1.Different metabolites of leaves between Tripterygium wilfordii and Tripterygium hypoglaucum based on UPLC-Q-TOF-MS.

To analysis the differences between Tripterygium wilfordii and T. hypoglaucum, specimens of their leaves were collected from five production regions and analyzed by ultra performance liquid chromatography coupled with quadrupole time of flight mass spectrometry (UPLC-Q-TOF-MS). The data were analyzed by multivariate statistical method, such as hierarchical cluster analysis (HCA) principal component analysis (PCA) and orthogonal signal correction partial least square discrimination (OPLS-DA). Potential markers with VIP values above 5.0 and corresponding r values above 0.85, were selected and further tested by combining mann-Whitney nonparametric. Those with P < 0.001 and AUC = 1 were confirmed as metabolite markers to discriminate them from each other. Results revealed that the two species were obviously different in their leaf metabolites. Based on their mass spectra, 23 potential metabolite markers were identified to distinguish T. wilfordii from T. hypoglaucum.

2.Echocaridiographic evaluation on early left ventricular remodeling after acute myocardial infarction: experimental study

Objective To investigate the onset of left ventricular remodeling (LVRM) after acute myocardium infarction (AMI) and its changes within 6 hours in dogs on echocardiography. Methods AMI was induced in 14 dogs by ligating the left anterior descending arteries. Eight myocardium infarcted models were successful and were sacrified for pathological study. The indices of LVRM: wall infarction thickness (WIT), the wall motion score index (WMSI), left ventricular end-diastolic volume (LVEDV), left ventricular end-systolic volume (LVESV) and left ventricular ejection fraction (LVEF) were evaluated before and 1 h, 2 h, 3 h, 4 h, 5 h and 6 h after operation. Results Compared with the pre-operation, WIT and LVEF were decreased (P<0.01), LVESV and WMSI were increased (P<0.01), and LVEDV was increased (P<0.05 or P<0.01) at every time point after operation. WIT had no significant difference at 1 h, 2 h, 3 h, 4 h, 5h and 6h after operation (P>0.05). LVEDV, LVESV were higher (P<0.05) and LVEF was lower (P<0.05 or P<0.01) at 4 h, 5 h, and 6 h than at 1 h, and 2 h after operation. WMSI was higher at 3 h, 4 h, 5 h, and 6 h than at 1h (P<0.05). Conclusions In our experiment, LVRM occurred at 1 h after AMI in dogs. Thus echocardiography may evaluate early LVRM.
3.Budd-Chiari syndrome:diagnosis with three-dimensional contrast-enhanced MR angiography

Objective To assess the various features of Budd-Chiari syndrome(BCS)on three- dimensional contrast-enhanced magnetic resonance angiography(3D CE MRA)and to evaluate the potential value of this new technique.Methods Thirty-three patients with BCS underwent 3D CE MRA examination. In 23 cases,BCS was secondary to hepatocellular carcinoma(21 patients)or right adrenal carcinoma (1 patient)or thrombophlebitis(1 patient).Ten patients had primary BCS.The patency of the hepatic veins,inferior vena cava(IVC)and portal veins were assessed.The presence of intra-and extrahepatic collaterals,liver parenchymal abnormalities and porto-systemic varices were evaluated.The diagnosis on 3D CE MRA was correlated with that on inferior vena cavography and right hepatic venography,which were available in 10 and 2 cases respectively.Results Various features of BCS were displayed on 3D CE MRA. Hepatic venous findings included tumor thrombosis(19 patients),tumor compression(2 patients), nonvisualization(4 patients)and focal stenosis(4 patients)of the hepatic veins.IVC findings were severe stenosis or occlusion(10 cases),tumor direct invasion(2 cases),tumor thrombosis(3 cases), thrombophlebitis(1 case)and web formation(3 cases).Intrahepatic collaterals were demonstrated in 9 patients including 2 with "spider web" sign.Detected extrahepatic collaterals included dilated azygos and hemiazygos veins(13 cases)and left renal-inferior phrenic-pericardiophrenic collaterals(2 cases).The occlusion of the left portal vein and the presence of porto-systemic varices were depicted in 2 and 10 patients respectively.Liver parenchymal abnormalities identified by 3D CE MRA consisted of caudate lobe enlargement(7 cases),heterogenous enhancement(18 cases)and associated tumors(18 cases). Compared with inferior vena cavography and hepatic venography,the accuracy of 3D CE MRA in the diagnosis of IVC obstruction or hepatic venous stenosis was 100%.Conclusion 3D CE MRA can display various features of BCS and has the potential to provide an accurate diagnosis.
4.Expression and bioactivity of OSF-1 in Pichia pastoris.

