1.Preparation and Curative Effects of Different Compound Scald Preparations for Deep Ⅱ? Scald

OBJECTIVE:To prepare different compound scald preparations and to investigate their repairing effects for deep Ⅱ? scald in rabbits.METHODS:The deep Ⅱ? scald model was established in depilated rabbits by infrared radiation.Then the rabbits in the trial groups were treated with the same drug at different dosage and different forms of medication:compound scalded gels,compound scalded ointments and the compound scalded films,with Moist Exposed Burn Ointment as positive control and 0.9% NaCl aqueous solutions blank.After a certain time of treatment,the contraction of the scald skin was observed and the average value of the diameter of the scald skin was calculated.RESULTS:In three trial groups and Moist Exposed Burn Ointment positive control group,no scald infection occurred,and the healing surfaces of the scalds were all smooth and even.However,the healing time was different between 3 trial groups and Moist Exposed Burn Ointment positive control group,with compound scalded gels showing the best efficacy in that the scald surfaces were free from infection,the healing surfaces were smooth and even,and the contracted diameter of the scalded skin was significantly lower than in other groups,showing significant differences as compared with other groups(P
2.Preparation of Rifampicin Suppositories and Solubility Determination

OBJECTIVE: To prepare rifampicin suppositories and to determine its dissolution in vitro. METHODS: Rifampicin suppositories were prepared with gelatin and glycerine as base material, the dissolution of rifampicin suppositories was determined by UV spectrophotometry and which were compared with rifampicin capsules sold in the market. RESULTS: The linear range of rifampicin was 10.0~60.0?g?mL-1(r=0.9 999), and the average recovery was 99.87%(RSD=0.42%, n=6). The dissolutions of rifampicin suppositories and rifampicin capsules in vitro at 45min were(89.9?0.97)% and(79.8?1.14)%, respectively. CONCLUSION: Rifampicin suppositories were simple in preparative method, well-formed, low in cost, and with high dissolution in vitro.
3.THE THERAPEUTIC EFFECT OF GINGER OLEORESINS ON HYPERLIPIDEMIA AND PLATELET AGGREGATION

Objective: To study the effects of ginger oleoresins(GO) on blood lipids in hyperlipidemic mice and the platelet aggregation in rats.Methods: The rat platelet aggregation was induced by ADP and AA in vitro, and the inhibition by GO was tested by light transmission. The animal model of hyperlipidemia was establised by feeding Kunmimg mice with high lipid feed stuff. The effects of GO on serum lipid and hepatic MAD in hyperlipidemic mice was investigated. Results: GO could effectively inhibit rat platelet aggregation induced by ADP and AA in vitro, which were 58.9%,97.5% respectively, and in dose dependant manner. Besides, GO could also significantly decrease serum TC, LDL, AS index and hepatic MDA in hyperlipidemic mice, which were 16.45%, 60.45%,37.30% and 54.27% respectively, and showed no side-effect on liver. Conclusion: Long-term consumption of GO can effectively modulate blood lipids, prevent thrombosis and AS.

5.R-value comprehensive evaluation of effect of three methods for lung-kidney comprehensive evaluation study on R value of three methods for regulating and invigorating lung and kidney in regulating lung inflammation signaling pathways in COPD rats.

