Objective In this study, we tested the ability of platelet-rich plasma to enhance bone consolidation during distraction osteogenesis in a rabbit model of leg lengthening. Methods Twenty New Zealand white rabbits, which were randomly divided into two groups, the PRP treated group and the control group (n=10). Osteotomy will be made under the tibiofibular junction of left tibia, and 1cm tibia shaft will be removed through a second osteotomy. The tibia will be shorted for 1cm and then fixed by Orthfix unilateral lengthener with 4 pins. Lengthening will start 7 days after the osteotomy surgery, at a rate of 1.0 mm/day, in two steps, for 10 days. 500ul PRP were respectively injected into the distraction gaps at the end of the 10-day lengthening and at 10 days before termination. No management in the control group. The animals were terminated at day 20 following final lengthening. Serial radiographs will be taken at the day of surgery, day 12, day 17, day 27, day 37, using a C-arm radiography system. The excised bone specimens were scanned by X-ray and Micro-CT, and the bone mineral content(BMC) and the bone mineral density(BMD) of the regenerates were calculated and compared. Results The regenerate of PRP treated group was significantly better than the control group. The X-ray figures showed that the regenerate was significantly more than the control group. The figures of micro-CT 3D reconstruction showed that the growth and remodel of the regenerate in PRP treated group was better than the control group. Objective To establilsh the quality standard for Qingfei Yihuo tablet. Methods TLC was performed to identify Sophora flavescens Ait, Peucedani and Cortex Phellodendri. HPLC was used to determine Baicalin. ODS column was used. The mobile phase consisted of methanol-water-phosphoric acid (47∶53∶0.2). The flow rate was 1.0 mL/min and column temperature was at 25 ℃. The detecting wave length was at 280 nm. Results The characteristic for identification by TLC was distinct and hightly specific. Baicalin could be determined by HPLC. The linearity of Baicalin was good in the range of 0.163～0.975 ?g (r=0.999 9). The average recovery of Baicalin was 97.43% and RSD was 1.97% (n=6). Conclusion The methods of identification and quantification is simple and reproducible. It can be used effectively for the quality control of Qingfei Yihuo table.

Objective To observe the expression of anti-cardiac myosin antibody(AMA) and the collagen volume fraction(CVF) in serum in mice with chronic viral myocarditis(CVMC),and to explore the preventive and protective function of oxymatrine on myocardium.Methods BALB/c mice(n=60) were infected with coxsackievirus B3(CVB3) of increased dose biweekly to establish CVMC model.Mice in normal control group(n=8) received equal-volume 9 g?L-1 saline without CVB3 at the same time.The surviving mice in CVMC model group were randomly divided into CVMC control group(n=8) and oxymatrine therapy group(n=8) at the 42th day.From then on,mice in oxymatrine therapy group were treated with oxymatrine at the dose of 100 mg?kg-1?d-1 by gastric perfusion once a day for 28 days,and meanwhile mice in CVMC control group and the normal control group received equal-volume 9 g?L-1 saline by gastric perfusion every day.Then all mice were sacrificed at the end of the experiment.The ratio between heart weight to body weight(HW/BW) was calculated.Myocardium slides were stained with collagen specific Picric acid-Sirius red staining,and the CVF was calculated with image analysis software.The serum level of AMA(optical density value,A value) was detected by enzyme-linked immunosorbent assay(ELISA).Results HW/BW(0.007 9?0.000 3),CVF [(15.30?1.08)%] and the A value of AMA(0.286?0.053) in mice of CVMC control group were increased significantly compared with those in normal control group HW/BW(0.005 5?0.000 2),CVF[(6.84?1.11)%],the A value of AMA(0.160?0.050)(Pa

OBJECTIVE:To provide reference for rational use of recombinant human granulocyte-colony stimulating factor (rhG-CSF) in cancer patients. METHODS:Referring to the expert advice and guideline of clinical application of rhG-CSF at home and abroad,DUE criteria on rhG-CSF for cancer patients was established;questionnaire was designed,and the medical re-cords of rhG-CSF in a third grade class A hospital was evaluated. RESULTS:The DUE criterion on rhG-CSF for cancer patients included 3 parts,such as drug indications,the course of medication and medication results,14 items in total. Through the imple-mentation of DUE,retrospective evaluation of 220 medical records showed that the application of rhG-CSF was up to the stan-dard in respects of drug indications,no contraindication,routine blood test and drug interaction monitoring at least twice every week,there still was irrational phenomenon. CONCLUSIONS:The established DUE criterion on rhG-CSF for cancer patients is with the characteristics of definite content,target and paracticabilitg,which can provide reference for the work development of clinical pharmacists.

