1.Application of ARIMA model to forecasting bacillary dysentery in Jianghan District of Wuhan City

Objective To explore the feasibility of the application of autoregressive integrated moving average(ARIMA) model to forcast the monthly incidence rate of bacillary dysentery in Jianghan District of Wuhan ,and to provide scientific references for prevention and control .Methods SAS 9 .2 software was used to fit the ARIMA model for the monthly incidence rate of bacil‐lary dysentery from 2005 to 2013 .Thereafter ,the fitted model was used to forecast the data from January to June in 2014 ,and was evaluated by comparing the actual incidence .Results The ARIMA(0 ,1 ,1)X(0 ,1 ,1)12 model had a better fitted effect ,it was pre‐dicted that the incidence of bacillary dysentery of Jianghan District of Wuhan City from January to June 2014 would increase slight‐ly ,and the forecast data of bacillary dysentery from January 2005 to December 2013 consisted with the actual situation .Conclusion ARIM A model can be used as short‐term forecasting model to predict the monthly incidence rate of bacillary dysentery in Jianghan District of Wuhan .
2.Progress in the study of small molecule inhibitors of HSP90.

HSP90, which is the biomarker of cell stress and endogenous protective protein, functions as a molecular chaperone. Many client proteins of HSP90, including EGFR, Met, Raf-1, IKK and p53, play important roles in the occurrence and development of tumor. Binding of HSP90 inhibitors triggers the deactivation of HSP90, resulting in client protein degradation, and hence inhibits the tumor growth by blocking multiple targets involved in signaling of tumor proliferation. This review summarizes recent development of small molecule inhibitors bound to N-terminal of HSP90.

5.Effect of Acupotomy on Spasticity after Cerebral Palsy

ObjectiveTo observe the effect of Acupotomy on spastic cerebral palsy. Methods105 cases of spastic cerebral palsy were random divided into group A (35 cases with acupotomy), group B (34 cases with physical therapy) and group C (36 cases with acupotomy and physical therapy). Modified Ashworth Scale (MAS) was used to assess spasticity before and a month after therapy. ResultsEffective rate of group A was 65.71%, group B was 29.41%, group C was 77.78%, which of group B was significantly lower than those of group A of C. ConclusionAcupotomy can reduce muscular tension of spastic cerebral palsy, which seems more effective than physical therapy.
7.Generation of an alpaca derived nanobody recognizing human Her2 antigen

Objective To obtain alpaca single domain antibody targeting Her2.Methods An alpaca was immunized with human recombination Her2 protein mixed with Freund's adjuvant.Total RNA was extracted from the alpaca's blood and was used to synthesize first strand cDNA.Single domain antibody variable region (VHH) gene of the alpaca was amplified by PCR and cloned into pMES4 vector for library construction.After screening, E.coli BL21 (DE3) was transformed with selected clones and was induced with IPTG for the expression of recombinant proteins.The nanobody was purified by nickel ion affinity chromatography column.The affinity of the nanobodies to Her2 was tested.Results After the second round of screening, two antibody clones were selected, H3 and H5.The affinity of H5 was 8.106×10-10mol/L.Histochemistry results showed that H5 could recognize Her2 antigen in breast tumor tissue.Conclusion An Her2 specific nanobody derived from alpaca is obtained through phage display library screening, which can recognize human Her2 antibody in human breast tumor tissue.

This article pointed out that G uang-dong Chinese patent medicines manufacturing implementation patent strategy faced huge opportunities and chal-lenges. It was suggested that the fully understanding, mastering and using of beneficial conditions as well as con-trolling and eliminating the adverse factors will contribute to the better implementation of Chinese patent medicines manufacturing patent strategy in G uangdong province .
9.Changes of auditory evoked potential P50 of sensory gating in model rats with Alzheimer disease

