1.Exploration of training mechanisms for young faculty's research capabilities

Young faculty are an essential part of a university's strength and is important for its sustainable development.And their scientific research capability is associated with the long-term development of the university.At present,there are 4 major problems with the development of young faculty's research capabilities,which include their relatively single academic knowledge structure,insufficient experience in writing proposals,insufficient opportunities for academic exchange,and inequity in resource allocation.Establishing a training mechanism may help solve some of the problems and promot their development.The research management department of a university must abandon erroneous traditional management concepts,provide research training seminars,develop a new mechanism to stimulate academic exchange,design a system for short-term laboratory rotations,and promote the participation of your faculty in major research projects.These may bring about marked improvement in the improvement of young faculty's research capabilities.
2.Efficacy Observation of Ultrasonic Conductometric Acupoint Penetration of Wenhua Zhitong Recipe for Adenomyosis Patients.

OBJECTIVETo observe the efficacy of ultrasonic conductometric acupoint penetration of Wenhua Zhitong Recipe (WZR) in treating adenomyosis.
METHODSTotally 40 adenomyosis patients with Shen-deficiency blood stasis syndrome were randomly assigned to the treatment group and the control group, 20 in each group. Patients in the treatment group received ultrasonic conductometric acupoint penetration of WZR from the 2nd day before menstruation to the end of menstruation, while those in the control group took WZR. All patients received 3 consecutive menstrual cycles. Endometrial contents of cyclooxygenase-2 (COX-2) and oxytocin receptor (OTR) were determined using SP immunohistochemical staining method before and after treatment. Clinical effect, efficacy of Chinese medical syndrome, changes of uterus volume, and contents of carbohydrate antigen 125 (CA125) were observed before and after treatment.
RESULTSThere was no statistical difference in the total clinical effect between the two groups (P = 0.357). Efficacies for relieving abdominal pain and lumbar soreness during menstruation were better in the treatment group than in the control group (P < 0.05). Uterus volume, endometrial levels and integrals of COX-2 and OTR decreased in the two groups after treatment (P < 0.05, P < 0.01).
CONCLUSIONUltrasonic conductometric acupoint penetration of WZR and oral administration of WZR both could effectively improve adenomyosis patients' symptoms such as Shen deficiency, and blood stasis, reduce the foci of uterus, and relieve the pain.

3.The relationship between the nerve growth factor level in tears in active thyroid-associated ophthalmopathy and tear film stability

Background Nerve growth factor (NGF) plays an important role in the maintenance of the tear film stability.It is well known that the damage of ocular surface causes dry eye in the patients with thyroid-associated ophthalmopathy (TAO).However,if NGF is helpful for the stability of tear film is unclear.Objective This study was to investigate the relationship of NGF concentration in tear with tear film stability in TAO patient before and after glucocorticoid treatment.Methods Prospective observational cohort study was designed.This study was approved by the Ethic Committee of Binzhou Medical College,and written informed consent was obtained from each subject before entering this trial.Thirty-eight patients with active TAO and thirty age-and gender-matched controls were enrolled in the study.Prednisolone acetate tablets were administered in TAO patients for 4 weeks at a back-titration regimen.The tear fluid was collected and NGF level was detected by ELISA.Clinical activity score (CAS) was evaluated under the slit lamp lamp.Schirmer Ⅰ test (S Ⅰ t) was performed and breakup time of tear film (BUT) was obtained in controls and patients with active TAO before and 4 weeks after steroid treatment.Results Baseline NGF levels in tear was higher in the patients with active TAO than in the controls (U=-5.026,P<0.01).NGF level decreased from 46.23 (42.34,51.42) ng/L to 27.45 (19.38,29.12) ng/L 4 weeks after steroid treatment (Z=-6.172,P<0.05),and CASdecreased significantly from 6 (5,7) to4 (2,5) (Z=-2.496,P<0.01).SⅠtvalue increased from 6 (4,11) mm to 11 (7,16) mm 4 weeks after steroid treatment (Z =-4.286,P<0.05),and the BUT prolonged from 5 (3,9) seconds to 11 (6,14) seconds(Z=-3.160,P<0.05).NGF levels in active TAO was significantly correlated with CAS (r=0.645,P<0.01),S Ⅰ t value (r =-0.530,P<0.01) and BUT (r =-0.430,P<0.01).Conclusions Tear NGF may stabilized the tear film.The application of steroid anti-inflammation drug can reduce the tear NGF level,increase the tear secretion and improve the tear film stability in patients with active TAO.
4.Effects of elicitors on growth of adventitious roots and contents of secondary metabolites in Tripterygium wilfordii Hook. f.

