Objective: To investigate the expressions of Slit2 and its receptor Robo1 in human gastric carcinomas. Methods: The expression of Slit2 protein, Robo1 protein and CD34-labeled microvessel density(MVD) were measured by immunohistochemical staining (SP) in 54 cases of gastric carcinomas and 28 cases of Para-cancer tissues. Objective To study the detection rate of early esophageal carcinoma using magnifying endoscopy,and to evaluate the relationship between the imaging patterns of microvasculature change and his- tological diagnosis.Methods Two hundred and fourteen patients with esophageal mucosa roughness,ero- sion,plaque,abnormal color and indentation in conventional endoscopy and 16 healthy volunteers were en- rolled.The magnifying endoscopy images were graded as four patterns by intraepithelial papillary capillary loop(IPCL)changes after iodine dyeing.The biopsies underwent pathologic evaluation.The comparison be- tween the imaging patterns of endoscopy and histological diagnosiswas was evaluated.Results 80.4%(90/ 112)esophagitis was type 2,and 85.7%(12/14)early esophageal carcinoma was type 3 and type 4.The difference was significant between early esophageal carcinoma and normal mueosa(?~2=27.32,P
5.Relationship between regional homogeneity of depressed patients and anxious symptoms: a resting-state fMRI study

Objective To investigate the differences of regional homogeneity(ReHo) in depressed patients with vary degrees of anxiety using resting-state function MRI,and the relationship vary degrees of anxiety symptoms.Methods 10 depressed patients with scores less than 1 in anxiety/somatization factor of Hamilton depression scale,10 depressed patients with scores above 2,and 10 age-,gender-,education-matched healthy controls were underwent resting-state fMRI scanning at 3.0 Tesla.Correlation analysis between ReHo with significant differences level of brain region and the total score of Hamilton depression scale,and anxiety/somatization factor were underwent respectively.Results The brain regions with significant difference level among three groups were the left posterior cerebellum(-39,-77,-29 ; K =15),the left angular gyrus (-56,-63,22 ; K =13) and the left anterior cingulate (-3,22,29 ; K =19) (P < 0.05,Alphasim) ; compared with healthy group,the mild anxiety group showed significantly decreased ReHo in the left posterior cerebellum,and increased ReHo in the left anterior cingulate; the moderate and severe anxiety group showed significantly decreased ReHo in the left posterior cerebellum,and increased ReHo in the left anterior cingulate and the left angular gyrus; no significant differences were found in patient groups.In the mild anxiety group,the left anterior cingulate positively correlated with the anxiety severity(r=0.646,P=0.043),and in the severe anxiety group positively correlated with the somatic anxiety(r =0.693,P=0.038).Conclusion Depressed patients showed abnormal neural activity in resting state,and the degree of abnormality is associated with clinical anxiety/somatization symptoms.
6.Quantitative detection of prostate specific membrane antigen and the splicing variant DNA in different tumor cell strains and prostate tissues

Objective To understand and illuminate the bionomic characteristics of prostate specific membrane antigen (PSMA) and splicing variant PSMA5, through detecting the DNA levels of them in different tumor cell strains and prostate tissues. Methods The fluorescent quantization reverse transcriptase PCR (FQ-RT-PCR) method built up by our research group was used to detect the PSMA and variant PSMA5 DNA levels in different tumor cell strains and prostate tissues. Results The PSMA and PSMA5 DNA levels in tumor cell strains and pathological prostatic tissues were obviously more than those of the normal prostatic tissues (F=3.40, 11.94, both P<0.05), and the PSMA5 DNA level was much higher than was the PSMA DNA level in prostatic carcinoma tissues (P<0.05). Conclusions The different expressions between PSMA and PSMA5 in different tumor cells and prostatic tissues show that PSMA5 is more specific than PSMA as a prostate carcinoma tumor marker.
7.Determination of Content and Entrapment Efficiency of VEC-5 Liposomes by HPLC

Objective:To establish an HPLC method for the determination of content and entrapment efficiency of HIV-1 virus in-fection factor Vif inhibitor VEC-5 liposomes. Methods:VEC-5 liposomes were prepared by a method of freeze-drying and reconstruc-tion. The separation of free drug from the liposomes was achieved by ultracentrifugation, and an HPLC method was used to determine the content and entrapment efficiency of VEC-5 liposomes. Results:The linear range of VEC-5 was 20-100 μg·ml-1(r=0. 999 0). The average recovery was 100. 25% and RSD was 0. 93%(n=9). The content of three batches of VEC-5 liposomes was 98. 63%, 100. 43% and 102. 65%, respectively within the range of 90%-110%, and the entrapment efficiency was 94. 89%, 93. 68% and 94. 56%, respectively, which was above 90%. Conclusion:The method is accurate and reliable, which can be used to determine the content and entrapment efficiency of VEC-5 liposomes.

