1.Effect of telomere seeding on the biological behaviors of gastric cancer cell SGC-7901

Objective To study the effect of telomere seeding on the biological behaviors of gastric cancer cell line and its mechanism. Methods The plasmid pSXneo-1.6-T2AG3, containing telomere fragment, was prepared in large scale and transfected into gastric cancer cell line SGC-7901 by LipofectAMINTM 2000. G418 was used to scan the positive clones and PCR was preformed to verify the stable seeding of the exogenous telomere in cell. MTT aassay and flow cytometry were used to determine the growth and cycle distribution of cells. The expression of cell cycle-related genes was detected by RT-PCR. Results Both pSXneo-1.6-T2AG3 eukaryonic expressing vector be inserted with telomere TTAGGG fragments and control vector were transfected into gastric cancer cell line SGC-7901 with LipofectamineTM 2000 and gain clones through G418 selection respectively, which were named as SGC-7901-telo and SGC-7901-neo. The cells were examined on DNA level by PCR to determine the correction of transfection. Flow cytometric analysis displayed an accumulation in G1M and G2M phase of cell cycle and an decreased percentage in S phase and proliferative index. Expression of p21 mRNA in 7901-telo cell line was increased (P0.05). Conclusion The pSX-T2AG3-neo eukaryonic expressing vector be inserted with 1600bp telomere TTAGGG fragments was successfully transfected into gastric cancer cell line SGC-7901 with the help of LipofectamineTM 2000. The seeding decreased malignant phenotypes of gastric cancer cell line and up-regulated the expression of p21, but the seeding did not distinctly influence on caspase3 mRNA expression.
2.Telomere position effect in gastric cancer cell SGC7901

Objective To determine the possibility of telomere seeding in human gastric cancer cell SGC7901 and the influence of telomere on the expression of luciferase reporter gene nearby and to explore the position effect of telomere in gastric cancer cell. Methods The TET-OFF regulatory system was prepared and then transfected to gastric cancer cell SGC7901 by Lipofectamine TM 2000. With doxycycline added, the activity of the fluorescence caused by the expression of the target gene (the luciferase gene) with TET-OFF regulatory system was analyzed. The luciferase reporter plasmid in which telomere fragment pBX was inserted and the control plasmid pBX-ntf (no teloemere fragment) were co-transfected to cell 7901 with pTET-OFF regulate plasmid respectively and the luciferase activity was determined with doxycycline's induction. Results With the inducing of Dox, the activity of the fluorescence caused by the expression of the luciferase gene was inhibited. Treated with deoxycyline, there was a 4-fold decrease in the activity of the fluorescence in the cells infected with pBX and pTET-OFF as compared with that in the cells infected with pBX-ntf and pTET-OFF. Conclusion With the help of Lipofect2000, the gastric cancer cell SGC7901 can be transfected by TET-OFF regulatory system successfully. Dox could repress luciferase gene expression. Telomere seeding in gastric cancer cell SGC7901 could decrease the expression of luciferase reporter gene.
3.Pilot Evaluating Informant Questionnaire of the Community Screening Instrument for Dementia in a Sample of Community Elderly Population

Objective: To evaluate feasibility of informant questionnaire of the Community Screening Instrument for Dementia(CSID) and related factors in a sample of community elderly population, and to investigate prevalence of suspectable dementia in the sampled people.Method: A total of 2267 people aged 65 and above in one rural area and one urban area in Beijing were recruited and their informants were assessed by the CSID informant questionnaire.Validity of CSID were evaluated according to the computerized diagnosis of 10/66 project and ICD-10.Result:The split-half reliability of CSID informant questionnaire was 0.85 and the consistency was more than 0.8.When the cutoff point of CSID was 6 or more, sensitivity was 86.2% and specificity was 93.2%.At meantime, the prevalence of suspectable dementia was 9.1%.Logistic regression analysis showed that the information obtained from informant as a spouse and from informant who lived in urban area was correlated with positive diagnosis of dementia.Conclusion:CSID informant questionnaire is valid as a screening instrument of dementia.Information from spouse is useful to identify the change of cognitive function of the elderly.
4.Early diagnostic value of combining non-specific indicators in neonatal septicemia