In order to research the biologic activity of osteoblast-stimulating factor 1 (OSF-1), the pPIC9K/osf-1 yeast expression vector was constructed to express and purify OSF-1. Firstly, the osf-1 gene sequence was obtained by artificial synthesis and cloned into Pichia pastoris expression vector pPIC9K to generate pPIC9K/osf-1. The recombinant plasmid was linearized by Sac I and transformed into P. pastoris GS115 by electroporation. Recombinant P. pastoris GS115/ pPIC9K/osf-1 was screened by MD and G418-YPD plates and further identified by PCR. The positive P. pastoris was induced with 1% methanol at 25 degrees C for 96 h. The target protein was analyzed by SDS-PAGE showing a special band about 18 kDa. The target protein was successfully purified from the supernatant of the broth using ion exchange chromatography of SP-Sephadex C-50. The purity of target protein was above 98%. Western blotting appeared a good antigenicity of the purified protein. Bioassay results show that the recombinant protein OSF-1 can promote the differentiation and proliferation of osteoblasts MC3T3-E1. We successfully expressed OSF-1 by recombinant P. pastoris for further development of anti-osteoporosis of research and industrial production of OSF-1.

5.Effect of vitamin D insufficiency on the quality of life in the elderly

Objective To assess the effect of vitamin D insufficiency on the quality of life in the elderly.Methods A total of 205 cases in health examination center of our hospital were randomly divided into elderly group (aged ≥ 60 years) and middle-aged g roup (aged 45-60 years).The SF-36 Form was adopted to investigate status of body and mental health.Serum 25 (OH)D3 levels were measured by enzyme-linked immunosorbent assay (ELISA).Serum 25 (OH) Ds ≤ 49.9 nmol/L was defined as vitamin D insufficiency. ResultsThere were 55 cases (53.9%) with vitamin D insufficiency and 16 cases (15.7%) vitamin D sufficiency in the elderly.The scores of vitamin D insufficiency group in the elderly were lower than of vitamin D threshold and sufficiency groups in fourdimensions,including physical function,physical role,body pain and general health.The scores ofvitamin D insufficiency group in the elderly were also lower than of vitamin D threshold and sufficiency groups in three dimensions,including social functioning,vitality and mental health (F=10.28,9.20,6.60,all P<0.05).Serum 25(OH)D3 played more important role in scores of the quality of life in the elderly than other factors by stepwise multiple linear regression. ConclusionsThe vitamin D insufficient status widely exists in the elderly,and it might influence their quality of life.
6.Nutrition survey of vitamin D in the elderly people from state organs and public institutions in Jinan urban district

Objective To study the vitamin D nutritional status in the elderly people from organs and institutions in Jinan urban district. Methods 564 cases were available from institutions enjoying a public medical care in Qianfoshan Hospital by stratified random sampling.The subjects aged (69.5±6.5)years were divided into two groups:60-74 years and≥75 years.The [25(OH)D] levels was measured by enzyme linked immunosorbent assay (ELISA).It was defined that [25(OH) D] levels ≥ 75 nmol/L was sufficient,50.0 74.9 nmol/L was critical,25.0-49.9 nmol/L were insufficient and <25 nmol/L was deficient.All data processing and statistical analysis were finished by SPSS13.0. Results The numbers of sufficient,critical,insufficient and deficient vitamin D in 564 subjects were 114(20.2%),180(31.9%),234(41.5%) aud 36(6.4%),in subjects aged ≥75years were 13(9.8%),36(27.3%),70(53.0%) and 13(9.8%),and in subjects aged 60-74 years were 101(23.4%),144(33.3%),164 (38.0%),23 (5.3%),respectively. There was significant difference in the vitamin D nutritional status between the two age groups (x2=19.097,P<0.01 ).The numbers of sufficient,critical,insufficient and deficient vitamin D in the 246 male were 57 (23.2%),91(37.0%),86 (35.0%%) and 12(4.9%),in the 318 female were 57(17.9%),89 (28.0 % ),148(46.5 % ) and 24(7.5 % ).The percentages of insufficient and deficient vitamin D in the female were higher than in the male (x2 =11.445,P<0.01 ). Conclusions Insufficient and deficient vitamin D nutritional states are common in the elderly people from organs and institutions in Jinan urban district and are increasing with age,especially in the female.

