Purpose:To explore the effects of stem cell factor(SCF) in combination with erythropoietin (EPO) on erythropoiesis in patients with anemia associated with non-Hodgkin's lymphoma (NHL). Methods:Bone marrow (BM) mononuclear cells (MNC) from NHL patients and normal simples were cultured for 7 consecutives days in the presence or absence of SCF, EPO, and SCF+EPO in liquid medium. Erythropoiesis was analyzed by calculating the fraction of recovered erythroid cells and by flow cytometry. 4.Effect of selective cyclooxygenase-2 inhibitor NS-398 on cell apoptosis in K562 cells and its underlying mechanism

Aim To investigate the effect of selective cyclooxygenase-2 inhibitor, NS-398, on cell apoptosis in leukemia cell line K562 cells and its underlying mechanism. Methods Apoptotic cell percentage was examined by flow cytometry(FCM). The protein expression of Bcl-2 and Caspase-3 was detected by Western blot, and the activity of Caspase-3 was checked by FCM. Results NS-398 induced the cell apoptosis in K562 cells in a dose-dependent manner. After NS-398 treatment, the expression of Bcl-2 protein was downregulated, whereas the expression of Caspase-3 protein was upregulated. Moreover,the activity of Caspase-3 was increased in a dose-dependent way after NS-398 treatment. Conclusion Selective cyclooxygenase-2 inhibitor,NS-398,significantly induced apoptosis in K562 cells. The underlying mechanism might be related to the downregulation of Bcl-2 and the activation of Caspase-3.
5.Etiology of recurrent diabetes ketoacidosis in children with type 1 diabetes

Objective To analyze the etiology of diabetes ketoacidosis(DKA) in children with type 1 diabetes.Methods Totally 850 person-time of type 1 diabetes children in recent 20 years in our hospital were selected as studied subjects. Two hundred and twenty-five person-time of them were hospitalized because of DKA.Fifty-six cases (131 person-time) were due to recurrent DKA.These patients were classified into 2 groups according to onset time: group 1(diagnosed from 1982 to 1991) and group 2(diagnosed from 1992 to 2001).Results The analysis of recurrent DKA suggested that 71.8 % of them was due to infection, 20.4 % of them did not obey diabetic diet and 9.2 % of them discontinued insulin injection. 【Objective】To observe the hemorrheological changes in lacunar cerebral infarction(LCI) patients with arteriopathy of middle and small arteries.【Methods】The hemorrheological changes of cerebral arteries in 85 LCI patients without arteriopathy of large artery confirmed by cervical vessels color ultrasonography and magnetic resonance image(MRI)were observed by transcranial Doppler ultrasonography(TCD).The correlation of the hemorrheological changes with age and blood pressure(BP) of arteries was investigated.The above indexes were compared with those in 25 healthy volunteers confirmed by encephalography in the same age group.【Results】The mean blood flow rates of cerebral middle,anterior and posterior artery in LCI patients were(62.09?16.90)cm?s~(-1),(50.42?13.11)(cm?s~(-1),)and(32.33?7.55)cm?s~(-1) respectively,lower than(69.65?19.20)cm?s~(-1),(57.75?16.75)cm?s~(-1),and(38.75?8.81)cm?s~(1) respectively in the healthy volunteers(P0.05).【Conclusion】The arteriopathy of small artery in LCI patients can cause the mild decrease of blood flow rate and the increase of pulsating index.There exists a correlation of pulsating index with age and arterial blood pressure.
7.The Toxic Effects of the Extracellular Active Components from One Algae-lysing Bacteria on Chlorella Pyrenoidosa

Toxic effects of L7 lyophilized powder of extracellular active components (L7-LPEAC), extracted from the Algae-lysing bacteria L7, on Chlorella pyrenoidosa were studied according to the changes of effective photosynthesis rate (EPR), the protein content, the chlorophyll a content and the MDA content of algae. The results showed that the growth of alga was promoted at low concentrations of L7-LPEAC (0.80 g/L, 1.25 g/L). The 96 h-EC50 and 120 h-EC50 upon Chlorella pyrenoidosa are 5.75 g/L and 2.55 g/L, respec tively. The chlorophyll a content increased firstly and then decreased at high concentrations of L7-LPEAC (≥2 g/L), so did the protein content. 8.Application of quality control chart in the determination of urinary iodine

