CTGF, a member of the CCN family of immediate early genes, is a recently discovered profibrotic growth factor, which is involved in many pathophysiologic procedures. CTGF acts as a downstream effector of TGF-? acting on interstitial cells to enhance the progression of fibrotic renal diseases. It has been shown that CTGF gene expression can be induced or blocked by some kinds of cytokine and drugs. It is an interesting candidate target for future intervention strategies of renal interstitial fibrosis. [

Objective:To establish an HPLC method for the determination of calcium disodium edetate in fosaprepitant dimeglu-mine for injection. Methods:The chromatographic column was Agilent Zorbax Eclipse XDB C18(250 mm ×4.6 mm,5μm). The mo-bile phase consisted of 0. 2% tetrabutylammonium hydroxide(adjusting pH to 4. 0 with phosphoric acid)-acetonitrile(85∶15). The flow rate was 1. 0 ml·min-1 . The detection wavelength was 254 nm. The chromatographic column temperature was 35℃. The injection volume was 20μl. Results:The calibration curve was linear within the range of 26. 06-104. 24μg·ml-1(r=0. 999 8) for calcium di-sodium edetate. The average recovery was 99. 51% (RSD=0. 25%, n=9). Conclusion: The method is simple, rapid, accurate, specific and reproducible, and can be used in the determination of calcium disodium edetate in fosaprepitant dimeglumine for injection.

Objective To study the automatic word segmentation scheme suitable for traditional Chinese medical record literature. Methods Hierarchical Hidden Markov Model was used as segmentation model. Totally 300 ancient medical record literature and 300 modern medical record literature were set as experimental subjects to establish the dictionary of traditional Chinese medicine and the test corpus, with a purpose to segment the words and evaluate of the results. Results Without using dictionary of traditional Chinese medicine, the word segmentation accuracy of two kinds of medical record literature was about 75%;the part-of-speech tagging accuracy of ancient medical literature was 56.74%, the modern medical literature accuracy was 64.81%. By using dictionary of tradition Chinese medicine, the word segmentation accuracy of ancient medical record literature was 90.73%, the modern medical record literature accuracy was 95.66%;the part-of-speech tagging accuracy of ancient medical record literature was 78.47%, the modern medical record literature accuracy was 91.45%, which was obviously higher than that of ancient medical record literature. Conclusion The current word segmentation scheme has initially solved the problem of word segmentation of traditional Chinese medical record literature and part-of-speech tagging of modern medical record literature. Part of speech tagging is basically correct, but part-of-speech tagging of ancient medical record literature tagging needs further study for many influencing factors.

Objective To explore the therapeutic effect of Xiefu Zhuyu decoction joint brain protein hydrolysate injection in the treatment of patients with traumatic brain injury and its effect on cerebrospinal fluid of endothelin-1 ( ET-1 ) .Methods 102 cases of patients with craniocerebral injury were divided into control group (n=51) and experimental group (n=51),the control group were treated with brain protein hydrolyzate injection, the experimental group based on the control group were treated with Xuefu Zhuyu decoction, the curative effect, ET-1, inflammatory factor, protein, blood rheology, S-100β, neuron specific enolase (NSE), National Institutes of Health Stroke Scale (NIHSS) and complications of two groups were compared.Results The total effective rate of experimental group was higher than the control group (94.11% vs.78.43%), the difference was statistically significant (P<0.05).The ET-1 of experimental group was lower than the control group[(32.45 ±4.05) ng/L vs.(28.11 ±3.50) ng/L] (P<0.05).The inflammatory factor, protein, blood rheology, NSE, S-100βof experimental group were better than those of control group (P<0.05). The NIHSS of experimental group was lower than that of the control group ( P <0.05 ) .The complication rate had no difference between two groups. Conclusion The clinical curative effect of Xiefu Zhuyu decoction joint brain protein hydrolysate injection in the treatment of traumatic brain injury , can reduce cerebrospinal fluid levels of ET-1 and can improve inflammatory factors, proteins and hemorheology.

