1.Optimization of β-Cyclodextrin Inclusion Process for Volatile Oil in Yingxinning Capsules by Orthogonal Test

Objective:To optimize β-cyclodextrin (β-CD) inclusion process for volatile oil in Yingxinning capsules. Methods:The content of volatile oil in theβ-CD inclusion compound as the evaluation index, the ratio of volatile oil toβ-CD, inclusion tempera-ture, inclusion time and stirring speed as the influencing factors, the inclusion process was studied using L9(34)orthogonal design. Re-sults:The best preparation conditions were as follows:the ratio of volatile oil toβ-CD was 1∶ 9, the inclusion temperature was 40℃, the inclusion time was 30min and the stirring speed was 800 r·min-1 . Conclusion: The preparation technology is simple, feasible and stable with high inclusion rate of volatile oil.
2.Diagnostic value of basophil activation test in food allergy in children

At present,diagnostic programs of food allergy in children mainly include medical history,skin prick test,detection of serum specific immunoglobulin E (sIgE) antibody,food challenge test,and so on.Among them,skin prick test and detection of sIgE antibody are used as primary screening tests with high diagnostic sensitivity while low specificity.Additionally,double-blind placebo-controlled food challenge test is the gold standard for the diagnosis,but it is faced with a potential risk of causing systemic allergic reactions,resulting in its rare application in clinic.Moreover,basophil activation test,as an in vitro test for allergen diagnosis,has significant value in diagnosis of food allergy in children due to its high specificity and safety.Besides,basophil activation test can also be used to evaluate the severity of food allergy in children,monitor immune therapy,detect trace allergens,and so on.
3.Effects of Batroxobin on intimal proliferation of graft veins

Objective To study the effects of Batroxobin( BX) on the intimal proliferation of graft veins. Methods In this study 25 dogs were selected and evenly divided into experimental group, control group and sham operation group. In experiment and control group, a segment of auto- femoral vein were grafted into femoral artery by clean microsurgery technique. In experimental group, BX was given at the dosage of 0. 1 BU/kg, dayly?2 preoperatively, and once a day for consecutive 6 days postoperatively. Plasma NO, ET was determined in the three groups. Computer image analysis system was applied to calculate the thickness of neointima and media in the vein grafts, immunohistochemistry was used to identify PCNA and C-myc. Result The experimental group had a higher level of NO and lower level of ET compared with control group and sham operation group(P 0. 05 ). 4.Expression changes in fractalkine induced by focal cerebral ischemia and reperfusion in rats

Objective To investigate the expression changes of fractalkine (FKN)in focal cerebral ischemia and reperfusion penumbra,and to explore its variation law and role in the inflammation of cerebral ischemia-reperfusion injury.Methods The cerebral ischemia reperfusionmodel was established by intraluminal thread occlusion in the middle cerebral arteries occlusion (MCAO).FKN protein expression in focal cerebral ischemia and reperfusion penumbra was detected by immunohistochemistry and Western blot.Results The results of immunohistochemistry stain showed that the chemokine FKN was expressed in a low level in the normal group and the sham operation group,and there were no significant differences among the two groups (P> 0.05).Compared with the humbers of FKN in normal group (37.03± 6.28) in focal cerebral ischemia and reperfusion penumbra,the expression of FKN in model group was increased after 3 h of reperfusion (48.58±7.29) (P<0.05),peaked at 24 h (112.08±8.26) (P<0.05],and then decreased gradually at day 7 after reperfusion,but had no significant difference (40.73 ± 4.02) (P> 0.05).FKN was expressed in a low level in the sham operation group (0.527±0.002),then up-regulated after 3 h of reperfusion [(0.598±0.004),P<0.05],peaked at 24 h [(0.833±0.005),P<0.05],maintained a high level till 48 h after reperfusion [(0.735±0.002),P<0.05],and return to baseline level at day 7 after reperfusion [(0.533±0.004),P>0.05].Conclusions Fractalkine is upregulated after focal cerebral ischemia/reperfusion,and has a dynamical change,which indicates that fractalkine might involve in the inflammatory process after cerebral ischemia-reperfusion injury.