BACKGROUND: Auditory evoked potential P50 (AEP-P50) is a cerebroeilectrophysiological index to directly reflect normal inhibitory function of brain.OBJECTIVE: To observe the changes of AEP-P50 of sensory gating (SG)in model rats with Alzheimer disease (AD).DESIGN: Randomized controlled animal study.SETTING: Neuroscience Institute of Kunming Medical College.MATERIALS: A total of 24 healthy female SD rats, aged 4-6 months,weighing 200-300 g, were randomly divided into experimental group, control group and normal group with 8 in each group. Morris water maze was consisted of round pool and lucite plant. Pool was divided into platform,left, right and bilateral quadrants. METHODS: The experiment was carried out in the Neuroscience Institute of Kunming Medical College from September 2003 to March 2005. ① Experimental group: Bilateral fimbria-fornix was transected to induce AD models; control group: Except fimbria-fornix, cortex and callus were transected; normal group: Rats were not treated with any operation. ② One week after modeling, all rats were tested with Morris water maze which contained localizing navigational ability and spatial seeking ability in order to determine successful models. Each rat was trained 4 times a day for successive 5 days. The duration from putting in pool to finding out platform was regarded as the escape latency (EL) which could reflect localizing navigational ability. Swimming trace of rats which searched platform within 1 minute could reflect spatial seeking ability. ③ AEP-P50 was recorded with "condition (C)-test (T)" auditory double clicks stimuli, additionally,differences of AEP-P50 among the three groups were compared at the same time. C-P50 amplitude, T-P50 amplitude, T/C and absolute value of difference of S2-S1 amplitude were calculated. Measurement data were compared with one-way analysis of variance among groups and with t test intergroup.MAIN OUTCOME MEASURES: ① Morris water maze, I.e., comparison between localizing navigational ability a nd spatial seeking ability; ② comparison of AEP-P50 of rats among three groups.RESULTS: All 24 rats were involved in the fin al analysis. ① results of Morris water maze: With increase of training time, mean latency was shortened. The latency was decreased in the first 3 days and then stabilized gradually. Latency in experimental group was longer than that in normal group and control group (P < 0.05). Swimming traces in normal group and control group were located at platform quadrant, and they were accounted for 45.23% and 39.7% of total quadrant, respectively. There was significant difference as compared with other quadrants (P < 0.01). Swimming traces of rats in experimental group were accounted for 28.31%, 29.84%,20.47% and 21.38% in platform, right, bilateral and left quadrants, respectively; however, there was not significant difference (P > 0.05). Swimming trace was randomly located in four quadrants. ② Results of AEP-P50 of SG: C-P50 amplitude and absolute value of difference of S2-S1 amplitude were (21.00±2.85), (15.26±4.07) μV in control group and (17.04±5.32), (10.85 ±4.24) μV in normal group, which were obviously higher than those in experimental group [(9.67±3.77), (2.89±2.61) μV, P < 0.01].T-P50 amplitude and C-P50 amplitude were 0.25±0.18 in control group and 0.39±0.16 in normal group, which were lower than those in experimental group (0.92±0.41, P < 0.01).CONCLUSION: ① Transection of bilateral fimbria-fornix can be used to establish AD animal models successfully. ② AD model rats whose bilateral fimbria-fornix is transected have deficiency of SG.
10.Hemodynamic Parameter Changes in Pulmonary Arterial Hypertension Treated by Transplantation of Bone Marrow Mesenchymal Stem Cells in Experimental Rats

Objective To explore hemodynamic parameter changes in pulmonary arterial hypertension(PAH)treated by transplantation of bone marrow mesenchymal stem cells(MMSCs)in the experimental rats.Methods MMSCs cells were collected from bone marrow of Sprague-Dawleye(SD)rat's femoral and tibial bones,cultured and passaged in vitro,then stained by Hoechst 33342 fluorescence dye stuff.Ninety healthy male SD rats were randomly divided into 3 groups(n=30):normal control group(group N),MMSCs transplanted group(group M),PAH model group(group H).The rats in the two latter groups were given a single subcutaneous crotaline(50 mg/kg)to establish the model of PAH.The rats of group N were injected respectively a single subcutaneous 9 g/L saline water(6 mL/kg).After 21 days,5?109 L-1 MMSCs cultured in 1 mL phosphate-buffered saline were infused into the rats respectively in group M by sublingual vein and 1 mL L-DMEM was given in group H.The indexalso of right ventricle systolic pressure(RVSP),right ventricle hypertrophy index,arterial blood gas analysis and the changes of small pulmonary blood vessel were observed after 28 days.Results The administration of MMSCs 28 days after PAH nearly completely prevented the increase in RVSP with PAH alone [(32.20?2.32)mmHg vs(48.30?1.56)mmHg P
