2.Investigation of drinking water fluoride and fluorosis in Shaanxi Province from 2005 to 2007

Objectives To investigate fluoride in drinking waters and fluorosis status and evaluate the effectiveness of fluoride-reducing projects in Shaanxi Province from 2005 to 2007. Methods In the Shaanxi province-wide, the 10 endemic areas of fluorosis were chosen according to historical data as focusing areas for investigation. The village was considered as investigation spot, 5 water samples were collected from each village for investigating of fluoride content. Four water samples were collected from each fluoride-reducing project for evaluating its effectiveness. Fluoride concentrations in drinking water were measured by fluoride-selective electrode method or speetrophotometry. When fluoride content in drinking water was greater than 1.00 mg/L, the epidemical study wasd conducted to investigate fluorosis patients, focusing on investigating of dental fluorosis prevalence in 8 to 12-year-old children and skeletal fluorosis prevalence in adults. Dental fluorosis of children was diagnosed by using Dean's method, and adult skeletal fluorosis was diagnosed according to National Standard for Clinical Diagnosis of Endemic Skeletal Fiuorosis(GB 16396-1996). Results The fluoride content in drinking water from 6390 villages was measured. The fluoride content of drinking water of 2619 villages ranged from 1.0 to<2.0 mg/L, where 1 654 998 people exposed. Additionally, the fluoride content of water of 845 villages ranged from 2.0 to<4.0 mg/L, where 355 623 people exposed. Moreover, the fluoride contents of water of 272 villages exceeded 4.0 mg/L, where 111 466 people exposed. The median of fluoride content in drinking water was 1.15% in the whole province, and fluoride content in drinking water exceeded 1.00 mg/L in Weinan, Xianyang and Yulin where were concentrated distribution areas of high fluoride water. Among 3115 fluoride-reducing projects, the fluoride content of drinking water of 1269 projects ranged from 1.0 to<2.0 mg/L, where 1 415 877 people exposed. Additionally, the fluoride content of drinking water of 120 projects ranged from 2.0 to<4.0 mg/L, where 43 888 people exposed. Moreover, the fluoride content of drinking water of 14 projects exceeded 4.0 mg/L, where 5960 people exposed. The detectable rate of dental fluorosis of 8 to 12 year-old children and skeletal fluorosis of adults reached 37.4%(16 489/44 081) and 5.1%(15 877/310 993), respectively. Conclusions The widely distribution of high-fluoride in drinking water still contributes to the prevalence of fluorosis in Shaanxi Province. The quality of fluoride-reducing projects should be further improved.
4.Expression of thioredoxin reductase in myeloid leukemia and its significance

Objective To explore the expression of thioredoxin reductase (TrxR) mRNA and protein,as well as the relationship between TrxR and myeloid leukemia.Methods Bone marrow samples from 20 acute myeloid leukemia (AML),20 chronic myeloid leukemia (CML),and 20 healthy persons as normal control group were collected.The human non small cell lung cancer A549 cells were selected as the positive control group.The expression of TrxR mRNA was detected by real-time quantitative polymerase chain reaction.The expression of TrxR protein level was detected by Western blot.Results The relative quantitation expressions of TrxR mRNA were 6.751 (13.459),4.321 (11.389),18.477 (2.089),1.045 (0.467) in AML,CML,positive control and normal control group,were 17.814±3.979 and 4.860±1.550 in the primary diagnosed/relapse group and complete remission (CR) group of AML patients,and were 19.552 (5.758) and 3.459 (2.047) in the primary diagnosed and treatment group of CML patients.The expression levels of TrxR mRNA in AML,CML and positive control group were significantly higher than that in normal control group (H =43.978,P < 0.001).For AML patients,the expression levels of TrxR mRNA in the primary diagnosed/relapse group,CR,positive control group were significantly higher than that in normal control group (F=246.793,P < 0.001),and the difference between the primary diagnosed/relapse group and positive control group was no significant (P > 0.05).The expression of TrxR mRNA were higher in the primary diagnosed/relapse group than that in CR group,and that in CR group than that in normal control group (both P < 0.05).For CML patients,the expression levels of TrxR mRNA in the primary diagnosed,treatment,positive control group were significantly higher than that in normal controls (H =38.222,P < 0.001),the difference between that in the primary diagnosed and that in positive control group was no significant difference (P > 0.05),but the expression of TrxR were significantly higher in the primary diagnosed group than that in treatment group,and that in treatment group than that in normal controls (both P < 0.05).The expression of TrxR mRNA in the primary diagnosed/relapse group of AML and the primary diagnosed group of CML showed no significant difference (P > 0.05).The positive levels of TrxR protein were 0.679,0.606,0.877 and 0.095 in AML,CML,positive control and normal control group.TrxR protein was highly expressed in myeloid leukemia patients.Conclusion The expression levels of TrxR mRNA and protein are increased in myeloid leukemia.The low expression of TrxR in normal hematopoietic stem cells is highly expressed in myeloid leukemia,and is downgraded after treatment,which may be used as one of the parameters to diagnosis,effect and prognosis of myeloid leukemia.
5.Clinical Applied of Radiopaque Marker's Method on Gastrointestinal Motility

