OBJECTIVE:To establish a method for the rapid identification of Sanjiu weitai granule and quantitative analysis of moisture. METHODS:Near-infrared (NIR) spectroscopy was adopted. 38 batches of Sanjiu weitai granule were collected,NIR spectrum was determined by integrating sphere diffuse-reflectance. Conformity test model was performed by comparing consistency index(CI)value and CI limit, and quantitative model of moisture was established using partial least squares(PLS)algorithm. RE-SULTS:When CI value was less than 6,the established model can accurately distinguish Sanjiu weitai granule, the model was proven feasible;the correlation coefficient (r2) of quantitative model was 97.44,the root mean square error of cross validation (RMSECV) was 0.453,the root mean square error of prediction (RMSEP) was 0.748. CONCLUSIONS:The method is simple and rapid without destroying, which can be applied to fast screening of Sanjiu weitai granule in site and fast determination of mois-ture content.

Objective:To analyze the significance of equipment custodian routine maintenance work and then offer some suggestion of countermeasure. Methods: Twenty four cases of maintenance cassette sterilizer were divided into test group(12 cases), equipment custodian had been trained; control group (12 cases)has not received training. In addition, we analyzed separately the maintenance work with the risk theory and cost-benefit.Results: The total number of failures in the test group was 13, obviously lower than 32 in the control. The risk of all devices under control, the maintenance of the benefits far outweigh the cost. Conclusion:Training equipment custodian, can reduce the incidence of failure, are worthy of promotion.

Objective To construct a luciferase gene expression vector containing full-length 3' untranslated region(3'UTR)of mouse vascular endothelial growth factor-C(VEGF-C)gene,and to observe the effects of VEGF-C 3'UTR on luciferase gene expression by a double-fluorescence report system. Methods Polymerase chain reaction(PCR)was used to amplify VEGF-C 3'UTR and a 312bp VEGF-C coding region(CR)fragment from full-length VEGF-C cDNA in mouse Lewis lung cancer cells. The luciferase expression vectors containing VEGF-C 3'UTR or VEGF-C CR were constructed by subcloning the PCR products to luciferase reporter vector pGL3-Promoter using gene engineering technology, and then they were transfected to mouse Lewis lung carcinoma cells by LipofectamineTM 2000.The activities and mRNA expression of luciferase were detected by Dual-Luciferase Reporter System and quantitative RT-PCR,respectively.Results Mouse VEGF-C 3'UTR(429bp)and VEGF-C CR(312bp)were successfully amplified by PCR.The VEGF-C 3'UTR and VEGF-C CR fragments were successfully inserted into pTA2 vector, and then subcloned to pGL3-Promoter vector at Xba Ⅰ site by using restriction endonucleases analysis. The DNA sequences and insertion orientation of PCR products were all correct by sequencing analysis. The resulted luciferase expression plasmids were named pGL3-VEGF-C 3'UTR and pGL3-VEGF-C CR,respectively.Dual-Luciferase Reporter System detection and quantitative RT-PCR showed that in Lewis lung carcinoma cells, the activities of luciferase and expression of luciferase mRNA in the pGL3-Promoter group were higher than that in the pGL3-VEGF-C 3'UTR group, and there was no significant difference between pGL3-VEGF-C CR group and pGL3-Promoter group. Conclusion VEGF-C 3'UTR can inhibit luciferase gene expression.

Objective To examine the heating capability of thermoseeds in isolated hepatic tissues. Methods Isolated hepatic tissues were heated uniformly to 37 ℃,meanwhile the thermoseeds and thermocouples were planted using a template paralleling with the magnetic line.Afterwards, the tissues were treated in an extracorporeal magnetic field of 120-123 gauss at a frequency of 113-116 kHz for 30 minutes. At the meantime, the temperature was recorded by a computer through thermocouples. Results The temperature of the tissues raised markedly with a largest change of 14 ℃.The heating area became hard and showed a gradual change of color(white-yellow-brown).The final temperature was negatively correlated with distance when using same number of the thermoseeds(r=-0.96017 to-0.99767,P=0.0023 to 0.0398),and positively correlated with the number of thermoseeds when measuring at a same point(r=0.93775 to 0.99270,P=0.0007 to 0.0185)except for the center group(r=0.93936,P=0.0606).Conclusions Thermoseeds inductive heating technology can achieve an appropriate temperature in isolated hepatic tissues. In certain extent, the temperature increase the number of thermoseeds and heating distance.
