Microbiology 2008;0(11):-
The mechanism of chromium accumulation and microstructure transformation of the fused yeast were studied in this paper. The result showed that the process of Cr6+ reduction and adsorption was accom-panied by the H+ consumption. The main adsorptive groups on the strain surface included amino, hydroxyl, phosphate group and amide, among which phosphate group played vital role in the chromium accumulation. The removal rate of chromium and reduction rate of hexavalent chromium declined 70% and 46%,respec-tively,when phosphate group was masked.During the adsorption process the chromium ions complexed on the surface of fused yeast was transported into the cell wall and combined with inclusion to form steady spe-cies and this took 90 min to reach the equilibrium. The biosorption and reduction of Cr on the cell surface would alter microstructure of cell surface, reduce cell membrane potential and increase cell membrane per-meability.

ObjectiveTo establish the maximum blood order schedule (MSBOS) for maxillofacial malignant tumor surgery of patients undergoing free flap reconstruction so as to guide clinicians in using blood rationally and prepare blood scientifically by the department of blood transfusion. MethodsBasic information and blood transfusion data of patients in the perioperative period of maxillofacial surgery in our hospital from January 1, 2010 to December 31, 2020 were collected, including age, gender, body mass index (BMI), preoperative hemoglobin value, ASA staging, T staging, and whether the free flap contains bone flaps, surgical methods (category 6), the amount of blood ordered for preparation, and the amount of blood used in the operation. A total of 1 953 cases of elective surgery in maxillofacial surgery were included. The univariate and multivariate logistic regression analyses of perioperative transfusion were performed to screen out independent risk factors for perioperative transfusion and establish scoring criteria. According to the surgical method, the patients were divided into 6 groups, then each group of patients was further divided into low-risk, intermediate-risk and high-risk groups in accordance with scoring standard. The cumulative percentage of the actual intraoperative blood transfusion frequency of each group of patients was calculated to establish the MSBOS of the maxillofacial surgery patients. Results①Regression analysis found that BMI, tumor T staging, ASA grade, preoperative hemoglobin (Male Hb < 130 g/L, female Hb < 120 g/L), and free flaps containing bone flaps were independent risk factors for perioperative blood transfusion. ②According to risk factor scores, it was found that the low-risk group had the lowest blood transfusion rate, but cross match to transfusion ratio was the highest. ③The MSBOS of maxillofacial malignant tumor patients who underwent free flap reconstruction in our hospital was established. The analysis found that after MSBOS was established, the total amount of blood prepared before the operation could theoretically be reduced by 54.35% than before the establishment. ConclusionThe establishment of MSBOS is not only beneficial for guiding the blood preparation more scientifically and reasonably, but also can optimize the management of blood inventory and save blood resources.

Objective To investigate the effect of HSP70/CD80 DNA vaccine on the airway inflammation and hyperresponsiveness of asthmatic mice. Methods Forty female healthy BALB/c mice were randomly divided into 4 groups; saline group, asthma group, pcDNA3. 1 plasmid control group, and prevention group with HSP70/CD80 DNA vaccine, with 10 mice in each group. The mice were immunized by intramuscular( i. m. ) injection with HSP70/CD80 DNA vaccine before sensitization and challenge with ovalbumin. Then, the murine model of allergic asthma was made with injection of ovalbumin intraperitoneal ( i. p. ) , and inhalation of ovalbumin. Before mice were sanctified, their airway hyperresponsiveness( AHR) was measured. After mice were sanctified, bronchoalveolar lavage fluid( BALF) was obtained and cytokine IL-13 and IFN-γ were measured. And the lung histology and histochemistry were examined. Results Compared with mice in asthma and pcDNA3. 1 group, mice in vaccine group showed significantly reduced airway inflammation (P<0. 05) and AHR (P<0. 05). IFN-γ content in BALF were increased in mice from vaccine group compared with the asthma group and the pcDNA3. 1 group ( P <0. 05) , and IL-13 content in BALF were decreased. Conclusion HSF70/CD80 DNA vaccine can reduce airway inflammation and hyperresponsiveness in asthmatic mouse and this chimerical plasmid could be a candidate vaccine to prevent asthma.

