2.Influence of multiple sedation of dexmedetomidine and midazolam on young rats′nervous injury and learning and memory ability after grow up

Objective To investigate the influence of multiple sadation of dexmedetomidine (Dex) and midazolam on young rats′nervous injury and the learning and memory ability after grow up .Methods 36 male SD rats were randomly allocated to 3 groups (n=12):control group(C) ,midazolam group (M ) and dexmedetomidine group(D) .The group C was given normal saline 0 .1 mL/kg by subcutaneous injection .The group M was subcutaneously injected by midazolam 2 .4 mg/kg ,0 .1 mL/kg .The group D was subcutaneously injected by dexmedetomidine 75 μg/kg ,0 .1 mL/kg .Each group received once injection for 3 d .Plasma S100β,NSE were measured at 6 h after medication .The rats′memory ability after grow up was assessed by the Morris water maze test .Results Compared with the group C ,the escape latency in the group M was extended and the times across the platform was decreased . The expressions of S100βand NSE in the group M were enhanced at 6 h after medication .There was no statistical difference be-tween the group C and D .The changes of various indexes in the group D had no significance .Conclusion Compared with midazo-lam ,using new type narcotic and sedative agent dexmedetomidine had little influence on the young rat nervous injury and the learn-ing and memory ability after grow up in rat .

5.Screening and Identification of Egg Mimotope of Schistosoma japonicum

Objective To explore egg mimotope of Schistosoma japonicum that can be used in the development of diagnostic reagents for schistosomiasis.\ Methods\ By performing three rounds of biopanning in the affinity selection before picking out single clones for identification, target specific phages were effectively enriched from a random 15\|peptides phage library with an immobilized mAb 6B12 which is specific to egg antigen of S\^japonicum as a bait. By using ELISA, competitive ELISA with natural egg antigen as competitor, and Western blotting, 13 phage clones with high affinity and specificity to 6B12 were obtained and amplified from 400 single clones. \ Results \ DNA sequencing revealed that all the 13 selected single clones were identical in displaying gene sequence. Serum samples were tested by ELISA for the presence of IgG antibodies to the phages with mimic epitope, and showed that there was a significant difference between healthy volunteers and schistosomiasis patients.\ Conclusion \ It is possible that the phage display peptide antigen, egg mimotope of S.japonicum may be a replacement of natural egg antigen for the diagnosis of schistosomiasis.
6.DNA vaccination encoding variable region gene of the T cell receptor induces regulatory immune response in BALB/C mice

Objective:To investigate regulatory immune response induced by DNA vaccination encoding T cell receptor V?5.2 or V?2.1 chain predominantly displayed on ovalbumin (OVA)-specific T cell clone.Methods:BALB/C mice were vaccinated with pcDNA3.1 encoding T cell receptor (TCR) V?5.2 or V?2.1 chain respectively.Using RT-PCR,transcription of the recombined plasmids was analysed. Cell proliferation was measured by 3H-TdR incorporation.CTL response was assayed by JAM test.Immuno-fluorescent assay was used to examine the anti-TCR antibody level and the number V?2 +T cells.Results:RT-PCR analysis showed that the recombined plasmids can be transcripted in vivo and in vitro.DNA vaccine with TCR variable chain effectively induced TCR-specific humoral and cellular immune response,V?2 +T cells was not depleted by V?2.1 TCR-DNA vaccination,but rather was anergy.Conclusion:Regulatory immune response can be induced by DNA vaccination encoding TCR V? or V? region in normal mice.
7.Multi-slice spiral CT diagnosis of complex congenital heart diseases

Objective To investigate scanning and reconstruction techniques of multi-slice spiral CT (MSCT) in patients with complex congenital heart diseases (CCHD).Methods One hundred eighty-four patients suffering from CCHD underwent 16-detector MSCT scanning without ECG-gating.Multi-planar reconstruction (MPR),maximum intensity projection (MIP),curved-planar reconstruction (CPR) and volume rendering (VR) were used to reconstruct images.CT findings were compared with those of surgical operation or angiocardiography.Results A total of 616 cardiac deformities were found with MSCT and proved by angiocardiograms or surgical operation.The diagnostic accuracy of extracardiac malformation with MSCT was 100%,of atrial septal defect was 54.65%,and of ventricular septal defect was 78.62%.MSCT failed to display heart valve disease well.Conclusion MSCT can accurately detect extracardiac malformations of CCHD.
8.Causal analysis of blood heat syndrome in psoriasis vulgaris