Objective To investigate the changes of buffy coat components after 25Gy 137Cs-irradiation.Methods Lymphocyte proliferation was detected by MTT,the fluorescent intensity of neutrophils was measured as phagocytotic rate by flow cytometry,O2-release was determined by cytochrome C reduction,and cytokine production pre-and post-irradiation was detected by ELISA.Results 25Gy is adequate to inactivate T cells.The percentages of phagocytosis of E.coli before and after irradiation were of no significant difference.The release of O2-increased significantly and monocytes maintained the ability to produce cytokines after irradiation.Conclusion Radiation does affect the components of buffy coat,and neutrophils isolated should be transfused as early as possible.

Objective To expand the NKT cells in vitro and to determine the cytokine releasing of NKT cells during proliferation.Methods Several kind of methods were established to expand TCRV ?24 +/TCRV ?11 + NKT cells from PBMC or purified T lymphocyte.The levels of IL 4,IFN ?and TNF ? of NKT cells were determined by flow cytometry.Results After 19 days' expansion,the largest number of NKT cells increased (23.0?16.7)?10 3 times and the largest ratio of NKT cells to total T lymphocyte was (25.5?7.2)%.The ratio of TCRV ?11 + NKT cells that secreted IL 4,IFN ?and TNF ? was higher than the TCRV?11 T lymphocytes.Conclusion TCRV ?24 +/TCRV ?11 + NKT cells can be expanded in vitro with ? Galcer presented by CD 1d molecule.The NKT cells can also be expanded from PBMC because the monocyte lineage cells within PBMC can express CD 1d molecule and present specific glycolipids like ? Galcer to them.

To generate recombinant adenovirus expression vector of human Sema4C gene and observe its expression in mouse myoblasts cell line C2C12 for ensuring easy access to investigate the role of Sema4C gene during myogenesis. The recombinant plasmid was packaged and amplified after being transfected in HEK293 cells through Lipofectamine. After infecting C2C12 myoblasts with recombinant adenovirus vector, the adenoviral infection efficiency was determined by confocal microscope which showed that the expression of green fluorescence could be detected at 12h and then reached peak at 24h after recombinant adenovirus infection. The infection efficiency was almost 100% confirmed by FACS examination. Objective:To assess the consequences of switching aspirin dosage from 100 mg/d to 40 mg/d on cardiovascular benefit,bleeding risk and platelet aggregation in very elderly patients. Methods:Arachidonic acid induced platelet aggregation(AA-Ag)was measured in 537 patients aged 80 or older treated with aspirin (100 mg/d).In the study,100 patients with low on-treatment platelet ag-gregation and at high risk of bleeding and low risk of cardiovascular events,were switched to aspirin (40 mg/d)and their platelet aggregation was measured again 7 days later.Their bleeding and upper gastroin-testinal symptoms were also recorded in following 3 months.Results:The study observed a heterogeneous distributed aspirin 100 mg/d AA-Ag (range:0.42% to 28.78%)in the 537 very elderly patients.Aspi-rin 100 mg/d AA-Ag before the switch in aspirin 40 mg/d group was 5.00% ±2.32% and the rate of the patients with low on-treatment platelet aggregation was 71.00%.The rates of melena or occult blood positive,other minimal bleeding,upper gastrointestinal symptoms and a history of gastrointestinal bleeding in 40 mg/d group were higher than those in 100 mg/d group.On a regimen of aspirin 40 mg/d,AA-Ag increased to 11.21% ±4.95%(range:2.12% to 28.84%)with 95.00%of the patients with AA-Ag<20%and the rate of the patients with low on-treatment platelet aggregation was 15.00%.Multiple vari-able analysis revealed that aspirin 40 mg/d AA-Ag was significantly influenced by aspirin 100 mg/d AA-Ag,BMI and platelet counts.The rate of gastrointestinal bleeding decreased from 12.00% to 5.00%, and upper gastrointestinal symptoms decreased from 59.00% to 21.00% after the switch in 40 mg/d group.Conclusion:Switching aspirin dosage from 100 mg/d to 40 mg/d reduces the bleeding events and improves upper gastrointestinal symptoms,thus inhibiting platelet aggregation effectively in very elderly patients.

Myopia is the most important cause of visual impairment in adolescents. However, its etiology is complex. In recent years, a large number of epidemiological studies have been done on risk factors of myopia. Most of these studies is cross- sectional study, not longitudinal cohort study. Overall, the incidence of myopia is the result of the interaction between genetic susceptibility and environmental exposure. This review is about the risk factors for myopia.