Objective To analyze the displacement of titanium clips for tumor bed localization after breast-conserving surgery for breast cancer and its influential factors. Methods A retrospective analysis was performed on the cone-beam computed tomography (CT) images of 14 patients with breast cancer who received radiotherapy after breast-conserving surgery from April to October,2016.The relative position of the chest wall and the errors of the titanium clips in radiotherapy were measured.A Pearson correlation analysis was used to analyze the correlation of the displacement of titanium clips with the relative position of titanium clips, the breast volume, the vertical distance between the titanium clips and the tangential line of the chest wall, and the maximum thickness of the breast. Results The system errors of the chest wall in left-right,superior-inferior,and anterior-posterior directions were 4.42,3.44,and 5.13 mm,respectively, and the random errors were 3.55,3.07,and 4.54 mm,respectively. The titanium clips had a large displacement relative to the chest wall, mainly in the left-right direction. The maximum system error was 4.39 mm and the random error was 2.42 mm.The displacement of titanium clips was not significantly correlated with the breast volume and the maximum thickness of the breast (P>0.05).However,the relative position of titanium clips in superior-inferior direction was significantly correlated with the displacement of the lowest, the most lateral, the most anterior, and the most posterior titanium clips (P<0.05).As to the uppermost clips, there was a significant difference in displacement between the clips close to the chest wall and the clips far from the chest wall (P=0.02).Conclusions Due to large setup error and displacement of titanium clips during radiotherapy, simultaneous integrated boost is not suitable for patients with breast cancer who are immobilized by vacuum cushion and received radiotherapy. The unstable immobilization may be the major influential factor for the displacement of titanium clips.

To investigate the effect of compound Chinese traditional medicine PC-SPES II I in inhibiting proliferation of human prostate cancer cell LNCaP based on the androgen receptor (AR) signaling pathway. The effect of PC-SPES II on LNCaP cell proliferation was detected by MTT assay. According to the findings, at the mass concentration of 180-1 440 mg x L(-1), PC-SPES II significantly inhibited the proliferation of LNCaP cells; the IC50 of PC-SPES II at 24 h and 48 h were 311.48, 199.01 mg x L(-1), respectively. The flow Cytometry detection showed 240 mg x L(-1) PC-SPES II arrested cells in G2/M phase, and an obvious apoptotic peak appeared before G0/G1 peak and rose over time. Meanwhile, Hoechst 33258 staining revealed apoptotic cellular morphology. Annexin V-FITC/PI staining manifested an increase in apoptotic cell ratio at the PC-SPES II concentration of 480 mg x L(-1) in a dose dependent manner. The prostate specific antigen (PSA) secretion of LNCaP cells was tested by PSA ELISA kit. Besides, compared with 25 mg x L(-1) Bic, 480 mg x L(-1) PC-SPES II significantly reduced the cell secretion of PSA. The AR and PSA mRNA and protein expressions were detected by qRT-PCR and Western blot. According to the results, after the induction of LNCaP cells with synthetic androgen 25 μg x L(-1) R1881, 240-480 mg x L(-1) PC-SPES II notably down-regulated the AR and PSA mRNA and protein expressions and inhibited the translocation of AR from cytoplasm to nucleus. In summary, PC-SPES II significantly can inhibit the in vitro proliferation of LNCaP cells and arrest cell cycle arrest in G2/M phase. Its mechanism may be associated with the down-regulation of the AR and PSA expressions and the inhibition of AR nuclear translocation.