Objective:To establish the quality control methods for Xinmeng Anshen granule. Methods:TLC was used to identify Salviae Miltiorrhizae Radix Et Rhizoma,Mori Fructus, Ziziphi Spinosae Semen( fried) ,Arachis Hypogaea L. and Schisandrae Chinensis Fructus in the granules. HPLC was used to determine the content of protocatechuic aldehyde in Salviae Miltiorrhizae Radix Et Rhizoma and spinosin in Ziziphi Spinosae Semen. Results:The TLC spots were clear and well-separated without any interference from the nega-tive sample. The calibration curves were linear within the range of 3.972-198.600 μg·ml-1(r=0.999 9) for protocatechuic alde-hyde and 7. 070-226. 240 μg·ml-1(r=0. 999 9) for spinosin. The average recovery was 98. 46% (RSD=1. 18%, n=9) for proto-catechuic aldehyde and 98. 20% (RSD=0. 90%, n=9) for spinosin. Conclusion:The established quality control methods are accu-rate, simple, repeatable and specific. It can be well used for the quality control of Xinmeng Anshen granule.

The high expression of ABCA1 and SR-BI/CLA-1 can decrease the danger of atherosclerosis. The purpose of the study is to find ABCA1 and CLA-1up-regulators for treating atherosclerosis by using cell-based high throughput screening models. In conclusion, E0869 could up-regulate ABCA1 and CLA-1 activity, and had good anti-atherosclerotic activity in vitro.

Objective To investigate whether the combination of hydroxyapatite/tri-calcium phosphase (HA/TCP) and distraction osteogenesis technique would greatly reduce the time for the treatment of bone defects and enhance bone consolidation.Methods Osteotomy was made in the left tibia of all the 36 rabbits (age 20-24 weeks,body weight 2.2-2.8 kg),and a 1.0 cm length of the tibial shaft was removed below the tibiofibular junction.All rabbits were randomly divided into 3 groups,12 per group.Group A:The 1.0 cm defect gap was immediately reduced with the tibia shortened for 1.0 cm.Group B:The 1.0 cm defect gap was immediately filled with 1.0 cm restorable porous HA/TCP cylindrical block.Group C:The 1.0 cm defect gap was immediately reduced with the tibia shortened for 0.5 cm,and the remaining 0.5 cm defect gap was filled with 0.5 cm restorable porous HA/TCP cylindrical block.Then all the tibia was fixed with unilateral lengthener.For Group A and Group C,the lengthening started on the seventh day postoperatively,and lasted for 10 days and 5 days respectively.Group B didn't perform lengthening.All the rabbits wore terminated on the 37th days postoperatively.Serial radiographs were taken on the day of surgery,12,17,27 and 37 d postoperatively.The excised bone specimens were subject to micro-CT,mechanical testing,and histological examinations.Results Compared with the other two groups,the results in Group C were better in the terms of bone mineral content 454.44 ±89.98 mg,tissue mineral content 454.40±89.97 mg,maximum torque,maturity of regenerate bone,and the speed of bone consolidation and remodeling.Complete bone healing was achieved in the Group C within 37 days,but not in the Group A and B within 37 days.Conclusion The combination of HA/TCP and distraction osteogenesis technique can reduce the treatment time and promote bone consolidation compared with single treatment.

The purpose of this study is to investigate the intracellular transporters effect and the cytotoxicity of carboxyl nanodiamond (CND) - podophyllotoxin (PPT). Nanodiamond (ND) was treated with mixed carboxylic acid and finally got 64 nm CND by centrifugation, and then it was reacted with PPT to form CND-PPT. UV spectrophotometry was used to calculate the content of PPT in CND-PPT, the particle size distribution and zeta potential were measured by Dynamic laser scattering instrument. CND, PPT, CND-PPT and CND + PPT (physical mixture of CND and PPT) were characterized by Fourier transform infrared spectroscopy, at the same time, thermal analysis and element analysis were used to estimate the content of the PPT in CND-PPT. The affect of CND, PPT, CND-PPT on HeLa cell was measured with MTT assay. The results showed that content of PPT combined with CND accounted for about 10%. MTT assay showed that CND has low cytotoxicity and CND-PPT can increase the water soluble of PPT. As a conclusion, CND as a hydrophilic pharmaceutical carrier combined with PPT is able to increase the water solubility of PPT, at low concentration, CND-PPT can enhance the antitumor activity in comparison with PPT, so CND can be used as a potential anticancer drug carrier.