Objective To investigate the clinical value of radiopaque markers method on gastrointestinal motility.Methods Using self-made markers,2 mm?2 mm,20 markers were put into a capsule.42 patients with functicnal dypepsia(FD),50 patients with chronic functional constipation(CFC)and 20 healthy controls were included in the study.Each subjects ingested a capsule together with standard food and plain abdominal films were made after 4 h,48 h and 72 h.Gastric emptying rate at the fourth hour,gastrointestinal emptying rate at 48 h and 72 h and emptying index at 72 h were calculated.Results 66.6%(28/42)FD patients had delayed gastric emptying rate.Gastrointestinal emptying rate at 48 h and 72 h were lower in CFC patients than in controls (?
6. Short-term clinical efficacy of radioactive125i seed implantation in treatment of pleural recurrence of thymoma

Objective To investigate the short-term clinical efficacy of radioactive125I seed implantation in the treatment of patients with thymoma recurrence. Methods Clinical data of patients with thymoma, who underwent radioactive125I seed implantation in Huadong Hospital of Fudan University during Sep. 2016 and Jun. 2017, were retrospectively collected. Inclusion criteria: local pleural recurrence lesions; intolerable to systemic chemotherapy or failure in previous chemotherapy; previously received high-dose chest radiotherapy; and normal coagulation function. All the patients were examined by computed tomography (CT), ultrasound and positron emission tomography-computed tomography (PET-CT) before operation. According to the Radiotherapy Planning System (TPS), the patients were inserted with the particle needles at the puncture point under the guidance of CT or ultrasound, and the distance between the sources was generally 0.5 cm. The clinical efficacy and adverse reactions were observed at 2 months after treatment. The patients were followed up for 3-9 months. Results A total of 13 patients underwent radioactive125I seed implantation, aged from 30 to 78 years, with 7 males and 6 females. The median number of implanted particles was 30, ranging from 20 to 58; the median dose of the particles was 1.9×107 Bq, and the range was (1.5-2.6)×107 Bq. The objective response rate was 100% (7 cases in complete remission and 6 in partial remission) after 2 months of follow-up, and all the 4 patients with chest wall pain were relieved. Pneumothorax occurred in 1 patient during particle implantation. During 3 to 9 months of follow-up, no patient developed disease progression at the site of particle implantation. Conclusion Radioactive125I seed implantation is a safe and effective method in the treatment of pleural recurrence of patients with thymoma, and can be used as a salvage therapy after the failure of traditional radiotherapy and chemotherapy, but its long-term effect needs further observation.
7.Clinicopathological analysis of central and extraventricular neurocytoma: A report of 17 cases.

Neurocytoma, a rare brain tumor, is characterized by a mass located mainly in cerebral ventricles. It is prone to be misdiagnosed as oligodendroglioma or ependymoma due to their similar histopathological features in clinical practice. This study aimed to examine the clinicopathological features and differential diagnosis of central and extraventricular neurocytoma. The clinical and histopathological data of 17 patients (male: female=7:10; age: 4-41 years; mean age: 27.4 years) with central or extraventricular neurocytoma were retrospectively analyzed. These patients showed typical radiological, histopathological and immunohistochemical features of neurocytoma. The tumor tissue was found to be composed of small uniform cells with round nuclei and clear cytoplasm resembling that of oligodendroglioma and ependymoma. Immunohistochemistry revealed the tumor tissues were positive for neuronal markers such as synaptophysin (SYN) and neuronal nuclear antigen (NeuN). It was concluded histopathological features of neurocytoma overlaps with some tumors in the central neural system. Immunopositivity for SYN and NeuN can help differentially diagnose neurocytoma.

9.Researching on fingerprint of Inulacappa by HPLC.

OBJECTIVEThis study is to establish the fingerprint and find out the common chromatographic peaks of Inula cappa by HPLC.
METHODThe HPLC analysis was performed on an Agilent Eclipse Plus C18 column (2.1 mm x 150 mm, 1.8 μm) with 0.1% fomic acid aqueous solution-0.1% fomic acid acetonitrile solution as mobile phase at a flow rate of 0.3 · mL(-1) · min(-1); The detective wavelength is 325 nm; The column temperature is 45 °C.
RESULTThe results indicated that 5 of 17 common peaks were identified . The similarity about 10 groups of Inulacappais is over 0.95.
CONCLUSIONThis method is able to be a scientific basis of quality assessment according to its convenient and reliable.

10.In vitro growth characteristics of bone marrow mesenchymal stem cells derived from patients with multiple myeloma and significance of TGF-β1 gene expression

Objective To investigate the expression and significance of transforming growth factor-β1 (TGF-β1) gene in bone marrow mesenchymal stem cell (BMMSC) derived from patients with multiple myeloma (MM).Methods BMMSC of 7 MM patients and 10 patients with iron deficiency anemia were cultured in vitro.The morphology of BMMSC was observed and the growth curve was portrayed according to the daily results of BMMSC proliferation.Total RNA was extracted from BMMSCs and transcription of TGF-β1 gene in BMMSC was measured by reverse transcription-PCR.Results The proliferative activity of BMMSC was not significantly different between the two groups,but expression of TGF-β1 gene of BMMSC was higher in MM patients (0.01241±0.00419) than the control group (0.00122±0.00030) (t =3.218,P < 0.05).Conclusion The abnormally high expression of TGF-β31 gene in BMMSCs could contribute to the pathogenesis of MM.
