1.Correlation of lymphatic vessel density with a high mobility group Box-1 protein and tumor-associated macrophages in cervical carci-noma

Objective: To detect the correlation of lymphatic vessel density (LVD) with a high mobility group box-1 Protein (HMGB1) and tumor-associated macrophages (TAMs) in cervical carcinoma and the effect on prognosis. Methods:Immunohistochem-istry was applied to detect HMGB1, CD68, and D2-40 expressions in cervical squamous cell carcinoma in 93 cases. t test,χ2 test, Spear-man rank correlation analysis, Kaplan-Meier method, and Cox regression were performed to analyze the expression levels, correlation, and prognosis. Results: HMGB1 protein, CD68, and D2-40 were highly expressed in CSCC. As HMGB1 and TAM expressions in-creased, lymphatic vessel density increased. As HMGB1 and TAM expressions decreased, lymphatic vessel density decreased. Positive correlations were also found between the HMGB1 protein, TAM content, and LVD. In the group with low HMGB1 and TAM expres-sions, the survival time of the group with a high LVD expression was significantly lower than that of the group with a low LVD expres-sion. A multivariate Cox regression model showed that HMGB1 and lymph node metastasis were independent prognostic factors. TAMs and LVD were not independent prognostic factors. Conclusion:HMGB1 proteins and TAMs were highly expressed in CSCC. Patients who exhibit increased HMGB1 expression or increased TAM count consequently show enhanced LVD expressions, increased lymph node metastasis, and poor prognosis.

4.Gene expression profiles of toxicated fetal rat testis exposed to flutamide

Objective To study the toxic effect of flutamide (Flu) on testis with DNA chip. Methods Flu was injected subcutaneously at doses of 6.25 mg?kg-1 ?d-1 to 25 F0 rats from gestation day (GD) 12 to 17, and the 25 rats of control group received same doses of normal saline. All 50 pregnant SD rats were sacrificed on GD 20.The cDNA were extracted respectively from fetal testis, then reversely transcribed to cDNA and labeled with cy5 and cy3 fluorescence. Subsequently, cDNA probes were hybridized to the Mouse40S cDNA microarray and fluorescence signals were analyzed. Testosterone level was determined by enzyme-linked immunosorbent assay (ELISA). Results Compared with control,a total of 31 identified gene was found to be expressed differently, including 14 down-regulated and 17 up-regulated. Among them, 5 down-regulated and 10 up-regulated genes were unknown in Genbank. ELISA indicated that testosterone levels were decreased in Flu toxic group. Conclusion Flu treatment to F0 pregnant rats can damage apparently to the reproductive development of embryonic period male SD rats. The obtained differential genes may provide reference for the molecular mechanism of reproductive development.
5.Research on predictive values of Philips 256-slice CT angiography in acute coronary events

Objective:To investigate the Philips 256-slice CT coronary angiography images on a variety of forms of coronary artery calcification and coronary stenosis in relation to predict the value of the study of acute coronary events by coronary angiography and coronary artery calcification score.Methods: From May 2014 to February 2015 in the Third Affiliated Hospital of Qiqihar in patients with suspected coronary artery disease, with256-slice spiral CT coronary artery enhancement scanning and DSA, and using software of automatic analysis for coronary artery calcium score analysis, and comparing the accuracy of the two methods of inspection, combined with the clinical data of patients with scientific prediction of acute coronary events.Results: Different forms of coronary artery calcified plaque resulting in local differences in the proportion of luminal stenosis were statistically significant. AS calcium score standard are in grouped in patients with coronary heart disease (CHD),along with increasing calcium score,the risk of coronary heart disease also increase.Conclusion: The diagnosis of coronary artery calcification score of coronary heart disease have a higher value. Calcium score for judging coronary artery stenosis to have high specific degrees, worthy of clinical popularization and application.
6.Preparation and identification of hPPAR?2 monoclonal antibody