To study the effects of the extract of fungal elicitor, AgNO3, MeJA and yeast on the growth and content of secondary metabolites of adventitious roots in Tripterygium wilfordii. The above elicitors were supplemented to the medium, the growth and the content of secondary metabolites were measured. When the medium was supplemented with the elicitor Glomerella cingulata or Collectotrichum gloeosporioides, the content of triptolide was increased by 2.24 and 1.93-fold, the alkaloids content was increased by 2.02 and 2.07-fold, respectively. The optimal concentration of G. cingulata was 50 μg/mL for accumulation of triptolide, alkaloids and for the growth of adventitious roots. AgNO3 inhibited the growth of adventitious roots and the accumulation of the alkaloids, whereas it (at 25 μmol/L) increased the accumulation of triptolide by 1.71-fold compared to the control. The growth of adventitious roots, the contents of triptolide and alkaloids were increased 1.04, 1.64 and 2.12-folds, respectively when MeJA was at 50 μmol/L. When the concentration of yeast reached 2 g/L, the content of triptolide increased 1.48-folds. This research demonstrated that supplementation of AgNO3 and yeast enhanced the biosynthesis of triptolide in adventitious roots and the synergism of G. cingulata and MeJA could promote the biosynthesis of both triptolide and alkaloids.

5.Study on the medical services monitoring under the concept of big data

Medical service utilization monitoring of county areas residents is one of the important contents of health policy management and research.Traditional service utilization monitoring, such as National Health Services Survey (NHSS), residents healthcare treatment (clinical) data from medical insurance, had problems of high cost, large deviation, slowly updating monitoring indicators, out of time in health management decisions.The data convergence and re-architecture of multi-source data provide a new idea for the monitoring of residents'' service utilization.Different linking data-sources would come true with the tools of Microsoft Access database administration and Excel programming techniques.At the same time, a new database would be accessed with the indices of residents'' annual medical service utilization, preference in medical services, unconscionable service utilization, and lead to the promotion of medical service utilization monitoring research development.
6.Quality Evaluation of Solid Lipid Nanoparticle of the Skin Extract of Bufobufo gargarizans

OBJECTIVE:To evaluate the quality of solid lipid nanoparticle of the skin extract of Bufobufo gargarzans. METH-ODS:The morphology of solid lipid nanoparticle of the skin extract of B. gargarzans was observed by TEM. The particle size was determined by laser scattering particle size analyzer. The contents of cinobufagin and resibufogenin, encapsulation efficiency, drug-loading amount and accumulative release rate of cinobufagin were determined by HPLC. The stability of nanoparticle was in-vestigated within 24 h at 60,25 and 4 ℃. RESULTS:The solid lipid nanoparticle of the skin extract of B. gargarzans were uni-form in particle size and showed round and spheroidicity shape;average particle size was(138.5±4.2)nm,The encapsulation effi-ciency of cinobufagin and resibufogenin were 90.60% and 91.51%,and drug-loading amount were 35.82% and 44.15%. The accu-mulative release rate of cinobufagin was 50%at 4 h and reached 88%at 48 h,which was in line with Weibull equation(r=0.9438). Under 3 kinds of temperature conditions,encapsulation efficiency decreased gradually as the holding time of nanoparticle pro-longed;the decrease degree was the smallest at 4 ℃.CONCLUSIONS:The quality evaluation results of solid lipid nanoparticle of the skin extract of B. gargarzans are in line with the standard,and prepared nanoparticles show sustained-release effects and should be kept under low temperature.
7.Effects of levetiracetam on the expression of SYN and GAP-43mRNA in the hippocampus of rats with epilepsy

