1.The imaging and differential diagnosis of sclerosing cholangitis

Sclerosing cholangitis includes a spectrum of chronic, variably progressive cholestatic liver disease characterized by inflammation, fibrosis and stricture of the intrahepatic and extrahepatic bile ducts. Sclerosing cholangitis can be divided into primary sclerosing cholangitis of unidentified etiology and secondary sclerosing cholangitis caused by various identifiable etiologies, including immunoglobulin G4-related sclerosing cholangitis, recurrent pyogenic cholangitis, ischemic cholangitis, acquired immunodeficiency syndromerelated cholangitis and eosinophilic cholangitis. In this article, we describe the imaging findings of the various spectrums of sclerosing cholangitis with anemphasis on a systemic approach in differential diagnosis. It is very important for magnetic resonance cholangiography and endoscopic cholangiography in imaging diagnosis and differential diagnosis of sclerosing cholangitis. We also discuss the clinical significance and therapeutic options for treating sclerosing cholangitis.
2.Progress of Gd-EOB-DTPA in diagnosis of hepatocellular carcinoma

The liver specific contrast agent gadolinium ethoxybenzyldiethylenetriaminepe ntaacetic acid (Gd-EOB-DTPA) is routinely used in clinical magnetic resonance imaging. It combines assessment of tissue perfusion during the vascular phase with hepatocyte specific imaging during the hepatobiliary phase. This combination of imaging properties has been shown to provide excellent results in the detection and differentiation of liver lesions. The enhancement characteristics of Gd-EOB-DTPA in hepatocellular carcinoma, regenerative nodules and degenerated nodules were summarized in this review. Liver cell specific contrast agent can further improve the diagnostic ability.
3.Correlation between ABCB1 Polymorphisms and Synergistic Effects of Wuzhi Capsules on Tacrolimus

Objective:To investigate the correlation of ABCB1 polymorphisms and the synergistic effect of Wuzhi capsules on ta-crolimus. Methods:The ABCB11236C>T(rs1128503), ABCB12677G>T/A(rs2032582) and ABCB13435C>T(rs1045642) genotypes were determined by restriction fragment length polymorphism ( RFLP) analysis. The whole blood levels of tacrolimus in renal transplant recipients were measured by chemiluminescent microparticle immunoassay. Analysis of covariance ( ANCOVA) was per-formed to determine the difference of tacrolimus C0/D among the various groups. Results: Whether co-administeration of Wuzhi cap-sules and tacrolimus or not, tacrolimus C0/D of ABCB11236C>T, ABCB12677G>T/A and ABCB13435C>T genotype and haplo-type was without significant difference. Conclusion:When combined with Wuzhi capsules, ABCB11236C>T, ABCB12677G>T/A and ABCB13435C>T mutation is not associated with tacrolimus C0/D.
4.Relationship of retinal lesions and postoperative visual acuity of cataract phacoemulsification

AlM: To study the relationship of retinal associated lesions and postoperative visual acuity of cataract phacoemulsification.
METHODS:From February 2013 to February 2014, 120 cases with given cataract phacoemulsification were divided into observation group and control group. Cases in observation group were with of retinal associated lesions, those in control group were without retinal associated lesions. The best corrected visual acuity, ision acuity, visual evoked potential, intraocular pressure were compared between two groups.
RESULTS:Two weeks after surgery, visual acuity in observation group was higher than those of before therapy. Visual acuity recovery rate of observation group was significantly slower than that of control group; best corrected vision, visual acuity in observation group was significantly lower than that in control group;observation group's amplitude (12. 01±4. 50ü V) was significantly lower than control group, incubation period (114. 29±11. 32ms) was significantly higher than control group; After 6, 12, 24h, intraocular pressure (23. 64 ± 4. 28, 24. 12 ± 5. 13 and 18. 28± 3. 22mmHg) were significantly higher than control group.
CONCLUSlON: Retinal associated lesions can lead to visual evoked potential change, elevated lOP, affect postoperative visual recovery level.
5.Significance of the expression of cyclooxygenase-2 in breast cancers

Objective To investigate the expression of cyclooxygenase-2(COX-2)in breast cancers, and understand its clinical significance.Methods The immunohistochemistry was used to determine the ex- pression of COX-2 in the 40 tissues of breast cancer and 18 adjacent noncancerous mammary tissues.Results The positive expression of COX-2 was detected in 52.5% of breast cancer tissues and in 11.1% of adjacent noncancerous mammary tissues.There was a significant difference in the expression of COX-2 between breast cancer tissues and adjacent noncancerous mammary tissues(P0.05).Conclusion COX-2 was over expressed in breast cancer and correlated with metastatic lymph nodes and HER-2/neu.The expression of COX-2 may play an important role in carcinogen- esis,progression and prognosis of breast cancer.
6.Expression and clinical significance of GSK-3β,PTEN,PLK1 in pediatric acute myeloid leukemia

