1.Preparation and Quality Control of Minoxidil Gel

OBJECTIVE:To prepare the minoxidil gel and to establish a quality control method for the gel.METHODS:Carbopol934was used as the gel base;the content of minoxidil was determined with UV-spectrogram and the stability test was performed.RESULTS:There was a good linearity of calibration curve of minoxidil in range of2~10?g/ml(r=0.9999).The average recovery was99.8%,and RSD was0.67%(n=4).The gel was stable.CONCLUSION:The gel is reasonable in for?mulation,simple and accurate in quality control,as well as satisfactory in stability.

4.Study of technical method of immunohistochemical staining in pathological tissue

Objective To study the technical method of immunohistochemical staining in pathological tissue.Methods 80 cases of hospitalized patients in our hospital from May 2015 to May 2016 were selected, they were randomly divided into immunohistochemical staining group(immunohistochemistry group, n=40)and biopsy group(n=40)two groups, then the AFP(alpha-Fetoprotein), Glypican-3 positive situation of the the two groups and diagnostic Resultsof patients with different types of disease in the immunohistochemistry group were statistically analyzed.ResultsThe positive rate of AFP(a-Fetoprotein), Glypican-3 of the immunohistochemistry group 92.5%(37/40)was significantly higher than the biopsy group 50.0%(20/40)(P<0.05), Among the 37 cases of positive patients, metastatic tumors 19 cases, hepatocellular carcinoma 9 cases, intrahepatic bile duct carcinoma 7 cases, hemangioma 3 cases, the positive rates were 100.0%, 75.0%, 100.0%, 100.0%, 26 cases died, the mortality rate was 70.3%, of which metastatic tumors 13 cases, hepatocellular carcinoma 6 cases, intrahepatic bile duct carcinoma 5 cases, hemangioma 2 cases, the mortality rates were 72.2%, 66.7%, 71.4%, 66.7%.Conclusion Immunohistochemical staining technic in pathological tissue can promote positive rate, and standard operation procedures can reduce errors, which is worthy of clinical attention.
5.Application of ARIMA model to forecasting bacillary dysentery in Jianghan District of Wuhan City

Objective To explore the feasibility of the application of autoregressive integrated moving average(ARIMA) model to forcast the monthly incidence rate of bacillary dysentery in Jianghan District of Wuhan ,and to provide scientific references for prevention and control .Methods SAS 9 .2 software was used to fit the ARIMA model for the monthly incidence rate of bacil‐lary dysentery from 2005 to 2013 .Thereafter ,the fitted model was used to forecast the data from January to June in 2014 ,and was evaluated by comparing the actual incidence .Results The ARIMA(0 ,1 ,1)X(0 ,1 ,1)12 model had a better fitted effect ,it was pre‐dicted that the incidence of bacillary dysentery of Jianghan District of Wuhan City from January to June 2014 would increase slight‐ly ,and the forecast data of bacillary dysentery from January 2005 to December 2013 consisted with the actual situation .Conclusion ARIM A model can be used as short‐term forecasting model to predict the monthly incidence rate of bacillary dysentery in Jianghan District of Wuhan .
6.Significance of DNA ploidy analysis in diagnosis of ASCUS

