1.Improvement of diabetic peripheral neuropathy using monochromatic infrared energy

Objective To observe the effect of monochromatic infrared energy(MIRE)on diabetic periphe- ral neuropathy(DPN).Methods Seventy-four subjects with diabetic peripheral neuropathy who were tested by Semmes-Weinstein monofilaments(SWM)were randomized into 2 groups:a conventional management group and a conventional management plus MIRE group.Then the patients'sensory function and other DPN symptoms were evalu- ated by the SWME and the score of Michigan Neuropathy Screening Instrument.Results After treatments,there was a decrease(P<0.01)in the number of the sites insensitive to SWME(grade 5.07),and MNSI scores were sig- nificantly decreased(P<0.01).The MIRE management was more effective than conventional management.Con- clusion Monochromatic infrared energy is perhaps a safe,non-pharmaceutical and non-invasive method for the treat- ment of diabetic peripheral neuropathy.
2.Analysis of complications after treatment of femoral intertrochanteric fractures with internal fixation

Objective To analyze causes for postoperative fixation-related complications of femoral in- tertrochanteric fractures treated with dynamic hip screws(DHS)and dynamic condylar screws(DCS).Methods From January 1995 to December 2004,215 cases of intertrochanteric fracture were treated with DHS/DCS.Their postoperative fixation-related complications were analyzed to find out the causes.Results All cases were followed up for 13 to 72 months(mean,32 months).The rate of excellent and good functional recovery of hip was 94.0%. Postoperative internal-fixation-related complications occurred in 31 cases,including hip varus in 14 cases who had posterior intertrochanteric fragments,delayed union of fracture in 12 cases who had fractured calcar and lesser trochanter and massive dissection of soft tissues,and cutting-out of lag screws in five cases,three of whom had to receive prosthetic replacement.Conclusions Unsuitable implant selection is an important factor leading to postoperative fixation-related complications.Unstable fracture,serious osteoporosis and unsatisfactory reduction will increase the incidence of complications.
3.Progress in research on bacterial HD-GYP domain proteins

Bis-(3′,5′) cyclic di-guanylate (c-di-GMP) is an almost ubiquitous intracellular second messenger in bacteria.Now it is known to regulate complex physiological processes, including mobility, adhesion, virulence and biofilm formation.The level of c-di-GMP is regulated by diguanylate cyclases (DGCs) containing GGDEF domains and phosphodiesterases (PDEs) containing EAL or HD-GYP domains.Recent studies have demonstrated that HD-GYP domain protein is a novel phosphodiesterase, which is also involved in the regulation of c-di-GMP degradation.This review highlights recent advances in the structure and biochemical functions of HD-GYP domain proteins, which might help to further clarify the mechanism of c-di-GMP signal system.
4. Optimization of extraction technology of Schisandrae Chinensis Fructus by Multi-objective Genetic Algorithm

OBJECTIVE: To study the application of Multi-objective Genetic Algorithm in optimization of micro-extraction technology of Schisandrae Chinensis Fructus. METHODS: Sub-target model of extraction rate, schizandrin and the total lignan was established based on micro-extraction Schisandrae Chinensis Fructus experimental data resulted from uniform design. Applying MOGA, the optimal extracting conditions were explored, and the results were compared with those of Single-objective genetic algorithm. Using SGALAB beta5008 of the Matlab2009a plug-in tool-box, the genetic algorithm optimization was achieved. SPSS13.0 software was used for statistical analysis. RESULTS: The optimal conditions were as follows; microwave power 270 W, ethanol 87%, extraction time 7 min, ethanol amount 1:4.5, grinding degree 70. Under these conditions, the extract rate was 21.40%, Schizandrin recovery was 4.72%, and the total lignan recovery was 10.84%. CONCLUSION: MOGA provides a reasonable pareto ensuring the optimal multi-objective, which provides a reasonable method for extraction of Schisandrae Chinensis Fructus. 5.Comparison of two surgical treatments for pediatric lower eyelid trichiasis

