1.Investigation on the short-term clinical application of two types of glass fiber posts.

OBJECTIVEThe clinical results of restoring defective teeth with two types of glass fiber prothetic systems is investigated to acquire clinical experience for further application of glass fiber posts with independent intellectual property rights.
METHODSA total of 120 out-patients with restored defective teeth were selected from the Department of Stomatology, Beijing Xuanwu Hospital of Traditional Chinese Medicine and Peking University School and Hospital of Stomatology and randomly divided into two groups. OUYA FIBER posts and Tenax Fiber White posts were used to restore defective teeth in the experimental group and the control group, respectively. Clinical evaluation was conducted one week and three months post-restoration.
RESULTSBoth clinical satisfactory rates of OUYA FIBER posts and Tenax Fiber White posts were higher than 98% one week post-restoration and higher than 96% three months post-restoration. No significant differences were observed between the clinical results of restoring defective teeth with the two types of fiber posts. Throughout the healing process, no root canal fracture or marginal staining were observed.
CONCLUSIONOUYA FIBER post and Tenax Fiber White post showed similar clinical outcomes during the period of observation in this study. However, the long-term effects should be observed in future studies.

2.Study on the effects of heat and ultraviolet fight radiation on hepatitis C virus JFH-1 strain inactivation

Objective To investigate the inactivating effect of heat and ultraviolet(UV) light on HCV JFH-1 strain using the cell culture system. Methods The HCV JFH-1 virus stock, with an initial titer of 2.5 × 104 FFU/ml, was exposed in 56℃ water bath or to UV light for varying durations of time for explo-ring their inactivating effects on the virus. The kinetics of virus titer reduction was determined by an indirect immuno-fluorescence assay (IFA). If the cells infected with the exposed virus stock were IFA negative after three blind passages, the virus stock was considered to be inactivated completely. Results After incubation of the HCV JFH-1 virus stock (2.5 × 104 FFU/ml)in 56℃ water bath for 10 min, 20 min and 30 min, the virus titers were reduced to 1.6 × 103 FFU/ml, 3.1 × 102 FFU/ml and 3.3 × 10 FFU/ml, respectively. The exposure of the virus stock to UV light (wavelength 253.7 nm, intensity ≥60 μW/cm2, 30 cm below the UV lamp) for 15 s, 30 s and 45 s resulted in virus fiter reduction to 1.0 × 103 FFU/ml, 1.1 × 102 FFU/ml and 2.7 × 10 FFU/ml, respectively. After 40 min incubation of the virus stock at 56℃, or 1 min exposure to UV light (wavelength 253.7 nm, intensity ≥60 μW/cm2) the virus infectious titer was reduced below the detection limit of IFA, and the IFA was still negative even after three blind passages, indicating that the virus was inactivated completely. Conclusion HCV is sensitive to heat and UV light treatment. For HCV JFH-1 virus stock containing 2.5 × 104 FFU/ml virus, heat treatment at 56℃ for 40 min, or UV light expo-sure at an intensity of ≥60 μW/cm2 for 1 min, resulting in complete virus inactivation.
3.The clinical analysis of patients with Turner syndrome and spondyloarthropathy

Objective To investigate the clinical characteristics of Turner syndrome in patients with spondyloarthropathy.MethodsSix patients from our hospital were included and the clinical manifestations,laboratory parameters and imaging presentations were analyzed retrospectively and the relevant literature were reviewed.ResultsThe mean age of these 6 cases was (27±5) years.Two cases were spondyloarthropathy,3 cases were ankylosing spondylitis,1 was psoriatic arthritis.Five cases were complicated with peripheral arthriris,2 cases were complicated with inflammatory back pain,4 cases had vertebral activity limitation,3 cases had joint deformity.The karyotype of 6 cases was 45,XO.Six cases were complicated with osteoporosis.Conclusion Turner syndrome can be presented with spondyloarthropathy,arthritis,osteoporosis.Dysplasia of bone is common.
4.MRI manifestations of dysplastic gangliocytoma of the cerebellum