Objective To investigate the significance of DNA ploidy analysis in diagnosis of atypical squamous cell of undetermined significance ( ASCUS).Methods From Jan.2009 to Jul.2011,875 women with ASCUS confirmed by liquid based thin layer cytology technique underwent DNA ploid analysis in Hubei Maternal and Child Health Hospital.Among 294 women underwent high risk HPV detection.All subjective were examined colposcopy directed biopsy at day 3 to 10 after menstruation.Results Among 875 ASCUS cases,553 cases with histologically as chronic cervicitis (63.2%),165 cases with cervical intraepithelial neoplasia (CIN) Ⅰ (18.9% ),45 cases with CIN Ⅱ (5.1%),79 cases with CIN Ⅲ (9.0%) and 33 cases with cervical invasive cancer (3.8%) were confirmed by colposcopy.Totally 532 cases were observed with DNA heteroploid,and 343 were not observed with DNA heteroploid.When DNA heteroploid negative and more than or equal to three ploid were used to predict CIN Ⅱ or more severe cervical diseases,the sensitivity,specificity,positive predictive values and negative predictive values were 98.7% and 90.3%,47.5% and 46.1%,29.1% and 40.8%,99.4% and 92.1%,respectively.The amount of heteroploid cells >2.5c and > 5c among every 100 detected cells in chronic cervicitis and CIN Ⅰ,CIN Ⅱ,CIN Ⅲ and cervix cancer were respectively 2.53 ± 1.99 and 0.10 ±0.07,2.24 ± 1.69 and 0.20 ±0.11,4.10 ± 1.91 and 0.28 ±0.19,7.97 ±7.33 and 1.27 ± 1.23,8.99 ±7.33 and 0.36 ±0.33,there was no statistical difference in amount of heteropolid cells between > 2.5c and > 5c at group of chronic cervicitis and CIN Ⅰ,CIN Ⅲ and cervix cancer (P > 0.05).However,the amount of heteroploid cells at >2.5c and> 5c at group of chronic cervicitis,CINⅠ,CIN Ⅲ and cervical were higher than that of CIN Ⅱ significantly (P <0.05).Among 294 cases with high risk (HR) HPV detection,216 cases were HR-HPV positive,and 78 cases were HR-HPV negative.The pathology result by colposcopy at group of negative heteroploid,heteroploid < 3,or ≥ 3 showed statistical distribution (x2 =115.2775,P < 0.01).ConclusionDNA ploidy analysis can be used for ASCUS diagnosis,which can avoid excessive biopsy under colposcopy,in the mean time,CIN and cervical cancer could decrease missed diagnosis.
7.Comparison of effects of general anesthesia versus combined epidural-general anesthesia on postoperative complications in patients undergoing thoracic surgery

Objective To compare the effects of general anesthesia versus combined epidural?general anesthesia on postoperative complications in the patients undergoing thoracic surgery. Methods Two hundred twenty?one patients of both sexes, aged 18-80 yr, weighing 36-100 kg, of American Society of Anesthesiologists physical status Ⅰ?Ⅲ, scheduled for elective thoracic surgery, were randomly divided into either combined epidural?general anesthesia group ( group GE, n=112) or general anesthesia group ( group GA, n=109) . The patients were further allocated to high?risk population and low?risk population. Anesthesia was induced with target?controlled infusion of remifentanil and iv injection of propofol, sufentanil and rocuronium. A double?lumen endotracheal tube was placed, and the patients were mechanically ventilated. Partial pressure of end?tidal CO2 was maintained at 30-40 mmHg. Anesthesia was maintained with inhalation of 1% sevoflurane, target?controlled infusion of remifentanil, iv infusion of propofol, and intermittent iv boluses of sufentanil. 2% lidocaine 4 ml∕h was infused epidurally in group GE, and normal saline 4 ml∕h was given in group GA. Bispectral index value was maintained at 40-60 during surgery. Patient?controlled epidural analgesia ( PCEA ) was performed after surgery. PCEA solution contained
0.125% ropivacaine and sufentanil 0.5μg∕ml ( in 250 ml of normal saline) . The PCEA pump was set up to deliver a 2 ml bolus dose with a 20?min lockout interval and background infusion at 4 ml∕h, and visual analogue scale ( VAS) score was maintained less than 4. When VAS score ≥ 5, sufentanil 5 μg was injected intravenously as the rescue analgesic. The requirement for rescue analgesics after surgery, length of hospital stay after surgery, complications during postsurgical hospital stay ( cardiovascular events, cardiac insufficiency, acute coronary syndrome, pulmonary infection, stroke, transient cerebral ischemic attack) , and death within 30 days after surgery were recorded. Results Compared with group GE, the incidence of postsurgical cardiovascular events and atrial fibrillation was significantly decreased in group GA ( P<0.05) . In the high?risk population, compared with group GE, the length of hospital stay after surgery was significantly shortened, and the incidence of postsurgical atrial fibrillation was decreased in group GA ( P<0.05) . In the low?risk population, compared with group GE, the length of hospital stay after surgery was significantly shortened (P<0.05), and no significant change was found in the incidence of postsurgical complications, requirement for rescue analgesics after surgery, and fatality rate in group GA (P>0.05). Conclusion Compared with combined epidural?general anesthesia, general anesthesia alone provides lower occurrence of postsurgical complications for the patients undergoing thoracic surgery, and offers advantage for the high?risk patients having underlying diseases before surgery.
8.Effect of Various Kind of Dyslipidaemia on Insulin Resistance and Pancreatic Islet Function in Newly Diagnosed Type 2 Diabetic Patients

