Objective To address angiotensin Ⅱ type 1 receptor (AT 1R) gene polymorphism and its relationship with plasma endothelin (ET) in PIH. Methods Gene polymorphism of AT 1R at site 1166 was detected with polymerase chain reaction (PCR), digestion of restriction enzyme and electrophoresis. Plasma ET was measured by radioimmunoassay. Results 1. The frequencies of genotype(AC and CC) at 1166 of AT 1R gene and C allele in PIH individuals were significantly higher than those of control group, (? 2=11.06, 9.73, respectively, P0.05). Conclusions 1. The variant (A→C) of 1166 polymorphism site of AT 1R is associated with the generation of PIH, and C allele may be a susceptible gene to PIH. 2.There is relationship between AT 1R gene and plasma ET level in PIH patients.

ObjectiveTo explore the impact of urinary incontinence on women's quality of life, and the effective measures to improve it.MethodsWe conducted a comprehensive literature review to analyze the present research about quality of life on urinary incontinent women and interventions for improving it.ResultsUrinary incontinence influenced patients' quality of life in many aspects, especially in their physical health, mental health, daily living and sexual life. Regular pelvic floor muscle training could prevent and control the incontinence symptoms and improve patients' quality of life.ConclusionsIt is necessary to carry on a series of activities to propagandize that urinary incontinence can be treated, effective measures should be taken to prevent and manage urinary incontinence in order to improve quality of life in urinary incontinent women.

Objective To observe the effects of combined penebyclidine hydrochloride-ketamine-propofoi intravenous anesthesia with local anesthesia in transcatheter occlusion of congenital heart diseases (CHD).Methods Eighty-six patients suffered in CHD scheduled for transcatheter Amplatzer occlusio were divided randomly and averagely into two groups with 43 cases each.Group A received combined ketamine--propofol ina'avenous anesthesia with local anesthesia. Group B received combined hydrochloride-ketamine-propofol intavenous anesthesia with local anesthesia.Results The rate of upper airway obstruction of child patient that was caused by increased oral secretion in group B (4.7%) was significantly lower than that in group A(14.0%) (P ＜ 0.05 ).The upper airway obsa-uction was removed by aspirating sputum and oxygen therapy in group A,while removed "by decreasing anesthetic depth in group B.The rate of arrhythmia in operation,the time of operation and wake-up time were not significantly different between two groups [37.2%,(2.65±1.85)h,(45.4±15.2)min in group A,but 34.9%,(2.58±1.74)h,(50.2±17.3)rain in group B (P＞0.05)].Conclusion The combined penehyclidine hydrochloride-ketamine-propofol intravenous anesthesia with local anesthesia is feasible and safe in transcatheter occlusion of congenital heart diseases.

Objective To evaluate the two methods for quantitative hepatitis B virus (HBV) DNA test. Methods The Hybrid Capture II system from Digene Co. and Real Time PCR fluorimetry quantitative HBV DNA test kit from PIJI Bio Technical Development Company Ltd. were used to detect sera HBV DNA in sera of patients with chronic hepatitis B. Results With the two quantitative methods, the positive rates of HBV DNA were 98 6% and 94.6% respectively. The concordant rate of these two methods was 93.2%. In assessing the sensitivity of the two kits with a serial of 10 fold diluted patients′s sera, PG kit could test 10 -7 of diluted serum, and HC II could test 10 -6 . The test value of HC II was more accurate in quantitation than PG kit especially in higher titer of HBV DNA. Whereas in the case of low titer of HBV DNA, the deviation of test value increased in both two methods. For monitoring the anti virus effect with quantitative HBV DNA in CHB patients, these two methods had same sensitivity, and the test value of HBV DNA had the same trend of change. Conclusion HC II system and PG test kit were sensitive and reliable, and they were worthy of monitoring the anti virus efficacy and of clinical practice.

Objective:To investigate the differences in related indices of ulcerative colitis (UC) respectively induced by free drinking and intragastric administration of dextran sodium sulfate (DSS) in mice to provide experimental reference for the optimization of UC model.Methods:Totally 30 C57BL/6 mice were randomly divided into the normal control group,free drinking group and intragastric administration group with 10 ones in each.The mice drank water freely with free drinking or intragastric administration of 3% DSS solution at the dose of 4 g·kg-1·day-1 for 7 days to establish the UC model.The differences in disease activity index (DAI),histological damage sore and activity of myeloperoxidase (MPO) among the groups were compared.Results:Two mice died during the experiment in the free drinking group,and DAI of survival mice was (8.8±1.6).There was no death of mice in intragastric administration group,and DAI was (9.0±0.8),and there was no significant difference in DAI between the groups (P>0.05),while the coefficient of variation in the free drinking group was higher than that in the intragastric administration group (18.7 vs 8.6).The colonic histological damage score of the free drinking group and the intragastric administration group was 24.8±4.2 and 27.0±2.8,respectively,which was typical inflammatory change with no significant difference (P>0.05),while the coefficient of variation of the free drinking group was higher than that of the intragastric administration group (16.9 vs 10.4).MPO of the normal control group,free drinking group and intragastric administration group was (0.41±0.03),(2.32±0.34) and (2.05±0.18) U·g-1,respectively.Compared with the normal control group,significant difference in MPO was shown in the free drinking group and the intragastric administration group (P<0.01),while there was no significant difference in MPO between the groups (P>0.05),and the coefficient of variation in the free drinking group was higher than that in the intragastric administration group (14.7 vs 8.8).Conclusion:Both free drinking and intragastric administration of DSS can successfully induce the UC model in mice.Compared with the free drinking group,the intragastric administration group has low mortality rate and low coefficient of variation.Therefore,intragastric administration has more advantages than free drinking in inducing the UC model in mice.

