1.Effect of total flavonoids of Chinese hawthorn leaf on expression of NF-?B and its inhibitor in rat liver with non-alcoholic steato-hepatitis

Objective:To study the effect of NF-?B and its inhibitor in rats liver during the development of non-alcoholic steato-hepatitis and observe the effect of total flavonoids of Chinese hawthorn leaf on expressions of NF-?B and its inhibitor,and explore the prevention and treatment mechanism of non-alcoholic steato-hepatitis. Methods:Non-alcoholic steato-hepatitis SD rats model were established by administering a high-fat diet for 12 weeks and were treated with total flavonoids of Chinese hawthorn leaf at dosages of 250mg/kg body weight /d,125 mg/kg body weight /d and with Essentiale at a dosage of 195.4mg /kg body weight /d. Pathological changes of liver tissues were observed with HE,the serum MDA,SOD and TNF-? were measured,the protein and mRNA expressions of NF-?B and I?B in rats liver were detected with RT-PCR and immunohistochemical method. Results:There were severe steatosis inflammatory cell infiltration and necrotic foci in non-alcoholic steato-hepatitis rat. Compared with the control group,contents of the serum MDA and TNF-? in non-alcoholic steato-hepatitis group obviously increased,the activity of SOD decreased,the mRNA and protein expressions of NF-?B P65 and I?B? in liver obviously strengthened. There was a positive relativity of the protein expression of NF-?B p65,I?B? with the contents of serum TNF-?,MDA,but a negative relativity with the activity of serum SOD. Conclusion:Total flavonoids of Chinese hawthorn leaf can obviously lower lipid peroxidation in rats,lower the harm of cytokine to hepatocytes,and regulate the expression of protein and mRNA of NF-?B and I?B. It maybe the important mechanism of total flavonoids of Chinese hawthorn leaf for preventing and treating the development of non-alcoholic steato-hepatitis.
2.Experimental Study on Treatment of Acute Radiation Sickness in Dogs

Seventy dogs were irradiated -with 300. 325, or 350 rad of gamma rays from a 60Co sourse. Half of them were used as controls and the other 35 animals were divided randomly into four groups and were experimentally treated with different regimes.In the first group there were 9 treated, animals and an equal number of control animals 5 all the animals were irradiated with 300 rad. The treatment consisted of a blood transfusion on the 16th day after radiation exposure as well as several antibiotics and other medications. 8 out of the treated animals and one of the controls survived the gamma irradiation.In the second group, ten animals were exposed to 350 rad and 8 to 325 radand they were divided equally into treated and control groups. In spite of early use of blood transfusion and antibiotics, all the treated animals as well as those of the control group died of severe mycotic infections.In the third group, 325 rad was given to the dogs. All 7 dogs of the controls died and two out of the 7 treated dogs survived. The treatment was the same as that of the first group but no blood transfusion was used.In the fourth grou'p?ten animals were irradiated with 350 rad and another ten with 325 rad. They were divided equally into control and treated groups. All the ten controls died. The treated group was given the same treatment as the first group. In addition, tetracycline was given in case there was fever in 'the critical phase of the sickness, and the dosage of tetracycline was doubled whenever fever persisted or recurred. All the treated animals survived.Early blood transfusion was found to be better in its therapeutic effect than late transfusion.
3.Differential Analysis of Two-dimensional Gel Electrophoresis Profiles of Gastric Carcinoma and Paired Normal Gastric Mucosa

Objective To establish two-dimensional electrophoresis profiles with high resolution and reproducibility from human gastric carcinoma and paired normal gastric mucosa tissues and to analyse the differential expression proteins between the two types of the tissues. Methods The total proteins of gastric carcinoma and paired normal gastric mucosa tissues were seperated by immobilized pH gradient-based two-dimensional gel electrophoresis (2-DE).After silver staining, the differential expression proteins were analyzed by using Imaging Master 2D analysis software. Results ⑴Average protein spots were 1049?67, 1097?73 in gastric carcinoma and paired normal gastric mucosa respectively,the matched spots were 835?48, 953?56 in both the two types of tissues respectively, and the average matching rate was 79.6%, 86.7% respectively. ⑵A total of 769?45 protein spots were matched between the electrophoretic maps of 5 gastric carcinoma and paired normal gastric mucosa tissues. The 81 differential protein spots were identified between the two types of the tissues. 17 protein spots were expressed only in the gastric carcinoma and 24 protein spots were expressed only in the normal gastric mucosa. The expression levels of 26 protein spots in the gastric carcinoma were higher than those in the normal gastric mucosa, and the expression levels of 14 protein spots in the gastric carcinoma were lower than those in the normal gastric mucosa. Conclusion In this study, the well-resolved,reproducible 2-DE patterns of gastric carcinoma and paired normal gastric mucosa tissues were established and some differential proteins between the two types of tissues were found. These data will be helpful to further screen specific biomarkers of gastric carcinoma.
4.Finite element analysis of femoral stress changes after total hip arthroplasty