AIM: To compare the clinical results of suture method and partial eyelash resection treating for pediatric eyelid trichiasis, and screen an effective method for the treatment of pediatric lower eyelid trichiasis.
METHODS: Fifty-six cases of pediatric patients with lower eyelid trichiasis were randomly divided into a control group and an observation group in accordance with the method of drawing lots, and each group was 28 cases. The control group was treated with suture method, and the observation group was treated with partial eyelash resection. The clinical efficacy, patient satisfaction before and after treatment, and the incidence of complications were compared.
RESULTS:(1) The clinically total effective rate was 74%of the control group, which was 89% of the observation group, and there were statistical differences of the clinical efficacy between the two groups ( P<0. 05 ); ( 2 ) the satisfaction scores were (3. 89±1. 22) points and (6. 27±1-86) points of the the control group before and after treatment, which were ( 4. 00±1. 34 ) points and ( 8-95±2-34) points of the observation group, and there were statistical differences of the two groups before and after treatment (P<0. 05), and the scores of the observation group were higher than that of the control group ( P<0-05);(3) complications such as swelling and stitches off appeared in both groups, and the rate was 21% in the control group, which was 11% in the observation group, and there were statistical differences between the two groups (P<0. 05).
CONCLUSION: The treatment of children with lower eyelid trichiasis, suture method is simple and can be performed under local anesthesia in collaboration with children, but with a higher relapse rate, some patients required reoperation;partial resection of eyelashes can be more thoroughly solve the problem of pediatric eyelid trichiasis with low recurrence rate, but children need to be under general anesthesia with some of big risk. So partial resection of eyelashes is unsuitable for using in clinical practice widely and can be used in special cases.
6.Value of prenatal ultrasonographic diagnosis in fetal pulmonary atresia

Objective To explore the clinical value of prenatal ultrasonographic diagnosis of fetal pulmonary atresia.Methods The data of echocardiography in 7 fetus with pulmonary atresia confirmed by autopsy were analysed retrospectively,the characteristics of abnormal ultrasound imaging were summarized.Results The dysplastic main pulmonary had been detected only in one fetus by two-dimensional echocardiography,five cases were shown ventricular septal defect with straddle of aortic root on the ventricular septum,two cases with intact ventricular septum had hypoplasis of right heart.Color Doppler flow imaging showed the reverse flow of ductus arteriosus in 5 cases and tricuspid regurgitation in 3 cases.Five cases were diagnosed pulmonary atresia,one case was hypoplasia of right heart,and one case was mistaken for persistent truncus arteriosus.Conclusions Prenatal ultrasonographic diagnosis of tetal pulmonary atresia has great clinical value,the reverse flow of ductus arteriosus is an important clue to diagnosis.
7.Construction and identification of prokaryotic expression system of staphylococcal enterotoxin A gene and expressed product

AIM:To construct a prokaryotic expression system of staphylococcal enterotoxin A(SEA)gene and determine the effects of the recombinant expression product rSEA in promoting lymphocyte proliferation and inhibiting tumor cell growth.METHODS:PCR was used to amplify entire SEA gene of S.aureus strain ATCC13565.The cloned SEA gene was sequenced after T-A cloning.SDS-PAGE was applied to measure the output of rSEA expressed by pET32a-SEA-E.coli BL21DE3.Ni-NTA affinity chromatography was performed to extract rSEA.Cytotoxicity of rSEA to Vero cells was detected using TCID_ 50 titration method and then the value of TCIC_ 50 was determined.MTT colorimetry was established to examine the effects of rSEA at different dosages on proliferation of mouse splenocytes and human peripheral blood mononuclear cells(PBMC)as well as on growth of HepG2 cells and HeLa cells in vitro.RESULTS:In comparison with the published corresponding sequences,similarities of the nucleotide and putative amino acid sequences of the cloned SEA gene were 100%.The output of rSEA was approximate 25% of the total bacterial proteins.rSEA had a cytotoxicity with TCIC_ 50 of 3.14 ?g to Vero cells.1.0-20.0 mg/L rSEA showed the significant effects of promoting proliferation of mouse splenocytes and human PBMC(P
8.Research on the electrophysiological mapping and ganglianated plexi ablation techniques during the minimally invasive atrial fibrillation surgery