Objective To discuss MRI characteristics of dysplastic gangliocytoma of the cerebellum.Methods MRI manifesta-tions of 13 cases with dysplastic gangliocytoma of the cerebellum verified by operation and pathology were analyzed retrospectively. Results All of 13 cases were located in the cerebellar hemisphere and vermis.The lesions presented striated pattern of iso-intensity signal and hypo-intensity signal on T1-weighted images.The sign of tiger stripes was the feature in dysplastic gangliocytoma on T2-weighted and FLAIR images.All of the tumors showed slightly striated enhancement after intravenous administration of contrast agent.The mass effects of dysplastic gangliocytoma of the cerebellum along with the forth ventricle and brain stem oppressed were seen in all cases.NAA was mildly reduced on magnetic resonance spectroscopy (MRS)in 7 cases of dysplastic gangliocytoma of the cerebellum.Conclusion There are distinctive manifestations in MRI images for dysplastic gangliocytoma of the cerebellum,which has crucial diagnostic value before operation.

6.The role of immune receptor NOD1 in insulin resistance of human adipocytes

Objective To investigate the role of nucleotide-binding oligomerization domain-containing protein 1 (NOD1) in inducing insulin resistance in differentiated human adipocytes.Methods Human preadipocytes obtained via liposuction were induced to differentiate into mature adipocytes.iE-DAP,a specific ligand for NOD1,was administered to human adipocytes in culture.NF-κB transcriptional activity and proinflammatory cytokines production were determined by luciferase assay and enzyme-linked immunosorbent assay.Glucose uptake in adipocytes was measured by 2-deoxy-D-[3 H] glucose uptake (P<0.05).The expression of phosphatidylinositol-3-kinase p85 (PI-3K p85),insulin receptor substrate-1 (IRS-1) and Akt phosphorylations were detected by Western blotting.Results NF-κB transcriptional activity and cytokines such as interleukin (IL)-6,IL-8,and monocyte chemotactic protein 1 secretion were markedly increased after stimulation with iE-DAP (P<0.05 or P<0.01).Insulin-induced glucose uptake was decreased with the activation of NOD1 in a dose-and time-dependent fashion.NOD1 activation weakened insulin signal transduction as being revealed by increasing IRS-1 Ser307phosphorylation,reducing protein expression of PI-3K p85,and attenuating insulin-induced phosphorylation of Akt on Ser473 and Thr308in human adipocytes.Conclusion These results indicate that NOD1 activation induces inflammatory response and insulin resistance via IRS-1/PI3-K/Akt signaling pathway in human adipocytes.
7.The role of human papillomavirus in the pathogenesis of pterygium

Background The pathogenesis of pterygium has been in the study.Relative molecular biology study showed that pterygium is a tumor-like lesion,and based on overseas literatures,human papillomavirus (HPV) is positively expressed in 100% patients with pterygium in some region.However,if this result is suitable for Chinese patients is unclear.Objective This study was to identify the role of human HPV in the pathogenesis of pterygia in Wuxi area.Methods Forty-eight pterygium specimens including 7 recurrent pterygia and 41 primary pterygia were collected during the operation,and these patients were from Wuxi area.Two cervical carcinoma specimens and 2 conjunctiva specimens from normal donors were obtained as positive control and negative control respectively.The fluorescent quantitative polymerase chain reaction (FQ-PCR) was used to detect HPV DNA of specimens.Results The amplified curves of HPV 6,11 of pterygium specimens,cervical carcinoma specimens and normal specimens were all below the positive quality control curve,but the amplified curves of HPV16,18 were above the quality control curve in cervical carcinoma specimens; while those of pterygium specimens and normal conjunctival specimens were all below the quality control curve.HPV16/18 was identified in 2 cervical carcinoma specimens,but no HPV6/11 was detected in 2 cervical carcinoma specimens.However,HPV DNA expression in primary and recurrent pterygias were absent.Conclusions According to these results,HPV is not the primary cause for the pathogenesis of pterygium in Wuxi region.
8.Association of TNF-αand HSP70 in serum with type 2 diabetes mellitus among Hui and Han nationalities in Ningxia

