4.Effects of RO20-1724 on repetitive ketamine administration-induced learning and memory impairment in immature rats

Objective To investigate the effects of RO20-1724 on repetitive ketamine administration-induced learning and memory impairment in immature rats.Methods Forty-eight 21-day-old SD rats of both sexes weighing 45-55 g were randomly divided into 4 groups (n =12 each):control group(group C); ketamine group (group K); ketamine + RO20-1724 group (group K+ R) and ketamine + vehicle (ethanol) group (group K+ A).Ketamine 70 mg/kg was injected intraperitoneally (IP) once a day for 7 consecutive days in groups K,K+ R and K+ A.RO20-1724 0.5 mg/kg and equal volume of ethanol were injected IP at 30 min after IP ketamine once a day for 7 consecutive day in groups K + R and K + A respectively.Morris water maze test was used to assess learning and memory ability.The escape latency and the number of times of passing the safe zone were recorded.The animals were killed after water maze test and their brains removed for microscopic examination of hippocampus and determination of p-CREB protein expression in hippocampus (by Western blot).Results Repetitive ketamine administration significantly prolonged the escape latency,decreased the number of times of passing the safe zone and down-regulated the expression of p-CREB protein in hippocampus on the 3rd and 4th day in group K as compared with group C.RO20-1724 significantly attenuated the above changes induced by repetitive ketamine administration in group K + R as compared with group K.Electron microscopic examination showed that RO20-1724 significantly ameliorated repetitive ketamine administration-induced hippocampal neuronal damage.Conclusion RO20-1724 can ameliorate cognitive dysfunction induced by repetitive ketamine administration.Up-regulation of cAMP /CREB signaling pathway is involved in the mechanism.
5.Effects of RO20-1724 on cognitive function in immature rats after ketamine anesthesia

Objective To investigate the effect of RO20-1724 on the cognitive function in immature rats after ketamine anesthesia.Methods Ninety-six SD rats of both sexes,aged 21 days,weighing 45-55 kg,were randomly divided into 8 groups ( n =12 each):control group (group C) ; ketamine group (group K) ; ketamine + normal saline group (group K + N) ; ketamine + anhydrous alcohol group (group K + A) ; ketamine + 4 different doses of RO20-1724 groups (group K + R1-4 ).The rats were anesthetized with intraperitoneal injection of kctamine 70 mg/kg in groups K,K+N,K+A and K+.R1-4.Normal saline 2 ml,anhydrous alcohol (in normal saline 2 ml),and RO20-1724 0.25,0.50,0.75 and 1.00 mg/kg (in anhydrous alcohol 8 μl and then in normal saline 2 ml) were injected intraperitoneally in groups K + N,K + A and K + R1-4 respectively 30 min later.Six rats from each group were randomly selected at 24 h after administration and Morris water maze was used to test the ability of learning and memory.Six rats from each group were sacrificed at 48 h after administration and hippocampus and cerebral cortex were removed for determination of the expression of CREB and phospho-CREB (p-CREB) by Western blot.Ressults Compared with group C,the escape latency was significantly prolonged at 2-4 days after administration,the number of animals' swimming across the platform decreased,and the expression of CREB and pCREB in hippocampus and cerebral cortex down-regulated in groups K,K+ N,K+ E,K+ R1 and K+ R2(P <0.05 ).Compared with group K,the escape latency was significantly shortened at 2-4 days after administration,the number of animals' swimming across the platform increased,and the expression of CREB and p-CREB in hippocampus and cerebral cortex up-regulated in groups K + R3 and K + R4 ( P < 0.05).Compared with groups K + R1 and K + R2,the escape latency was significantly shortened at 2-4 days after administration,the number of animals' swimming across the platform increased,and the expression of CREB and p-CREB in hippocampus and cerebral cortex up-regulated in groups K+ R3 and K+ P4(P < 0.05).There were no significant differences in the escape latency,the number of animals' swimming across the platform,and the expression of CREB and p-CREB in hippocampus and cerebral cortex between groups K + R1 and K + R2,and between groups K + R3 and K + R4 ( P > 0.05 ).Conclusion RO20-1724 0.75-1.00 mg/kg can improve ketamine-induced cognitive dysfunction by up-regulating CREB and p-CREB expression in hippocampus and cerebral cortex in immature rats.
6.The clinical application value of multi-slice spiral CT angiography