OBJECTIVE: To isolate and purify the reference compounds and improve the quality standard of Korean medicinal Herba Artemisiae sacrorum. METHODS: The chemical constituents were isolated from the aerial parts of Korean medicinal herb Artemisia sacrorum by silica gel, ODS column chromatography and preparative HPLC, and the structures were identified by NMR and MS. RESULTS: Fourteen known compounds were isolated and identified as follows:1α-acetoxyeudesm-4-en-6β,11βH-12,6-olide(1),(11S)-3-oxoeudesma-1,4(15)-dieno-12,6α-lactone(2), 1-epi-dehydroisoerivanin(3), 11-epi-taurin(4), chrysanthemolide(5), 1α-acetyl-gallicadiol(6), erivanin(7), 1α, 4α-dihydroxyeudesm-2-en-5α, 6β, 11βH-12, 6-olide(8), vulgarin(9),(+)-dehydrovomifoliol(10), isoevodionol(11),(+)-epi-pinoresinol(12), lariciresinol-4'-O-β-D-glucopyranoside(13), and lariciresinol-4-O-β-D-glucopyranoside(14). CONCLUSION: All compounds are obtained from this plant for the first time. These compounds can be used as reference substances for the quality control of this ethnic medicine.

OBJECTIVE: To establish a quantitative method for determining the contents of uracil, cytidine, hypoxanthine, xanthine, uridine, inosine, and guanosine in Holotrichia diomphalia Larvae by HPLC. METHODS: The HPLC analysis was performed on Waters HSS T3 column(4.6 mm×250 mm, 5 μm). The mobile phase was composed of acetonitrile(A) and water(B), and gradient elution was carried out. The flow rate was 1.0 mL•min-1. The column temperature was maintained at 30 ℃. The detection wavelength was 260 nm. RESULTS: The correlation coefficients of the seven components ranged from 0.999 1 to 1.000 0. The average recovery rates(n=6) were between 91.2% and 97.4%, and the RSDs were between 1.5% and 2.3%. CONCLUSION: The proposed method is simple, accurate and reliable, thus providing basis for comprehensive quality control of Holotrichia diomphalia Larvae.

Objective To investigate the effect of fusidic acid cream on inflammatory reaction caused by skin barrier damage.Methods Eight male SKH-1 hairless mice were included in this study.The back of each of these mice were equally divided into six regions measuring 1 cm × 2 cm in size,which were then assigned into six groups:blank control group remaining untreated,barrier-impaired group,barrier-impaired and fusidic acid-treated group,barrier-impaired and vehicle-treated group,barrier-unimpaired and fusidic acid-treated group,barrierunimpaired and vehicle-treated group.Stratum corneum was removed by adhesive tape stripping to establish an animal model of acute skin barrier damage in the corresponding skin regions of these mice,and fusidic acid cream or vehicle was topically applied to the corresponding regions once.Twelve hours later,skin surface swab samples were collected from the back of these mice followed by bacterial culture and colony counting.Mice were then sacrificed,and skin tissue specimens were resected from these mice,and subjected to real-time fluorescence-based quantitative PCR for the measurement of the mRNA expressions of myeloid differentiation factor 88 (MyD88),interleukin-1α (IL-1α),IL-6,epidermal antibacterial peptides S100a8 and S100a9.Statistical analysis was carried out by repeated-measures analysis of variance (ANOVA) and least significant difference (LSD) test.Results The mRNA expressions of MyD88,IL-1α,IL-6,S100a8 and S100a9 were all significantly higher in the barrier-impaired group than in the blank control group (all P ＜ 0.05).Specifically,the mRNA expression level of MyD88 in the barrier-impaired group was 8 times that in the blank control group (8.3 ± 3.0 vs.0.8 ± 0.4).Compared with the barrier-impaired group,the barrier-impaired and fusidic acid-treated group showed a significant decrease in the mRNA expressions of IL-1α (2.8 ± 0.3 vs.20.1 ± 10.0,F =47.11,P ＜ 0.01),IL-6 (1.6 ± 2.3 vs.9.4 ± 4.0,F =16.18,P＜ 0.01),S100a8 (1.5 ± 1.4 vs.5.0 ± 1.6,F=59.71,P＜ 0.05) and S100a9 (1.2 ± 0.7 vs.3.4 ± 1.6,F=21.94,P ＜ 0.05).Conlusions Fusidic acid cream could attenuate the inflammatory reaction caused by acute skin barrier damage,which might partly explain its action mechanism in the treatment of inflammatory skin diseases.

This paper reported the suitable medium of L S-1 stain in detail,which could yield natural blue pigment.Single-factor exper imental design shows that the best carbon source was 2% glucose and nitrogen was 0 1% KNO 3.Orthogonal experimental design shows that the most suitable fermen tation medium was consisted of 4% glucose,0 1% KNO 3,0 075% salt and 10?g/m L FeSO 4.The best cultivation temperature was 30℃ and pH7 4.The dissolved oxyg en on the process of fermentation,as well as the variety of pH and the utilized condition of carbon and nitrogen were measured and analyzed.The separation of th is blue pigment by HPLC shows that this material contains actinorhordin and at l east other four ingredients.