Objective:To analyze the cause of the blood heat syndrome in psoriasis.Methods: Made the investigation form about information of psoriatic patients through DME method and reference both Chinese and foreign documents,analyzed main component of data by SAS software.Results: The first main component of blood heat syndrome in psoriasis are infection,drinking,dietetic problem,and emotion;the second main component is family history;the third main component is dampness;the forth main component is the allergy to the food;the fifth is outside irritation.Through the corresponding investigation we found that food include hot food,beef,sheep and sea food,emotion problem include over work and stress,dampness include work and living in humid place for long time,outside stimulus meant chemical stimulus.Conclusion: The main cause of the blood heat syndrome in psoriasis are infection,drinking,dietetic problem,and emotion,family history,humidity,allergy to the food and chemical stimulus in order.
9.Clinicopathological and prognostic analyses of primary ovarian small cell carcinoma:A report on four cases and a review of the literature

Objective:This study analyzes the clinicopathological characteristics, pathological diagnosis, treatment, and prognosis of ovarian small cell carcinoma (SCCO). Methods:The medical records of SCCO patients in the Cancer Hospital of Peking Union Medical College between 2005 and 2012 were reviewed. Results:The mean age of patients was 43. 75 years old (ranging from 17 to 57), two cases were postmenopausal, and one case was less than 30 years old. Twenty-one patients had FIGO stageⅠ, whereas three cases had stageⅢto Ⅳ. Three cases were classified as pulmonary type. Forty-three patients received cytoreductive surgery, and one underwent fertili ty-conserving surgery. All patients were postoperatively treated with platinum-based chemotherapy, whereas three cases received Paclitaxel plus Carboplatin. No patient received adjuvant radiotherapy. One patient died nine months after the initial diagnosis, and one died 12 months after the initial diagnosis. The other two cases remain alive with no evidence of recurrence after follow up at 7 and 30 months after diagnosis. Conclusion:SCCO is a rare ovarian tumor with high malignancy potential and thus has poor prognosis. The clinical manifestations of SCCO resemble those of epithelial ovarian cancer. Immunohistochemistry can be used for differential diagnosis. The standard SCCO treatments are cytoreductive surgery and adjuvant platinum-based chemotherapy. The optimal chemotherapy regimen requires further research.
10.Effect of immune enteral nutrition support on the patients in early stage after severe multiple injury

Objective To explore the recuperative effect of immunological function and nutritional status on the patients treated by immune enteral nutrition in early stage after severe multiple injury (SMI). Method The patients with SMI,in department of Trauma Surgery,Tongji Hospital,Tongji Medical College, Huazhong University of Science and Technology,between January 2006 to May 2007 were randomly divided into 2 groups: immune enteral nutrition group (IEN group, 20 cases), enteral nutrition group (EN group, 20 cases). The health persons served as the control group(15 cases) .Since 1st postinjury day, all patients were treated with nutritional support. The T-cell subgroup in periphera blood were detected by FCM and the level of PA, RBP, IL-2 and IL-4 in blood serum were detected by ELASA on the 1st, 3rd,5th, 8th postinjury day. Results After the treatment of IEN and EN,the serum levels of PA, RBP and the proportion of T-cell subgroup were significantly increased on the 8th postinjury day compared with on 1st postinjury day (P < 0.01), but there were no differences between IEN group and EN group. The level of IL-4 were significantly decreased and the level of IL-2 were significantly increased in each group on 8th postinjury day, at same time, the level of IL-2 were significantly increased in IEN group compared with EN group (P < 0.05), and the level of IL-4 were significantly decreased in IEN group compared with EN group (P < 0.05). The duration of SIRS was transient and the infected complication was low on the patients treatment by IEN than EN. Conclusions On the patients with severe multiple injury, IEN was most ascendant than EN to improve the immunosuppression and clinical prognosis.
