Objective To investigate the ratios and the distribution of subjects with indeterminate results of HIV antibody in different populations who were positive for HIV screening test in Fujian and to ana-lyze the features of Western blot bands for further understanding the relationships between the Western blot band patterns and HIV infection. Methods The subjects who were diagnosed with indeterminate results of HIV antibody at the first examination were re-tested with Western blot assay every month until the results turned into HIV antibody-positive or HIV antibody-negative. Results The detection rate of indeterminate results of HIV antibody was 3. 69% in Fujian in 2015. Higher rates were detected in blood donors, pregnant woman and other clinical patients. The most common band patterns of HIV antibody-indeterminate samples were p24, gp160+p24 and gp160. Results of the follow-up confirmatory tests showed that 76% of the sam-ples of p24 band pattern were HIV antibody-negative and 76. 47% of the samples of gp160+p24 band pattern were HIV antibody-positive. The nucleic acid test showed that positive results were detected in 29 samples whose band patterns were gp160+gp120+p24,gp160+gp120+p66andgp160+gp120+p31. Conclusion The detection rates of indeterminate results of HIV antibody varied in different populations. The most com-mon band patterns of HIV antibody-indeterminate samples were p24, gp160+p24 and gp160. The more bands the samples presented themselves in Western blot assay, the more likely they would be HIV antibody-positive. Follow-up should be strengthened to prevent the spread of HIV infection. Samples with the band pattern of gp160+gp120+p24 showed a low probability of false positive and they were more likely to be end-stage AIDS with immunosuppression.

Journal of China Pharmaceutical University 2010;41(1):76-80
Aim: To explore the relationship between structure and immunological activity of marine polysaccha-ride YCP,the physicochemical property and immunological activity of the YCP-derived fragment were studied. Methods: YCP was hydrolyzed by α-amylase from human saliva. The hydrolysate was purified to obtain an polysaccharide fragment by gel filtration chromatography. The physicochemical properties of this YCP-derived fragment was characterized by HPLC,FT-IR and TLC.In addition, changes of phagocytic activity, production of reactive nitrogen and macrophage binding were investigated. Results: The relative molecular weight of YCP-de-rived fragment was approximately 6.6 × 10~3.The monosaccharide composition and FT-IR of the YCP-derived frag-ment were identical to YCP.No significant effect of the YCP-derived fragment on NO production and murine mac-rophage phagocyte were observed. And this fragment was not able to compete the binding between YCP and mac-rophages.Conclusion: The remarkable decrease of immunological activity of YCP-derived fragments degraded byα-amylase of human saliva suggests that the complete structure and high molecule weight of YCP are essential for its immuno-modulatory activity.

Objective · To investigate the effects of insulin on high glucose-cultured humanmononuclear cell line THP-1 and macrophage phenotype transformation in diabetic wounds. Methods · THP-1 cells were cultured with normal (5.6 mmol/L) and high (25 mmol/L) glucose, respectively, stimulated with PMA for differentiation, and induced to M1 macrophages with LPS. After treated with insulin for 6 h, expression changes of M1 type macrophage markers inducible nitric oxide synthase (iNOS), tumor necrosis factor α (TNF-α), and interleukin-1β (IL-1β), as well as M2 type macrophage markers arginase1 (Arg1) and IL-10 were detected using real-time PCR andWestern blotting. High fat diet feeding plus multiple intraperitoneal injections of low dose streptozotocin (STZ) were used to induce type II diabetes rat model. After blood glucose level has been stable for five weeks, two fullthickness skin wounds with the diameter of 1cm were made on the back of DM rats. Wounds were randomly assigned to being treated with insulin (0.2 U insulin /20 μL saline) or saline (20 μL saline) using the random number table. Characteristics of macrophagephenotypes were observed 3, 7, and 25days after wounds were made. Normal rats (n=3) served as controls. Results · After being cultured with high glucose, the mRNA levels of M1 markers iNOS and TNF-α were up-regulated in LPS-induced THP-1 cells, while the mRNA levels of M2 markers Arg1 and IL-10 were down-regulated.Afterbeing treated with insulin for 6 h, mRNA levels of iNOS and TNF-α weredown-regulated, protein levels of iNOS, IL-1β were down-regulated too, while mRNAand protein levels of Arg1 and IL-10 were up-regulated. In addition, the expression level of phosphorylated NF-κB-p65 was significantly increased after high glucose culture and was significantly decreased after insulin intervention. Compared to normal rat skin wounds, the expression of iNOS in macrophages was significantly increased in wounds of diabetic rats. The expression of iNOS in macrophages was high in saline treated wounds 3 and 7 days after the wounds were made and the expression of Arg1 was low 25 days after the wounds were made. In insulin treated wounds, the expression of iNOS started to decrease on day 7 after the wounds were made and the expression of Arg1 was significantly higher than that in saline treated wounds on day 25 after the wounds were made. Conclusion · Insulin can induce macrophage phenotype transformation from M1 to M2 under high glucose condition and the mechanism may be associated with the phosphorylation of NF-κB-p65.