Reverse transcriptase-PCR experiments suggest that the two clusters of genes potentially involved in the oxidation of reduced sulfur compounds are organized as operons in strain of the acidophilic, chemolithoautotrophic bacterium Acidithiobacillus ferrooxidans ATCC 23270, the two clusters of genes including such the ORF of putative sulfate-thiosulfate-molybdate binding proteins, the ORF of putative thiosulfate: quinone oxidoreductase and the ORF of the rhodanese-like protein (P21). Bioinformatic analyses have predicted the possible promoters sequences and the possible +1 start site of transcription for the doxDA operons.
9.Flow cytometry immunophenotyping and minimal residual disease evaluation in multiple myeloma

Objective To investigate the characteristics of immunophenotyping and clinical significance of MRD analysis in MM patients. Methods Multi-parameter flow cytometry was applied to analyze the immunophenotyping of malignant plasma cells from 172 MM patients, and normal plasma cells from 16 healthy individuals. MRD was analyzed in 32 MM patients with remission. Meanwhile, the effects of MRD status on the disease relapse and patient disease free survival ( DFS ) time was evaluated by following up patients. Results The immunophenotyping of normal plasma cells were CD38, CD138, CD19 and CD45 positive, while the predominant phenotype of MM cells were CD+38( 100.0% ), CD+138( 100.0% ), CD-19 ( 167/172, 97. 1% ), CD+56( 152/172, 88.4% ) and CD-45( 166/172, 96. 5% ). The characteristic markers for MM cells were CD+38, CD-138, CD-19, CD-45 and CD+56. MRD analysis revealed that, among 32 MM patients with remission, 14 patients were MRD negative and 18 patients were MRD positive. During follow-up of 2 to 16 months, the relapse rate in MRD negative patients was significantly lower ( 4/14, 28.6% ) than that of MRD positive patients ( 13/18, 72. 2% ;χ2 =6. 03, P <0. 05 ). Furthermore, the DFS time was significantly longer in MRD negative patients[ 16. 23( 10. 37-21.62 )months ] than that of the MRD positive patients [ 10. 07( 3. 79-16. 20 )months,χ2 =7. 53,P <0. 05 ]. Conclusions CD+38, CD+138, CD-19, CD-45 and CD+56 are the characteristic markers of MM cells compared to those of the normal plasma cells. MRD analysis is a valuable prognostic factor for MM patients.
10.Relationship between preoperative FIB-4 and hepatocellular carcinoma prognosis after curative resection

Objective: To investigate the correlation between FIB-4 and the clinicopathological characteristics and prognosis of patients with hepatocellular carcinoma (HCC) after curative resection. Methods: From January 2009 to December 2012, the clinicopathological and follow-up data of 245 patients with HCC after curative resection were retrospectively studied. Their survival was calculated using the Kaplan-Meier method. The Cox proportional hazard regression model was used for the multivariate analysis. Results: According to FIB-4 index, patients were divided into two subgroups: FIB-4Ⅰ(≤3.25) and FIB-4Ⅱ(>3.25). FIB-4 could predict liver cirrhosis severity (Ishak grade, Grade 1-5 vs. Grad 6, r=0.681, P<0.001). It was associated with liver function such as:aspartate transaminase (P<0.001)、total bilirubin (P=0.009)、albumin (P=0.001) and platelet count (P<0.001) other than tumor clinicopathologic features. Both univariate and multivariate analysis showed FIB-4 could predict the prognosis of HCC patients (Overall survival: P=0.037 and 0.011; Recurrencefree survival: P=0.027 and P=0.043, respectively). Conclusion: The preoperative FIB-4 index could be used as a prognostic marker for the prognosis of HCC after curative hepatectomy.
