Background Proliferative vitreo-retinal diseases include all the overly proliferative diseases of eyes.Many kinds of cytokines play important roles in their pathogenesis.Hypoxia-inducible factor 1α(HIF-1α)participates in the pathogenesis of a lot of ischemic diseases.HIF-1α may play an important role in the pathogenesis of proliferative vitreo-retinal diseases. Objective Present study was to observe the expression of HIF-1α in vitreous body in the eyes with proliferative vitreo-retinal diseases.The vitreous samples were collected during the vitrectomy from 71 eyes of 69 patients(experimental group)including 39 eyes with proliferative diabetic retinopathy(PDR)and 32 eyes with proliferative Vitreoretinopathy(PVR).Sixteen eyes of 16 patients as the cases control group,including 14 eyes with macular hole(MH)and 2 eyes with epiretinal macular membrane(ERM).What is more,there were 8 vitreous fluid samples of the normal control group.The concentrations of HIF-1α in vitreous body were detected by enzyme-linked immunosorbent assay(ELISA).The written informed consent was obtained from each patient before any medical procedure. Results The concentrations of HIF-1α in vitreous body in the eyes of experimental group,the cases control group and the normal control group were(294.08±2.97)、(260.41±8.29)、(16.38±3.56)mg/L respectively,showing a significant difference among the three groups(F=248.77,P=0.00).The concentrations of HIF-1α in vitreous body were obviously increased in the eyes of experimental group in comparison with the normal control group and the cases control group respectively(t=22.25,P=0.00;t=2.70,P=0.00 respectively).The concentrations of HIF-1α in vitreous body were increased in the eyes of the cases control group in comparison with the normal control group eyes(t=14.21,P=0.00).There was inferior correlation between the concentrations of HIF-1α in vitreous body in the eyes of experimental group and the course of ophthalmopathy. Conclusion Intravitreous HIF-1α level regulates in the eyes with proliferative vitreo-retinal diseases. Expression of HIF-1α in vitreous had inferior correlation with the course of ophthalmopathy. HIF-1α may play an important role in the pathogenesis of proliferative vitreo-retinal diseases.

9.Effects of lactoferrin on activity of PKG in spinal dorsal horn in a rat model of neuropathic pain

Objective To investigate the effects of lactoferrin on activity of PKG in spinal dorsal horn in a rat model of neuropathic pain(NP).Methods Thirty-two male SD rats weighing 200-250 g were randomly divided into4 groups(n = 8 each): sham operation group(group S),NP group,lactoferrin group and KT5823(an inhibitor of PKG)group.Neuropathic pain was produced by placing loosely constrictive ligatures around the common sciatic nerve in group NP,lactoferrin and KT5823,while the sciatic nerve was only exposed but not ligated in groupS.In group S and NP,normal saline 10 μl + 50% dimethyl sulfoxide(DMSO)10 μl were injected intrathecally.Lactoferrin 100 μg + 50% DMSO 10 μl were given intrathecally in group lactoferrin.Lactoferrin 100 μg + KT5823 10 μl were given intrathecally in group KT5823.The paw withdrawal latency(PWL)to a thermal nociceptive stimulus was measured every 30 min within 180 min after administration.The rats were then sacrificed and the spinal cord was removed.The activity of PKG in the spinal dorsal horn was determined by immunofluorescence.Results Compared with group NP and KT5823,the PWL was significantly prolonged after administration in group lactoferrin and the PKG activity was significantly increased in group lactoferrin(P < 0.05).There was no significant difference in the parameters mentioned above between group NP and group KT5823(P > 0.05).Conclusion Lactoferrin reduces NP by inhibiting the activity of PKG in spinal dorsal horn in rats.
10.Enrichment of caffeate in Cirsium setosum by macroporous adsorption resins

Objective To search for the method used in enriching caffeates in Cirsium setosum by macroporous adsorption resin, 23 types of macroporous adsorption resin were optimized, which could provide the industrial production of caffeates with the theory basis. Methods The adsorption and elution ratios, product purity and yield, as total indices, were comprehensively evaluatued by 23 types of macroporous adsorption resin in enriching the extracts in C. setosum. Results The product purity and yield of caffeates by the HPD-100 type macroporous adsorption resin were the highest and up to 52.2% and 87.6%. Conclusion The HPD-100 type macroporous adsorption resin shows better comprehensive adsorption property. It is available for the enrichment of caffeates in C. setosum.