Objective To investigate the expression level of tumor necrosis factor α( TNF-α) , heat shock protein 70 (HSP70) in serum and to analyze the correlation of TNF-αand HSP70 with the incidence of type 2 diabetes mellitus (T2DM) among Hui and Han nationalities in Ningxia .Methods Using the method of case-control study and according to the inclusion and exclusion criteria of case and control groups , 225 patients with T2DM who were treated by Department of Endocrinology and 600 healthy people during health checkups were enrolled in the study between October 2011 and October 2012 .A double antibody sandwich enzyme-linked immunosorbent assay ( ELISA) was used to determine the protein expres-sion levels of TNF-αand HSP70 in serum.Results The expression levels of TNF-αin the total population investigated and Hui population in T2DM group were significantly higher than in the control (t=-3.512,-3.791,all P<0.01).The difference in expression levels of TNF-αbetween the Han people of T 2DM group and control group , and that in the expression levels of HSP70 between the case group and control group were not statistically significant ( t =-0.565,-0.020,-1.251,-0.809,all P>0.05).The elevated serum levels of TNF-αincreased the risk for T2DM(OR=1.500, 95%CI:1.075-2.093,P=0.017).The expression levels of TNF-αin the total people, case and control of Hui people were significant lower than the homologous Han people (t=9.373,2.223,9.734, all P<0.01 or 0.05).Pearson analysis showed that the high expression of TNF-αwas positively correlated with blood glucose level (r=0.110,P=0.001),but there was no correlation between the high expression of HSP 70 and blood glucose level (r=0.023, P=0.504).The result of multiple logistic regression analysis indicated that gender and waist-to-hip ratio(WHR) were risk factors for high levels of TNF-α(P<0.01 or 0.05)while HSP70 and nationalities were protective factors.Systolic blood pressure(SBP) and WHR were risk factors for the high level of HSP70(all P<0.01), but TNF-αwas protective factors.HSP70, TNF-α, gender, family history, BMI, SBP, triglycerides(TG), low density lipoprotein cholesterol (LDL-C), alanine trans-aminase(ALT), and psychological stress were risk factors for T2DM, but high density lipoprotein cholesterol (HDL-C) was a protective fac-tor.Conclusion The high expression of TNF-αin serum is closely related to elevated fasting plasma glucose ( FPG) and T2DM onset.The expression level of TNF-αin serum in Hui people is lower than in Han people ,possibly correlated with ethnic heredity.The high expression of HSP70 in serum may induce T2DM onset.
9.Association of HSP70 gene polymorphisms with type 2 diabetes mellitus in Hui and Han people in Ningxia

Results There was no significant difference in the distribution of genotypes and allele frequencies of HSP 70-1 and HSP70-hom between case group and control group (P>0.05), but there was significant difference in the distribution of genotypes and allele frequencies of HSP70-2 between the two groups(P<0.05).There was no significant difference in the distribution of genotypes and allele frequencies of HSP70-1,HSP70-2 and HSP70-hom between Hui and Han people (P>0.05)or in the distribution of genotypes of HSP 70-2 between males and females in case group, but there was significant difference in the distribution of allele frequencies of HSP 70-2 in case group .There was no significant difference in the distribution of genotypes and allele frequencies of HSP 70-1 and HSP70-hom between males and females in case or control group .Logistic analysis confirmed that the WC, TG, TC, LDL-C, SBP, family history of diabetes and G allele were risk factors of T 2DM. Conclusion GG genotype and G allele of HSP 70-2 (+1267 ) SNP may be genetic markers for the susceptibility of type 2 diabetes.There is no significant difference in the distribution of genotypes and allele frequencies of HSP 70-1,HSP70-2 and HSP70-hom between Hui and Han nationalities .Family history of diabetes, LDL-C, TC, and G allele of HSP70-2 (+1267) SNP are the main risk factors of T2DM while HDL-C is a protective factor.
