1.Effect of linoleic acid on lipopolysaccharide-induced release of inflammatory factors in macrophages of mice

Objective To evaluate the effect of linoleic acid on lipopolysaccharide (LPS)-induced release of inflammatory factors in the macrophages of mice.Methods The peritoneal macrophages obtained from C57BL/C mice were seeded in 24-well plates at a density of 4× 105 cells/well and in 6-well plates at a density of 2× 106cells/well.The cells were incubated and attached to the wall overnight in a 5% CO2 incubator in humidity at 37 ℃.The experiment was performed in 2 parts.Part Ⅰ The cells in 24-well plates were randomly divided into 5 groups (n =8 each) using a random number table:control group (group C);LPS group;3 different concentrations of linoleic acid groups (LA1-3 groups).The sterile anhydrous alcohol 1 μl was added in group LPS,0.1,0.5 and 1.0 mol/ml linoleic acid 1 μl were added in LA1-3 groups,respectively,and 30 min later 100 μg/ml LPS 1 μ,l was added in LPS and LA1-3 groups.The culture medium was collected at 6 h after LPS administration to measure the concentrations of tumor necrosis factor-alpha (TNF-α) and interleukin-6 (IL-6) in the supernatant by enzyme-linked immunosorbent assay.PartⅡ The cells in 6-well plates were randomly divided into 3 groups (n =6 each) using a random number table:control group (group C);LPS group;0.5 mol/ml linoleic acid group (group LA).The sterile anhydrous alcohol 1 μl was added in group LPS,0.5 mol/ml linoleic acid 1 μl was added in group LA,and 30 min later 100 μg/ml LPS 1 μl was added in LPS and LA groups.At 1 h after administration of LPS,the expression of Toll-like receptor 4 (TLR4) was determined by flow cytometry,and the expression of phosphorylated nuclear factor kappa B (NF-κB) p65 (p-NF-κB p65),phosphorylated extracellular signal-regulated protein kinase (p-ERK) and phosphorylated p38 mitogen-activated protein kinase (p-p38 MAPK) was determined by Western blot.Results Part Ⅰ Compared with group C,TNF-α and IL-6 concentrations in the supernatant were significantly increased in LPS and LA1 3 groups (P < 0.05).Compared with group LPS,TNF-α and IL-6 concentrations in the supernatant were significantly decreased in LA1 3 groups (P<0.05).Compared with group LA1,TNF-α and IL-6 concentrations in the supernatant were significantly decreased in LA2 and LA3 groups (P<0.05).Compared with group LA2,TNF-α and IL-6 concentrations in the supernatant were significantly decreased in group LA3 (P < 0.05).Part Ⅱ Compared with group C,the expression of TLR4,p-NF-κB p65,p-ERK and p-p38 MAPK in macrophages was significantly up-regulated in LPS and LA groups (P<0.05).Compared with group LPS,the expression of TLR4,p-NF-κB p65,p-ERK and p-p38 MAPK in macrophages was significantly down-regulated in group LA (P<0.05).Conclusion Linoleic acid can inhibit LPS-induced release of inflammatory factors in the macrophages of mice,and the mechanism may be related to the inhibition of TLR4 signaling pathway activation.
2.Synthesis and identification of artificial antigens of paneoniflorin.

Oxidation method with sodium iodide was used to synthesize immunogenic antigen (PF-BSA) and coating antigen (PF-OVA) of paeoniflorin. UV spectroscopy showed that paeoniflorin was successfully conjugated with BSA and OVA. After immunized by PF-BSA, the mice can produce anti-paeoniflorin antibodies specifically. The ELISA test results showed the high titer (1:12 800) and specificity (IC50 = 0.791 mg x L(-1)) of the antiserum from mice injected with PF-BSA. Also, the antiserum showed low cross activities against nine traditional Chinese medicine (TCM) of small molecules. These artificial antigens were successfully synthesized and the anti-paeoniflorin antibody well prepared, which provides the experimental basis for the further study of ELISA and its kit.

3.Effects of the Culture Method on the Construction of Dermal Substitutes in vitro

Culture environment is the key factor in the construction of dermal skin.It was investigated that the effects of the culture methods,including the static culture and spinner flask culture,and stir speeds on the cells proliferation,metabolism and distribution within collagenchitosan sponges.A higher cell density and specific growth rate was obtained with spinner flask culture versus static culture,especially,the 80 r/min spinner flask culture.The cell distribution in dermal substitutes from stirred culture system was more uniform than static culture,as well as that with increase of stir speeds in spinner flask.In summary,the spinner flasks culture with proper stir speed shows promise for the construction of dermal substitutes in vitro.
4.Mechanism of Chromium Accumulation by Fused Yeast