Objective To determine the biomechanical variation in full-length femoral stress pat-tern and periprosthetic femoral stress distribution after implantation either with Charnley Elite cemented prostlesis or Summit proximal porous cementless femoral prosthesis. Methods Three-dimensional finite element models of intact femur, Charnley Elite and Summit femoral prostheses were developed. The stress distributions on the femur and the implants were measured. Applied with hip joint loading and related muscles strength, the stress changes of an intact femur and those implanted with either Charnley Elite or Summit protheses, especially changes in proximal femur, were respectively quantitatively analysed. Results Com-pared with intact femur, there was no change of the full-length femur stress pattern in implantation of both implants, the areas of peak values were all presented at middle and lower regions of the femur, and femoral stress value levels were decreased with both implants. Both prostheses induced significant decrease of stress in the periprosthetic bone tissue, the most serious loss of stresses all appeared at femoral calcar, the stress-shielding rates of Elite and Summit prostheses were 90.8% and 95.3% respectively. The distributions of stress shielding of both prostheses were consistent each other, the Summit proximal porous cementless pros-thesis had more serious stress decrease than the Charnley Elite cemented prosthesis. Conclusion Implan-tation of both implants may cause significant stress shielding in the proximal femur. This may be related with periprosthetic bone loss and aseptic loosening of stems, and can be used to explain the mechanism of post-operative femoral fracture and thigh pain. Both implants designs need further improvement to reduce stress changes in proximal femur.
5.Study of the expression level of FKBP-12 in Alzheimer's disease model mice's brains.

Objective To observe the difference of FKBP-12 expression level between in Alzheimer's disease model mice's brains and in control mice's brains.Methods Taking the Senescence Accelerated Mouse imported from Japan(SAM-P/8)as the Alzheimer's disease model mouse,and SAM-R/1 mouse as control,observe the distribution difference of FKBP-12 between in Alzheimer's disease model mice's brains and in control mice's brains both in mRNA level and in protein level with the methods of RT-PCR and immunohistochemistry staining.The mice were divided into four groups of 4,6,8,10 months old.Results There was no distribution difference of FKBP-12 between in Alzheimer's disease model mice's brains and in control mice's brains both in mRNA level and in protein level.Conclusion FKBP-12 probably has no relativity with the accelerated aging process of the Alzheimer's disease model mice imported from Japan.
6.The Change and the Immpact of IFHL on Oxidative Stress in the Formation of NASH in Rats

Objective To study the change of oxidative stress in the formation of NASH in rats induced by high-fat diet,and observe the preventive and therapeutic effect of TFHL on NASH and explore its mechnism.Methods 110 rats were randomly divided into the normal group(40 rats),the model group(40 rats),the A.Nattermam group(10 rats) and high dosage and low dosage TFHL(10rats each).The normal rats were fed with the standard feedstuff the other rats fed with the high-fat diet.At the time of model-making,the rats in the A.Nattermam were fed the A.Nattermam suspension by daily gavage by 195.4mg/(kg?d),the rats in the high dosage and low dosage TFHL group were fed TFHL by daily gavage in 250、125mg/(kg?d),the rats in the normal and model groups were fed isometrical distilled water by daily gavage.At the ends of 2nd,4th,8th weeks of the experiment,10 rats in the nomal and model were killed respectively,at the end of experiment,all the rest were killed.pathological changes of liver tissues were observed,the levels of serum ALT,AST,the amount of TG,CHOL of livers tissue,the amount of MDA of serum and liver,the activity of OD、GSH-PX、T-AOC were measured.Results 1.the rats were fed with high-fat diet from 2 to 12w,the fatty degeneration of liver tissues were gradually serious,the amounts of TG,CHOL of livers were gradually increased.at 12th w the inflammatory cells in livers infiltrated obviously,the serum ALT,AST were obviously increased,it apeared a progressive process of NASH.2.from 4th w,after feeding with high-fat diet,the amounts of MDA of serum and liver were gradually decreased,the activity of SOD were gradually increased,the amount of T-AOC were gradually decreased as well and,increase or decrease with the development of inflammation.3.the inflammation degree of liver,the amount of liver CHOL and the serum ALT,AST in FMCL groups were lower than those in the corresponding model group;the amounts of MDA of serum and liver were decreased,the activity of SOD,GSH-PX and T-AOC were inceased than those in the corresponding model group,their oxidationresisting activity was better than those in the A.Nattermam group.Conclusions oxidation stress/Lipid Peroxidation plays an important role in the occurrence and development of NASH,TFHL can effectively treat and prevent the development of NASH,obviously ease oxidation stress,decrease Lipid Peroxidation of NASH,markedly enhance oxidationresisting abilily of body,which is the important mechanism of prevention and treatment for NASH.
7.The Relationship between the Bcl-2/Bax Ratio and Drug Resistance in Acute Myelogenous Leukemia

