1. Determination and comparison of luteoloside, apigenin-7-O-β-D-glucosidase, and luteolin in Humulus scandens from different habitats in Hebei province

Objective: To establish a method for the determination of luteoloside, apigenin-7-O-β-D-glucosidase, and luteolin by high performance liquid chromatography (HPLC) method, and the determination of luteoloside, apigenin-7-O-β-D- glucosidase, and luteolin in Humulus scandens female and male plants from different origins and different harvest months in Hebei province. Methods: HPLC method was used for simultaneous determination of luteoloside, apigenin-7-O-β-D-glucosidase, and luteolin. System suitability conditions were as follows: column: Agilent 5 TC-C18 (250 mm × 4.6 mm, 5 μm); mobile phase: acetonitrile-0.2% phosphoric acid; flow rate: 1.0 mL/min; detection wavelength: 340 nm; column temperature: 25 ℃; injection volume: 10 μL. Results: The linear ranges were 4.640-74.24 μg/mL of luteoloside (r = 0.999 2), 1.510-30.20 μg/mL of apigenin-7-O-β-D-glucosidase (r = 0.999 9), and 0.796 3-12.74 μg/mL of luteolin. RSD of precision, stability, and repeatabillity tests were ≤ 1.99%. The average recoveries were 102.69% (RSD = 1.64%, n = 9), 100.49% (RSD = 1.93%, n = 9), and 100.39% (RSD = 1.65%, n = 9). Conclusion: The method is simple, reproducible, and accurate, and reliable measurement results can be used for simultaneous determination of luteoloside, apigenin-7-O-β-D- glucosidase, and luteolin in H. scandens female and male plants from different origins and different harvest months in Hebei province. There is greater difference between luteoloside, apigenin-7-O-β-D-glucosidase, and luteolin in H. scandens female and male plants from different origins and different harvest months in Hebei province.
3. Comparative study on HPLC characteristic spectra of Humuli Scandentis Herba from different habitats in Hebei province

Objective: To establish and compare the HPLC characteristic spectra of Humuli Scandentis Herba form different habitats in Hebei province. Methods: HPLC was performed on Agilent 5 TC-C18 (250 mm × 4.6 mm, 5 μm) column, with the mobile phase of acetonitrile -0.2% phosphoric acid at flow rate of 1.0 mL/min; Detection wavelength was 340 nm; The column temperature was 25 ℃ and the sample size was 10 μL. Eleven batches of Humuli Scandentis Herba samples form different habitats in Hebei province were determined and the characteristic spectra of those were established. The fingerprint evaluation software (2004 edition) for Chinese materia medica (CMM) was used to evaluate the similarity of the 11 batches of samples. Results: There were nine characteristic peaks identified in the characteristic spectra of Humuli Scandentis Herba samples. Peak 7 was luteoloside and Peak 8 was apigenin-7-O-β-D-glucosidase. The similarities of seven batches of Humuli Scandentis Herba samples were proved to be higher than 0.900 and four batches of them were proved to be less than 0.900. Conclusion: The method is simple, accurate, and reproducible, and can provide the scientific evidence for controlling the internal quality standards effectively.
4.Effect of Comprehensive Therapy on Forearm Extensor Myotenositis: 72 Cases Report

ObjectiveTo observe the effect of comprehensive therapy on forearm extensor myotenositis.Methods72 cases were divided into two groups: a control group of 36 cases who were given routine treatment,and an experiment group of 36 cases who were given thermotherapy,computerized medium-frequency electrotherapy,physiotherapy,and ADL instruction,etc.After two courses,a simple grading score(for forearms) was used to assess the effect.ResultsOf the control group,22 cases were cured,10 remarkably effective,4 effective;of the experiment,30 cured,4 remarkably effective,2 effective(u=2.04, P<0.05).The difference of average score for forearms before and after the treatment were(6.58±3.17) points for the control and(8.19±3.55) for the experiment(t=2.03,P<0.05).The average days of cure were(5.60±2.54) d for the experiment group,shorter than those for the control(7.00±2.27) d(t=2.05,P<0.05).ConclusionComprehensive therapy is effective on forearm extensor myotenositis.

