1.Application of thiocyanate elution in the selection of phage antibodies with high affinity

Objective:To establish a method for the selection of phage antibodies(Phabs) with higher affinity.Methods:Simulant libraries were constructed using anti keratin and anti HBsAg Phabs with different affinities.During the panning procedure,thiocanate solutions of different concentrations were used as elution buffers to favor the recovery of Phabs with higher affinities.The recovered Phabs were identified by restriction enzyme mapping.Results:All the tested Phabs were proved tolerant to thiocyanate treatment.By panning the simulant libraries,the ratio of Phabs with higher affinity in the obtained clones were increased alonging with the increase of NH 4SCN concentrations.Conclusion:Thiocyanate elution is a powerful and effective method to help obtain the Phabs with high affinity from phage antibody library.
2.Evaluation of the effect of neonatal hepatitis B vaccine and the prevention of low and no responders

Objective To evaluate the immunological effect of neonatal hepatitis B vaccine and to prevent the immunization effect of low and no responders. Methods A total of 1,200 infants were selected from Yantai City, Shandong Province. All the infants were immunized for the first time after birth, and were inoculated with brewer's yeast hepatitis B vaccine 5μg. The venous blood of the infant was taken to separate the serum and the geometric mean concentration of anti-HBs in serum by using chemiluminescent microparticle immunoassay. Of which 128 cases were low, no response were immunized again: 71 cases were treated with brewer's yeast hepatitis B vaccine 5μg and 57 cases were inoculated with 10%Hansenula hepatitis B vaccine again and again, three times. The results of the re-immune were evaluated . Results There were 80 cases (62.5%) wtith normal response, 23 cases (17.9%) with high response, 22 cases (17.2%) with low response, 3 cases (2.3%) with no response The immunization response rate of HBsAg positive in fathers and mothers was significantly lower than that in county, city and above, and the fathers' mother HBsAg was negative. There was no significant difference in the positive rate of antibody transfection between the first and the third of the above two yeasts, but the geometric mean concentration of anti-HBs was significantly lower for both vaccines, and the brewer's yeast vaccine was significantly lower. Conclusion The immune response rate of hepatitis B vaccine in children with HBsAg-negative mothers at the county, municipal and above hospitals is relatively high. Compared with the brewer's yeast vaccine, the effect of the hepatitis B vaccine is obviously better .
3.Genetical Engineering of Humanized Anti-keratin Antibodies with High Affinity

Objective To produce humanized anti-keratin antibodies with high affinity. Methods Panning of phage antibody libraries against human epidermal keratin was conducted to select specific antibodies. Random mutation method and chain-shuffling technique for the gene of the antibodies were employed to improve antibody affinity. The binding activity, specificity and affinity of the obtained antibodies were de-termined and analyzed. Results Four clones of either IgG or IgM anti-keratin antibodies were seleeted from semisynthetic or natural antibody libraries. One of them was seleeted to undergo affinity maturation. Random mutation method for the gene did not result in any improvement in antibody affinity. By chain-shuffling technique, however, affinity level was markedly increased and an antibody with affinity of 8?1010/M was isolated. Conclusion It is feasible to generate humanized antikeratin antibodies using genetical engineering technique.

Medicinal materials research is one of the main content of the pharmaceutical research of new drug of TCM, and it is also the focus of the new medicine pharmaceutical evaluation content. This article through the analysis of the present problems existing in the development of TCM research of new drug of TCM, from medicine research concept, quality stability, quality standard, etc are expounded, including medicine research idea value medicine study should focus on the important role and from the purpose for the top-level design of new drug research problem. Medicinal materials quality stability should pay attention to the original, medicinal part, origin, processing, storage, planting (breeding), and other aspects. 5.Clinical Observation of Locally Advanced Gastric Cancer Treated by Compound Matrine Injections Combined with Concurrent Chemoradiotherapy