8.Expression of cellular fibronectin mRNA in adult periodontitis and peri-implantitis: a real-time polymerase chain reaction study.

Cellular fibronectin (cFn) is a type of bioactive non-collagen glycoprotein regarded as the main substance used to maintain periodontal attachment. The content of cFn in some specific sites can reflect the progress of periodontitis or peri-implantitis. This study aims to evaluate the expression of cFn messenger RNA (mRNA) in tissues of adult periodontitis and peri-implantitis by real-time fluorescent quantitative polymerase chain reaction (PCR) and to determine its clinical significance. A total of 30 patients were divided into three groups of 10: healthy, adult periodontitis and peri-implantitis. Periodontal tissue biopsies (1 mm×1 mm×1 mm) from each patient were frozen in liquid nitrogen. Total RNA was extracted from these tissues, and the content, purity and integrity were detected. Specific primers were designed according to the sequence, and the mRNA expression levels of cellular fibronectin were detected by real-time PCR. The purity and integrity of the extracted total RNA were both high, and the specificity of amplified genes was very high with no other pollution. The mRNA expression of cFn in the adult periodontitis group (1.526±0.441) was lower than that in the healthy group (3.253±0.736). However, the mRNA expression of cFn in the peri-implantitis group (3.965±0.537) was significantly higher than that in the healthy group. The difference revealed that although both processes were destructive inflammatory reactions in the periodontium, the pathomechanisms were different and the variation started from the transcription level of the cFn gene.

9.Effect of β-sodium aescinate on hypoxia-inducible factor-1α expression in rat brain cortex after cardiopulmonary resuscitation

BACKGROUND: This study was undertaken to investigate the expression of hypoxia-inducible factor-1α (HIF-1α) in rat cerebral cortex and the effects of β-sodium aescinate (SA) administration after return of spontaneous circulation (ROSC).METHODS: Sixty rats were divided into three groups: SA group, injected intraperitoneally with SA instantly after ROSC; control group, injected intraperitoneally with normal saline; and sham-operated group, without cardiac arrest or SA. The cardiac arrest model was established using asphyxiation and intravenous potassium chloride. Blood was sampled 1, 6, 12, and 24 hours after ROSC. Protein and mRNA levels of HIF-1α, VEGF and EPO were detected in the cerebral cortex by immunohistochemistry and real-time RT-PCR; serum levels of NSE and S100β were determined by enzyme-linked immunosorbent assays.RESULTS: Serum S100β and NSE were signifi cantly increased in the control group versus the sham-operated group 1, 6, 12 and 24 hours after ROSC (P<0.05). Protein and mRNA levels of HIF-1α, VEGF and EPO were signifi cantly increased in the control rats (P<0.05). Serum NSE and S100β were significantly decreased in the SA group versus the control group 1, 6, 12 and 24 hours after ROSC (P<0.05). Protein and mRNA levels of HIF-1α, VEGF and EPO were signifi cantly increased in the SA group (P<0.05).CONCLUSIONS: The expression of HIF-1α is increased in rat cerebral cortex after ROSC, and SA up-regulates the expression of HIF-1α. The up-regulation of HIF-1α improves the resistance of the cortex to ischemia and hypoxia and contributes to neuroprotection, possibly because of up-regulation of EPO and VEGF expression.
10.The lipidsolubility components analysis and antioxidant activity research of Caryopteris tangutica

Objective:To analyze the liposoluble components and antioxidant capacity of Caryopteris tangutica. Methods:By using GC-MS, this paper analyzes liposoluble components of different positions of Caryopteris tangutica. By making the removal rate of free radicals with liposoluble components as the index, and making IC 50 as reference indicators to evaluate the antioxidant capacity of lipidsoluble extracts of different parts. Results:There are 31, 40 and 62 compounds identified in the liposoluble components of the stems, leaves and flower, among which unsaturated fatty acids account for the most. The different parts of Caryopteris tangutica all play the role of clearing DPPH and ABTS + free radicals, and the clearing rate of leaves was greater than that of flowers and stems. Conclusion:The unsaturated fatty acids in lipidsoluble components of Caryopteris tangutica are beneficial to the human body, and have antioxidant activity.
