1.Preparation and Quality Control of Minoxidil Gel

OBJECTIVE:To prepare the minoxidil gel and to establish a quality control method for the gel.METHODS:Carbopol934was used as the gel base;the content of minoxidil was determined with UV-spectrogram and the stability test was performed.RESULTS:There was a good linearity of calibration curve of minoxidil in range of2~10?g/ml(r=0.9999).The average recovery was99.8%,and RSD was0.67%(n=4).The gel was stable.CONCLUSION:The gel is reasonable in for?mulation,simple and accurate in quality control,as well as satisfactory in stability.

3.Significance of DNA ploidy analysis in diagnosis of ASCUS

Objective To investigate the significance of DNA ploidy analysis in diagnosis of atypical squamous cell of undetermined significance ( ASCUS).Methods From Jan.2009 to Jul.2011,875 women with ASCUS confirmed by liquid based thin layer cytology technique underwent DNA ploid analysis in Hubei Maternal and Child Health Hospital.Among 294 women underwent high risk HPV detection.All subjective were examined colposcopy directed biopsy at day 3 to 10 after menstruation.Results Among 875 ASCUS cases,553 cases with histologically as chronic cervicitis (63.2%),165 cases with cervical intraepithelial neoplasia (CIN) Ⅰ (18.9% ),45 cases with CIN Ⅱ (5.1%),79 cases with CIN Ⅲ (9.0%) and 33 cases with cervical invasive cancer (3.8%) were confirmed by colposcopy.Totally 532 cases were observed with DNA heteroploid,and 343 were not observed with DNA heteroploid.When DNA heteroploid negative and more than or equal to three ploid were used to predict CIN Ⅱ or more severe cervical diseases,the sensitivity,specificity,positive predictive values and negative predictive values were 98.7% and 90.3%,47.5% and 46.1%,29.1% and 40.8%,99.4% and 92.1%,respectively.The amount of heteroploid cells >2.5c and > 5c among every 100 detected cells in chronic cervicitis and CIN Ⅰ,CIN Ⅱ,CIN Ⅲ and cervix cancer were respectively 2.53 ± 1.99 and 0.10 ±0.07,2.24 ± 1.69 and 0.20 ±0.11,4.10 ± 1.91 and 0.28 ±0.19,7.97 ±7.33 and 1.27 ± 1.23,8.99 ±7.33 and 0.36 ±0.33,there was no statistical difference in amount of heteropolid cells between > 2.5c and > 5c at group of chronic cervicitis and CIN Ⅰ,CIN Ⅲ and cervix cancer (P > 0.05).However,the amount of heteroploid cells at >2.5c and> 5c at group of chronic cervicitis,CINⅠ,CIN Ⅲ and cervical were higher than that of CIN Ⅱ significantly (P <0.05).Among 294 cases with high risk (HR) HPV detection,216 cases were HR-HPV positive,and 78 cases were HR-HPV negative.The pathology result by colposcopy at group of negative heteroploid,heteroploid < 3,or ≥ 3 showed statistical distribution (x2 =115.2775,P < 0.01).ConclusionDNA ploidy analysis can be used for ASCUS diagnosis,which can avoid excessive biopsy under colposcopy,in the mean time,CIN and cervical cancer could decrease missed diagnosis.
5.Comparison of effects of general anesthesia versus combined epidural-general anesthesia on postoperative complications in patients undergoing thoracic surgery

Objective To compare the effects of general anesthesia versus combined epidural?general anesthesia on postoperative complications in the patients undergoing thoracic surgery. Methods Two hundred twenty?one patients of both sexes, aged 18-80 yr, weighing 36-100 kg, of American Society of Anesthesiologists physical status Ⅰ?Ⅲ, scheduled for elective thoracic surgery, were randomly divided into either combined epidural?general anesthesia group ( group GE, n=112) or general anesthesia group ( group GA, n=109) . The patients were further allocated to high?risk population and low?risk population. Anesthesia was induced with target?controlled infusion of remifentanil and iv injection of propofol, sufentanil and rocuronium. A double?lumen endotracheal tube was placed, and the patients were mechanically ventilated. Partial pressure of end?tidal CO2 was maintained at 30-40 mmHg. Anesthesia was maintained with inhalation of 1% sevoflurane, target?controlled infusion of remifentanil, iv infusion of propofol, and intermittent iv boluses of sufentanil. 2% lidocaine 4 ml∕h was infused epidurally in group GE, and normal saline 4 ml∕h was given in group GA. Bispectral index value was maintained at 40-60 during surgery. Patient?controlled epidural analgesia ( PCEA ) was performed after surgery. PCEA solution contained
0.125% ropivacaine and sufentanil 0.5μg∕ml ( in 250 ml of normal saline) . The PCEA pump was set up to deliver a 2 ml bolus dose with a 20?min lockout interval and background infusion at 4 ml∕h, and visual analogue scale ( VAS) score was maintained less than 4. When VAS score ≥ 5, sufentanil 5 μg was injected intravenously as the rescue analgesic. The requirement for rescue analgesics after surgery, length of hospital stay after surgery, complications during postsurgical hospital stay ( cardiovascular events, cardiac insufficiency, acute coronary syndrome, pulmonary infection, stroke, transient cerebral ischemic attack) , and death within 30 days after surgery were recorded. Results Compared with group GE, the incidence of postsurgical cardiovascular events and atrial fibrillation was significantly decreased in group GA ( P<0.05) . In the high?risk population, compared with group GE, the length of hospital stay after surgery was significantly shortened, and the incidence of postsurgical atrial fibrillation was decreased in group GA ( P<0.05) . In the low?risk population, compared with group GE, the length of hospital stay after surgery was significantly shortened (P<0.05), and no significant change was found in the incidence of postsurgical complications, requirement for rescue analgesics after surgery, and fatality rate in group GA (P>0.05). Conclusion Compared with combined epidural?general anesthesia, general anesthesia alone provides lower occurrence of postsurgical complications for the patients undergoing thoracic surgery, and offers advantage for the high?risk patients having underlying diseases before surgery.
6.Exposure in complicated total knee arthroplasty using tibial tubercle osteotomy

