1.HPLC versus Gravimetric Analysis in the Determination of Sodium New Houttuyfonte Injection

OBJECTIVE:To compare between the HPLC method and gravimetric analysis in the determination of the content of sodium new Houttuyfonte injection.METHODS:The content of sodium new Houttuyfonte injection was determined by HPLC and gravimetric analysis respectively.HPLC condition: Zorba extent-C18 column(250 mm?4.6 mm,5 ?m) detected by UV(286 nm),and the mobile phase consisted of methanol-water-tetrabutyl ammonium hydroxide(70:30:0.3).The procedure of gravimetric analysis was in line with state drug standard.RESULTS: The determination results were notably different between the two methods.CONCLUSION:Gravimetric analysis showed no specificity in the determination of sodium new Houttuyfonte injection.
2.HISTOLOGICAL AND HISTOCHEMICAL STUDIES OF EFFECTS OF THE HIGH DOSE OF LRH-A ON THE CORPUS LUTEUM OF PREGNANT RATS

In the present experiment the high dose of LRH-A was injected into pregnant rats on Day 9 of pregnancy. Morphology and histochemistry of corpus luteum were studied. It was found that 24 hours after injection LRH-A could effectively cause the corpus luteum to degenarate. A great many large lipid droplets occured in the luteal cells, and the activity of 3?-HSD and of SDH of these cells decreased. At the same time the activity of AcP increased in a few of luteal cells. 48 hours after injection the effects outlined above became more distinct, and the activity of G-6-PD also began to decrease. The activity of 20 ?-HSD in both control and experimental groups was negative 24 hours after injection, while the activity of the AIP increased distinctly in endothelium of the capillary network of corpus luteum. These changes indicated that the high dose of LRH-A had damaged corpus luteum and reduced its synthetic and secretory function of progesterone which, we believe, plays a main role in terminating pregnancy.
3.Research on Formulation Process of Fengshilin Capsule

Objective: To improve the physical properties of micromeritics of the extracts of Fengshilin capsule, such as anti moisture. Methods: Critical relative humidity, absoption curve of moisture, angle of repose were used as the norms of the research. The suitable excipients and formulation process of Fengshilin capsule have been sieved. Results: It was feasible that the combined excipients of microcystalline cellulose and starch (in a ratio of 1∶1) were mixed with the extract powder. The absorption of moiture of the extract powder was improved. The humidity of mass production and store was lower than 45%. Conclusion: The research has been found to be an effective in the process of Fengshilin capsule.
4.Determination of Salicylic Acid and the Related Substances in Salicylic Acid Ointment by HPLC

OBJECTIVE:To determine the contents of salicylic acid and the related substances in salicylic acid ointment by HPLC.METHODS:The determination was performed on Kromasil ODS-C18 chromatographic column with mobile phase consisted of methanol-water(adjust pH=2.5 using acetic acid,50∶50)at a flow rate of 1.0 mL?min-1.The sample size was 20 ?L and the detection wavelength was set at 270 nm.RESULTS:The main peak and the related substances were well-separated.The linear range of salicylic acid was 10.014~100.14 ?g?mL-1(r=0.999 9)with an average recovery of 99.35%(RSD=0.99%)and a lowest detectable limit of 5 ng?mL-1,the labeled amout of related substances all less than 1.01%.CONCLUSION:The method developed in our study is simple in operation,accurate in results,specific and suitable for the content determination of salicylic acid ointment.
5.Feasibility of real-time triplane strain rate imaging for quantitative assessment of left atrial function in normal subjects

Objective To evaluate the feasibility of real-time triplane strain rate imaging for the quantitative assessment of left atrial function in normal subjects. Methods Totally 40 healthy volunteers were enrolled. Left atrial expansion index (LAEI), left atrial passive emptying fraction (LAPEF) and left atrial active emptying fraction (LAAEF) were measured using real-time triplane volume analysis. Mean systolic peak strain rate (MSRs), mean early diastolic peak strain rate (MSRe) and mean late diastolic peak strain rate (MSRa) were measured using real-time triplane strain rate imaging. The results were compared respectively. Results MSRs correlated positively with LAEI (r=0.61, P<0.01). MSRe correlated inversely with LAPEF (r=-0.67, P<0.01), so did MSRa with LAAEF (r=-0.78, P<0.01). Conclusion Real-time triplane strain rate imaging is feasible for the quantification of left atrial function in normal subjects.
6.HISTOLOGICAL AND HISTOCHEMICAL STUDY OF THE HUMAN PLACENTA FROM RIVANOL-INDUCED ABORTION DURING MIDPREGNANCY

24 cases of human placentas were obtained from rivanol-induced abortion, and 6 cases from cesarean section, curettage, and water saccule-induced abortion were chosen for comparative study. All the specimens were proccessed under the same histological and histochemical procedures.In all the specimens from rivanol-induced abortion we observed a reduction in the activity of the ?~5-3?-hydroxysteroid dehydrogenase and of the number of fine lipid droplets in the syncytiotrophoblasts, and in these cells we also observed an increased activity in acid phosphatase in most of the specimens and a decrease of RNA content in a few specimens. A large number of decidual cells and a few trophoblasts showed necrotic changes. The chorionic plate also showed necrosis and distinct inflammatory reactions, and on the fetal surface of the placenta the blood circulation showed disturbance. These changes indicated that rivanol had demaged the placenta and reduced its synthetic function of progesterone which we believed played a rote in terminating pregnancy and caused abortion eventually. It was suggested that the intraamniotic injection of rivanol is preferable to extraamniotic injection.
7.THE IMMUNOHISTOCHEMICAL CHANGE OF PITUITARY GONADOTROPIC CELLS IN THE PREGNANT RATS AFTER INJECTION OF A HIGH DOSAGE OF LUTEINIZING HORMONE RELEASING HORMONE ANALOGUE (LHRH-A)