Objective To promote the idea and practice of truthful conduct of research among fellow researchers by creating a research integrity training mode for Chinese postgraduates.Methods Based on the RCR (Responsible Conduct of Research) training module,a common research education initiative adopted by many American research institutes,and through literature review and social science study,this paper explored the idea of develop a research integrity training mode for Chinese postgraduates.Results A thorough explanation and analysis was given to the five major aspect of research integrity training mode,including theoretical basis,training objectives,operational procedure,neces sary conditions for application and training evaluation.Conclusion Research integrity training is one of the most important contents of graduate education,and a well designed training mode is the first and foremost step of developing a responsible and credible environment in academia of China.
6.Prognostic value of CDKN2A mRNA level in glioblastoma

Objective To investigate the prognostic value of CDKN2A mRNA in glioblastoma (GBM).Methods CDKN2A gene mRNA data were obtained from three different GBM database online (TCGA,REMBARNDT and GSE16011).The correlations between overall survival (OS) and CDKN2A expression were analyzed by Kaplan-Meier method and multivariate Cox regression analysis.Results In the TCGA database (n =358),patients with high CDKN2A mRNA level got longer OS than those with low expression level [median OS:18.0 months (95 % CI 15.0-21.0 months) vs 13.9 months (95 % CI 12.4-15.4 months),P =0.001].In another two validation datasets,patients with high CDKN2A mRNA level had longer OS than those with low expression level [median OS in REMBRANT:16.6 months (95 % CI 13.3-19.8 months) vs 11.8 months (95 % CI 7.3-16.4 months),P =0.019; in GSE16011:11.9 months (95 % CI 8.3-15.6 months) vs 8.4 months (95 % CI 6.2-10.5 months),P =0.005].CDKN2A mRNA level was an independent prognostic factor for GBM.The combination of CDKN2A mRNA expression with MGMT promoter methylation status or G-CIMP status/IDH1 mutations provided an optimized prognostic factor in GBM patients.Conclusion The CDKN2A mRNA has prognostic value in GBM patients,which provided an optimized stratification strategy based on multiple biomarkers.
7.Immunological activity change of regulatory T cells and its relation with outcome of patients with severe burn

Objective To investigate the immunological activity change of regulatory T cells (Treg) and discuss its significance in the outcomes of patients with multiple organ dysfunction syndrome (MODS) and severe burn. Methods A total of 106 patients with total burn surface area (TBSA) larger than 30% were included in the study and randomly divided into three groups according to the burn area: Group Ⅰ (TBSA of 30%-49%, n = 41), Group Ⅱ (TBSA of 50% -69%, n = 34) and Group Ⅲ (TBSA of 70%-99%, n = 31). According to the development of MODS, patients were divided into MODS group (n =21) and non-MODS group (n =85). The patients with MODS were further divided into non-survival group (n = 16) and survival group (n = 5) based on their outcomes. Healthy volunteers were served as normal control (n = 25). Peripheral blood samples were collected at days 1,3,7, 14 and 21 postburn. The immunomagnetic separation technique was applied to separate and purify CD4+ CD25+Tregs in peripheral blood, and phenotypes (CTLA-4) were analyzed by flow cytometry and the contents of interleukin-10 released in the supernatants were determined by ELISA. Results Expression of CTLA-4 and level of IL-10 were significantly increased in burn patients compared with normal control group, with statistical differences. The expression of CTLA-4 and level of IL-10 were significantly increased in patients with severe burns at all time points. The expression of CTLA-4 and level of IL-10 in MODS group were much higher than those in non-MODS group at days 3-21 postburn (P < 0.01). Among the MODS patients, the expression of CTLA-4 and level of IL-10 in the survival group were obviously lower than those in the non-survival group at days 3-21 postburn (P < 0.05 or P < 0.01). Conclusions After severe burn injury, expressions of the markers on CD4 + CD25 + Treg surface and secretion of cytokines produced by CD4 + CD25 + Tregs show significant difference in patients with different born areas, MODS development and survival state. CD4 + CD25 + Treg may play an important role in the pathogenesis of immunoregulation, MODS and mortality of burn patients through secretion of inhibitory cytokines.
8.Effect of miR-21 on albuminuria in db/db mouse with early stage of diabetic nephropathy

