BP).Methods HLA-DRB1,DQA1,DQB1were determined by polymerase chain reaction se-quence specific oligonucleotide probe methods in56patients with BP and150normal controls from Shanghai area.Results It was shown that HLA-DRB1*10(*1001)was linked to DQB1*0501,and its frequency in-creased significantly in BP group compared to normal controls.There was a positive correlation between DRB1*1001and mucosal involvememt as well as autoantigen BP230.HLA-DRB1*04was linked to DQB1*0302,and its frequency increased significantly in BP group compared with normal control.There was a positive correlaton between DRB1*04and autoantigen BP180.The frequency of HLA-DRB1*12(*1201,*1202)decreased in BP group compared to normal controls.Conclusions It is suggested that HLA-DRB1*1001and DRB1*04be the susceptible genes while DRB1*1201,and*1202be the protective genes of BP patients in Shanghai area.
3.Preparation of total alkaloids from Sophora alopecuroides freeze-dried powders and investigation of its antitumor effects.

OBJECTIVETo investigate molding technology of total alkaloids from Sophora alopecuroides freeze-dried powders and observe its inhibition effects on liver transplantation tumor in mice.
METHODWith color, clarity, water-soluble and formability as indexes, single factor tests were adopted to screen type and amount of filler, the concentration of total alkaloids in drug liquid, pH in order to determine optimum prescription of total alkaloids from S. alopecuroides freeze-dried powders, the lowest melting point was determined and freeze drying curve was drafted. Mice hepatoma H22 ascites tumor strain was sterile inoculated in right axillary subcutaneous of mice, and antitumor effect of total alkaloids from S. alopecuroides freeze-dried powders on liver transplantation tumor H22 in mice.
RESULTWhen selected 80 g x L(-1) as mannitol as filler, the concentration of total alkaloids in drug liquid was 25 g x L(-1) and pH 6.5-7.5, freeze-dried effect was optimum with fast reconstitute speed. Average inhibition rate of the big (120 mg x kg(-1)) and medium (60 mg x kg(-1)) dose group of total alkaloids from S. alopecuroides freeze-dried powders on liver transplantation tumor H22 in mice were 56.08% and 35.49%, respectively.
CONCLUSIONPreparation technology was reasonable, reproducible and stable, total alkaloids from S. alopecuroides freeze-dried powders had significant antitumor effect and showed a dose-effect relationship.

5.Application value of serum cystatin C for diagnosing early kidney injury of diabetes and hypertension

Objective To evaluate the significance of cystatin C (CysC) in the diagnosis for early kidney injury of diabetes and hypertension by testing serum CysC and serum creatinine concentration and urinary albumin excretion (UAE) in patients with type 2 diabetes and primary hypertension.Methods Sixty-six patients with type 2 diabetes were selected,among whom the total 38 patients whose UAE were 30-300 mg/24 h were classified into early diabetic nephropathy (DN) group,and 28 patients whose UAE were < 30 mg/24 h were classified into non diabetic nephropathy (NDN) group.Fifty-two patients with primary hypertension were selected,among whom the total 25 patients whose UAE were ≥ 30 mg/24 h were classified into hypertension with kidney injury group,and 27 patients whose UAE were < 30 mg/24 h were classified into hypertension without kidney injury group.Sixty health people were selected as control group.The UAE,CysC and creatinine levels were detected.Results The CysC in early DN group was significantly higher than that in control group and NDN group [(1.84 ± 0.83)mg/L vs.(0.41 ± 0.62) and (0.66 ± 0.59) mg/L],and there was statistical difference (P < 0.05).There was no statistical difference in CysC between NDN group and control group (P > 0.05).There were no statistical differences in creatinine and UAE among the DN group,NDN group and control group (P > 0.05).The CysC in hypertension with kidney injury group was significantly higher than that in control group and hypertension without kidney injury group [(0.93 ± 1.04) mg/L vs.(0.41 ± 0.62) and (0.69 ± 0.57) mg/L],and there was statistical difference (P < 0.05).There was no statistical difference in CysC between hypertension without kidney injury group and control group (P > 0.05).There were statistical differences in creatinine and UAE among hypertension with kidney injury group,hypertension without kidney injury group and control group (P > 0.05).Conclusion CysC can be regarded as the value to judge early kidney injury,and it is very important inunderstanding patient's condition,by which appropriate treatment can be carried out to prevent or decrease the occurrence of chronic renal failure.
6.MicroRNA-215 is a potential prognostic marker for cervical cancer.