Objective To evaluate the significance of routine blood test and C‐reaction protein(CRP) ,procalcitonin(PCT ) in early diagnosis of neonatal septicemia .Methods Totally 95 cases of confirmed septicemia ,158 cases of the clinical diagnosis of sep‐ticemia and 160 cases of non‐septicemia who were admitted to Newborn Diagnosis and Treatment Center of Children′s Hospital of Chongqing Medical University from January 2013 to May 2014 were enrolled for this study .The routine blood test ,CRP and PCT concentration of the three groups were analyzed .Taking blood culture as the gold standard ,receiver operating characterristic (ROC) curve was drawn ,sensitivity and specificity of single or combined indicator in early diagnosis of septicemia was analysed .Results There was a significant difference between confirmed septicemia group and non‐septicemia group in PCT ,CRP ,WBC ,I/T ,PLT (P< 0 .05) ;the area under the ROC curve(AUC) was 0 .892 ,0 .847 ,0 .828 ,0 .700 ,0 .681 ,respectively ;the five indicators for con‐firmed the diagnosis positive rate of septicemia group and non‐septicemia group was statistically significant (P < 0 .05) ;the single indicator PCT had better diagnosis value ,its sensitivity and specificity was 84 .2% and 66 .9% .With the application of regression of binary Logistic the AUC of combining PCT ,CRP ,WBC was 0 .974 ,its sensitivity and specificity was 94 .7% and 87 .5% ,which was higher diagnosis value than the only one indicator alone .Conclusion The combination of PCT ,CRP and WBC have contribute to early diagnosis of neonatal septicemia .
5.Effect of Qizhu Oral Liquid on radiation injury in mice

AIM: To observe the protective effect of Qizhu Oral Liquid on radiation injury in mice. METHODS: The model was made in mice by X-ray radiation. The white blood cells(WBC), bone marrow nucleated cells (BMNC) were measured. The weight of thymus and spleen were measured. RESULTS: Qizhu Oral Liquid could increase WBC and BMNC remarkably. It could increase the weight of thymus and spleen of mice signifcantly. CONCLUSION: Qizhu Oral Liquid is effective in protecting the X-radiation injury mice.

7.Effect of Paridis Rhizoma total saponins on apoptosis of human gastric cancer cell MKN-45 and Fas/FasL signaling pathway.

OBJECTIVEThe study aimed to test if Paridis Rhizoma total saponins (PRTS) could induce apoptosis of human gastric cancer cell MKN-45.
METHODBased on the previous researches, PRTS was set by different concentrations to treat human gastric cancer cell for 12 h (5, 10, 20 mg x L(-1)). Fluorescent staining methods were adopted to observe apoptotic morphological changes of MKN-45. The apoptosis rates were analyzed by flow cytometry with Annexin V-FITC/PI staining. The enzymatic activities of caspase-3 and caspase-8 were measured by ELISA. The protein levels of Fas and FasL were detected by Western blotting.
RESULTUnder a fluorescence microscope, MKN-45 treated by PRTS was seen typical apoptotic morphological features. PRTS significantly increased the rate of apoptosis. Compared with the control group, there exsited significant differences in apoptosis rate of PRTS concentration of 20 mg x L(-1) (P < 0.01); besides, the enzymatic activities of caspase-3 and caspase-8 were promoted obviously after the effect of PRTS on MKN-45 cells for 12 h (P < 0.01). The protein levels of Fas and FasL in the MKN-45 were upgraded significantly.
CONCLUSIONPRTS can induce apoptosis of human gastric cancer cell MKN-45 , which is concerned with caspase-3 and caspase-8 and upgraded Fas and FasL.

8.Acupuncture for premature ovarian failure: a prospective cohort study.