Previous studies showed that three methods for regulating and invigorating lung and kidney (lung invigorating and spleen strengthening, lung invigorating and kidney tonifying, and Qi supplementing and kidney nourishing) could regulate inflammatory signaling pathways of chronic obstructive pulmonary disease (COPD) in rats, so as to alleviate inflammation. In the present study, R-value comprehensive evaluation method was used to evaluate the comprehensive effect of three methods for regulating and invigorating lung and kidney on inflammatory signaling pathways. Rats were randomly divided into control, model, lung invigorating and spleen strengthening, lung invigorating and kidney tonifying, Qi supplementing and kidney nourishing and aminophylline groups. The COPD rat models were established by cigarette smoking combined with bacterial infection, and orally administered with drugs between the 9th and 20th week. Afterwards, efforts were made to observe the long-term effects between the drug withdrawal and the 32rd week and detect indicators in two batches in the 20th week and 32th week. Specifically, (1) Linking JAK/STAT signaling pathway: JAK2 mRNA, and protein expressions of STAT-1, STAT-3, STAT-5, JAK-2; (2) NF-kappaB signaling pathway: Smad2 mRNA and protein expressions of I-kappaB, NF-kappaB, TGF-beta1; (3) PPARgamma and antioxidant signaling pathway: SOD, PGE mRNA, PPARgamma protein. According to the results, 5 indicators in JAK/STAT pathway, 4 indicators in NF-kappaB pathway, and 3 indicators in PPARgamma pathway were significantly rectified by three methods for regulating and invigorating lung and kidney in between the 20th week and 32nd week. Between the 20th and 32nd week, the recipes for rectifying JAK/STAT pathway with intensity from high to low were recipes for lung invigorating and spleen strengthening, Qi supplementing and kidney nourishing, lung invigorating and kidney tonifying, aminophylline, particularly those for lung invigorating and spleen strengthening; The recipes for rectifying NF-kappaB pathway with intensity from high to low were recipes for lung invigorating and spleen strengthening, lung invigorating and kidney tonifying, Qi supplementing and kidney nourishing and aminophylline, particularly the first three types of drugs. The recipes for rectifying PPARgamma and antioxidant signaling pathway with intensity from high to low were recipes for lung invigorating and kidney tonifying, Qi supplementing and kidney nourishing, lung invigorating and spleen strengthening and aminophylline. Therefore, three methods for regulating and invigorating lung and kidney showed better long-term effects in regulating COPD lung inflammation signaling pathways. Specifically, recipe for lung invigorating and spleen strengthening showed a better effect in JAK/STAT and NF-kappaB pathways, while recipe for lung invigorating and kidney tonifying and Qi supplementing and kidney nourishing showed better effects in PPARgamma and antioxidant signaling pathways. In conclusion, R-value comprehensive evaluation method can evaluate the comprehensive effect of medicines and define the ranking of multiple drugs and their main targets.

6.Inhibitory effect of RGDRGD-endostatin on the expression of vascular endothelial growth factor and its receptor in corneal neovascularization

BackgroundEndostatin (ES) is currently the strongest endogenous angiognesis inhibitor,and it can inhibit the occurrence of neovascularization.Various studies demonstrated that the poly RGD sequence can enhance the function of the ES gene.ObjectiveThis study was to evaluate the use of gene therapy of modified ES for alkaline burn-induced corneal neovascularization (CNV).MethodsOne hundred and two clean SD rats were randomly divided into the normal control group,the pCI empty vector group,the pCI-ES group,and the pCI-RGDRGDES group.Corneal neovascularization models were established by placing a piece of 3 mm filter paper with 1 mol/L NaOH at the central cornea for 40 seconds.3 μg of the pCI blank vector,ES-tranfected pCI blank vector,or RGDRGD-ES-transfected pCI vector was injected into the superior bulbar conjunctiva after the alkali burn twice at 1-week intervals.Area of CNV and edema of the cornea in the various groups of rats were examined daily under the slit lamp biomicroscope.1,4,7 and 14 days after operation,the rats were sacrificed by the excessive anesthesia method and corneal tissues were obtained to evaluate pathological changes.The expression of CD34 in vascular endothelial cells was detected by immunochemistry to calculate the corneal neovascular density.The expressions of VEGF mRNA and Flk-1 protein in the corneas were detected by reverse transcription polymerase chain reaction (RT-PCR) and Western blot analysis.The use and maintenance of animals followed the Statement of ARVO.Results Seven to fourteen days after corneal alkali-burning,the corneal neovascular area was smaller in the pCI-ES group and pCI-RGDRGD-ES group compared with the normal control group and pCI blank vector group (P<0.05,P<0.01 ),and nevascular area in the pCI-RGDRGD-ES group was smaller than that in the pCI-ES group (P<0.05).The expression level of CD34 was significantly lower in the pCI-ES group and pCI-RGDRGD-ES group than that in the normal control group and pCI blank vector group (P<0.05,P<0.01 ),and the expression level of CD34 was further declined in the pCI-RGDRGD-ES group compared with the pCI-ES group (P<0.05 ).Compared with the normal control group and pCI vector group,the expressions of the Flk-1 protein and VEGF mRNA were decreased in the pCI-ES group and pCI-RGDRGD-ES group on the fourth day after corneal alkali-burning (P<0.01,P<0.05 ),and those in the pCI-RGDRGD-ES group were less than the pCI-ES group (P< 0.05,P< 0.05 ).Conclusions Subconjunctival injection of both ES and modified RGDRGD-ES genes result in significant suppression of CNV in vivo,and modified RGDRGD-ES appears to be more effective than native ES.The main mechanism of ES in inhibiting neovascularization is to downregulate the expression of VEGF and Flk-1.
7.Long-term clinical study of modified posterior double-door cervical laminoplasty, more than ten years follow-up