Objectives To observe the effects of levetiracetam (LEV) on spatial learning and memory ability in epileptic rats and the expression of synaptophysin (SYN) and growth-associated protein 43 (GAP-43) in the hippocampus of rats with epi-lepsy induced by lithium pilocarpine (Li-pilo). Methods SD rats were randomly divided into normal control group (normal saline group), LEV group (treatment group), Li-pilo group (model group) and LEV control group (normal dose group). Twenty-four hours after the establishment of model, LEV (150 mg/kg per day) was given to rats in LEV group and LEV control group, and equivalent normal saline was given to rats in normal control group and Li-pilo group. At 7, 14 and 28 days after successful model building, the expression levels of the SYN and GAP-43mRNA in hippocampus of rats in each group were detected by RT-PCR. In the 4th week, the ability of learning and memory of rats in each group was evaluated by the Morris water maze. Results There was no significant difference in escape latency and the frequency of crossing platform between LEV control group and normal control group in Morris water maze test. Compared with normal control group, the rats in Li-pilo group had a longer latency to escape and reduced frequency of crossing platform (P<0.05). Compared with Li-pilo group, the rats in LEV group had shorter latency to escape and more frequency of crossing platform (P<0.05). There was no significant difference in the expressions of SYN and GAP-43mRNA in hippocampus of rats between LEV control group and normal control group. The expressions of SYN and GAP-43mRNA in Li-pilo group were lower than those of normal control group and LEV group (P<0.05). Conclusions Levetiracetam may improve learning and memory ability of rats with epilepsy through the regulation of SYN and GAP-43mRNA expressions in hippocampal tissue.
8.The clinical significance of dynamic determination of serum procalcitonin levels in judgement of infection and prognosis in patients with severe craniocerebral injury

Objective To explore the clinical significance of dynamic determination of serum procalcitonin(PCT)levels in judgment of infection and prognosis in patients with severe craniocerebral injury.Methods A total of 80 patients with severe craniocerebral injury were selected and they were divided into infection group and non-infection group.Infection group was divided into improved group and non-improved group.Acute physiology and chronic health evaluation Ⅱ(APACHE Ⅱ)score and Glasgow coma scale(GCS)score was compared between infection group and non-infection group.The level of serum PCT,white blood cell count(WBC),C-reactive protein(CRP)and interleukin(IL)-6 was compared on the first,third,fifth day of admission in intensive care unit among these groups.Results Infection group in 48 patients,and non-infection group in 32 patients.There was no significant difference in age,sex and GCS score between two groups(P >0.05).APACHE Ⅱ score in infection group was higher than that in non-infection group [(21.71 ± 7.13)scores vs.(15.32 ± 6.17)scores],and there was significant difference (P< 0.05).The level of serum PCT on the first day of admission in intensive care unit in infection group was higher than that in non-infection group[(2.15 ± 1.79)ng/L vs.(0.56 ± 0.47)ng/L],and there was significant difference(P< 0.05).There was no significant difference in WBC,CRP and IL-6 between two groups(P>0.05).The level of serum PCT,WBC,CRP,IL-6 on the third day of admission in intensive care unit in infection group was higher than that in non-infection group,and there was significant difference(P< 0.05).The level of serum PCT on the fifth day of admission in intensive care unit in infection group was higher than that in non-infection group [(3.09 ± 1.98)ng/L vs.(1.06 ± 0.63)ng/L],and there was significant difference (P<0.05).WBC,CRP,IL-6 on the fifth day of admission in intensive care unit was(15.13 ±6.32)×109/L,(103.56 ±40.62)mg/L,(230.41 ± 121.83)ng/L in improved group(38 cases),and (18.62 ±8.89)× 109/L,(134.38 ± 34.16)mg/L,(268.73 ± 138.12)ng/L in non-improved group(10 cases),and there were significant differences between two groups(P < 0.05).The level of serum PCT on the first,third,fifth day of admission in intensive care unit in improved group was lower than that in non-improved group,and there was significant difference(P < 0.05).Conclusion The alteration of serum PCT level may be used as one of the effective predictors for infection in patients with severe craniocerebral injury and for their prognosis.
9.Effect of inhaled arsenic trioxide and moxa leaf oil on eosinophil in asthmatic guinea pigs