Objective To explore the expression and clinical significance of GSK-3β,PTEN and PLK1 in pediatric AML . Methods Experiment group was bone marrows of 33 cases incipient children with AML .Control group was 10 cases normal bone marrows .GSK-3β,PTEN and PLK1 gene expressions in BMMNC of the two groups were tested using semi quantitative reverse transcriptase polymerase chain reaction(RT-PCR)technique .GSK-3βprotein and P-GSK-3βexpressions were tested by ELISA .Re-sults The expression of GSK-3βmRNA ,GSK-3βprotein ,PLK1 mRNA in BMMNC of children with AML was higher than that of control group(P=0 .012 ;P= 0 .014 ;P= 0 .040);The expression of PTEN mRNA ,P-GSK-3β was lower than the control group (P=0 .012 ;P=0 .002);GSK-3βprotein had a negative correlation with PTEN mRNA (r= -0 .415 ,P=0 .016);GSK-3βmRNA , GSK-3βprotein had a positive correlation with PLK 1 mRNA(r=0 .388 ,P=0 .026;r=0 .427 ,P=0 .013) .The expression of GSK-3βprotein was high in which had high peripheral white blood cell counts ;both the expressions of GSK-3βmRNA and GSK-3βpro-tein were high in which had high risk ;but the expression of P-GSK-3βwere low .Conclusion In pediatric AML ,GSK-3βand PLK1 may play a role of oncogene and PTEN may play a role of tumor suppressor gene .
7.Clinical Study on Treatment of 50 Cases of Prospermia with Chinese Medicine Combined with Antidepressant

Objective:To investigate the therapeutic effect of Chinese medicine combined with small dose of antidepressant on prospermia.Methods:150 cases of prospermia conformed to the diagnostic criteria were divided into 3 groups according to their wishes,50 cases in each group.Group I were treated with Zhibai Gujing Decoction,group Ⅱ with Sertraline hydrochloride,and group Ⅲ with Zhibai Gujing Decoction plus Sertraline hydrochloride,for 4 consecutive weeks.The ejaculatory latency,satisfaction degrees of sexual life,and the adverse effect of the medicine were observed.Results:After treatment,the ejaculatory latency was significantly prolonged and the satisfaction degree of sexual life was significantly increased in the 3 groups(P
8.Study analgesic effect site of eupatorium odoratum L and its mechanism

Objective To study the analgesic effect of Eupatorium odoratum L and its mechanism .Methods Mice were random‐ly divided into 6 groups .Swinging tail method in spinal cord destroyed mouse ,hot‐plate and twisted body method in naloxone blocked mouse ,and formalin‐induce inflammatory pain method were done to investigate the analgesic effect of Eupatorium odoratum L .Each pain index under each experiment condition was determined after intra gastric administration of each group of mice .The concentration of NO and PGE2 in serum and brain were measured to study its mechanisms .Results High‐dose of Eupatorium odor‐atum L significantly enhanced pain threshold in either spinal cord destroyed or naloxone blocked mouse with pain induced by hot and acetic acid(P<0 .05) .High and middle dosage groups of Eupatorium odoratum L markedly reduced the first‐and second‐phase re‐sponses of formalin‐induced inflammatory pain mouse(P< 0 .05) ,and decreased the writhing times and licking rear feet times caused by twisted body method(P<0 .05) .The level of NO and PGE2 in serum and brain of mouse induced by formalin were both decreased(P<0 .05) .Conclusion Eupatorium odoratum L has notable analgesic effect ,which may be related to reduce the level of NO and PGE2 in central and peripheral site .
9.effect of treatment from the lung and treatment from the intestine on vasoactive intestinal peptide contents of ulcerative colitis rats: a comparison study.