Objective:To prepare and identify the monoclonal antibody against hPPAR?2.Methods:cDNA of hPPAR?2 was obtained from pMD18-T/hPPAR?2.hPPAR?2 was expressed in prokaryocyte and purified successfully.The anti-hPPAR?2 monoclonal antibodies (mAbs) 1B4,3H2,3H10,10D6 and 10D8 were produced by immunization with purified hPPAR?2.The specificity of the antibodies was identified by ELISA,Western blot assay and immunocytochemistry.Results:Five novel murine monoclonal antibodies only against hPPAR?2 were harvested,and they were all specific to hPPAR?2.Conclusion:The antibodies obtained provide more useful tools for following research and settling the basis for screening new drugs and mechanism.
7.Design and Synthesis of New γ-Secretase Inhibitors

Objective By virtual screening in MDL,to search for a novel γ-secretase inhibitor.Methods A series of compounds were designed,synthesized,and evaluated based on pharmacophore model of γ-secretase inhibitors by virtual screening in MDL.Results The drug-likeness analytic data synthesized indicated that target compounds had drug-likeness.Each svnthesized compound was checked by IR spectroscopy,~1H and ~(13)C-NMR spectroscopy.Conclusion The designed compounds had better activity by model prediction.And the optimal compound showed a significant estimated activity value of 0.025 nmol/L and can be used as a lead for further drug development.
8.Establishment of mice breast cancer model in BALB/c mice with 4T1 cells

Objective:To establish a model of mice breast cancer of 4T1 cells in BALB/c mice and choose the best model making method.Methods:Ninety mice were divided into three groups randomly ,with 30 in each group injected by 4T1 cells suspension of 1×106 ml-1 ,1×107 ml-1 ,1×108 ml-1 respectively.Each group of mice were randomly divided into two groups which were inoculated on the chest wall and lateral abdominal wall respectively.Tumor formation time ,tumor growth rate and the 8 week survival rate in each group were compared ,and pathological character was observed by C-erbB-2 immunohistochemistry staining.Results:Tumor growth rate in cells suspension of 1×107 ml-1 was high and grow steadily.Tumor growth rate wasn′t correlative with 4T1 cells injection in different parts.The result of C-erbB-2 immunohistochemistry staining was positive.Conclusion: Injection of 4T1 cells suspension with 1 ×107 ml-1 is the best way to male tumor model in three suspension.
9.Comparison of placement of peripherally inserted central catheters using vascular ultrasound guidance system and traditional method in 938 tumor patients

Objective To compare the placement of peripherally inserted central catheters(PICC)by using vascular ultrasound guidance system and traditional method.Methods Totally 938 patients undergoing PICC were divided into the traditional method group and the vascular ultrasound system and microintroducer techniques group according to the puncture time.The differences in the one-attempt success rate and the overall success rate of the placement and the incidences of complications were compared between the two groups.Results The one-attempt success rate was 93.01% in the traditional method group and 98.76% in the vascular ultrasound system and microintroducer techniques group(P =0.005).The successful rate of PICC placement was 100%.The incidence of complication was 11.29% in the traditional method group and 2.47% in the vascular ultrasound system and microintroducer technique group(P =0.000).Conclusion The PICC placement using vascular ultrasound guidance system and microintroducer techniques can increase one-attempt success rate and decrease complications.

Cardiac arrhythmia is one of the most common cardiovascular diseases,with high morbidity and mortality,which threatens human health and lives. With the development of medicine, natural products are revealing ever-greater anti-arrhythmic benefits and potential. However,their targets have not been fully clarified. In the recent ten years ,scientists have been studying the molecular mechanisms of natural products that have been found to inhibit INa,ICa-L,Ito,IK1,IKr,IKM3,HERG channel current and steady state K+ current,promote IKs and IKATP,inhibit microRNA-1 expression and change cardiac microRNA expression profile,affect Na+-K+-ATPase and superoxide dismutase activity,inhibitβ receptor and angiotensinⅡ receptor,and regulate lipid metabolism,thus affecting cardiac rhythm and exerting anti-arrhythmic effect. This paper revies the research advances in the antiarrhythmic tar?gets of natural products.
