1. Determination and comparison of luteoloside, apigenin-7-O-β-D-glucosidase, and luteolin in Humulus scandens from different habitats in Hebei province

Objective: To establish a method for the determination of luteoloside, apigenin-7-O-β-D-glucosidase, and luteolin by high performance liquid chromatography (HPLC) method, and the determination of luteoloside, apigenin-7-O-β-D- glucosidase, and luteolin in Humulus scandens female and male plants from different origins and different harvest months in Hebei province. Methods: HPLC method was used for simultaneous determination of luteoloside, apigenin-7-O-β-D-glucosidase, and luteolin. System suitability conditions were as follows: column: Agilent 5 TC-C18 (250 mm × 4.6 mm, 5 μm); mobile phase: acetonitrile-0.2% phosphoric acid; flow rate: 1.0 mL/min; detection wavelength: 340 nm; column temperature: 25 ℃; injection volume: 10 μL. Results: The linear ranges were 4.640-74.24 μg/mL of luteoloside (r = 0.999 2), 1.510-30.20 μg/mL of apigenin-7-O-β-D-glucosidase (r = 0.999 9), and 0.796 3-12.74 μg/mL of luteolin. RSD of precision, stability, and repeatabillity tests were ≤ 1.99%. The average recoveries were 102.69% (RSD = 1.64%, n = 9), 100.49% (RSD = 1.93%, n = 9), and 100.39% (RSD = 1.65%, n = 9). Conclusion: The method is simple, reproducible, and accurate, and reliable measurement results can be used for simultaneous determination of luteoloside, apigenin-7-O-β-D- glucosidase, and luteolin in H. scandens female and male plants from different origins and different harvest months in Hebei province. There is greater difference between luteoloside, apigenin-7-O-β-D-glucosidase, and luteolin in H. scandens female and male plants from different origins and different harvest months in Hebei province.
2. Comparative study on HPLC characteristic spectra of Humuli Scandentis Herba from different habitats in Hebei province

Objective: To establish and compare the HPLC characteristic spectra of Humuli Scandentis Herba form different habitats in Hebei province. Methods: HPLC was performed on Agilent 5 TC-C18 (250 mm × 4.6 mm, 5 μm) column, with the mobile phase of acetonitrile -0.2% phosphoric acid at flow rate of 1.0 mL/min; Detection wavelength was 340 nm; The column temperature was 25 ℃ and the sample size was 10 μL. Eleven batches of Humuli Scandentis Herba samples form different habitats in Hebei province were determined and the characteristic spectra of those were established. The fingerprint evaluation software (2004 edition) for Chinese materia medica (CMM) was used to evaluate the similarity of the 11 batches of samples. Results: There were nine characteristic peaks identified in the characteristic spectra of Humuli Scandentis Herba samples. Peak 7 was luteoloside and Peak 8 was apigenin-7-O-β-D-glucosidase. The similarities of seven batches of Humuli Scandentis Herba samples were proved to be higher than 0.900 and four batches of them were proved to be less than 0.900. Conclusion: The method is simple, accurate, and reproducible, and can provide the scientific evidence for controlling the internal quality standards effectively.
4.Effect of Comprehensive Therapy on Forearm Extensor Myotenositis: 72 Cases Report

ObjectiveTo observe the effect of comprehensive therapy on forearm extensor myotenositis.Methods72 cases were divided into two groups: a control group of 36 cases who were given routine treatment,and an experiment group of 36 cases who were given thermotherapy,computerized medium-frequency electrotherapy,physiotherapy,and ADL instruction,etc.After two courses,a simple grading score(for forearms) was used to assess the effect.ResultsOf the control group,22 cases were cured,10 remarkably effective,4 effective;of the experiment,30 cured,4 remarkably effective,2 effective(u=2.04, P<0.05).The difference of average score for forearms before and after the treatment were(6.58±3.17) points for the control and(8.19±3.55) for the experiment(t=2.03,P<0.05).The average days of cure were(5.60±2.54) d for the experiment group,shorter than those for the control(7.00±2.27) d(t=2.05,P<0.05).ConclusionComprehensive therapy is effective on forearm extensor myotenositis.
5.Current status of iodine deficiency disorders in Henan Province in 2011