Recently, microRNAs (miRNAs) have been shown to be involved in multiple biological pathways that can influence tumor progression and metastasis and they can serve as prognostic biomarkers in many cancers. The present study examined the prognostic significance of miR-215 in cervical cancer. The paraffin-embedded paired cervical scrape samples and tumor tissue samples from 302 patients with stage II cervical cancer were detected for the expression of miR-215 by using qRT-PCR. A miR-215-based classifier was established by using the Cox regression model. The prognostic and predictive accuracy of this classifier was determined in both the internal testing group of 138 patients, and the external independent group of 280 patients. Moreover, cervical cancer HeLa cells overexpressing miR-215 (HeLa-miR-215) were constructed and subcutaneously injected into the nude mice to examine the effect of miR-215 on tumor growth and metastasis in vivo. The results showed that the expression level of miR-215 was significantly higher in cervical cancer tissues than in paired normal tissues (P<0.0001). When patients were classified into high- and low-risk cancer progression groups according to miR-215 level, the 5-year disease-free survival in high- and low-risk groups were 43% (95% CI: 32.1-51.6) and 67% (95% CI: 48.6-77.3) (hazard ratio [HR] 2.02, 95% CI: 1.16-3.52; P=0.013) respectively. Moreover, the expression level of miR-215 was negatively associated with survival rate in patients at TNM stage T3 (HR: 3.317; 95% CI: 1.18-5.14, P=0.017) and TNM stage T4 (HR: 3.48; 95% CI: 1.49-4.45, P=0.008). Tumor volume in nude mice injected with HeLa-miR-215 cells was significantly larger than that in mice injected with control HeLa cells. It was concluded that the expression level of miR-215 is associated with cervical tumor progression and worse survival rate, suggesting that it may serve as a potential prognostic marker to identify patients at higher risk of recurrence.
7.The finger lateral local flaps to repair clinical application of finger tip defect

Objective To discuss the clinical influence and advantages of applying the local flap of finger tip lateral skin to repair the fingertip defective injuries.Methods From August,2013 to December,2015,used the way of local the flap of finger tip lateral skin to repair the first-fourth fingertip defects for 32 cases and 32 fingers,among which there were 20 males and 12 females;finger categories:11 thumbs,9 index fingers,8 middle fingers and 4 ring fingers.The defective area of fingertips was 0.5 cm×0.8 cm-1.0 cm×1.5 cm,all with bone exposure,23 cases had defects at the end part of phalanx,32 cases had different extent of nail bed defects.Reasons for the injuries:13 cases of grind injuries,11 cases of punch pressing injuries,8 cases of sharp tool cutting injuries.The time from injury to the treatment was 0.5-3.0 h,all of them had no associated injuries.Results Thirty-two cases of finger flaps all survived,with the follow-up for 3-24 months and average follow-up of 6 months,the flaps were slightly bloated after 3 months comparing with the normal skin,there was no contracture in the flap area and pedicle scars,two-point discrimination was 4-5mm,the farinterphalangeal joint flexion was normal.Conclusion The local flap of fingertip lateral skin can repair the defects of fingertip with less scar,not bloated flap,satisfying appearance,operation is simple,and good function,so it's the ideal way to repair the defects of fingertip.
8.Preparation and Drug Sustained Release Behavior of Poly(?-caprolactone)/Poly(ethylene glycol)/Poly(?-caprolactone) Amphiphilic Block Copolymeric Microspheres Containing Biological Macromolecule