Objective To explore the effect of miR-21 on 24 h urine albumin excretion (UAE), glomeruli morphology, and its molecular mechanism in db/db mouse with early stage of diabetic nephropathy. Methods Twenty-four 5-week-old male mice were randomized into 4 groups: control group (untreated db/m mice, n=6),miR-21-treated db/db group (n=6), control empty plasmid treated db/db group (n =6), and untreated db/db group (n = 6). Mice were injected intraperitoneally using a hydrodynamics-based procedure with plasmids (30 mg · kg-1 · d-1 of miR-21 or 30 mg· kg-1 · d-1 of control plasmid) until albuminuria was detected in the untreated db/db mice. The glomeruli were observed under the light microscope, and measured by the software in the computer. The expression of miR-21 was tested by real-time RT-PCR, the expression of PTEN, p-Akt (Ser 473),and phosphatidylinositol-3-kinase (PI3K) p85α protein levels were examined by western blot and immunohistochemistry. Results Male db/db mice of 9-week-old with hyperglycemia and significant elevation of urinary albumin excretion showed features similar to the early stage of diabetic nephropathy. Real-time RT-PCR showed that miR-21 was increased in the miR-21-treated group. By measurement of glomeruli acreage, the glomeruli were smaller in miR-21-treated group than in the control empty plasmid treated and untreated groups (all P<0.01). By Western blot and immunohistochemistry, PTEN were reduced in the miR-21-treated group, whereas PI3K p85α and phospho-Akt (Ser 473 ) were increased in miR-21-treated group (all P < 0.01). Conclusions miR-21 ameliorates glomerular hypertrophy in db/db mice with diabetic nephropathy, and it may be a novel protecting factor of diabetic nephropathy.
9.Clinical study on the second surgical procedure of adjacent segment degeneration after the first time spinal fusion

Objective To discuss the effectiveness of the second surgical procedure of adjacent segment degeneration after the first spinal fusion.Method 35 patients who had been performed spinal fusion in our hospital or had symptoms recurred or aggravated after 12 ~ 114 (42 ±35) months of the prior surgery were enrolled in this study.A second surgery was performed and intraoperative the intradiscal pressure of adjacent segments of degeneration and normal segments was measured.The VAS score systems were compared among prior surgery,3 months later and 2 years after the second surgery.Result The intradiscal pressure of adjacent segments after the cervical vertebra and lumbar vertebra fusion were [ (15 ± 4.6)cmH2O,(23 ±5.2)cmH2O],much higher than normal segments [ (3 ±2.3)cmH2O,(8 ±4.1)cmH2O](P <0.01).The VAS score systems of 3 months later and 2 years after the second surgery were [ (2.9 ±0.7),(2.0 ± 0.6) ],which were dramatically lower than the prior (7.8 ± 1.2) (P < 0.01).In 12 ~ 46(31 ± 12) months of follow-up after the second procedure,X-ray and MRI showed that fusion segments reached nearly bone fusion,well decompressed and without nerve compression or other complications.Conclusion If recurrence of symptoms after spinal fusion were caused by adjacent segment degeneration,reoperation would guarantee good clinical outcome.
10.Preliminary clinical study on contrast-enhanced ultrasound for the peripheral pulmonary tumors

Objective To study the feasibility and characteristic findings of contrast-enhancedultrasound(CEUS)on peripheral pulmonary tumors.Methods Twenty patients with peripheral pulmonarytumors proven by pathology were studied.They were divided into two groups:primary pulmonary tumors (n=15),and metastatic pulmonary tumors(n=5). The dynamic enhancement images were stored and Strip enhancement was dominant in the primary pulmonary tumors,while dot enhancement was dominant in ascended quickly and descended slowly,while ascended slowly and descended slowly in majority of metastaticThe modes of enhancement and time-intensity curves were all different in primary and metastatic pulmonary tumors.Contrast-enhanced uhrasound is promising in the diagnosis of peripheral pulmonary tumors.
