3.Pediatric Drug Counseling Telephone Service:Analysis of 542 Cases

OBJECTIVE:To probe into the pediatric drug counseling service and the characteristics of adverse drug reactions (ADR). METHODS:The telephone records of drug consultation from Feb. 2006 to Apr. 2008 in our hospital were statistically analyzed and the problems related to adverse drug reactions were classified. RESULTS:Among the 542 cases of drug consulting telephone records,the top 3 consulting questions were the usage and dosage of drugs (29.15%),recommendation and choice of drugs (13.28%) and drug interactions (10.52%). Of which,41 cases of ADR problems were about the use of antibacterials and Chinese traditional patent drugs. The most common manifestation of ADR was the lesion of digestive system. CONCLUSION:Drug consulting telephone service can not only enhance children patients' medication compliance and promote rational use of drugs but also improve the learning of clinical pharmacists.
4.Recent research findings on non-invasive diagnosis of liver fibrosis

Early diagnosis of liver fibrosis and dynamic monitoring of relevant changes have great implications for the treatment and prognosis improvement of chronic liver diseases. So far, liver biopsy remains the “golden standard” for the diagnosis and staging of liver fibrosis. However, due to its inherent limitations, a great effort has been made to develop more accurate non-invasive diagnostic methods, including serum fibrosis markers and mathematical models, ultrasound, contrast-enhanced ultrasonography, ultrasonic elastography, computed tomography, magnetic resonance imaging, and nuclear medicine. The advantages and disadvantages of relevant methods are discussed. Furthermore, proper selection of the non-invasive diagnostic methods for clinical application and the means for mutual verification are analyzed. As for the future direction, it is expected to employ the above methods for combined analysis and comprehensive assessment, in order to enhance the clinical value of non-invasive liver fibrosis diagnosis.

6.Cardiopulmonary Quantitatively Assessment for Spinal Cord Injury during Aerobic Exercise

Objective To quantitatively assess the cardiopulmonary exercise function of spinal cord injury (SCI) patients and observe the effect of aerobic exercise on cardiopulmonary function, motor function and activities of daily living. Methods From December, 2014 to June, 2016, 34 incomplete SCI patients (ASIA C and D) and 23 healthy controls received cardiopulmonary exercise test (CPET). SCI pa-tients were randomly divided into conventional rehabilitation group (n=17) and aerobic exercise group (n=17). The aerobic exercise group received aerobic exercise for four weeks. They were assessed with CPET, motor and sensory function, walking index for spinal cord injury II (WISCI II) and spinal cord independence measure (SCIM) before and four weeks after training. Results Oxygen uptake (VO2)peak, anaerobic threshold (AT), metabolic equivalent of energy (METpeak), VO2/heart rate (HR)peak, respiratory exchange rate (RER)peak, minute ventilation (VE)peak, work rate (WR)peak and systolic blood pressure (SBP)peak were lower in the patients than in the controls (t>2.714, P<0.05). VO2peak、AT、METpeak、VO2/HRpeak、WRpeak increased in the aerobic exercise group after training (t>2.431, P<0.05). METpeak and WRpeak improved in the conventional rehabilitation group after training (t>3.282, P<0.01). The scores of motor in ASIA and SCIM improved in both groups after training (t>2.985, P<0.05). Conclusion The cardiopulmonary function decreased in incomplete SCI patients, which could be improved by moderate intensity aerobic exercise.
7.Implication of budding uninhibited by benomyl gene in spontaneous abortion embryos with numerical chromosomal abnormality

Objective To explore the mechanism of hBUB1 gene in the developing of spontaneous abortion embryos with numerical chromosomal abnormalitv.Methods Quantitative real-time RT-PCR and Western blot were used to determine the mRNA and protein level of hsMAD2 gene both in spontaneous abortion embryos with numerical chromosomal abnormality(experimental group)and with numerical chromosomal normality(control group).Recombinant shRNA plasmids targeting hBUB1 gene was constructed to inhibit the expression of endogenous hBUB1 genes in embryonic cells.Interference efficiency was demonstrated by fluorescent quantitative PCR and Western blot.The inhibitory rate of cell proliferation was measured by MTT assay and cells-cycle was assessed by flow cytometry.Resuns Western blot analysis showed that protein level of hBUB1 in the experimental group was decreased signifieandv(the rate of positivity and strong positivity were 8%and 93.5%,respectively,P<0.05)compared with the control group.The expression of hBUB1 gene in embryonic cells was significantly and specially inhibited by shRNA plasmids (the mRNA level before and after treatment witll RNAi were 0.196±0.067 and 0.042±0.006,respectively,P<0.05).The inhibitory rate of cell proliferation was increased to 62%from 4%at 48 h after transfection.The rate of G2/M phase cells was decreased after transfection with efficient shRNA(control group:40.2%and 41.3%,test group:21.3%).Conclusions Down-regulation of hBUB1 gene leads to the inhibition of cell proliferation and the arrest of cell-cycle.It also probably plays an important role in the development of spontaneous abortion embryos with numerical chromosomal abnormality.The clinical relevance warrant further investigation.
8.Visualization of serotonin 1A receptor trafficking in neuron-like PC12 cells

