To investigate the changes of immunological function of LAK,NK cell activity and T lymphoctyes subsets before and after operation in patients with hepatic cancer.The activity of Lymphokine-activated Killer Cell (LAK|,Natural Killer Cell(NK) and T lymphocytes subsets were detected in 31 patients with hepatic cancer of preoperation and postoperation,so did the 30 normal volunteers(as control group).The results showed that the activities of LAK,NK cell and T lymphocytes subsets of CD3,CD4 and the ratio of CD4/CD8 in patients with hepatic cancer were lower than that in normal control group(P

4.Effect of Overexpression of NAD+-dependent Malic Enzyme on Anaerobic Mixed Acid Fermentation of E.coli FMJ39

In order to investigate the effect of malic enzyme on anaerobic mixed acid fermentation,the gene encoding NAD+-dependent malic enzyme was amplified from E.coli DH5? genome by using PCR and cloned into vector pTrc99a to give an expression vector pTrc99a-sfcA.Malic enzyme was overproduced by E.coli FMJ39(ldh,pfl)harboring pTrc99a-sfcA under the condition of IPTG induction.The results of anaerobic fermentation indicated overexpression of malic enzyme would have influence on the pathway of formate,acetate and succinate of FMJ39.The concentration of formate and acetate was 17.58% and 15.27% higher than FMJ39,the succinate concentration was reduced by 26.87%.There was no obvious citrate concentration change.The experimental results also indicated that high concentration of L-Thr and L-Ser would induce the Tdc operon to convert pyruvate into formate and acetate even when pfl gene encoding the pyruvate formate lyase was inactivated.
7.Analysis of clinical characteristics of colorectal related multiple primary colorectal cancer

Objective To observe and compare the clinical characteristics of colorectal related multiple primary carcinoma (MPC) and colorectal carcinoma (CRC) for promoting early diagnosis and treatment of colorectal related MPC. Methods Pathological and clinical documents of clearly staged CRC and colorectal related MPC cases from Jul. 1997 to Nov. 2007 were retrospectively analyzed. Results 573 colorectal carcinoma cases were analyzed, including 45 MPC (7.85 %). Parenteral multiple carcinoma originated most frequently from stomach, and then breast, ovary, lung, small intestine and other sites. Among all multiple primary colorectal carcinomas (MPCC), ascending colon carcinoma was most frequent (34.0 %).While among CRC cases, rectal cancer cases was most frequent(36.5 %). Comparing CRC and MPCC, there were no significant difference in terms of tumor family history. Median morbidity age was 57 years and 63 years respectively. Cases with previous colonic polyps accounted for 20.0 % of all MPC cases, while only 0.9 % of all CRC cases. The mOS of CRC and MPC was 93.7 month and 64.8 month respectively. Most frequent pathological type of CRC and MPC were both well-moderately differentiated adenocarcinoma, but more mucinous adenocarcinoma cases were observed in MPC. Conclusion Colorectal related MPC are relatively common among colorectal carcinoma patients. More patients with MPC especially MPCC has colonic polyp. mOS of MPC is shorter than that of CRC, indicating the poor prognosis of MPC compared with CRC.MPCC has multiple colonic polyps, shorter interval of secondary carcinomas, and shorter mOS, worse prognosis than MPC with parenteral tumor.
8.Analysis of Survival and Functional Outcome after Nerve-Sparing Surgery with Extraperitoneal Lateral Lymphadenectomy for Lower Rectal Carcinoma

Objective To investigate the survival rate, voiding, sexual function after nerve-sparing surgery with extended systematic extraperitoneal lateral lymphadenectomy in lower rectal cancer. Methods Three hundreds and ninety-two cases with advanced lower rectal cancer who underwent nerve-sparing surgery with radical dissection from 1996 to 2000 were reviewed. Among them,173 cases only cleared in abdominal cavity,219 cases coupled with extraperitoneal lateral lymphadenectomy. 9.Application of human leukocyte antigen and cross-reactive group matching in clinical renal transplantation

Objective To investigate the application and significance of human leukocyte antigen (HLA) and cross reactive groups(CREGs) matching in clinical renal transplantation. Methods A total of 312 cases of kidney transplantation were divided into two groups.In one group of 149 cases of kidney transplantations,ClassⅠCREGs matching criteria were applied instead of conventional HLA A,B matching which had two A,B mismatches(MM).In the other group of 163 cases of kidney transplantation there were A,B 0~2MM by the conventional criteria.The graft survival rate at 1 year and incidence of acute rejection within 1 month after transplantation were compared between the 2 groups. Results The percentages of HLA Ⅰ antigens 0,1,2MM were 16.7%,41.6% and 34.2% by CREGs matching criteria,and were 6.7 %,21.5% and 71.8% by conventional matching criteria.The matching rates in CREGs 0,1MM group were significantly higher than those in corresponding conventional matching group ( P 0.05);however,the incidence rate of CREGs 0MM group was obviously lower than that of A,B 2MM group ( P 0.05 ),but the rate of CREGs 0MM group was obviously higher than that of A,B 2MM group ( P
10.Association between vascular endothelial growth factor + 936C/T gene polymorphism and age-related macular degeneration

Background Vascular endothelial growth factor(VEGF) has been shown to be associated with the pathogenesis of age-related macular degeneration (AMD),therefore VEGF is a target for the treatment of wet AMD.However,the mechanism of VEGF in the pathogenesis of AMD is not clearly understood.Studying the correlation between VEGF gene polymorphism and AMD is becoming a new research hotspot,but relevant studies on Han Chinese have not been performed.Objective This study was to investigate the association between the VEGF +936C/T gene polymorphism and AMD in the Chinese population.Methods A pilot prospective and nonrandomized controlled trial was designed.This protocol complied with Declaration of Helsinki and was approved by the Ethic Committee of Chinese PLA Second Artillery General Hospital.Written informed consent was obtained from each subject prior to entering the study.Two hundred AMD patients and 200 age-and gender-matched normal controls were enrolled in this study.The genomic DNA was extracted from the peripheral blood samples of the subjects,and analysis of the VEGF polymorphisms at the +936 position in the promoter and 3'-untranslated regions was performed by the restriction fragment length polymorphism method.Frequencies of the VEGF+936C/T genotype were compared between the two groups,and the risk of the VEGF+936C/T gene polymorphism in pre-disposing AMD was evaluated.Results No significant differences were seen in the incidence rates of smoking(P = 0.76),hypertension(P = 0.84),hyperlipidemia (P=0.71),diabetes mellitus (P=0.86) and cardiovascular disease(P=0.89) between the AMD group and the normal control group,and BMI was matched between the two groups (P =0.18).The prevalence of the TT genotype was 9.0% (18/200)in the AMD group,but that in the normal control was 3.5% (7/200),showing a significant difference between the two groups (P =0.03).The odds ratio (OR) was 2.73 with a 95% confidence interval(CI) of 1.11 to 6.68 for AMD in this genotype.The CC and CT genotypes were not significantly different between the two groups (P =0.52,P =0.57).The genotype frequency and allele frequency conformed to HardyWeinberg equilibrium law.There were no significant differences found in the CC,CT,TT genotype frequencies among the early AMD,geographic atrophy AMD and choroidal neovascular AMD (all at P>0.05).Conclusions The VEGF+936TT genotype is associated with AMD in Han Chinese population.