Objective To study the recent clinical efficacy of compound matrine injections combined with concurrent chemoradiotherapy in the treatment of locally advanced gastric cancer. Methods Totally 65 cases of untreated locally advanced gastric cancer were randomly divided into treatment group (33 cases) and control group (32 cases). The control group was treated by the conformal intensity modulated radiotherapy combined with chemotherapy. On the basis of the control group, the treatment group was given compound matrine injections 20 mL/time, once a day for 5 weeks. The recent curative effect, the quality of life and adverse reaction of the two groups were observed. Results The recent curative effect rate of the treatment group was 66.67% (22/33), the control group was 40.63% (13/32), the difference was statistically significant (P=0.031). On hematologic toxicity, leukopenia occurred in the treatment group was 30.30% (10/33), the control group was 65.63% (21/32), the difference was statistically significant (P=0.004). KPS of 26 cases (78.79%) improved in the treatment, and 18 cases (56.25%) improved in the control group. Conclusion The effects of compound matrine injections combined with concurrent chemoradiotherapy for locally advanced gastric cancer are good, they can improve short-term efficacy, reduce side effects, and improve the quality of life.
6.The role of CD2 in acute rejection after small bowel transplantation in rats

Objective To investigate the expression of CD2 in peripheral blood T lymphocytes in acute rejection after small bowel transplantation (SBT) in rats.Methods Three groups were set up: Group A (n=18), receiving sham operation and feeding of common feedstuff; Group B (n=18) receiving SD rat to SD rat iso-series SBT and the treatment of routine liquid infusion and antibiotics after operation; Group C (n=18) receiving SD rat to Wistar rat taniso-series SBT and the same post- operation treatments as in group B. Samples of grafts and peripheral blood in the three groups were collected on the day 3, 5 and 7 respectively after SBT for detection of flow cytometry. The intestinal grafts were obtained for pathological examination.Results The survival time of rats in group C, group B and group A was 7.0? 2.1, 33.3? 2.3 and more than 90 days, respectively. Compared to traditional paper & pencil test,computer-based test has obvious advantages in improving reliability and validity of test.With the increasing popularity of computer-basedtest,making ICO examinations(International Basic Science and Clinical Sciences Assessment for Ophthalmologists) computerized can be realized in our country for the reason that we own hardware and software required by computerized exam,including standardized examination room,professional test personnel,examinee with computer skills,etc.Currently,we can rely on available resources to carry out pilots in the areas meeting the condition of computer-based test,take the form of paper and pencil and computer at the same time,and then promote the process of computer-based ICO examinations.
8.Clinical analysis of 12 cases of primary synovial chondromatosis of hip joint

Objective To study the clinical, imageological and pathologic characters of primary synovial chondromatosis of hip joint, and effect of open surgery. Method 12 cases of primary synovial chondromatosis of the hip joints admitted from May 1973 to May 2004 were retrospectively studied in respect to clinical manifestations, X-ray picture, and pathological findings. The effects of operation were followed up. Results Intermittent pain, swelling, limitation of hip joints were the main complaints. Mass and myatrophy near involved hip joint were discernible in a few patients. Although there was no obvious image of calcification, swelling of articular capsule was seen in X-ray film in 4 patients. CT scan showed arthroedema, loose bodies and bone erosion of the femoral head in the hip joint. The X-ray films of the other 8 patients showed obvious calcified loose bodies. All cases were diagnosed by pathology after operations. 8 cases were categorized Milgram Ⅱstage, and 4 cases Milgram Ⅲ stage. Eight patients were followed-up for a period of 1 to 7 years (mean 5 years). Recurrence occurred in 2 cases. Conclusion The diagnosis should be based on clinical characters, X-ray films and pathologic findings, and the pathology is most reliable. In order to decrease recurrence, CT or MRI examinations should be taken before the operation. X-ray monitoring should be carried out during the operation of the patient in whom loose bodies were found in X-ray film before the operation.
9.Study on differentially expressed genes in Jurkat cells treated with phosphonoformate using DNA microarray

Objective To screen the genes differentially expressed in gene expression in human lymphoma cell line Jurkat cells treated with phosphonoformate (PFA). Methods cDNA microarray technique was employed to detect the mRNA expressed in Jurkat cells treated with PFA or 0.9 percent sodium chloride, respectively. Results Among 1 152 genes there were 94 genes with different expression, of which 38 genes were upregulated and 56 genes were downregulated in Jurkat cells treated with PFA compared with those treated with 0.9 percent sodium chloride. Conclusions cDNA microarray technique was successfully used to screen the genes with different expression in Jurkat cells treated with PFA, and the results brought some new clues for the study of the immune regulation mechanism of PFA.