6.Effectiveness of enteral nutrition support and growth hormone treatment in critically ill patients

Objective To explore the effectiveness of early enteral nutrition (EN) support and growth hormone (GH) treatment in critically ill patients.Methods Seventy critically ill patients were randomly divided into early EN support group and early EN support plus GH treatment group.The nutrition intakes were isonitrogenic and isocalorie in these two groups.Body weight,blood biochemistry,nutritional statues,and lactulose/mannitol levels were measured or analyzed before and after nutrition support.Immunologic functions were analyzed after ten days.Nitrogen balance was measured daily.Results The changes of body weight,albumin level,and transferrin level were more obvious in the EN + GH group than those in the EN group without significant difference (P >0.05).The changes of prealbumin and fibronectin in the EN + GH group were significantly higher than those in the EN group (P < 0.05).The level of IgA in the EN + GH group was significantly lower than that in the EN group,while the levels of CD4 and NK in the EN + GH group was significandy higher than those in the EN group (P <0.05).The gut barrier function in the EN + GH group was superior to that in the EN group during nutrition support (P <0.05).Nitrogen balance was positive in the EN + GH group and negative in the EN group (P < 0.05).Conclusions Early EN can improve the nutritional status and reduce complications in critically ill patients.GH treatment may strengthen the immune function and remarkably decrease the disability and mortality in critically ill patients.
7.Clinic study of the adaptability of peripheral nerve to limb lengthening

Objective To observe if subclinical lesion exists during and after tibia lengthening and investigate the repair mechanism of the nerve. MethodsTwenty-three cases(with 42 tibia) were lengthened by traction with the velocity of 1mm/d. The somatosensory evoked potentials (SEP) induced by stimulation of tibial nerve were recorded before operation and 2,3 months and 2 years after operation. The tibia was lengthened for a total of 4~10cm. ResultsAll cases had no significant symptoms except for 1 case with mild symptoms of nerve lesion when his tibia was lengthened for 9.5cm. However, the subclinical lesion was found by SEP record in all the cases, and this lesion could be repaired mostly in 3 months and completely recovered in 2 years. ConclusionThe peripheral nerves have good adaptability to tibia lengthening with a velocity of 1mm/d and the tibia could be lengthened to a great extend as long as there were no clinical symptoms manifestated.
8.Related investigation and thinking on the course of Laboratory diagnostics in general medical train-ing from the Navy physicians

Objective To investigate and analyze the application and effect of the course of Labo-ratory diagnostics in general medical training from the Navy physicians. Methods 37 students attending the general medical training in our university in September 2015 were selected as the subject. At the end of the course, the “Questionnaire Survey on Teaching Feedback” was designed and conducted among them to understand the students'!opinions on the arrangement, teaching content and teaching methods of the experi-mental diagnostics. A total of 37 questionnaires were issued and retrieved effectively and the survey data were collected and summarized. Results Among 37 questionnaires, 100%of trainees (n=37) thought it was necessary to set up the Laboratory diagnostics course in the general medical training, 72.9%(n=27) suggested 3 to 4 teaching hours, and on the demand side of teaching content settings, 91.9%(n=34) chose analyses of blood, urine, stool routine tests and then tests of liver and kidney function. The proportions of students to recommend theory teaching, case discussion and practice operation were 24.3% (n=9), 51.4% (n=19) and 48.6% (n=18) respectively. Conclusions The setup of the Laboratory diagnostics course plays an active role in the general medical training from navy physicians. In the future, according to the demand of the general medical training, teachers should optimize the teaching contents and learning hours, improve the teaching methods and enhance the training quality.

10.Expression of EZH2 mRNA and protein in human prostate carcinoma and its relationship with cell proliferation

Purpose To investigate the expression of EZH2 mRNA and protein, and its relationship with tumor cell proliferation in human prostate carcinoma (PCa).Methods A tissue microarray was constructed, which contained 68 dots of formalin-fixed,paraffin-embedded tissue samples, including 48 cases of human PCa.Immunohistochemical markers, including EZH2 and Ki-67,were used on the tissue microarray sections by the immunohistochemical staining method.In situ hybridization (ISH) using an EZH2 oligonucleotide probe was also performed on the tissue microarray sections.Additional 15 cases of benign prostate hyperplasia (BPH) and 12 cases of high grade prostate intraepithelial neoplasia (HGPIN) were used as controls.Results The positive rates of EZH2 protein and mRNA expression were 87.50% and 81.25% in the PCa, 16.67% and 16.67% in the HGPIN,13.33% and 6.67% in the BPH,respectively. There were statistical difference between PCa, HGPIN and BPH, respectively (P<0.05).The expression of EZH2 protein and mRNA had no statistical difference (P>0.05).The expression of EZH2 protein was related to Gleason score, TNM stage (P<0.05), but not to age and serum prostate-specific antigen (PSA) level (P>0.05). The expression of EZH2 mRNA was related to TNM stage (P<0.05),but not to age, PSA and Gleason score (P>0.05).The staining intensity of EZH2 was positively correlated with the Ki-67 indexes (r=0.746,P<0.05).Conclusions Over-expression of EZH2 which is involved in accelerating proliferation may play an important role in the pathogenesis and progression of human prostate carcinoma. As a molecular marker of prostate carcinoma, EZH2 may serve as a new index for estimating the level of malignancy and progression of the prostate cancer.