Objective To analyze the present situation of iodine deficiency disorders (IDD) in Henan Province,and to promote implementation of sustainable control strategies.Methods In 2011,a stratified proportion to population probability sampling (PPS) method was used to survey 1200 children aged 8 to 10 in 30 counties of the province.One primary school was selected in each chosen county.Goiter,intelligence quotient (IQ),urinary iodine and salt iodine level were studied.Meanwhile,12 families per capita salt intake was investigated.In each school,30 5th-grade students and 30 pregnant and lactating women in the school townships and adjacent neighboring townships were selected to carry out questionnaire survey on health education with unified papers.Results ①The goiter rate of children aged 8 to 10 by B ultrasound was 4.5% (54/1201) ; the IQ of 1080 children was 107.75 ± 16.81 ; median urinary iodine level of 358 children was 201.4 μg/L.②The median of salt iodine content was 28.6 mg/kg,the coverage rate of iodized salt was 98.8% (1186/1200),and qualified rate of iodized salt was 93.0% (1116/1200).③The residents average daily salt intake was 10.5 g.④Average score of the questionnaire survey of 1084 5th-grade students was 4.2 points.Average score of 961 housewives was 4.4 points.Conclusions Various technical indicators show that IDD is in a sustained elimination state in Henan Province.Strengthen health education,enhance public awareness of disease prevention is still the important work ahead.
6.External quality control of iodine deficiency disorders laboratory in Henan province from 1999 to 2011

Objective To analyze the examination results of external quality assessment(EQA) at all levels of iodine deficiency disorders(IDD) laboratories in Henan province and the network operation to further standardize and improve the laboratory,and to provide reliable laboratory quality assurance for surveillance and control of IDD.Methods The examination results of EQA at all levels of IDD laboratories in Henan province were statistically analyzed in accordance with the National Reference Laboratory (NRL) of IDD (1999-201 1).Results The survey results showed that the provincial level laboratory was all qualified in testing urinary iodine and salt iodine in the past 13 years.In prefectural level,the laboratory response rates were 100.0%,and through participation in EQA,laboratory capacity had been significantly increased and stabilized.From 1999 to 2001,the passing rate of check up of urinary iodine was 22.2% (4/18),72.2% (13/18),94.4% (17/18),respectively,and the rate was stable at 100.0%(18/18) from 2002 to 2011 except 94.4% (17/18) in 2003.Since 2000,the prefectural level laboratory began to take part in the salt iodine EQA,and the laboratory response rate was 100.0% (18/18) from 2000 to 2011.Except 88.9%(16/18) in 2003,the passing rate of check up of urinary iodine was 100.0%(18/18)from 2000 to 2011.In 2003 and 2004,6 to 7 county-level laboratories participated in the EQA of urinary iodine in Zhengzhou city,respectively,and all qualified.The number of county-level laboratories that participated in the salt iodine quality control network increased from 29 in 1999 to 148 in 2011.Response rate was 94.4%(68/72),96.7%(58/60) and 92.3%(144/156)in 2003,2006 and 2007,respectively,and the rate remained stable at 100.0% in the remaining 10 years.In 1999,the passing rate was 69.0% (20/29),then increased significantly,except 86.7% (26/30) in 2001 and 84.6%(132/156) in 2007,the rates were all above 90.0% in other years,especially in 2000 and 2009,the passing rates were both 100.0%.Conclusions The accuracy of test results of external quality controls and the normal operation of the network at all levels of laboratories is closely related to the IDD laboratory conditions and detection techniques.
7.Effects of Medical Treatment of Urinary Calculi in 112 Infants under Three Years Old with A History of Fee-ding Melamine-Contaminated Milk Powder

Objective To summarize the effects of medical treatment of urinary calculi in 112 infants under 3 years old with a history of feeding melamine-contaminated milk powder.Methods All the infants with a history of feeding melamine-contaminated milk powder[73 male,39 females;aged(14.43?8.63) months] were diagnosed with type-B ultrasonic,and through the main treatment procedures were 50 g/L sodium bicarbonate(0.5 mL/kg,intrarenous injection) and fluid infusion.The dosage of drugs were timely adjusted according to the monitored uine pH.Hemodialysis was performed in the patients with renal failure.These were decided on the basis of the effect of medical treatment.Results During their hospitalization of 3 to 25 days,56 cases obtained full response,49 cases showed partial response and 7 cases demonstrated no response.The mean diameters of calculi were(4.77?3.16) mm,(8.13?3.79) mm and(6.92?2.65) mm,and the average pH values were 5.64?0.11,5.75?0.10 and 6.38?0.31 in full response,partial response and no response groups,respectively.The average pH value in no response group was significantly higher than that in full response group(P0.05).Conclusions Medical treatment can help to obtain considerable therapeutic effects for infants under 3 years old with urinary melamine calculi.Treatment response has shown partial relationship with the size of urinary calculi and pre-treatment urine pH,while no relationship is found to be related to infants′ age.
8.CT imaging of coexisting pulmonary tuberculosis and lung cancer