Objective To conclude the results of electrophysiological (EP) mapping and ganglianated plexis (GPs) ablation during the minimally invasive atrial fibrillation (AF) surgery.Methods During the period of Sep 2007 to May 2010 to-tally 185 paroxysmal atrial fibrillation cases were recruited receiving the minimally invasive AF surgery with video assistance.The EP mapping was used to test the dual direction block after ablation and the distribution of GPs was recorded.The sinus rhythm rate was followed up after surgery.Results All the minimally invasive surgery were performed successfully withoutdeath in the hospital.There was no permanent pacemaker implant and other serious complications.EP mapping results showed92% pulmonary vein (PV) potential to the left atrium (LA) could be isolated with the dual direction block for the paroxysmal atrial fibrillation cases.And there were 89% GPs positive,higher rate than the persistent AF.There were more GPs positive in the right PV area than the left and 86% GPs lied at the LA domain,not the PV orifice.At least 5 or 6 ablation lesions were needed to get the dual direction block and denervation of the GPs.The sinus rhythm rate after surgery after 3,6,12 and 24months is 83.7%,82.4%,85.4% and 83.9%.The 12 months follow up data showed the cases with more GPs positive (>6) had 81.3% sinus rate and the less GPs positive (≤2) group with 52.6%.Conclusion The EP mapping and GPsablation techniques during the minimally invasive AF ablation surgery could get good sinus rhythm rate during the long term follow up.
9.Expression of Beelin-1 and LC3 in adriamycin induced cardiomyopathy rats

Objective To explore the expressions of autophagy-related gene Beclin-1 and LC3 in adriamycin induced eardiomyopathy rats,to prnve that autophagy might take part in the development of adriamycin induced eardiomyopathy in rats,so as to provide experimental and theoretical evidence for preventing and treating adriamycin induced cardiomyopathy.Methods Forty-five male SD rats were randomly divided into 3 groups,control groups,ADR group and ADR+3-MA group.The model of adriamycin induced cardiomyopathy rats was established.The tissue sample taken from the left ventricle wall was checked for the morphons of autophagosome by electron microscope. The expressions of Beelin-1 and LC3 of myocardium were detected.Results The morphons of autophagosome in ADR group was significantly increased compared with that in control and ADR +3-MA groups. The expression of Beclin-1 in myocardium of ADR group was significantly inereased compared with that in control and ADR +3-MA groups (P < 0.05).The level of LC3 in myocardium of ADR group was significantly increased compared with that in control and ADR+3-MA groups (P < 0.05).Conclusions Autophagy plays an important role in adriamycin induced cardiomyopathy.
10.Effect of RNA Interference Targeting HIF-1αGene on the Proliferation of Human Tongue Cancer Cells

Objective To explore the effect of RNA interference targeting hypoxia-inducible factor-1α(HIF-1α) gene on the proliferation of human tongue cancer cells (Tca8113 and Tscca). Methods HIF-1α-specific short hairpin RNA (shRNA) expression vector and empty shRNA vector were respectively transferred into two kinds of cultured human tongue cancer cells. The expression of HIF-1αgene was detected by RT-PCR and Western blot method. The clone forma-tion assay was used to observe the cell proliferation. The cell counting was used to monitor the number of living cells. Flow cytometry was applied to detect cell cycle distribution. Additionally, the cells were injected into nude mice, and tumor form-ing condition was observed. Results After the shRNA HIF-1αplasmid was transfected into tongue cancer cells, the expres-sions of both HIF-1αmRNA and protein were significantly decreased in tongue cancer cells under hypoxia condition. The number of colony formation was significantly lower in HIF-1αsmall interfering RNA (siRNA) group than that in empty and negative control group. The cell counting revealed that the number of living cells was remarkably lower in HIF-1αsiRNA group than that in negative control group. Also, the cell cycle in HIF-1α siRNA group was arrested at G0/G1 phase after transfection and 48 hours hypoxia culture. Besides, the tumor volume was remarkably smaller in HIF-1αsiRNA group than that in negative control group after the tongue cancer cells transplanted into nude mice for several days. The statistical analy-sis indicated that the difference was significant (P<0.05). Conclusion RNA interference targeting HIF-1α gene could suppress the proliferation of human tongue cancer cells effectively. HIF-1αgene could be researched as a new therapeutic target of tongue cancer in the future.