Objective To evaluate the effect of various kinds of dyslipidaemia on insulin resistance and pancreatic islet ? cell function in newly diagnosed type 2 diabetic patients. Methods 263 patients with newly diagnosed type 2 diabetes were divided into normal lipidemia group and dyslipidaemia group according to ATPⅢ standard. The dyslipidaemia group included 3 subgroups as the following: high TG group, high TC group, and mix group with high TG and TC. Homeostasis model assessment (HOMA) was applied to estimate insulin resistance (IR) and pancreatic islet ? cell function. Objective To summarize the meticulous nursing experience of indwelling urinary catheter between the children with hypospadias during perioperative. Methods 317 cases of indwelling urethral only in the silicone Foley tube, and without other urinary diversion with hypospadias were retrospectively analyzed, summarized the fine points of nursing, including preoperative children with comprehensive risk assessment, standardized preoperative skin preparation and postoperative protective constraint, effective properly fixed catheter keep the urine drainage unobstructed, close observation of urine drainage characteristics, maintain urine collection system tightness, maintain perineal skin clean and improved extubation nursing method, and do a good job after extubation nursing. Results 266 cases were cured with a cure rate of 83.9%(266/317);the incidence of urine leakage in 42 cases, the incidence rate was 13.2%(42/317); 7 cases of urethral stenosis, the incidence rate was 2.2%(7/317); 2 cases of urethral closing, the incidence was 0.6% (2/317), none of the patients had accidental extubation. Conclusions The meticulous nursing has a better effect in hypospadias patients during the perioperative period and it can reduce the incidence of postoperative complications;shorten the time of hospitalization and alleviate the suffering of children;improve the quality of nursing service, which is worthy of clinical application.
10.The effect of visible light on apoptosis of cultured human retinal pigment epithelium cells

Objective To observe the effect of visible light on apoptosis of cultured human retinal pigment epithelium (RPE) cells. Methods Being the light source,500lx,(2 000?500)lx and (3 400?200)lx cold white light were used. The duration of exposure was 0,6,12 and 24 hours respectively. Apoptosis was detected by terminal deoxynucleotidyl transferase mediated dUTP nick end labelling, Annexin V flunorescein isothiocyanate/Propidium iodium labelling and flow cytometry. Results Apoptosis and necrosis were found in cultured human RPE cells which were exposed to visible light.(1)A significant increase in apoptotic and necrotic percentages was consistent with a higher light intensity.(2)Apoptosis was the main response to shorter (6 h and 12 h) exposure duration,while necrosis was more pronounced correlated to the prolongation of post exposure culture ( P 500 lx) increases the proportion of apoptosis and necrosis of human RPE cells in vitro.The extent is related to exposure intensity and duration. It demonstrates that the lower intensity and the shorter duration of exposure to light are, the more pronounced apoptotic percentages are observed,otherwise necrosis.