OBJECTIVETo observe effects of treatment from the lung and treatment from the intestine on the level of vasoactive intestinal peptide (VIP) in the lung and intestine of ulcerative colitis (UC) rats.
METHODSThe UC rat model was established in 52 rats by using rabbit intestine mucosa tissue allergen combined TNBS-ethanol model (with the model successful rate of 78.0%). Eight rats randomly selected from 40 successfully modeled rats and 8 of 16 rats from the normal group were recruited as the model group and the normal control group before intervention (at week 0). The rest 32 successfully modeled rats were randomly divided into the model group, the Western medicine treatment group (salazosulfapyridine), the treatment from lung group (Huangqi Jiegeng Decoction), and the treatment from intestine group (Huangqi Huanglian Decoction), 8 in each group. Rats in each treatment group were administered with corresponding medication 8 times the dose of a 60 kg adult human. Another 8 normal rats were recruited as the normal group. Equal volume of pure water was given to rats in the model group and the normal group by gastrog avage, once per day. Contents of VIP in the lung tissue and the intestinal tissue were detected at week 0 and 4 after 4-week consecutive intervention. Pathomorphological changes of the lung tissue and the colon tissue were observed under light microscope.
RESULTSCompared with the normal control group at week 0, evenly distributed diffuse inflammation could be seen in the pulmonary interstitial tissue; the bronchial wall was thickened; a huge amount of infiltration surrounded bronchi and blood vessels; a large area of necrosis of intestinal mucosa and inflammatory cell infiltration could also be seen in the model group. Pathological injuries of the lung and the colon were more alleviated in each treatment group than in the model group at the same time point. Compared with the normal control group at the same time point, VIP contents in the lung tissue significantly decreased in the model group at the end of week 4 (P<0.05); VIP contents in the colon tissue significantly increased in the model group at the end of week 0 and 4 (P <0.05). Compared with the model group, VIP contents in the lung tissue significantly increased in the Western medicine treatment group and the treatment from lung group at the end of week 4 (P<0.01); VIP contents in the colon tissue significantly decreased in the treatment from lung group and the treatment from intestine group (P<0.05, P<0.01).
CONCLUSIONTreatment from the lung and treatment from the intestine showed predominant advantage in improving local inflammation of the lung and the intestinal tract, alleviating pathological injuries, promoting repair of injuries through regulating VIP contents in the lung tissue and the colon tissue.

10.Biological characteristics of canine bone marrow mesenchymal stem cells in vitro under non-inducing conditions

BACKGROUND: Research on seed cells is the most important aspect in the filed of tissue-engineering research, and because of their various ad vantages, bone marrow mesenchymal stem cells (BMSCs) have been taken as the ideal bone tissue-engineering seed cells. OBJECTIVE: To observes the growth characteristics of in vitro cultured BMSCs and their osteogenetic characteristics under non-inducing conditions. DESIGN: A single sample experiment.SETTING: Implanting Center of Stomatological Hospital of Fujian Medical University MATERIALS: This experiment was conducted at the Central Laboratory of Stomatological Hospital of Fujian Medical University, between May andDecember 2003. BMSCs are derived from the primary passage to the 3rd passage of in vitro cultured cells from 4 one-year old dogs. METHODS: Under the aseptic conditions, bone marrow puncture was made at bilateral femur trochanter and 5 mL of heparin anticoagulatedbone marrow was obtained. Then it was placed in 50 mL of anti-Dulbecco's modified Eagle's culture medium in centrifuge tube for monocyte isolation. The BMSCs single cells were primarily isolated and placed in culture box for subculture. After 48 hours, culture medium was removed and the medium was cha1ged once every 3 days. Then subculture was carried on continuously to observe BMSCs in morphology under inverted the phase contrast microscope with the assistance of HE staining. The number of cells was counted daily to calculate doubling time and to draw a growth curve. Alkaline phosphatase was detected by using calcium-cobalt method,and chinalizarin staining was used to detect the growth state of calcification tubercle. MAIN UTCOME MEASURES: ①Optical microscopic structure of dog BMSCs; ② The growth curve of dog BMSCs; ③Observation of osteogenetic index- alkaline phosphatase and calcification tubercle. RESULTS: ① Morphological observation indicated that BMSCs were adherent to the walls, clonogenic and appearing fibroblastic phenotype, and they presented morphological changes without exposing to osteogenetic inducer. ② The expression of Alkaline phosphatase in primary cells was stronger, and it was strong in the 1st passage cell, but weak in the 2nd and 3rd passage cells. ③Calcium deposition was observed, which was stronger in primary cells than in subcultured cells. CONCLUSION: ①BMSCs massively proliferated during in vitro culture,capable of differentiating into osteoblasts and considered as the optimal seed cells for bone tissue-engineering reconstruction. ② BMSCs derived from the 3rd passage has osteogenic activity, but the osteogenic activity of the primary cultured cells was stronger than thatr of the subcultured cells.
