1.Proteomic analysis of human ovarian cancer cell lines and their platinum-resistant clones

Objective To perform comparative proteomic analysis of human ovarian cancer cell lines for detecting platinum-resistance associated proteins.Methods The total proteins of two sensitive (SKOV3 and A2780)and four resistant(SKOV3/CDDP,SKOV3/CBP,A2780/CDDP and A2780/CBP) human ovarian cancer cell lines were separated by two-dimensional gel electrophoresis(2-DE).The differentially expressed proteins were analyzed using image analysis software,stained with Coomassie Brilliant Blue,then identified using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry(MALDI-TOF-MS)and database searching.The mRNA and protein levels of the differentially expressed protein which was most significant in all of the four resistant cell lines were validated by RT-PCR and western blotting,respectively.Results Five proteins were found to be significant in four cell lines. Annexin A3 and destrin were up-regulated and nicotinamide-adenine dinucleotide phosphate(NADP)- dependent isocitrate dehydrogenase 1 was down-regulated in all the four resistant samples.Glutathione transferase omega 1 had an increased expression in the other three resistant cell lines except for SKOV3/CBP in which its expression was not changed.However,cofilin 1 represented a different trend.In the two resistant sublines of SKOV3,eofilin 1 had a down-regulation,but it had an up-regulation in the cell lines induced from SKOV3.The expression of annexin A3 was up-regulated by 3-20 fold and the results of RT- PCR and western blotting showed complete consistency with that by 2-DE.Conclusions Proteomic techniques are useful to the identification of the resistance-associated proteins in ovarian cancer platinum- resistant cell lines and five candidates have been found.The five differential proteins might become hopeful candidate biomarkers for resistance.
2.The anti-itching and anti-hypersensitivity effects of Doxepin hydrochloride cream

Aim To study the anti-itching and anti-hypersensitivity effects of Doxepin hydrochloride cream. Methods The effects of Doxepin hydrochloride cream on the pruritus of skin in mice due to 4-AP and guineapigs due to histamine phosphate were observed. The inhibitory actions of it on the increase of abdominal cavity capillary permeability in mice due to acetic acid and the angiectasis of skin in rats due to histamine were also observed. Its effects on the delayed-type hypersensitivity (DTH) of mice due to 2, 4-dinitrochlorobenzol (DNCB) and the homologous passive anaphylaxis of skin in rats were used to investigate its anti-hypersensitivity actions. Results The application of Doxepin hydrochloride cream (50, 125, 250 mg?kg -1) on the skin of mice had significant inhibitory actions on the pruritus of skin caused by 4-AP, and the application of Doxepin hydrochloride cream (10, 25, 50 mg?kg -1) on the skin of guinea pigs also had significant inhibitory actions on the pruritus of skin caused by histamine phosphate. The application of Doxepin hydrochloride cream (10,25,50 mg?kg -1 and 50,125,250 mg?kg -1) on skin inhibited the increase of skin capillary permeability in rats caused by histamine phosphate and the increase of abdominal cavity capillary permeability in mice caused by acetic acid. The application of Doxepin hydrochloride cream (50, 125, 250 mg?kg -1 and 10, 25, 50 mg?kg -1) on skin also significantly inhibited the delayed-type hypersensitivity in mice due to 2, 4-dinitrochlorobenzol and the homogeneous cutaneous anaphylaxis in rats. Conclusions Doxepin hydrochloride cream can relieve itching,inhibit the increase of capillary permeability caused by inflammatory substances. It also has significant inhibitory actions on TypeⅠ and Type Ⅳ allergic reactions.
3.Perioperative nursing of patients with planted soft tissue expander after breast cancer surgery

Objective To explore the key points of perioperative nursing patients with planted soft tissue expander after breast cancer surgery.Method The clinical data of 55 patients with planted soft tissue expander after breast cancer surgery were reviewed to summarize the nursing measures.Result Operative process in 55 pattents were succesful,surgery time ranged from 3 to 5 hours.16 of 55 patients developed with complications and all of them were recovered and discharged.Conclusions Perioperative nursing intervention for the patients with planted soft tissue expander after breast cancer surgery can reduce the incidence of complications,improve the life quality and help them build up their confidence in social and family life.
4.Dexamethasone improves expression of basic fibroblast growth factor in neurons of spinal cord following sciatic nerve injury in rats