Objective:To explore the long-term clinical efficacy and complications of modified spinous process splitting laminoplasty using coralline hydroxyapatite (SLAC).Methods:The patients who underwent SLAC surgery in this hospital from 2005.5 to 2011.7 were included retrospectively. The perioperative data were collected and followed up. A total of 165 cases were included. Among them, there were 115 males and 50 females; the average age was 56.5±11.4 years (range: 26-84 years), and the average follow-up period was 136.5±23.2 months (range: 108-182 months). The modified Japanese Orthopedic Association (mJOA) score, the neck disability index (NDI) score were used to evaluate the clinical symptoms of patients, and follow-up imaging to observe the changes in cervical spine curvature and mobility.Results:In terms of functional score: mJOA score increased from 11.4±2.9 before surgery to 15.0±1.8 in the early postoperative period but dropped to 14.0±2.5 at the last follow-up ( F=77.096, P<0.001), and NDI score decreased from 19.2%±14.4% before surgery to 13.0%±15.0% in the early postoperative period, but it rose to 14.0%±14.9% at the last follow-up ( F=6.915, P<0.001). The improvement rate of mJOA was 63.5% (early postoperatively) and 50.8% (last follow-up). Regarding the curvature of the cervical spine, the C 2-C 7 Cobb angle decreased from 14.8°±9.1° before surgery to 11.1°±10.5°, and it was maintained at the last follow-up (11.0°±10.1°) ( F=1.083, P=0.342). The cervical spine range of motion decreased significantly, mainly because the Cobb angle in the flexion position decreased from -19.8°±13.6° before the operation to -3.7°±10.6° at the last follow-up ( P<0.001). Seventy-two patients (44.0%) had complications after operation. Six patients (3.6%) developed neurological symptoms aggravated during follow-up, and their mJOA decreased by more than 3 points. 62 patients (37.6%) developed axial symptoms, of which 7 cases were relieved early (within 6 months post-op), 55 patients had persisted symptom and 17 cases (10.3%) remained painful. C 5 nerve root palsy occurred in 10 cases (6.1%) after the operation, 9 cases resolved within two years and the last patient did not resolve. Conclusion:SLAC surgery can effectively alleviate the patient's neurological symptoms and maintain long-term efficacy and cervical lordosis. The occurrence of persistent axial symptoms and the loss of cervical flexion range are long-term complications after SLAC surgery.
8.Preparation and evaluation of enteric-coated and taste masking clarithromycin granules.

The study is to prepare taste masking and enteric-coated clarithromycin granules by melting and fluid bed coating technology. Clarithromycin and matrix materials were melted at a certain temperature, and then made into particles by fluidized bed coating. X-ray powder diffraction and scanning electron microscopy were used to identify the crystal and morphology of drug loading granules. In vitro dissolution method was used for the observation of the drug release behavior. The results showed that the drug particles size range was 0.2 - 0.6 mm; the crystal form of clarithromycin in the granule did not change; enteric-coated granules accumulated release in 0.1 mol L(-1) hydrochloric acid in 2 h was less than 10%, while in pH 6.8 phosphate buffer in 1 h was more than 80%. The taste masking and enteric-coated clarithromycin granules not only have good taste masking effect, but also have a good release behavior. It is expected to have better clinical application.

10.Effects of acupuncture on gastrointestinal responses after renal transplantation.

OBJECTIVETo observe the effect of acupuncture on gastrointestinal responses after renal transplantation.
METHODSSixty cases with gastrointestinal responses after renal transplantation were randomly divided into an acupuncture group and a medication group, 30 cases in each group. The acupuncture group were treated with acupuncture at Neiguan (PC 6), Zusanli (ST 36), Sanyinjiao (SP 6), etc.; the medication group were treated with oral administration of Weilexin. The symptoms of abdominal pain, nausea, gastric distention, and other gastrointestinal responses were observed in the two groups.
RESULTSThe effective rate was 93.3% in the acupuncture group and 76.7% in the medication group with a very significant difference between the two groups (P < 0.01), the acupuncture group being significantly better than the medication group.
CONCLUSIONAcupuncture has a good effect of promoting recovery of gastrointestinal function after renal transplantation.