The mechanism of chromium accumulation and microstructure transformation of the fused yeast were studied in this paper.The result showed that the process of Cr6+ reduction and adsorption was accom-panied by the H+ consumption.The main adsorptive groups on the strain surface included amino,hydroxyl,phosphate group and amide,among which phosphate group played vital role in the chromium accumulation.The removal rate of chromium and reduction rate of hexavalent chromium declined 70% and 46%,respec-tively,when phosphate group was masked.During the adsorption process the chromium ions complexed on the surface of fused yeast was transported into the cell wall and combined with inclusion to form steady spe-cies and this took 90 min to reach the equilibrium.The biosorption and reduction of Cr on the cell surface would alter microstructure of cell surface,reduce cell membrane potential and increase cell membrane per-meability.
5.Impacts of different nutrition support methods on short-term outcome in critically ill adult patients

Objective To explore the impacts of different nutrition support methods on short-term outcome in critically ill adult patients. Methods We retrospectively reviewed the clinical data of 1503 critically ill adult patients who needed nutrition support in an mixed intensive care unit (ICU) of a tertiary care university hospital from January 1994 to December 2009. The complication of nutrition support, length of stay (LOS) in ICU, morbidity of infection and multiple organ dysfunction syndrome (MODS), and mortality among different nutrition support methods were compared. Results The Nutritional Risk Screening (NRS) scores of the enrolled patients were no less than 3. LOS in early enteral nutrition (EN) patients were significantly shorter than those in not-early EN patients (P < 0. 001) and parenteral nutrition (PN) patients (both P < 0. 001). Infection rate (P < 0. 001),morbidity of MODS (P < 0. 001) and mortality (P < 0. 001) were significantly lower than those in not-early EN and PN patients. LOS of patients receiving PN rich in glutamine (Gln) was significantly shorter than that in conventional formula (P = 0. 0000). Morbidity of infection (P= 0. 0252) and MODS (P = 0. 0030), mortality (P =0. 0305) were significantly lower than that of conventional patients. LOS of patients receiving intensive insulin therapy (IIT) was significantly shorter than that of controlled group (P = 0. 0000). Morbidity of infection (P =0. 0001) and MODS (P = 0. 0237) and mortality (P =0. 0427) were significandy lower than those of controlled group. Conclusions Nutritional risk is prevalent among critically ill adult patients. Early EN, PN of rich in Gln,and receiving IIT can shorten LOS, decrease morbidity of infection and MODS, and improve prognosis among these patients.
6.Preparation and evaluation of novel mesoporous molecular sieve of baicalin surface molecularly imprinted polymers.

Taking mesoporous molecular sieve MCM-41 as a substrate, baicalin (BA) as template, acrylamide (AM) as the functional monomer, ethylene glycol dimethacrylate (EGDMA) as a cross-linking agent, ethanol as solvent, under thermal polymerization initiator of azobis isobutyronitrilo (AIBN) , a kind of selective recognition of baicalin surface molecularly imprinted polymer was synthesized. The surface morphologies and characteristics of the MIPs were characterized by infrared spectroscopy (IR) and transmission electron microscope (TEM). The adsorption properties of polymer microsphere for the template were tested by the dynamic adsorption equilibrium experiments and static adsorption equilibrium experiments. The experiment showed that the imprinting process was successfully and the well-ordered one-dimensional pore structure of MCM-41 was still preserved. Furthermore, molecularly imprinted polymers had higher selective ability for BA, then provided a new method for the efficient separation and enrichment of baicalin active ingredients from medicinal plants Scutellaria baicalensis.

7.Value of APACHEⅡfor predicting the nursing workload in EICU

Objective To investigate the predictive value of acute physiology and chronic health evaluation Ⅱ(APACHE Ⅱ) for nursing workload in emergency intensive care unit (EICU).Methods A retrospective cohort study was conducted. Critical patients except for trauma admitted to EICU of West China Hospital of Sichuan University from June 2012 to July 2016 were enrolled. APACHE Ⅱ score and therapeutic intervention scoring system (TISS-28) score were calculated in each case. The relationship between APACHE Ⅱ score and TISS-28 score was analyzed by correlation analysis and curve estimation.Results A total of 1148 patients were enrolled, including 689 male (60.0%), and age of (56±19) years old, APACHE Ⅱ score was 19 (13, 25), and TISS score was 19 (13, 25). The Spearman correlation coefficient between APACHE Ⅱ score and TISS-28 score was 0.849 (P < 0.001), and that among the linear model quadratic model, composite model, growth model, logarithm model, cubic model and exponential modelR2 were 0.669, 0.710, 0.792, 0.764, 0.765, 0.765, respectively (allP < 0.01), and the most suitable description for relationship between APACHE Ⅱ score and TISS-28 score curve was cubic model. The nursing workload was significantly increased when APACHE Ⅱ score ≥30, and it was decreased when APACHE Ⅱ score < 30.Conclusion APACHE Ⅱ was a useful score for predicting the nursing workload in EICU.
8.Comparison of the value of LDL-C and non-HDL-C in differential diagnosis of familial hypertriglyceridemia and familial combined hyperlipidemia

