1.Study on laboratory information system setting of microbiology specimen types

Objective The microbiology specimen types were various and complex ,the system of main specimen types in micro-biological detection was established under the laboratory information system (LIS) for realizing the management target of quality control before microbiological analysis .Methods A total of 3304 submitted microbiological samples were collected from January 1 to 31 in 2015 .After setting the microbiological item application procedure of main specimen types in LIS ,1532 submitted microbio-logical specimens from June 20 to 24 were performed the statistics .The error rates of specimen types were compared before and af-ter setting .Then 1635 and 1340 submitted microbiological specimens were re-collected form July 9 to 13 and August 10 to 15 ;the change of error rates was continuously observed for comparing whether the statistical difference of error rates existing between be-fore and after setting .Results The error rate of submitted microbiological specimens before setting was 4 .6% (152/3304) ,which after setting was 1 .3% (20/1532)(χ2 =31 .224 ,P<0 .001) ,which during the continuous observation period maintained the lower level of 1 .04% (17/1635) ,χ2 =39 .658 ,P<0 .001) and 0 .9% (13/1340 ,χ2 =34 .673 ,P<0 .001) .Conclusion Re-setting the LIS reduces the error rate of microbiological specimen type ,effectively increase the working efficiency and reaches the quality control in-dex before microbiological analysis .

Vertebral artery hypoplasia is a congenital vessel variation. Its incidence is from 1. 9 to 26. 5% . In recent years, studies have shown that vertebral artery hypoplasia may be a potential risk factor for posterior circulation infarction, especialy when it coexists with other cerebrovascular risk factors. Vertebral artery hypoplasia may also cause regional hypoperfusion and complex neurovascular regulation, and it also has a certaln link with migralne.
7.Insulin induces PKC-dependent proliferation of mesenteric vascular smooth muscle cells from hypertensive patients

Background and objectives Proliferation of human vascular smooth muscle cells (VSMCs) induced by hyperinsulinemia is a very common clinical pathology. Extensive research has focused on PKC (Protein kinase C)-MAPK (mitogen-activated protein kinase)intracellular signal transduction and the phenotypic modulation accompanied by reorganization of intracellular F-actins in VSMCs.Methods DNA synthesis, signaling of ERK1/2 MAPKs, and changes in α-smooth muscle (SM) actin and F-actin were studied in hypertensive and normotensive human arterial VSMCs exposed to insulin and PMA with and without the PKC inhibitor, GF109203X.Results Differences among cell types in MAPK signaling, α-SM actin, and F-actin isoforms in VSMCs harvested from the arteries of patients with essential hypertension (EH) and normotension (NT) were identified in response to insulin treatment. Proliferation and activation of MAPK were more pronounced in EH VSMCs than in NEH VSMCs. Insulin exposure decreased expression of α-SM actin and was accompanied by rearrangement of intracellular F-actins in VSMCs, especially in the EH group. These effects were reversed by treatment with the PKC inhibitor. Conclusions Human mesenteric VSMCs of EH and NT patients differed in proliferation, MAPK signaling, and degree of changes in α-SM actin and F-actin isoforms immediately following insulin exposure in vitro.
8.Construction of the High Throughput Technology for Screening Osmotic Stress Relevant Genes

With the development of functional genomics, high throughput analysis of genes’ function has been the mainstream of research, and exogenous gene's over expression via Agrobacterium-mediated transformation is the most commonly used method in gene functional analysis.The versatile plant expression vector cassette named pBHT-5 was constructed by the method of site-specific mutagenesis based on pBI121. First of all, the restriction enzyme SfiI recognition site in trfA gene (X00713) which was relevant to plasmid replication and stability was replaced without changing its amino acid composition. And then the SfiIA,SfiIB sites were added between promoter CaMV35s and terminator NOS. The versatile plant expression vector cassette can be directly used to construct plant expression vector containing the full-length genes cloned by Clontech SMARTTM technology, which will raise the efficiency of vector construction. The result will provide basis of new genes’ high throughput screening and functional analysis, then get the new genes functioning in plant osmotic stress resistance.
9.Establishment of Two-dimensional Electrophoresis System for Proteome of Monascus ruber

Two-dimensional electrophoresis(2-DE) is an important technique in proteomics research.The 2-DE system for proteome of Monascus ruber was established by comparing and analyzing the infection caused by different kinds of mediums,lysis buffer and the condition of rehydration.By cultivating the Monascus ruber with YES for 6 days,extracting total protein by TCA-acetone,lysis buffer with 8 mol/L urea,2 mol/L thiourea,4 % CHAPS,1 % DTT and 2 % Bio-lyte,an ideal 2-DE map with higher resolution and better legibility was obtained,which laid a foundation for the further studies on proteome of Monascus ruber.