Objective To explore the mechanisms of trafficking and signaling of serotonin 1A receptor(5-HT_(1A))and its spatiotemporal distribution in living cells.Methods The mouse 5-HT_(1A) gene amplified by RT-PCR was recombined into pEGFP-N1 vector and the EGFP coding sequence was located in-frame at the C-terminal end of the 5-HT_(1A) receptor.The 5-HT_(1A)-EGFP was transfected into neuron-like PC12 cells as well as HEK293.The transfected cells were visualized using confocal microscopy,the mobility of 5-HT_(1A)-EGFP was monitored by live measurements and fluorescence recovery after photobleaching.Results The 5-HT_(1A) gene was identitical with the published gene sequence NM_008308.4 and a 5-HT_(1A)-EGFP fusion construct was created.After stable transfection of the plasimd into a PC12 cell line and analysis with a confocal laser scanning microscopy,the EGFP-tagged 5-HT_(1A) was predominantly associated with the plasma membrane,but some intracellular vesicles in the perinuclear region also contained the fusion protein.The predominant localization of 5-HT_(1A)-EGFP at the plasma membrane was confirmed in transiently transfected HEK293 cells.Bleached fluorescence was partialy recovered in 100 seconds,indicating that the 5-HT_(1A)-EGFP was mobiled on the membrane.Conclusion Spatiotemporal distribution and mobility of 5-HT_(1A) tagged with EGFP can be monitored in the 5-HT_(1A)-EGFP stable PC12 cell line,which could be an excellent neuron-like experimental cell model for research of 5-HT_(1A) trafficking and signaling.

10. Predicting value of forearm bone density on osteoporosis in patients with postmenopausal rheumatoid arthritis and its influencing factors

Objective:
To explore the predictive value of forearm bone density on osteoporosis in patients with postmenopausal rheumatoid arthritis (RA) and its influencing factors.
Methods:
The clinical data of 98 postmenopausal RA patients in Department of Endocrinology and Hematology, the Third People′s Hospital of Hangzhou from January 1, 2016 to December 31, 2018 were retrospectively analyzed. The bone density of the forearm (left) and central axis (lumbar spine and left hip) were measured using dual-energy X-ray absorptiometry (DXA), and the score of health assessment questionnaires (HAQ) was evaluated. The relationship between central axis bone density and forearm bone density, the optimal T value of forearm bone density for the diagnosis of lumbar spine and left hip osteoporosis, and the influencing factors of forearm bone density were analyzed.
Results:
In 98 patients with postmenopausal RA, lumbar vertebrae bone density was (0.92 ± 0.17) g/cm2, and T value was -0.7 ± 0.4; left hip bone density was (0.80 ± 0.15) g/cm2, and T value was -1.2 ± 1.1; forearm bone density was (0.36 ± 0.13) g/cm2, bone mineral content was (0.72 ± 0.23) g/cm, and T value was -2.3 ± 1.8. The central axial bone density measurement result showed that normal bone density was in 15 cases (15.31%), and abnormal bone density was in 83 cases (84.69%). The lumbar spine bone density measurement result showed that normal bone density was in 36 cases (36.73%), and abnormal bone density was in 62 cases (63.27%). The left hip bone density measurement result showed that normal bone density was in 19 cases (19.39%), and abnormal bone density was in 79 cases (80.61%). The forearm bone density measurement result showed that normal bone density was in 24 cases (24.49%), and abnormal bone density was in 74 cases (75.51%). The age was significantly younger, the forearm bone density, bone mineral content and T value were significantly higher in patients with normal central axial bone, lumbar spine and left hip bone density, compared with those in patients with abnormal central axial bone, lumbar spine and left hip bone density respectively, and there were statistical differences (