Objective To investigate the influence and mechanism of erythropoietin (EPO) in renal blood flow after limb ischemia reperfusion (LIR). Methods Thirty male SD rats were randomly divided into control group, LIR group and EPO+LIR group with ten in each group. The values of renal blood flow, plasma creatinine (Cr), urea nitrogen (BUN) content in plasma, kidney tissue wet to dry ratio (W/D), nitric oxide (NO), nitric oxide synthase (NOS) and endothelin-1 (ET-1) in re-nal tissue were detected in three groups. The immunohistochemistry assay was used to detect the expression of intercellular adhesion molecule (ICAM-1) and vascular cell adhesion molecule (VCAM-1) in renal tissue. The morphological changes of renal tissue were observed with light microscope. Results The renal blood flow was significantly decreased, while the val-ues of Cr, BUN, W/D, NO, ET-1, NOS, expressions of ICAM-1 and VCAM-1 was significantly increased in LIR group than those of control group (P<0.05). Broaden interstitial and infiltration of inflammatory cells were observed in the renal tissue under light microscope. In the EPO+LIR group, the renal blood flow increased, the values of Cr, BUN, W/D, NO, ET-1 and NOS, expressions of ICAM-1 and VCAM-1 decreased significantly compared with those of LIR group (P<0.05). The patho-logical changes were alleviated in EPO+LIR group. Conclusion EPO can improve renal function, increase renal blood flow in rats after LIR. The mechanism may be related to the decreased edema, changed renal vasomotor function and decreased in-flammation.

【Objective】To evaluate the role of serum antimullerian hormone (AMH) concentration in predicting clinical outcomes in controlled ovarian stimulation and intrauterine insemination (IUI). 【Methods】 A retrospective analysis of IUI data from patients with polycystic ovary syndrome and infertility in our reproductive center from January 2014 to August 2017. According to the clinical outcomes, data was divided into three groups:clinical pregnancy group,non-pregnant group, and cancellation cycle group due to multiple follicular or no dominant follicular develop. The ovarian function evaluation indexes were compared such as serum AMH,basal FSH and basal sinus follicle(AFC). The cutoff values of the AMH to predict multi- follicular development or no dominant follicular development which IUI cycles were cancelled, and to predict obtain cumulative clinical pregnancy outcomes were calculated according to the ROC curve to cancel the cycle. 【Results】 The clinical pregnancy rates of PCOS patients were from 15.9% to 17.1% ,while the cumulative clinical pregnancy rate increased in repeat cycles (≤3 cycles) was 21.6% ,significantly higher than the average clinical pregnancy rate(16.7%)during the first cycle. AMH levels in the cancelled cycle group were significantly higher than those in the non-cancelled cycle group[(14.1±6.5)vs(10.3±4.3)ng/mL,(14.1±6.5)vs(9.3±4.3)ng/mL, P<0.025]. Compared with the clinical pregnancy group, cancel cycle group and non- pregnant group, the mean AFC of the former two groups were significantly higher[(34.5±11.4)vs(30.7±11.3),(7.8±10.8)vs(30.7±11.3),P<0.025]. In addition,we found that women′ s age, baseline FSH were negatively correlated with the cumulative pregnancy rate of repeated IUI cycles, and women′s baseline BMI,baseline LH,AFC,and AMH were positively correlated with cumulative pregnancy rates. The number of mature follicles on the HCG triggering day,as well as the value of AMH,may be the influencing factor of IUI cumulative clinical pregnancy. By ROC curve analysis,we assume that AMH is more suitable than the AFC to predict IUI cycle cancellation rate and the cumulative pregnancy rate.【Conclusion】Repeated IUI cycles can be improved cumulative pregnancy rate of PCOS infertile couples. AMH,as an important index to measure ovarian reserve function, can be used to predict the the IUI cumulative clinical pregnancy outcomes and cancelled cycle rates:as AMH increased to more than 6.56 ng/mL,the pregnancy rate increased;but when AMH ≥14.72 ng/mL,the risk of cancelled IUI cycles was increased, becasues of multi-follicular developing and absence of dominant follicle development.