Objective: The regeneration and repair of peripheral nerves are focuses of neurological studies.The(bFGF) may play an important role in the development of the neural system.Dexamethasone is usually employed in the management of neural injuries.The aim of this study is to establish a rat model,and investigate the effect of Dexamethasone on the expression of bFGF in the neurons of the spinal cord following sciatic nerve injury.Methods: Fifty-six male Wistar rats were randomly divided into 4 groups: a nerve injury group,with sciatic nerve injury induced by clamping the nerve in the right;a dexamethasone group,immediately injected with dexamethasone [0.5 mg/(kg/d)] in the injury side;a saline group,given saline only;and a control group,rats without sciatic nerve injury.Changes of bFGF in the motor neurons of the L4-L6 spinal cords were observed by immunohistochemistry.Results: Dexamethasone obviously upregulated the expression of bFGF in the motor neurons of the spinal cord.Conclusion: Dexamethasone can improve the expression of bFGF in the motor neurons of the spinal cord after sciatic nerve injury,and contribute to the recovery of the function of the injured neurons.
5.Effects of Xiaoyingqiangji Decoction on Mitochondria in Skeletal Muscle of Hyperthyroid Myopathy

Objective To observe the effect of Xiaoyingqiangji decoction on mitochondria in skeletal muscle of hyperthyroid myopathy. Methods Healthy Wistar rats were randomly divided into 4 groups: the group of hyperthyroid myopathy were given L-thyroxine intraperitoneal injection for 42 d; Chinese medicine treated group were treated with Xiaoyingqiangji decoction for 30 d after L-thyroxine injection; the saline control group were given saline solution for 30 d after L-thyroxine injection; normal control group were given saline solution. Then the skeletal muscles were obtained and the succinate dehydrogenase (SDH) were measured. The ultrastructural changes were observed with TEM. Results The activity and content of SDH in the group of hyperthyroid myopathy decreased, and the cristae of mitochondria ruptured and even disappeared. After given Chinese medicine, the activity and content of SDH increased, while the mitochondria showed normal ultrastructural appearance. Conclusion Xiaoyingqiangji decoction can improve the function of skeletal muscles in hyperthyroid myopathy by recovering enzyme activity and mitochondria structure.
6.Effect of Xiaoyingqiangjitang on the Motor End-plate of Skeletal Muscle in Hyperthyroid Myopathy

Objective To observe the effect of Xiaoyingqiangjitang on the ultrastructure of motor end-plate of skeletal muscle in rats with hyperthyroid myopathy, and changes of acetylcholin esterase (AchE).Methods The 36 healthy female Wistar rats were randomly divided into the normal control group (n=6), and model group, Chinese medicine treated group and saline control group (all n=10). All animals in the model group, Chinese medicine treated group and saline control group were treated with group given L-thyroxine intraperitoneal injection for 42 days to establish the animal model of hyperthyroid myopathy. The animals of the Chinese medicine treated group were treated with Xiaoyngqiangjitang for 30 days after L-thyroxine intraperitoneal injection. The enzyme activity of AchE and ultrastructure of motor end-plate were observed by light and electron microscopes.Results The enzyme activity and content in the group of hyperthyroid myopathy was decreased, the reaction products of AchE located in synapse became weaker and distributed heterogeneously, the junction infolding of motor end-plate became smaller and shorter, the mitochondria in axon terminal became vacuolated. After given Chinese medicine, the enzyme activity and content increased, the junction folding, the mitochondria showed normal ultrastructural appearance.Conclusion Xiaoyingqiangjitang can improve the function of skeletal muscle in hyperthyroid myopathy by recovering the enzyme activity of AchE and ultrastructures of motor end-plate.
7.Effect of puerarin on myocardial perfusion and ventricular wall motion in patients with acute coronary syndrome

Objective To investigate the effects of puerarin(Pur)on myocardial perfusion and ventricular wall motion in patients withacute coronary syndrome(ACS).Methods Thirty-seven patients with ACS were randomly divided into two groups:conventionaltreatment group(n=17,11 males,range of age:32-80 years,average age:60.9±4.9 years)and Purtreatment group(n=20,12 males,rangeof age:40-76 years.average age:62.7±3.5 years).Patients in the conventional treatment group received standard treatment according tothe current guidelines,while patients in the Pur treatment group received intravenous administration of Pur(500 mg/day)for 10 daysplus conventional treatment.Real-time myocardial contrast echocardiography (RT-MCE) was performed to evaluate the change inmyocardial perfusion index (MPI)and veiltricular wall motion index(VWMI)at admission and 10 days after treatment.Results At10 days after treatment,MPI was significantly higher(P<0.01)and VWMI significantly lower(P<0.01)in the Pur group comparingwith those in the conventional group.Conclusions Puerarin might improve myocardial microcirculation perfusion and ventricularwall motion in patients with ACS.
8.Prognostic significance of natural killer cell infiltration in hepatocellular carcinoma