Objective:To investigate Bcl-2 and Bax gene expression in acute myelogenous Leukemia and the relationship between Bcl-2/Bax ratio and drug resistance.Method:Expression of Bcl-2、Bax was detected by immunohistochemical method while drug resistance of AML was detected by cell culture and MTT assay.Results:A high expression of Bcl-2 and a low expression of Bax were detected in AML,respectively,and the ratio between Bcl-2 and Bax was significantly higher than that in normal control (P<0.01). The ratio in those who are resistant to chemotherapy was significantly higher than that in those sensitive to chemotherapy (P<0.05).The responsiveness to chemotherapy of those with a high ratio was poorer than thos with a low ratio (P<0.05).Conclusion:The alteration of Bcl-2 and Bax played certain role in the development of drug resistance in AML,the test of Bcl-2/Bax ratio may have great significance in choice of chemotherapeutic agents and also provide important information for outcome prediction.
8.Efficacy of PFNA and Gamma nail in treatment of intertrochanteric fractures in elderly

Objective to observe the efficacy and application value of PFNA and Gamma in the treatment of intertrochanteric fractures in elderly. Methods 100 patients with intertrochanteric fractures were divided into PFNA group and Gamma group, who received PFNA treatment and Gamma treatment respectively. And the operation status of patients at different ages,postoperative recovery,and complications were observed. Results The operation time,intraoperatve blood soss of PFNA group were lower than those of Gamma group(P<0. 05). There was no significant difference in fracture healing time. The Harris score of the over 75s in PFNA group was higher than that in Gamma group(P<0. 05),and the complications were less than Gamma group(P<0. 05). Conclusion PFNA was suitable for the patients at differ-ent ages with the advantages of more rigid fixation,good anti-rotation,fewer trauma and fewer complications.
9.Research status of conbercept treating age-related macular degeneration

Age-related macular degeneration( AMD) is one of the major reasons of blindness among the elderly in the developed countries. As AMD patients are increasing year by year, AMD has become one of the important topics of ophthalmic research to prevent blindness. Its pathogenesis is not fully understood, but many studies have shown that vascular endothelial growth factor ( VEGF ) plays an important role in the pathogenesis. With the development and application of anti - VEGF drugs, there are a variety of drugs applied to the disease. This article introduces conbercept for the treatment of AMD.
10.Effects of trichostatin A on HDAC8 expression, proliferation and cell cycle of molt-4 cells.

The effects of Trichostatin A (TSA) on histone deacetylase 8 (HDAC8) expression, proliferation and cell cycle arrest in T-lymphoblastic leukemia cell line Molt-4 cells in vitro were investigated. The effect of TSA on the growth of Molt-4 cells was studied by MTT assay. Flow cytometry was used to examine the cell cycle. The expression of HDAC8 was detected by using immunocytochemistry and Western blot. The results showed that proliferation of Molt-4 cells was inhibited in TSA-treated group in a time- and dose-dependent manner. The IC50 of TSA exposures for 24 h and 36 h were 254.3236 and 199.257 microg/L respectively. The cell cycle analysis revealed that Molt-4 was mostly in G0/G1 phase, and after treatment with TSA from 50 to 400 microg/L for 24 h, the percents of G0/G1 cells were decreased and cells were arrested in G2/M phase. Treatment of TSA for 24 h could significantly inhibit the expression of HDAC8 protein in Molt-4 cells (P<0.01). It was concluded that TSA could decrease the expression of HDAC8 in Molt-4 cells, which contributed to the inhibition of proliferation and induction of cell cycle arrest in Molt-4 cells.