[Objective]To explore the efficacy and results of tibial tubercle osteotomy used in exposure in complicated total knee arthroplasty.[Methods]During the period from Apr.2005 to Apr.2007,the tibial tubercle osteotomy were used in 16 cases of complicated total knee arthroplasty.The mean follow-up time were 20 months(6~26 months).Knee society score(KSS) and radiography were used to evaluate the clinical results.[Results]The mean KSS improved from 46 points preoperatively to 91 points postoperatively.The mean ROM improved to from 53?preoperatively 105?postoperatively.At 3 months after surgery the radiography examines showed all 16 cases had achieved satisfactory healing.The tubercle fragment slided toward proximal 0.7 cm occurred in one case,and finally healed at that position.[Conclusion]Exposure of the knee may be difficult in the total knee arthroplasty,but tibial tubercle osteotomy is a safe and reliable procedure which affords excellent exposure.
7.Mediating effect of loneliness on relationship between interpersonal adaptation and mobile phone addiction in college students

Objective:To explore the mediating effect of loneliness on the relationship between interpersonal adaptation and mobile phone addiction in college students.Methods:Totally 368 college students (176 males and 192 females )were surveyed with the College Student Adaptability (CSAI),University of Califomiaat Los Angels (UCLA)and Mobile Phone Addiction Index (MPAI).Then AMOS18.0 was used to establish structural equation modeling (SEM)among interpersonal adaptation,loneliness and mobile phone addiction,and test the mediating effect of loneliness on the relationship between interpersonal adaptation and mobile phone addiction in college students.Results:The MPAI scores were significantly higher in the low interpersonal adaptation group (the score of CSAI less than 21 )than in the high interpersonal adaptation group (the score of CSAI higher than 32)(P<0.001).The scores of interpersonal adaptation were negatively correlated with scores of loneliness and mobilephone addiction (r =-0.71,-0.25,P﹤0.01).The scores of loneliness were positively correlated with scores ofmobile phone addiction (r =0.32,P ﹤ 0.01).College students'interpersonal adaptation did not have significantnegative predictability for mobile phone addiction (β=﹣0.08,P =0.32),but it had significant negative predictabilityfor loneliness (β=﹣0.71,P <0.001).Loneliness had significant positive predictability for mobile phone addiction(β=0.29,P <0.001).Conclusion:The results show loneliness completely plays a mediating effect betweeninterpersonal adaptation and mobile phone addiction.
8.The effect of bFGF on apoptosis of cultured human retinal pigment epithelial cells exposed to visible light

Objective To observe the effect of exogenous basic fibroblast growth factor (bFGF) on apoptosis of cultured human retinal pigment epithelial (RPE) cells exposed to visible light,and determine the role of bFGF, fibroblast growth factor receptor 1 (FGFR1),bcl 2 and caspase 3. Methods (2000? 500) lx cold white light was used. Exogenous bFGF was utilized during culture. Annexin annexin V fluorescein isothiocyanate/propidium iodium (V FITC/PI) labeling,flow cytometry, Immunocytochemical staining, enzyme associated absorb examing and reverse transcriptional polymerase chain reaction (RT PCR) were used to determine the apoptosis, the expression levels of bFGF, FGFR1, bcl 2, as well as the activity of caspase 3. Results No protective effect of bFGF was observed under the concentration 5 ng/ml. 9.Changes of expression of estrogen receptor ? in the paraventricular nucleus of mice during pregnancy and lactation

Objective To study the expression of estrogen receptor beta (ER ?) in the hypothalamic paraventricular nucleus (PVN) of maternal mice during pregnancy and lactation. Methods Nickel ammonium sulfate intensified immunocytochemical method was employed to investigate the expression of ER ? in the postnatal developing PVN of female mice. Results ER ? immunopositive materials were predominantly localized in the magnocellular division of PVN, and sparse positive cells were found in the parvocellular division. Most of the positive materials were found in the whole cell nuclei, but no obvious cytoplasma or process immunopositive cells were detected. In the pregnant female brain, generally, the ER ? level was lower. The lowest levels of expression were found at mid pregnancy, and then peaked at peripartum (from gestational days 18 to postpartum day 1), followed by another decrease to normal adult level from postpartum day 4. Conclusion The above results suggest that during pregnancy and lactation, ER ? may be predominantly involved in the PVN regulation of parturitional process.
10.Integration and Expression of BGL1 Gene from Saccharomy-copsis fibuligera in Industrial Saccharomyces cerevisiae

The recombinant plasmid pPIC-gpd-bgl-hyg was constructed, which contained GPD2 promotor and terminator from industrial yeasts Saccharomyces cerevisiae, ?-glucosidase gene (BGL1) from Sac-charomycopsis fibuligera and hyg from hygromycin as the selected marker. With the yeast’s high efficiency of homologous integrated, the BGL1 gene was successfully integrated into industrial yeasts S. cerevisiae. The recombined yeast could grow on the cultures with the cellobiose as a sole carbon source, and the ?-glucosidase activity achieved 0.764 U/mL after 48 hours’ cultivation. In the experiments of VHG ethanol fermentation, the cellobiose concentration in broth of recombined yeast was 80% lower than that of indus-trial yeast.
