1.A cross sectional survey on the relationship between intolerance to eggs and body mass index in Beijing, China

Objective To explore the prevalence of intolerance to eggs and its relationship with body mass index in Beijing,China.Methods A cross sectional survey which included 12 766 adults from health sciences center of general hospital of the Chinese People's Liberation Army from August,2008 to July,2009 was carried out.Data was entered computer and organized by EpiData 3.0 software for epidemiological analysis.Nonconditional logistic regression model was used for odd ratio (OR) and 95%CIcalculation,and statistics analysis was carried out by SPSS 13.0 software.Results Positive rate of intolerance to eggs in the cohort was 28.5%.Grades of intolerance to eggs were associated with sex(P<0.05 ).Positive rate of intolerance to eggs in females(36.5% ) was higher than those in males (24.9%).Data from multiple regression analysis showed that intolerance to eggs was associated with sex and body mass index.Compared with males,the OR(95%CI) of female was 1.732 (1.590-1.887).Compared with normal weights,the OR (95% CI) of low weights was 1.443 ( 1.018-2.045 ).Conclusion Sex and body mass index might be associated with intolerance to eggs.
2.The relationship between occupational stress and serum glycosylated hemoglobin A1c level.

OBJECTIVETo explore the relationship between occupational stress and serum glycosylated hemoglobin A1c (HbA1c) level.
METHODSA cross-sectional epidemiological study was conducted in 115 dispatchers. The occupational stressors, personality, occupational strain and coping with stress were measured using Occupational Stress Instrument. Serum HbA1c levels were measured by stratigraphic analysis of cation exchange resin.
RESULTSSerum HbA1c level (6.63% ± 1.93%) in the shiftwork group was significantly higher than that (5.73% ± 1.57%) in the daily relay work group (P < 0.05). Spearman correlation analyses showed the positive correlation between type A behavior and serum HbA1c level (r = 0.221), the negative correlation between organizational commitment and serum HbA1c level (r = -0.218). Variance analyses revealed that serum HbA1c levels among high, middle and low job requirement groups and job control groups were significantly different (P < 0.05). There were significant differences of serum HbA1c levels among different type A behavior, internal control, susceptibility of stress and organizational commitment groups (P < 0.05). Stepwise regression analyses indicated that shiftwork affects obviously serum HbAlc level, internal control and job future were the predictors of serum HbA1c level (R2 = 0.082).
CONCLUSIONSerum HbA1c may be a potential biomarker in field investigation of the effects of occupational stress.

3.The improvement of determination method of carbon disulfide in workplace air with gas chromatography.

OBJECTIVEEstablishment of determination method of carbon disulfide in charcoal tube with low toxicity solvents desorption-gas chromatography.
METHODSFour types of solvent with low toxicity are applied respectively as substitution of benzene to desorb the carbon disulfide in samples of charcoal tube. The signal strength and desorption efficiency of the detector are compared by using different solvents.
RESULTSChloroform has been considered as the best alternative solvent of benzene. Carbon disulfide has a good linearity (R = 0.9997) over the concentration of 0 ∼ 54.7 µg/ml, detection limit can reach 0.2 µg/ml. When the sampling volume is 3.0 L, the minimum detectable concentration is 0.07 mg/m(3).
CONCLUSIONWith the use of chloroform, the health hazard to laboratory personnel and environment pollution as well as the costs of experiments are reduced.

4.Effect of silencing HlF-1αby siRNA on the expression of erythropoietin in malignant melanoma of the choroid cell under hypoxia

AlM: To silent hypoxia inducible factor-1α ( HlF-1α) gene in malignant melanoma of the choroid cell by small interference RNA ( siRNA ) and investigate its effect on the expression of matrix metalloproteinase-2 ( MMP-2 ) in the choroid cell line human uveal melanoma cell (OCM-1) in hypoxia environment.METHODS:OCM-1 cells cultured on culture flask were divided into normal group and hypoxia group. Hypoxia group were divided into five groups: simple hypoxic group, and interference group, and negative control group, and positive control group, and liposome group. Normal group cells were cultured on DMEM culture flask with 10% FBS, 100U/mL penicillin, 100μg/mL streptomycin as well as high concentration of glucose. The cells were maintained at 37℃ in a humidified 5% CO2 incubator. Cells in good condition were selected for experiment. For hypoxia group, chemical hypoxia inducer CoCl2 was added into nutrient medium at the concentration of 100μmol/L to simulate hypoxia microenvironment. We designed and synthesised siRNA ( siRNA + negative control+positive control ) , the target sequences of the HlF-1α to transfect hypoxic malignant melanoma of the choroid cell. SiRNA including HlF-1α siRNA, β-actin siRNA and negative control group synthesized in vitro transfected hypoxic OCM - 1 cell through Lipofectamine2000. The expression of HlF-1α, MMP-2 gene and the protein were detected by RT-PCR and Western blot. RESULTS: Compared with the normal group, the expression of HlF-1α mRNA was not obviously changed (P>0. 05), but the expression of HlF-1α protein and MMP- 2 mRNA protein was significantly higher ( P<0. 05) . Compared with the other hypoxia groups,β-actin mRNA expression of positive control group decreased ( P< 0. 05 ) , which proved successful transfection. The expression of HlF-1α mRNA and the expression of its protein and both MMP-2 mRNA and its protein was significantly lower ( P < 0. 05 ). The negative control group, liposome control group had no significant difference in the detection of factors (P>0. 05).CONCLUSlON: Hypoxia status may upregulate the HlF-1α in OCM-1 cells by increasing the expression of protein. Hypoxia can also inactivate MMP-2, resulting in upregulation of MMP-2 RNA and the expression of its protein. The expression of HlF-1α and MMP-2 mRNA can be down-upregulated by transfecting OCM-1 with HlF-1α siRNA.
5.Studies on the Screening of a Cyanide-Degradation Strain and Its Cyanide-Transformation Characteristics*