OBJECTIVE: To characterize morphology and drug release kinetics of protein-loaded poly(?-caprolactone)/poly(ethylene glycol)/poly(?-caprolactone) amphiphilic block copolymeric (PCE) microspheres, and elucidate the mechanistic details regarding protein release. METHODS: BSA-loaded PCE microspheres were prepared using a water-in-oil-in-water, followed by solvent evaporation. Morphology of the polymer microspheres was observed using scanning electron microscopy. Protein loading capacity and encapsulation efficiency were determined by extracting the proteins from the microspheres and measured using MicroBCA method. Protein release kinetics was characterized by cumulative release against the date of release incubation. RESULTS: The protein-loaded PCE microspheres were spherical and possess smooth surface under SEM. Protein loading capacity and encapsulation efficiency in the microspheres were independent of PCE molecular weight. However, the kinetic features of the protein release varied significantly with PCE molecular weight, suggesting a diffusion-degradation combined release mechanism. For the microspheres made of larger molecular weight PCE 4000, the portion of protein release attributed to diffusion from the polymer matrix was remarkably less than that from microspheres of small molecular weight PCE. In vitro release profile can be simulated using a diffusion-degradation model(Q=k1t1/2+k2t+k3t2+k4t3)(r=0.997). CONCLUSION: PCE microsphere formulation can offer sustainedrelease of proteins with initial burst and incomplete release reduced to acceptable level.
9.Expression of BMP-2/4 and BMPR-IA in oral squamous cell carainoma

Objective: To study the relationship between bone morphogenetic proteins(BMPs),BMP receptor type IA (BMPR IA) and the oncogenesis of squamous cell carcinoma in oral epithelia. Methods: Expression of BMP 2/4,BMPR IA was immunohistochemistry analyzed in the specimens obtained from buccal mucosa, including 18 cases of normal mucosa(NB), 24 cases of chronic inflammation(NCI) and 58 cases of oral squamous cell carcinoma(SCC). Result: Weak staining of BMP 2/4 and BMPR IA was observed in normal and chronic inflammation samples,while stroug staining was found in 52 out of 58 cases of oral SCC. No significant difference was found among the heighly,intermediately and poorly differentiated SCC groups.Conclusion: BMP 2/4,BMP IA might be involved in oncogenesis and development of squamous cell carcinoma of oral epithelium.
10.Application of the calculated electrophysiological parameters in early diagnosis of carpal tunnel syndrome

Objective: To investigate the application of the calculated electrophysiological parameters in early
diagnosis of carpal tunnel syndrome (CTS). Methods: 44 patients (60 hands) with a diagnosis of
CTS and 31 healthy volunteers (44 hands) were enrolled. Acquired indicators include median nerve
distal motor latency (DML), complex muscle action potential (CMAP) amplitude, conduction velocity
(MCV), median nerve sensory nerve action potential (SNAP) amplitude, and conduction velocity
(SCV). Then the terminal latency index (TLI), the residual latency (RL), and the difference in peak
sensory latencies between the median and ulnar nerves (ΔPSL) were calculated. Results: The two
groups were matched in age and gender distribution. The CTS group showed significant difference
in SCV, DML, SNAP, and CMAP compared with the control group. The sensitivity, specificity, cutoff value, Youden index, and area under the curve of each indicator are respectively as follows: TLI
(0.733, 0.932, 0.622, 0.629, and 0.877), RL (0.750, 0.977, 1.334, 0.727, and 0.907), ΔPSL (0.950,
0.841, 0.150, 0.791, and 0.942), SCV (0.950, 0.796, 56.5, 0.746, and 0.946), DML (0.867, 0.932,
3.55, 0.799, and 0.930), SNAP (0.683, 0.932, 21.68, 0.615, and 0.844), and CMAP (0.683, 0.773,
8.76, 0.456, and 0.758).
Conclusion: The calculated electrophysiological parameters have higher sensitivities and specificities
relative to a single electrophysiological parameter, which could greatly improve the accuracy of early
diagnosis of CTS.