Objective To evaluate the value of low-density lipoprotein-cholesterol (LDL-C) and non-high-density lipoprotein-cholesterol (non-HDL-C) in differential diagnosis of familial hypertriglyceridemia (FHTG) and familial combined hyperlipidemia (FCHL).Methods We recruited 9 FHTG pedigrees (94 subjects) and 24 FCHL pedigrees (94 subjects) and then divided them into affected groups and non-affected groups according to lipid abnormality.Another 10 normal control pedigrees (57 subjects) served as controls.We compared the routine lipid levels such as triglyceride (TAG),total cholesterol (TC),HDL-C and LDL-C and non-HDL-C between the groups.After stratification based on TAG level,we observed the relationship between LDL-C and non-HDL-C.Last we confirmed and analyzed the cut-off value of differential diagnosis between FHTG and FCHL with receiver operating characteristic (ROC) curve.Results The levels of TAG,TC,and non-HDL-C were significantly higher in the affected group of FHTG than in the non-affected group of FHTG and the normal group (P<0.01 or P<0.05).The levels of TAG,TC,HDL-C,LDL-C and non-tHDL-C wcrc significantly higher in the affected group of FCHL than in the non-affected group of FCHL and the normal group (P<0.01 or P<0.05).The levels of TAG were significantly higher (P<0.01) while TC,HDL-C,LDL-C and non-HDL-C levels were significantly lower (P< 0.01 or P<0.05) in the affected group of FHTG than in the affected group of FCHL.The association between LDL-C and non-HDL-C was positive both in FHTG and FCHL,but the relationship became weaker as TAG level increased.The cut-off value of LDL-C and non-HDL-C was 3.575 mmol/L and 4.525 mmol/L,respectively.Conclusion In addition to the routinely used lipid indexes,non-HDL-C may be a new index for differential diagnosis of FHTG and FCHL,and may be superior to LDL-C in this regard.
9.Liver Function of Lymphoma Patients with Chronic HBV Infection after Chemotherapy

Objective. To observe the effect of chemotherapy on hepatic function of lymphoma patients with chronic HBV infection. Methods; We used ELISA to detect the serum markers, of HBV and liver function in 207 lymphoma patients and 207 patients with other types of cancer (except pdmary hepatocellular cacinoma). Results: The incidence of HBV infection was higher in lymphoma cancer cases than that in the controlled cases (19.8% vs 9.7%, P=0.004). The incidence of abnormal liver function was higher in lymphoma patients with positive HBsAg than in lymphoma patients without HBsAg (58.5% vs 27.7%, P=0.000). The incidence of ab-normal liver function in lymphoma patients with postive HBsAg was higher than that in patients with other types of cancer with positive HBsAg (58.5 vs 30.0%, P=0.036). The abnormal liver function in lymphoma patients after chemotherapy was associated with HBV infection (P=0.000) but not correlated with age, sex, histological subtype, immune subtype, stage, ECOG PS, and hormone administration. Conclusion: Lymphoma patients with HBV are more likely to have liver function damage after emotherapy.
10.Investigation on malaria knowledge and demands on related training for CDC staff in Qinghai Province,China

Objective To investigate the malaria knowledge of CDC staff and their demands on related training in malaria non⁃endemic areas,so as to provide the reference for planning the appropriate curriculum. Methods All the participants who were the staff of county CDCs all over Qinghai Province and attended the provincial training workshop were surveyed. A self⁃administered questionnaire survey was carried out and the data was statistically analyzed. Results A total of 115 participants were involved in this survey. They were mostly(85.21%)from county CDCs. The general knowledge of malaria among the respondents was well,and the average rate of correct answers was 70.35%. However,the answers to the general knowledge of malaria and anti⁃malaria treatment were not well enough. The rates of correct answers were 61.96% and 48.99% respectively. The differences among the groups of job title ranking,department of working and level of CDC were not significant(F = 0.13-2.02,all P > 0.05). The number of correct answers was significantly increased after the training course. The average score after the training was 79.20±15.16 while the pre⁃training score was 70.34±17.46(t = 3.86,P < 0.05),especially in the answers to general malaria knowledge and malaria surveillance and response(t = 4.30,4.97,both P < 0.05). The general knowledge of malaria was considered as the most need of training as 80% of the respondents voted“Yes”,according to the demand analysis. There was no significant difference among the different groups(F = 0.61-3.11,both P > 0.05). Conclusion The malaria knowledge is well mastered by the staff of CDCs in Qinghai Province,and the further training courses are requested and addressed in the target areas such as general malaria knowledge,anti⁃malaria treatment,malaria surveillance and response.