Background and Objective: Several studies have Shown the correlation of high numbers of tumor-infiltrating natural killer (NK) cells with a good prognosis for cancer patients.This study was to investigate the impact of NK cell infiltration on the survival and prognosis of patients with hepatocellular carcinoma (HCC)after resection. Methods: The proportion of infiltrating NK cells of HCC patients was measured using flow cytometry,and the expression of CD56~+ (NK) cells was investigated using immunohistochemistry. Prognostic values of intratumoral and peritumoral NK cell densities were evaluated by Kaplan-Meier method and Cox regression. Results: The level of NK cells was significantly lower in tumor-infiltrating lymphocytes (TIL) of HCC patients than in nontumor-infiltrating lymphocytes (NIL) [(11.8±8.1 )% vs. (18.0±7.9)%, P=0.002]. The density of NK cells was also significantly lower in cancer nests than in peritumoral lesions (2.3±2.6 vs. 8.5±4.5 cells per field, P<0.001).Patients with low intratumoral NK cells had shorter disease-free survival (P=0.027) and overall survival (P=0.005) than patients with high intratumoral NK cells. In contrast, NK cells in the peritumoral area showed no prognostic significance for either disease-free survival or overall survival. Multivariate Cox proportional hazards analysis showed that intratumoral NK cell density was an independent prognostic fator of prolonged overall survival (hazard ratio=2.658,P=0.019). Conclusion:Low NK cells infiltration could predict poor prognosis in patients with HCC.
9.Simultaneous determination of amlodipine, benazepril and benazeprilat in human plasma by LC-HESI/MS/MS method.

The study aims to develop a rapid, sensitive and specified method of liquid chromatography with heated electrospray ionization tandem mass spectrometry (LC-HESI/MS/MS) for simultaneous determination of amlodipine, benazepril and benazeprilat in human plasma using amlodipine-d4 and ubenimex as internal standards (ISs). Selected reaction monitoring (SRM) with heated electrospray ionization (HESI) was used in the positive mode for mass spectrometric detection. Analytes and ISs were extracted from plasma by simple protein precipitation. The reconstituted samples were chromatographed on a C18 (100 mm x 4.6 mm, 5 microm) column with mixture of methanol-acetonitrile-5 mmol.L- ammonium acetate-formic acid (30 : 30 : 40 : 0.1) as mobile phase at a flow rate of 0.6 mL.min-1. The standard curves were demonstrated to be linear in the range of 0.02 to 6.00 ng.mL-1 for amlodipine, 0.2 to 1,500 ng.mL-1 for benazepril and benazeprilat with r2>0.99 for each analyte. The lower limit of quantitation was identifiable and reproducible at 0.02, 0.2 and 0.2 ng mL-1 for amlodipine, benazepril and benazeprilat, respectively. The intra-day and inter-day precision and accuracy results were within the acceptable limit across all concentrations. The plasma samples were stable after four freeze-thaw cycles and being stored for 93 days at -20 degrees C. The method was applied to a pharmacokinetic study of a fixed-dose combination of amlodipine and benazepril on Chinese healthy volunteers.

10.Studies on chemical constituents of Zhuang medicine Excoecaria venenata and their cytotoxic activity.

Fourteen compounds were isolated from 95% ethanol extract by silica gel, MCI, and ODS column chromatography. These compounds were respectively identified as quercetin (1), kaempferol (2), (+)-catechin (3), fraxin (4), protocatechuic acid (5), gallic acid (6), methyl gallate (7), ethyl gallate (8), apocynol A (9), baccatin (10), cerevisterol (11), ellagic acid (12), 3, 3',4'-tri-0-methylellagic acid(13) and N-benzoyl-L-phenylalaninyl-N-benzoyl-L-phenylalaninate(14) by analyzing their spectral data and comparing with the previously reported literatures. Except for gallic acid (6), all other compounds were isolated from this plant for the first time. Compounds 1, 2 and 6 showed moderate anti-proliferation activities on tumor cells.
