Fungal polyketide synthases are responsible for the biosynthesis of secondary metabolites such as pigments, mycotoxins, and they are very important in pharmacology , food science and agriculture. The recent advances in the methods for the isolation and manipulation of multiple classes of polyketide synthase genes from fungi were introduced. It is useful for discovery of novel fungal polyketide synthase gene clusters. These methods can also be useful for revealing the genetic potential of fungi to produce multiple types of bioactive polyketide.

5.Roles of ROS and TGF-?1 in aldosterone-induced production of PAI-1

Objective To explore the roles of reactive oxygen species(ROS) and TGF-?1 in aldosterone-induced PAI-1 production.Methods Quiescent rat mesangial cells (MCs) were treated by aldosterone.The level of ROS in MCs induced by aldosterone was measured by confecal laser scanning microscopy and the TGF-?1 activity in the supematant of culture was measured by mink lung epithelial cell (Mvllu) proliferation inhibition MTT assay.Then,before the addition of aldosterone,MCs were pretreated with NAC or TGF-?1 neutralizing antibody to decrease cellular ROS or inhibit activity of TGF-?1 induced by aldosterone respectively.PAI-1 mRNA was examined by semi-quantification RT-PCR and PAI-1 protein by Western blotting.Results The intracellular ROS induced by aldosterone increased by 5-fold compared to that of control group,and the activity of TGF-?1 stimulated by aldosterone increased markedly.TGF-?1 neutralizing antibody and NAC effectively decreased aldosterone-induced PAI-1 mRNA expression by 30% and 32%,and PAI-1 protein expression by 21% and 11%,respectively.However,neither TGF-?1 neutralizing antibody nor NAC alone could regulate aldosterone-induced PAI-1 mRNA and protein expression to normal level in 24 hours.Conclusions ROS and TGF-?1 play important roles in up-regulation of aldosterone- induced PAI-1 in MCs.ROS and TGF-?1 are not the exclusive pathway of PAI-1 expression induced by aldosterone in MCs.
6.Intergrated analysis of CYP3A5 and ABCB1 gene polymorphisms in renal transplanted recipients treated by Tacrolimus

Objective To approach the effect of CYP3A5 6986A>G (rs776746) and ABCB1 3435C>T (rs104564) on plasma tacrolimus(FK506) dose adjusted blood trough concentrations(C0/D) in kidney transplanted patients.Method Totally 125 renal transplanted recipients were recruited,and gene sequencing method were used to detect the genotype of CYP3A5 6986A>G and ABCB1 3435C> T.FK506 trough concentration of 125 patients was detected by enzyme multiplies immunoassay technique (EMIT) after the surgery over 6 month.The relevance between the ratio of trough concentration/dosage and polymorphisms was analyzed.Result In 125 renal transplanted receipients,the allele frequency of CYP3A5 and ABCB1 genetic variants was 74% and 34.8%,respectively.After 6 months,the C0/D ration in the homozygous mutant CYP3A5 * 3/* 3 receiptients was significant higher than that in the homozygous wild type * 1/* 3 and heterozygous mutant * 1/* 1 receiptients (P < 0.05).And there was no significant difference of the Co/D ration between CYP3A5 * 1/* 1 and CYP3A5 * 1/* 3 receipients(P>0.05).The the C0/D ration in homozygous wild type ABCB1 T/T and homozygous mutant T/C receipients was significant higher than that in heterozygous mutant C/C receipients (P<0.05),and there was no significant difference of the C0/D ration between T/C and T/T receipients (P> 0.05).Conclusion The study shows that the the polymorphism of CYP3A5 and ABCB1 genes was significantly correlated with the trough C0/D value of the recipients after renal transplantation who were treated by Tacrolimus for more than half a year.The detection of the SNP of the CYP3A5 and ABCB1 will be useful for FK506 dosage adjustment.
7.CT findings of pediatric hepatoblastoma and correlation with pathological subtypes

Objective To explore the CT findings of pediatric hepatoblastoma and its correlation with pathological subtypes.Methods CT findings and pathological characteristics of 110 patients with hepatoblastoma confirmed by pathology were retrospectively analyzed.Results The locations of the tumor were right lobe (67/110,60.90%),right and left lobe (25/110,22.72%),left lobe (15/110,13.63%) and caudate lobe (3/110,2.72%).The average tumor size was (287.14±272.36) cm3.Most cases were solitary (94/110,85.45%) and round shape (96/110,87.27%).Most cases were well defined (86/110,78.18 %).Necrosis were irregular in 102 cases,calcification were in 60 cases.Lung metastases were common (23/110,20.90 %) and vessel involvement were common (56/110,50.90 %).Lymph node metastasis was in 5 cases.Calcification and homogeneous enhancement in different pathological types had statistically significance (both P <0.001).Conclusion CT is helpful for pre-operation and pathological diagnose.

?Optic neuritis ( ON) is one of the most common causes of vision loss by neural eye diseases in youth and middle-aged. In the past, the diagnosis simply according to the risk position, which did not distinguish from the pathogenesis and clinical characteristics, can not meet the current clinical diagnosis and treatment needs. Combining with the etiology, clinical characteristics and prognosis, the latest classification of the current international diagnosis of ON are typical and atypical ON. Typical ON relates to multiple sclerosis ( MS ) or demyelinating disease of the central nervous system, it has a relatively good therapeutic effect and prognosis. Rather than, atypical ON has complex etiology, clinical manifestation, and the treatment and prognosis are also different. At present there are many international ON treatment guidelines with level I evidence-based medical evidence, but with different genetic background, geographical environment and ethnic groups, they are not been determined. China lacks of such a multicenter large sample, a wide range of research evidence. In this paper, we will summarize the progress of the diagnosis and treatment about ON, especially about the atypical ON, in order to provide some suggestions to further improve the standardization and individualization for clinical diagnosis and treatment on ON.
9.Study on Detection Value of Platelet Activation Indexes and Platelet Parameters in Patients with Hepatitis B-induced Cirrhosis

Objective:To study the detection value of platelet activation indexes and platelet parameters in patients with hepatitis B-induced cirrhosis.Methods:40 patients with hepatitis B-induced cirrhosis in our hospital from November 2015 to August 2016 were selected as the observation group.40 healthy subjects went through physical examination during the same period were chosen as the control group.The platelet activation indexes and platelet parameters of subjects with same age in two groups were detected and compared.The detection results of the two items in patients with different stages of hepatitis B-induced cirrhosis were compared,too.Results:The PLT of observation group was lower than that of control group,while the other platelet parameters and platelet activation indexes were all higher than those of control group.The detection results of two items in patients with different stages of hepatitis B-induced cirrhosis were with significant differences,all P<0.05.Conclusion:The platelet activation indexes and platelet parameters in patients with hepatitis B-induced cirrhosis are obviously abnormally expressed.There are significant differences among the detection results in patients with different stages.Therefore,surveillance of those indexes in patients with hepatitis B-induced cirrhosis should be strengthened.
10.DNA methylation of glioma related gene and its significance

DNA methylation is closely related to the genesis and development of glioma.The CpG islands hypermethylation in promoter region of DNA repair genes,cell cycle controlling genes,apoptosis related genes,tumor suppressor genes and invasion related genes are found in glioma.Finding the specific methylation profile and molecular marker of glioma is helpful for the pathology class,early diagnosis and prognostic evalua-tion.Meanwhile,DNA methylation has the characteristic of reversibility,which may provide new ways for the treatment of glioma.