Objective To explore the clinical application value of multi-slice spiral CT angiography.Methods 28 patients who were suspected cerebrovascular disease were taken cerebrovascular enhanced scan by 16-slice spiral CT,and VR,MIP and MPR post -processing were uset to reconstmct the cerebrovascular image.Results In 28 patients,11 cases were normal,17 cases were abnormal,and among these patients there were 8 cases of abnormal blood vessels,4 cases of aneurysms,1 case of cerebral arteriovenous malformation,4 cases of cerebral artery stenosis.Conclusion The multi-slice CT compared with ordinary spiral CT had the advantage of faster and more secure.It had an important role in guiding the screening and clinical treatment programs in cerebral infarction patients with suspected cerebral artery stenosis.It had a lot of advantages in diagnosis vascular disease and clinical observation after treatment.
7.Ultimate cold preservation time in Ringer's solution for murine cardiac graft

AIM: To determine the ultimate preservation time of murine cardiac grafts in 4℃ Ringer's solution. METHODS: Murine cardiac grafts were implanted to the abdominal vessels heterotopically 2, 4, 6, 8 and 10 hours after cold preservation. Graft survival rate and histological morphological changes, as well as the neutrilphil, T cell, macrophage infiltration, ICAM expression were determined by immunohistochemistry. RESULTS: The cardiac function-recovery rate and 1-week graft survival rate were 100% and 83.3% in 6-hour preservation group. Compared with non-preservation control group, no more apparent histological damages, cell infiltration and ICAM expression were found. CONCLUSION: The ultimate preservation time of murine cardiac graft in 4℃ Ringer's solution was 6 hours. [
8.The effect of body weight on the induction of mild hypothermia in a rabbit model of asphyxia cardiac arrest

Objective To investigate the effect of body weight on the induction of mild hypothermia in a rabbit model of asphyxia cardiac arrest. Methods Twenty-four rabbits were randomized into two groups: the ice bag group and the intravenous 4℃ saline group. Cardiac arrest was induced and after 3 minutes of cardiac arrest, cardiopulmonary resuscitation was begun. Simultaneously, mild hypothermia was induced by putting an ice bag over the abdomen or infusion of 4℃ saline via an ear vein. A 2℃ decrease of rectal temperature was considered as the completion of hypothermia induction. Induction times were recorded, compared, and analyzed with respect to body weight. Results All rabbits had restoration of spontaneous circulation (ROSC) and ROSC lasted during the experiment. Induction time in the ice bag group was significantly shorter than that in the intravenous 4℃ saline group (22.8±4.7 min VS 42.5±4.0 min, P< 0.001). Induction time significantly correlated with body weight in the ice bag group (Pearson Correlation: r = 0.725, P = 0.029), but not in the intravenous 4℃ saline group (Pearson Correlation: P = 0.418). Conclusions In a rabbit model, induction of mild hypothermia with an ice bag is faster than with intravenous 4℃ saline; induction time positively correlates with body weight when an ice bag is used, but not when intravenous 4℃ saline used. The effect of body weight should be considered when choosing an appropriate method to achieve early induction of mild hypothermia.

Melanin is formed by oxidative polymerization of phenolic compounds,basically different kinds of melanin come from different organisms.DOPA melanin and DHN melanin have same physical and chemical characters although they have different biosynthetic pathway.DHN melanin is common in plant fungi and plays an important role on infection.The melanin accumulates in the fungal cell walls and prevents organic and inorganic molecules penetrating out,that insures appressorium's pressure and infection ability.This paper has reviewed the kinds and characters,especially discussed the role of melanin during pathogen infection based on our some research.
10.Study of synergistic mechanism in the combination ofberberine and evodiamine from the perspective of miRNA

Aim To explore the mechanisms of action (MOA) of synergistic anticancer function in the combination of berberine and evodiamine.Methods We first analyzed the action of suppression in the drug combination from the cell level and validated the dose scope as well as ratio of concentration in synergistic effects of drug combination.Then, the miRNA chip of liver cancer cell BEL-7402 under different treatment was analyzed.By building the miRNA-mRNA network, the MOA of the synergistic drug combination was illustrated.Results Berberine and evodiamine used in combination could significantly synergistically suppress the proliferative ability of liver cancer cells.The special differentially expressed miRNAs (DEmiRNAs) mainly participated in some cancer proliferation-related pathways and biological processes, such as MAPK signaling pathway, endocytosis pathway and insulin signaling pathway.The special target genes influenced by the drug combination not only covered three kinds of membrane receptors, but also took part in the regulation of downstream pathways.Conclusions From the regulation of miRNAs, it is clear that berberine may play a primary role in the synergistical suppression activity of the drug combination in cancer cells.The discovery of synergistic MOA in the combination of berberine and evodiamine from the miRNA level will provide a new guidance to explore more synergistic drug combinations in the future.
