2. The establishment of the dysmenorrhea model in mice

AIM: To establish a dysmenorrhea model in mice. METHODS: The mice were given with some kinds of oestrogens once a day for 3-25 days. On the last day, the mice were injected intraperitoneally with oxytocin and the number of twisting body was recorded to evaluate the intensity of dysmenorrhea. The optimum conditions to establish the model was analysed statisticly. RESULTS: The optimum oestrogens was stilbestrol. Stibestrol should be given for 12-15 days. The regression equation of the dose-effect curve of stibestrol was Y = 0.03±0.04 X, r = 0. 9688. The optimum dosage of oxytocin was 20 U·kg-1. The dysmenorrhea model in mice could be preserved for about 7 days. 90% of the twisting body reactions concentrated in 0-30 minutes after oxytocin was given. The effect of oxytocin (20 U·kg-1) had significent difference with that of prostaoglantin (1.3 mg·kg-1). The test of uterus in vivo showed that stilbestrol could increase the uterine contraction frequency and strengthen the contractility. The dysmenorrhea model in mice was testified by some anti- dysmenorrhea drugs. CONCLUSION: Compared with the hypodermic implantation in rats, the dysmenorrhea model in mice was simple, reliable, economical and testifiable, etc.

6.Effects of IL-1beta on inducible nitric oxide synthase-nitric oxide system activity in arginine vasopressin-induced rat cardiac fibroblasts.

AIMTo explore the effects of interleukin-1beta (IL-1beta) on inducible nitric oxide synthase (iNOS)-nitric oxide (NO) system activity in arginine vasopressin (AVP)-induced rat cardiac fibroblasts (CFs).
METHODSCFs were isolated by trypsin digestion method. Nitric acid reductase method, spectrophotometry and reverse transcription-polymerase chain reaction (RT-PCR) were used to detect NO contents, NOS activity and iNOS mRNA expression.
RESULTSAVP significantly increased iNOS mRNA expressions, NOS activity and NO contents (P < 0.05) in CFs. IL-1beta enhanced the effects of AVP on iNOS-NO system activity in a concentration-dependent manner, moreover the iNOS mRNA expressions, NOS activity and NO contents of AVP + 3 ng/ml, AVP + 5 ng/ml IL-1beta group were both significantly higher than those of AVP group (P < 0.05). But when IL-1beta concentration increased to 5 ng/ml, the iNOS mRNA expressions, NOS activity and NO contents did not increase accordingly, slightly decreased instead.
CONCLUSIONWithin certain range of concentrations IL-1beta cooperates with AVP to increase iNOS-NO system activity in CFs.

7. Curative effect of cortical bone trajectory screw combined with pedicle screw in internal fixation for senile osteoporosis lumbar degenerative disease

Objective To evaluate the curative effect of cortical bone trajectory (CBT) screw in combination with pedicle screw in lumbar posterior decompression. bone graft and internal fixation for the treatment of osteoporosis lumbar degenerative diseases. Methods From February 2014 to April 2015. 32 patients with osteoporosis lumbar degenerative diseases underwent posterior decompression, bone gratt and internal fixation after invalid conservative treatment. Internal fixation was performed by pedicle screws in 20 patients (A group) and by CBT screws combined with pedicle screws in 12 patients (B group). The recovery, operation time, intraoperative blood loss and VAS scores before and after operation, and JOA score were recorded in the two groups. Results The symptoms were improved in all the 32 patients. After surgery. JOA scores were significantly increased and VAS scores were significantly decreased. There were no significant differences in terms of operation time. blood loss. JOA score or VAS score between the two groups (P>0. 05). In A group. one side pedicle screw placement failed due to osteoporosis in 2 cases and unilateral fixation was subsequently carried out No poor screw holding force. nailing failure. or fracture breakage appeared in B group. No vascular or nerve damage caused by nailing was found in B group. The postoperative radiographic follow-up showed no screw loosening, fall-off or collapse; and the screw. connector, and fusion cage were all at the right position. There were no any adverse events in the two groups. Conclusion CBT screws in combination with pedicle screws in lumbar internal fixation have the advantage of less muscle and sott tissue injuries. strong screw holding force. and satisfied early curative effect in treating senile osteoporosis lumbar degenerative diseases.
8.Expressions of injury-related gene in cultured developing neurons following seizures