A bacterial strain DN25, effective on cyanide-degradation, was isolated from contaminated soil and identified as Alcaligenes sp. on the basis of phenotype analysis and 16S rDNA sequence analysis. It showed great tolerance to the cyanide, which can grow in the medium containing 500mg CN -/L. The suitable condition for the cell growth and boitransformation was pH8.0 and 30oC and the transformation rate for 500mg CN - /L could achieve 99% in 10 h. It has also been found that the screened strain had the ability of K 4Fe(CN) 6 transformation with 96% of transformation rate at 12 h for the concentration of 500 mg CN /L.

9.Cyclin E overexpression and centrosome amplification in squamous cell carcinoma of oral cavity.

OBJECTIVETo study the correlation between cyclin E protein overexpression and centrosome amplification in oral squamous cell carcinoma (OSCC).
METHODSFormalin-fixed, paraffin-embedded tissues from 12 normal oral epithelium cases and 46 cases of OSCC were studied. Their centrosome status was analyzed by indirect immunofluorescence double staining with antibodies to centrosome protein gamma-tubulin and cytokeratin. The expression of cyclin E protein was studied by immunohistochemical methods. The correlation between cyclin E protein expression and centrosome amplification in OSCC was statistically analyzed by SPSS 12.0.
RESULTSThirty-seven of the 46 OSCC cases (80.4%) studied showed evidence of centrosome amplification, as signified by enlargement and/or increase in number of centrosomes, while normal oral epithelium possessed centromeres of normal size and number. Positive staining for cyclin E protein was observed in 30 of the 46 OSCC cases (65.2%), while all the normal oral epithelium cases were cyclin E protein-negative. The percentage of centrosome amplification in OSCC with positive cyclin E protein staining (90.0%, 27/30) was higher than that in OSCC with negative cyclin E protein staining (62.5%, 10/16) (chi(2) = 5.014, P < 0.05). Centrosome amplification showed positive correlation with cyclin E protein overexpression (r = 0.330, P < 0.05).
CONCLUSIONUp-regulation of cyclin E protein may represent one of the possible mechanisms for centrosome amplification in OSCC.

10.The relationship between occupational stress and cardiovascular disease risk factor.

OBJECTIVETo explore the relationship between the occupational stress and cardiovascular disease risk factors.
METHODSThe subject was 93 male freight train dispatchers, the occupational stressors, personalities, and occupational strain response were measured by using questionnaires. Blood pressure, heart rate, the concentrations of the cholesterol, triglyceride, and glycosylated hemoglobin in blood serum were also measured.
RESULTSSocial support score were significantly positively related to systolic pressure (r = 0.22) and diastolic pressure (r = 0.30) (P < 0.05), while job satisfaction negatively related to them and concentration of triglyceride (r = -0.37, -0.47 and -0.28 respectively, P < 0.05, P < 0.01), and self-esteem negatively to systolic pressure (r = -0.21, P < 0.05). The differences in diastolic pressure [(78.5 +/- 13.1), (83.6 +/- 8.2), (88.1 +/- 12.3), (85.8 +/- 9.8) mm Hg, P < 0.05] among groups of social support score, body mass index (BMI) (P < 0.01) among groups of job difficulty the differences in systolic pressure [(124.9 +/- 14.4), (129.7 +/- 13.1), (118.4 +/- 16.4), (133.5 +/- 23.1) mm Hg] and diastolic pressure [(85.5 +/- 11.3), (87.0 +/- 9.8), (80.1 +/- 10.1), (88.9 +/- 12.0) mm Hg] and cholesterol level in serum [(4.96 +/- 1.18), (5.39 +/- 0.85), (5.00 +/- 1.15), (4.34 +/- 0.91) mmol/L] among groups of vulnerability to stress (P < 0.05), as well as the difference in systolic pressure and glycosylated hemoglobin level in serum among groups of competition score (P < 0.05) were all statistically significant. Stepwise regression analysis showed that job time demands and negative coping affected the change of cholesterol (R(2) > 0.05); the job relation decision latitude, social support, job difficulty, personality (self-esteem and anxiety trait) and negative coping were the predictors of smoking (R(2) > 0.05). Heart rate was related to home income and competition factor of Type A Behavior (R(2) = 0.06).
CONCLUSIONThe psychosocial stress aspects of work may be related to some cardiovascular risk factors.