The specific anti-rat LH, anti-rat FSH and anti-hCG sera were used in the peroxidase antiperoxidase (PAP) immunohistochemical method to observe the change of pituitary gonadotropic cells in pregnant rats after being injected by a high dosage of LHRH-A. It was observed that 24 hrs after injection the immunohistochemical reaction of gonadotropic cells weakened strikingly; 48 hrs after injection the immunohistochemical reaction began to recover; and 168 hrs after injection, the recovery was still in progress. The number of LH cells of different immunoreactive intensity and their percentage in total LH cells of the control and experimental groups were calculated and tested by statistical method. It was found that the results were the same as the above. Our observations support the view that the high dosage of LHRH-A can stimulate gonadotropic cells in rat to release a great amount of LH rapidly. It inhibits the synthesis of progesterone in the ovary of pregnant rat and results in termination of pregnancy. Moreover it is believed that the effect of high dosage of LHRH-A on the secretory function of gonadotropic cells can recover themseleves.
8.Clinical observation on acupoint pressure plus long-snake moxibustion for upper-limb spastic hemiplegia after cerebral infarction

Objective: To observe the clinical efficacy of acupoint pressure plus long-snake moxibustion for upper-limb spastic hemiplegia after cerebral infarction. Methods: A total of 100 patients were randomized into a control group and an observation group, with 50 cases in each group. Both groups were treated with the same conventional internal medicine and rehabilitation training. The control group was treated with additional acupoint pressure therapy, and the observation group was treated with long-snake moxibustion on the basis of the treatment given to the control group. The Ashworth grade, Fugl-Meyer assessment upper limb scale (FMA-UL) and Barthel index (BI) were evaluated, and the root mean square (RMS) values of biceps brachii and flexor carpi radialis on the affected side were measured before and after treatment. The efficacy was evaluated after treatment. Results: After treatment, the total effective rate of the observation group was significantly higher than that of the control group (P<0.05). After treatment, the Ashworth grade of the observation group was superior to that of the control group (P<0.05). The scores of FMA-UL and BI in both groups increased compared with those before treatment (all P<0.05), and the scores of FMA-UL and BI in the observation group were higher than those in the control group (both P<0.05). The RMS values of biceps brachii and flexor carpi radialis in both groups decreased compared with those before treatment (all P<0.05), and the RMS values of biceps brachii and flexor carpi radialis in the observation group were lower than those in the control group (both P<0.05). Conclusion: Based on conventional internal medicine and rehabilitation training, acupoint pressure plus long-snake moxibustion has great therapeutic efficacy for upper-limb spastic hemiplegia after cerebral infarction. It can improve the degree of spasticity of the affected upper limb, reduce the muscle tone of biceps brachii and flexor carpi radialis on the affected side, and enhance the mobility of the affected limb and the activities of daily living.
9.Egr-1 promoter regulating effect on granulocyte-macrophage colony-stimulating factor (GM-CSF) gene expression induced by doxorubicin and ionizing radiation

Objective To explore the regulating effects of Egr-1 promoter activated by ionizing radiation (IR) and doxorubicin (ADM) on the expression of granulocyte-macrophage colony-stimulating factor (GM-CSF) genes. Methods The human GM-CSF cDNA and enhanced green fluorescent protein (EGFP) cDNA were linked together with IRES(internal ribosome entry site) and then inserted into the expression vector pCIneo under control of the Egr-1 promoter(Egr-EG). The vector was transferred into human bone marrow stromal cell line HFCL by liposome transfection. And the cells were exposure to ADM and IR. The activity of EGFP in HFCL/EG cells were detected by FACS. The effect of N-acetylcysteine on the expression of EGFP following exposure to ADM and IR was examined. The amounts of GM-CSF in HFCL/EG after chemotherapy or radiation were measured with ELISA. The effects of GM-CSF in HFCL/EG cultural supernatants on expansion of CFU-GM derived from cord blood were also studied. RT-PCR analysis for the expression of GM-CSF mRNA in HFCL/EG after exposure to ADM or IR. Results The percentage of EGFP+ HFCL/EG cells exposed to ADM and IR was increased compared with non-treatment group (1.2 % and 15.2 % vs 18.2 %, t = 5.11, P < 0.01). The levels of secreted GM-CSF in HFCL/EG cells exposed to ADM and IR was increased (P < 0.01), but no difference between ADM group and IR group (P 0.05). The expression of EGFP by HFCL/EG treated with ADM and IR was significantly decreased by N-acetylcysteine. The effects of GM-CSF in HFCL/EG cultural supernatants on expansion of CFU-GM in ADM group and IR group were significantly higher than that in HFCL group and non-treatment group. However, The CFU-GM count of IR group was higher than that of ADM group. The expression of GM-CSF mRNA in HFCL/EG cells exposed to ADM and IR was significantly increased(t = 4.37, P < 0.01). Conclusions GM-CSF gene expression regulated by Egr-1 promoter induced by ADM and IR could help the recovery from hematopoietic injury.