Objective To study the CT characteristics of coexisting pulmonary tuberculosis and lung cancer.Methods One hundred and four patients of coexisting pulmonary tuberculosis and lung cancer proved by histology,cytology or clinical underwent CT examination.All patients were divided into two groups,group Ⅰ were the patients with the lung cancer after tuberculosis or both found simultaneously (group Ⅰ a with peripheral lung cancer and group Ⅰ b with central lung cancer),group Ⅱ with tuberculosis during lung cancer chemotherapy (group Ⅱ a with peripheral lung cancer and group Ⅱ b with central lung cancer).Imaging characteristics of tuberculosis and lung cancer were compared.x2 test and t test were used for the statistical analysis.Results Of 104 patients,there were 92 patients (88.5%) in group Ⅰ and 12 patients (11.5%)in group Ⅱ.Seventy patients (76.1%) of lung cancer and tuberculosis were located in the same lobe and 22 patients (23.9%) in the different lobes in group Ⅰ.There was no significant difference in distribution of tuberculosis between group Ⅰ and group Ⅱ (x2 =4.302,P =0.507).The fibrous stripes,nodules of calcification and pleural adhesion of tuberculosis were statistically significant between the two groups (x2 =22.737,15.193,27.792,P <0.05).There were 33 central lung cancers and 71 peripheral lung cancers.In group Ⅰ a (64 patients of peripheral lung cancers),39 patients (60.9%) had typical manifestations and most of the lesions were ≥ 3 cm(n =49,76.6%),solid lesions showed variable enhancement.Conclusions Secondary tuberculosis during lung cancer chemotherapy has the same CT characteristics with the common active tuberculosis.The morphology,enhancement pattern of lesion and follow-up are helpful for the diagnosis of lung cancer after tuberculosis.
9.Prognostic value of CDKN2A mRNA level in glioblastoma

Objective To investigate the prognostic value of CDKN2A mRNA in glioblastoma (GBM).Methods CDKN2A gene mRNA data were obtained from three different GBM database online (TCGA,REMBARNDT and GSE16011).The correlations between overall survival (OS) and CDKN2A expression were analyzed by Kaplan-Meier method and multivariate Cox regression analysis.Results In the TCGA database (n =358),patients with high CDKN2A mRNA level got longer OS than those with low expression level [median OS:18.0 months (95 % CI 15.0-21.0 months) vs 13.9 months (95 % CI 12.4-15.4 months),P =0.001].In another two validation datasets,patients with high CDKN2A mRNA level had longer OS than those with low expression level [median OS in REMBRANT:16.6 months (95 % CI 13.3-19.8 months) vs 11.8 months (95 % CI 7.3-16.4 months),P =0.019; in GSE16011:11.9 months (95 % CI 8.3-15.6 months) vs 8.4 months (95 % CI 6.2-10.5 months),P =0.005].CDKN2A mRNA level was an independent prognostic factor for GBM.The combination of CDKN2A mRNA expression with MGMT promoter methylation status or G-CIMP status/IDH1 mutations provided an optimized prognostic factor in GBM patients.Conclusion The CDKN2A mRNA has prognostic value in GBM patients,which provided an optimized stratification strategy based on multiple biomarkers.
10.Effect of miR-21 on albuminuria in db/db mouse with early stage of diabetic nephropathy

Objective To explore the effect of miR-21 on 24 h urine albumin excretion (UAE), glomeruli morphology, and its molecular mechanism in db/db mouse with early stage of diabetic nephropathy. Methods Twenty-four 5-week-old male mice were randomized into 4 groups: control group (untreated db/m mice, n=6),miR-21-treated db/db group (n=6), control empty plasmid treated db/db group (n =6), and untreated db/db group (n = 6). Mice were injected intraperitoneally using a hydrodynamics-based procedure with plasmids (30 mg · kg-1 · d-1 of miR-21 or 30 mg· kg-1 · d-1 of control plasmid) until albuminuria was detected in the untreated db/db mice. The glomeruli were observed under the light microscope, and measured by the software in the computer. The expression of miR-21 was tested by real-time RT-PCR, the expression of PTEN, p-Akt (Ser 473),and phosphatidylinositol-3-kinase (PI3K) p85α protein levels were examined by western blot and immunohistochemistry. Results Male db/db mice of 9-week-old with hyperglycemia and significant elevation of urinary albumin excretion showed features similar to the early stage of diabetic nephropathy. Real-time RT-PCR showed that miR-21 was increased in the miR-21-treated group. By measurement of glomeruli acreage, the glomeruli were smaller in miR-21-treated group than in the control empty plasmid treated and untreated groups (all P<0.01). By Western blot and immunohistochemistry, PTEN were reduced in the miR-21-treated group, whereas PI3K p85α and phospho-Akt (Ser 473 ) were increased in miR-21-treated group (all P < 0.01). Conclusions miR-21 ameliorates glomerular hypertrophy in db/db mice with diabetic nephropathy, and it may be a novel protecting factor of diabetic nephropathy.