OBJECTIVETo explore the clinical efficacy and action mechanism of acupuncture for premature ovarian failure (POF).
METHODSAccording to prospective cohort study design, 30 cases were included. Based on theory of regulating Chong and Conception Vessels as well as soothing the liver and calming the nerves, acupuncture at Guanyuan (CV 4), Dahe (KI 12), Taixi (KI 3), Taichong (LR 3), Baihui (GV 20) was applied, three times per week and 3 months were considered as one session. Totally two sessions were performed. The menstruation condition, estradiol (E2), follicle-stimulating hormone (FSH) and scores of the clinical perimenopausal symptoms were taken as treatment outcomes.
RESULTSThe total effective rate was 86.7% (26/30) and the cured rate was 16.7% (5/30). The recovery rate of menstruation was 16.7% (5/30) and the regain rate of menstruation was 56.7% (17/30). After the treatment, the level of E2 was significantly increased from (45.41 +/- 18. 09) pmol/L to (59.07 +/- 24.21) pmol/L (P < 0.05), and the score of perimenopausal symptoms was obviously reduced from 14.28 +/- 8. 30 to 4.04 +/- 3.28 (P < 0.01). However, no statistical differences could be seen in FSH before and after treatment (P > 0.05).
CONCLUSIONAcupuncture has certain effect on improving menstruation and perimenopausal symptoms in POF patients, which is likely to he related with increasing the level of E2.

9.Quality assessment on guidelines of clinical practice in acupuncture and moxibustion: a study based on AGREE.

The quality of guidelines of clinical practice in acupuncture and moxibustion was evaluated with AGREE II tool. After retrieval of officially-issued guidelines of clinical practice in acupuncture and moxibustion from 2004 to 2013, totally 5 types of evidence-based guidelines of clinical practice in acupuncture and moxibustion were acquired, including depression, herpes zoster, pseudobulbar palsy after stroke, migraine and Bell's palsy. With AGREE II tool, independent score in six areas, including guideline's scope and purpose, participants and personnel, preciseness, clarity and readability, applicability and editorial independence were analyzed to perform a comprehensive evaluation in the end. The assessment results indicated that the score of editorial independence in evidence-based guidelines of clinical practice in acupuncture and moxibustion was the highest, averagely 97.9%, which was followed by clarity and readability (83.3%), participants and personnel (78.1%), preciseness (75.6%), the scope and purpose (68.1%) and the applicability (62.5%). The total evaluation score was all 6 points in the five guidelines with recommended as comprehensive evaluation advice. This study results indicate that although the guideline of clinical practice in acupuncture and moxibustion is low in number, the total quality is considerable.

10.Clinical observation of tuina manipulations for tic disorders in kids

Objective: To observe the clinical efficacy of tuina manipulations in treating different types of tic disorders (TD). Methods: Eligible TD patients were classified into three types, transient tic disorders (TTD), chronic multiple tic disorders (CMTD) and Tourette syndrome (TS), according to their disease duration and severity. The three types of children were treated with the same tuina manipulations. Changes in the Yale global tic severity scale (YGTSS) score, effective rate for tic, and cervical spine imaging examination results (including asymmetry of the lateral atlanto-dental interval, broadened anterior atlanto-dental interval, C2 spinous process deviation, occipito-atlanto-axial flexion/ extension instability) were observed after 1-month and 3-month treatments respectively. Results: The YGTSS score changed significantly after 1-month and 3-month treatments compared with that before treatment (both P<0.01); the effective rate for TD was 46.6% and 86.7% respectively after 1-month and 3-month treatments; there were significant differences comparing the effective rate for tic between different types of TD after 1-month and 3-month treatments (all P<0.05); comparing the effective rate for tic after 1-month treatment with that after 3-month treatment for the same type, the intra-group differences were statistically significant [TTD group (P<0.01), CMTD group (P<0.01), TS group (P<0.05)]; the abnormal parameter rates in neck imaging examination after 3-month treatment were significantly different from those before treatment (all P<0.01). Conclusion: Tuina manipulation is effective for TTD, CMTD and TS. It can correct the abnormal alterations of patients' cervical vertebrae, and its efficacy for TTD is most significant.
