4.Pharmaceutical research progress of rhynchophylla based on chemical stability.

Rhynchophylla is a Chinese herb commonly used in clinical practice. It's also the primary herb of some famous Chinese herbal compound such as Tianma Gouteng decoction, and Lingyang Gouteng decoction. According the record from many previous materia medica literatures, rhynchophylla should be added later during decoction. Pharmaceutical research showed that rhynchophylla alkaloids were not stable. Which has resulted in many problems in the research and its application. For example, there was not a quantitative determination method in "Chinese Pharmacopoeia" of past and present versions, which seriously impacted its quality control and product application. Firstly, records from previous materia medica literatures and "Chinese Pharmacopoeia" were systematically sorted based on the chemical stability of rhynchophylla. Secondly, pharmaceutical research including chemical compositions and their stability, pharmacological effects, extraction process and quality analysis, was reviewed after reference of literatures published at home and abroad in recent decades. Positive reference and evidence for further research and development of rhynchophylla will be provided in the article.

5.The Development of Endogenous CO and its Mechanism in Asthma

Objective To explore the development of endogenous CO and its mechanisms in asthma.Methods 50 guinea pigs were equally separated 5 groups.After the models of asthmatic guinea pigs were established in 4 groups,3 groups of them were treated with dexamethasone,the specific stimulator(hemin)and inhibitor(Sn-PP)of HO-1 respectively.The rest of groups were asthmatic one and normal control one(NCG).From all tested animals,the COHb in blood and lung HO-1 activity were examined,the lung tissues were pathologically observed and immunohistochemically stained.Results In asthmatic and hemin stimulated groups,the levels of COHb with enhanced expression of lung HO-1 and their activity were significantly increased compared with NCG(t=4.43~9.97,P<0.01).For example,the lung HO-1 activities in asthmatic group were (881±361)pmol/(mg*pro*h),COHb (4.94±2.15)% respectively.In dexamethason prevented and Sn-PP inhibited groups,however,all determined levels were markedly reduced compared with asthmatic group(P<0.01).Conclusion The increased HO-1 had resulted in the growth of endogenous CO in asthma.
6.Misdiagnosis Analysis of 31 Cases with Anaplastic Large Cell Lymphoma in Children

Objective To make clinical doctors better understanding of anaplastic large cell lymphoma(ALCL) and reduce the misdiagnosis of ALCL at an earlier stage.Methods Retrospective analysis of clinical features in 31 children with ALCL from Sep.2002 to Jan.2008,who had been misdiagnosed as other diseases at first and latterly been confirmed as ALCL by pathological study.The reasons for mis-diagnosis and the symptoms of the disease were analyzed and reviewed.Results ALCL clinical misdiagnosis rate reached to 91.2%.The reasons for misdiagnosis were:1.The fact that extra-nodal involvement present in earlier stage,which caused ALCL to have diversified clinical symptoms,and the initial symptoms were diverse.2.ALCL cells produce multiple cytokines such as IL-6,IL-9,IL-4,and others.They might make patients have symptoms like inflammation.3.The morphology of ALCL was quite variable.4.The clinical doctors and pathologists did not have frofound understandings of ALCL.5.Inapropriate usage of steroid before diagnosis was mode.Conclusions Clinical doctors should be aware of the diversity of ALCL clinical symptoms and use steroid carefully,while pathologists should pay attention to morphological varieties of ALCL and choose appropriate immunohistiochemical stain markers to avoid misdiagnose.Pathologic diagnosis should be made by more than 2 oncology centers.
7.Study on Pharmacokinetics of Ferulic Acid in Ordinary Guipi Pills and Ultramicro Guipi Pills in Rabbits

OBJECTIVE: To study the pharmacokinetic parameter and the relative bioavailability of ferulic acid in ordinary Guipi pills and ultramicro Guipi pills in rabbit.METHODS: HPLC-MS/MS was used to identify the concentration of ferulic acid in the blood sample.RESULTS: Plasma concentration of ferulic acid in ordinary pills was different from that in ultramicro pills.The main pharmacokinetic parameters of ferulic acid in ordinary pills vs.that in ultramicro pills were as follows:t1/2a:(17.59?2.57) min vs.(23.51?4.49) min;t1/2?:(300.91?38.74) min vs.(167.17?45.66) min;tmax: (9.61?1.69) min vs.(12.33?2.27) min;Cmax:(58.92?8.42) ng?mL-1 vs.(112.35?31.29) ng?mL-1;AUC: (5 851.67?895.49) ng?min-1?mL-1 vs.(8 586.13?1 497.62) ng?min-1?mL-1.There were significant difference in Cmax and AUC between ordinary pills and ultramicro pills.CONCLUSION: The application of super fine crushing technique in the preparation of Guipi pills could improve the bioavailability of Guipi pills.

9.Cloning and Characterization of 5′Flanking Region of Nitrate Reductase Gene Derived from Dunaliella salina

Aim: Clone and characterize of the 5′- flanking region of the nitrate reductase (NR) gene derived from Dunaliella salina(D. salina). Methods : The genomic DNA from D. salina was respectively digested with BamHI, EcoRI, HindIII, Pst I, Sal I and Xba I. A genomic walking cassette was ligated to the ends of the digested DNA fragments, and then genomic walking libraries comprising BL, EL, HL, PL, SL and XL were constsucted. The 5′- flanking region of the NR gene from genomic walking libraries of D. salina was amplified by LA-PCR. The DNA sequences were analyzed with the software - Promoter Predictions. Isolated 5′-flanking regions fused to the GUS gene were tested for transient expression in the alga. Results: A single specific PCR product of about 1200bp in length from the HL library was generated. Also, several conserved motifs, such as CAAT-box, GAGA-box were found, which are related to regulation of transcription, and the putative binding sites of transcriptional factors such as EBP, EFII, NF-E1 and LV. BLAST showed that the DNA sequences shared high homology with 5′-upstream region of the NR gene from Dunaliella viridis. The isolated 5′-flanking regions were able to strongly drive GUS reporter gene expression, suggesting that it contains the promoter elements necessary for the transcription of the NR gene. The expression pattern of the GUS gene and the NR gene were similar, both ware induced by nitrate and repressed by ammonium. Conclusion: The cloned 5′- flanking sequences of NR gene derived from D. salina might be a specific promoter with the ability to“switch on or off” an expression of the heterologous gene in transgenic D. salina.
10.Study on index for formulation optimization of sustained or controlled released dosage forms.

It is not only the important indexes for evaluation of bioequivalence, also the important parameter of formulation optimization, study of the stability and quality control within producing of sustained or controlled release dosage forms. The review is the study on index for formulation optimization of sustained or controlled released dosege forms in China since 1999, including the application of similarity factor and deviation.