Objective Establish a laboratory quality control system to ensure accurate and reliable test data and to contrapose the influence of error factors in current detection methods for urinary iodine measurement. Methods The results of reagent blank absorbance value and uric iodine standard materials were collected, then their relevant technical indexes such as mean, standard deviation, control limit, auxiliary line were worked out. Then the quality control chart of blank test and the relative error control chart were made base on these technical indexes. And different iodine concentrations (high, middle and low concentration) were tested and their mean,relative reduction difference value, weighted mean value and critical limit Rc value were calculated, and then critical limit Rc value precision control chart was made. Results The range of absorbance of blank control test was 1.183 to 1.553. And the limit of the accuracy control Rc value was 0.0883, 0.0572, respectively, when the concentrations of urinary iodine was 0~ < 150 μg/L and 150 ~ 300 μg/L. The control limit of the relative error was 9.3%. Conclusions The method of quality control chart could be satisfactorily applied to identify the quality of the analytical results of urine iodine, and ensure the results of the urine iodine reliable and authentic.
9.Modulatory effect of curcumin on the adhesion and migration of Raji cells

Objective To study the effect of SDF1/CXCR4 and FN/?1 integrin modulated by curcumin on the adhesion and migration of B lymphoma Raji cell and to explore the mechanism of curcumin to inhibit the invasiveness of lymphoma.Methods The experiment consisted of eight groups: Control,FN,SDF,FN+anti-?1 integrin,SDF+anti-CXCR4,curcumin+FN,curcumin+SDF.Adhesion assay and migration assay were used to detect the difference of invasiveness among different groups.Results In vitro,SDF1 and FN can increase the adhesion and migration of Raji cells,which can be blocked by ?1 integrin antibody and CXCR4 antibody.Curcumin can inhibit the adhesion and migration of Raji to FN in a dose-dependant manner.Low dosage curcumin had minimal effect on the adhesion and migration of Raji induced by SDF except that 25?mol/L curcumin can inhibit the adhesion and migration of Raji.Conclusion FN/?1 integrin and SDF1/CXCR4 pathway play important roles in the adhesion and migration of Raji cells.Curcumin can decrease the invasiveness of Raji cells through inhibiting FN/?1 integrin.It has good prospect of clinical application.
10.Inhibitory effects of triptolide on cell proliferation and metastasis in Raji cells in vitro

AIM:To investigate the inhibitory effects of triptolide on cell proliferation and metastasis in Burkitt's lymphoma cell line Raji cells.METHODS: The effects of triptolide on the growth of Raji cells were studied by 3-(4,5-Dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium(MTT) assay.The effects of triptolide on the cell apoptosis of Raji cells were detected by using Annexin Ⅴ/PI double-labled cytometry.The effects of triptolide on CXCR4 expression on Raji cells were studied by flow cytometric analysis.Chemotaxis assays were performed to observe the effects of triptolide on migration of Raji cells towards recombinant human SDF-1?(rhSDF-1?)in vitro.RESULTS: Triptolide inhibited the proliferation of Raji cells in a dose-and time-dependent way with a 24 h IC50 value of 43.06 nmol/L and a 36 h IC50 value of 25.08 nmol/L.Following the treatment of triptolide,the cell apoptosis rate was increased as the treatment concentration increased and the culture time extended.The effects were dose-and time-dependent.Triptolide could downregulate the expression of CXCR4 on Raji cells in a dose-dependent manner.Moreover,chemotaxis assay suggested that triptolide could block the migration of Raji cells to rhSDF-1? in vitro,and the inhibition was dose-dependent.CONCLUSION: Triptolide could inhibit the cell proliferation and induce the cell apoptosis of Raji cells.Furthermore,it could block the cell metastasis of Raji cells in vitro and the underlying mechanism might be related to the inhibition of the SDF-1/CXCR4 axis.