Objective The experiment was designed to observe the effect of inhaled arsenic trioxide(As2O3)and in combination with inhaled moxa leaf oil on eosinophils(EOS)and lung tissue's morphology in asthmatic guinea pigs, and to explore its mechanism of relieving wheezing. Methods The ovalbumin(OVA)-induced asthmatic guinea pig model was established. Sixty guinea pigs were randomly divided into As2O3 atomization inhalation group B1 As2O3 low dose group:2.0 mg/(kg·d), B2 As2O3 high dose group:4.0 mg/(kg·d), combined treatment group C1 low dose group:2.0 mg/(kg·d)+moxa leaf oil 0.05 ml, C2 high dose group: 4.0 mg/(kg·d)+moxa leaf oil 0.10 ml, saline aerosol inhalation group(group A) asthma model group)and normal control group(group D)according to random permutation table method. After 7 days medicated, the EOS in bronchoalveolar lavage fluid(BALF)were compared. Guinea pig lung, heart, liver and kidney pathological specimens were prepared and the change of the lung tissue's inflammation and heart, liver and kidney tissue were investigated. Lung tissue’s electron microscopy specimens were prepared and the apoptosis of acidophilic cells and repair of alveolar epithelial cells were observed. Results Compared with asthma model group's EOS number[(58.08±19.01)×105], the difference in As2O3 low dose group、As2O3 high dose group、combined treatment low dose group、combined treatment high dose group and normal control group[(26.37±1.12)×105, (11.50±1.61)×105, (14.16±4.88)×105, (5.03±1.66)×105 and(0.35±0.16)×105, respectively] were statistically significant(P<0.01); there was statistical difference between As2O3 low dose group and As2O3 high dose group(P<0.01);There was statistical difference between As2O3 low dose group and combined treatment low dose group(P<0.01);There was statistical difference between As2O3 high dose group and combined treatment high dose group(P<0.05). Compared with combined treatment high dose group, difference in As2O3 low dose group、As2O3 high dose group and combined treatment low dose group was statistically significant(P<0.05). The difference in B1, B2, C1, C2 and D group was statistically significant compared with group A (P<0.01). The difference in B1 and B2, B1 and C1 group was statistically significant (P<0.01). The difference between B2 and C2 group was statistically significant(P<0.05). The difference in C2 group was statistically significant compared with group B1, B2 and C1(P<0.05). Conclusion EOS abnormal apoptosis was an important pathogenesis of bronchial asthma. The mechanism of arsenic trioxide anti-asthma was that arsenic trioxide could reduce acidophil infiltration and promote acidophilic cell apoptosis. The mechanism of moxa leaf oil anti-asthma was it could reduce acidophil infiltration. Smaller doses of the combination of inhaled As2O3 and moxa leaf oil was safe and effective to achieve the effect of relieving wheezing and the combined use had synergy.
10.Effect of berberine in treating type 2 diabetes mellitus and complications and its relevant mechanisms.

Berberine (BBR) is a type of alkaloids isolated from Coptidis Rhizoma and Phellodendri Chinensis Cortex and has been used to treat bacterial gastroenteritis, diarrhea and other digestive diseases for more than 1 000 years. According to recent studies, berberine has been found to have multiple pharmacological activities, including lowering blood glucose and lipid, anti-inflammation, antioxidation, relieving type 2 diabetic nephropathy (DN), diabetic cardiovascular disease, diabetic peripheral neuropathy ( DPN) and other complications. In this article, the authors summarized the literature reports about the effects of BBR in lowering blood glucose and preventing and treating the above type 2 diabetes and its complications, in order to provide reference to further studies and promotion of BBR's application.