Objective To study interleukin-1 receptor(IL-1R) and connexin(Cx36) gene expression following Mg 2+-free-induced seizures in cultured developing neuron. Methods Rat embryo cortical neurons cultured for 6 and 17 days were exposed to Mg 2+-free media to induce seizure. At different time after Mg 2+-free treatment, real-time RT-PCR was used to detect IL-1R and Cx36 mRNA expression. Results 1. IL-1R mRNA expression transiently decreased after Mg 2+-free treatment in neurons cultured for 6 and 17 days in vitro. Then the levels of IL-1R mRNA expression recovered in neurons cultured for 6 days, but IL-1R mRNA expression were increased in neurons cultured for 17 days compared with control group and the peak was at 24 hours. 2. In neurons cultured for 6 days in vitro, Cx36 mRNA expression increased after Mg 2+-free treatment compared with control group, the peak was at 24 hours. But in neurons cultured for 17 days in vitro, Cx36 mRNA expression decreased at 6 hours after Mg 2+-free treatment compared with control group, the peak was at 24 hours. Conclusions IL-1R mRNA and Cx36 mRNA expression following Mg 2+-free-induced seizures are different between the neurons cultured for 6 and 17 days in vitro. This is possibly related to the different neuron injury between 6 and 17 days in vitro following seizures.
9.Effects of 1,25-Dihydroxyvitamin D_3 on Cell Proliferation,Differentiation and Expression of Vitamin D Receptor in Mouse Osteoblast

Objective To study the effect of different concentration of 1,25-dihydroxyvitamin D3[1,25(OH)2D3] on cell proliferation,differentiation and the expression of vitamin D receptor (VDR) in mouse MC3T3E1 osteoblast.Methods Osteoblast were cultured in medium with different concentrations of 1,25(OH)2D3.Incubated for 48 h,cell proliferation of osteoblast were examined by MTT reduction assay (mono-nuclear cell direc cytotoxicity assay),the osteocalcin (OC) levels in cell medium were detected by ELISA,and the expression of VDR mRNA and protein were examined by using SYBR Green real-time PCR and Western blot,respectively.Results 1.After incubation with 1,25(OH)2D3 for 48 h,the number of MC3T3E1 osteoblast was significantly less than that in control group(P0.05).3.SYBR Green real-time PCR and Western blot results showed that the expression of VDR mRNA as well as VDR protein of osteoblast in 10-8,10-9 mol/L experimental groups were significantly higher than those in control group (Pa0.05).Conclusions Cell proliferation of mouse osteoblast can be inhibited,while the cell differentiation was promoted by 1,25(OH)2D3.1,25(OH)2D3 up-regulated the expression of VDR in mouse osteoblast,which suggested that the VDR signal pathway may play some role in proliferation and differentiation of osteoblast.
10.Curative effect of nasal type extranodal NK/T-cell lymphoma by sequential chemotherapy combined radiotherapy compared with chemotherapy.

OBJECTIVE:
To explore the curative effect of nasal type extranodal NK/T-cell lymphoma by sequential chemotherapy combined radiotherapy compared with chemtherapy.
METHOD:
Fifty-seven cases of nasal type extranodal NK/T-cell lymphoma confirmed by pathological morphology and immuno-histochemistry were divided into chemotherapy combined radiotherapy group (34 cases) or chemotherapy group (23 cases). Twenty-three patients in the chemotherapy group alternately applied CHOP, VDLP and MEOP regimen after each two treatments into the clinical observation; Chemotherapy combined radiotherapy group of 34 patients, in addition to the above chemotherapy, applied three-dimensional conformal radiation therapy of the primary site by linear accelerators. Then all of patients were ceased treatment and followed up 3-5 years.
RESULT:
(1) After treatment, effective rate of two groups was 91.2% and 87.0%, there was no obvious difference (P > 0.05); After follow up about 1 year, effective rate of two groups was 76.5% and 73.9%, there was no obvious difference (P > 0.05); (3) After follow up about 3 years and 5 years, disease free survival (DFS) of two groups was 61.3%, 47.6% and 43.5%, 21.4%, there was obvious difference (P < 0.05). (4) Long-term survival is closely related to treatment mode. (5) B symptoms, advanced (III, IV) stage, the International Prognostic Index (IPI), KPS scores were correlated with prognosis, and were independent prognostic factors.
CONCLUSION
Treatment with chemotherapy and radiotherapy for nasal type extranodal NK/T-cell lymphoma had obvious curative effect and may improve long-term survival efficiently compared with chemotherapy alone.
