1.Study on the diagnostic value of serum miRNA-182 in colorectal cancer

Objective To explore the diagnostic value of serum miRNA-182 in colorectal cancer(CRC).Methods A total of 53 CRC patients were enrolled in this study,serum miRNA-182 was detected by quantitative real time polymerase chain reaction(qRT-PCR).The results were compared with those of patients with benign intestinal diseases and healthy controls.Results The content of serum miRNA-182 in CRC patients was significantly higher than those in patients with benign intestinal diseases and healthy group(P<0.05).The ROC curve showed that the area under the curve of serum miRNA-182 was 0.846,the confidence interval was 0.768-0.925,the diagnostic sensitivity was 77.1%,and the specificity was 81.1%.Serum miRNA-182 levels were correlated with TNM stage(P=0.011) and distant metastasis(P=0.002) in patients with CRC,but no significant correlation with other clinical parameters(P>0.05).Serum miRNA-182 level in CRC patients after surgery was significantly lower than that before operation(P<0.01),and which was significantly higher in patients with recurrence/metastasis after surgery(P=0.019).Conclusion Serum miRNA-182 level increased significantly in CRC patients,and which was closely related to the disease and malignant progression,serum miRNA-182 could be used as a potential marker for clinical diagnosis and disease evaluation of CRC.

Due to its effectiveness and specificity,RNA interference (RNAi) technology is widely used in the treatment of tumor,viral infectious and metabolic diseases.Noninvasively tracing the delivery of therapeutic RNA molecules in vivo and evaluating the therapeutic effects are important to promote the clinical application of RNAi technology.Molecular imaging allows noninvasive visualization of biological processes at the cellular and molecular level in vivo.It can be used to monitor the biodistribution of therapeutic RNA molecules and to assess the effects of the RNAi.This article reviews the advance of molecular imaging in monitoring the delivery of RNAi molecules and evaluating the therapeutic effects of RNAi.
3.Clinical Significance of Serum Lactate Dehydrogenase and Its Isoenzymes in the Retreated Patients with Multiple Myeloma

Objective To investigate the clinical significance of serum lactate dehydrogenase (LDH) and its isoenzymes,M or H subunits in the retreated patients with multiple myeloma(MM).Methods 141 medical records of retreated patients with multiple myeloma were collected,and the relationship of LDH and its isoenzymes,subunits with DS staging system,ISS staging system,creatinine,bone marrow plasma cell percentage,immunoglobulin and microglobin was analyzed.Results Serum total LDH activity was not significantly related to DS staging system and ISS staging system.LDH1 percentages was negatively correlated with ISS staging system (P<0.01),and LDH4 percentages positively associated with ISS staging system (P<0.05).LDH activity,absolute value of LDH1 ,LDH2,LDH3,LDH4,and H subunits were positively linked to serum creatinine in myeloma kidney disease patients with renal inadequacy (P<0.05).Both percentage and absolute value of LDH5 were negatively correlated with bone marrow plasma cell percentage (P<0.01 and P<0.05 respectively).Conclusion The detection of serum LDH isoenzyme is more valuable than total LDH activity in retreated patients with multiple myeloma.
4.Surgical treatment of the complicated fractures of the tibial plateau

Objective To investigate the therapeutic effect of the complicated fracture of tibial plateau treated by open reduction and internal fixation. Methods Surgical treatment in 68 cases with complicated fractures of the tibial plateau were analyzed retrospectively, T type plate, L type buttress plate, double plates, anatomical plate,locking plate and less invasive stabization system were adopted for different fractures,iliac or artificial bone grafted in the reconstruction of bone defect after the depression replaced. The time of operation and the effect of functional exercise in the postoperative patients to recover the function of the knees were analyzed at the same time. Results 52 patients of 68 cases were followed-up. Based on Rasmussen rating system,the recovery of knee joint function were excellent in 42cases(67%) ,good in 12 cases(19%) ,fair in 6 cases(10%) and poor in 2 cases(4%) ,the excellent-and-good rate was 86%. Conclusion Surgical treatment of the complicated fractures of the tibial plateau depended on the kind of fracture, therapeutic principles by stages. Moreover, bone grafte after the depression replaced in bone defect was neeassary,good internal fixtion oould meet functional exercise,soft tissue injuries like ligament and meniscus should be well treated,follow the principles of early exercise and delay weightbearing.
5.Prokaryotic Expression of eBCMA-Fc Fusion Protein, a Soluble BLyS Receptor, with Computer-aided Molecular Design in E. coli

B cell maturation antigen (BCMA) is a receptor of B lymphocyte stimulator (BLyS). Human IgG1Fc fusion proteins with the extracellular domain of BCMA(eBCMA), also called decoy receptors, have beenused as a potential BLyS antagonists to block BLyS activities. In order to design novel BLyS antagonistpeptides, computer-aided homologue modeling was used to construct an eBCMA-Fc fusion protein based on thecrystal structures of BCMA and Fc fragmant. To ensure the activity of eBCMA not to be interfered by Fcfusion, the root mean square distance (RMSD) for eBCMA and Fc were calculated to be 0.036 nm and 0.064nm, respectively, based on molecular docking modeling. An eBCMA-Fc fusion gene was constructed andintroduced into E. coli for expression. As expected, the purified 36 kD eBCMA-Fc fusion protein was able tobind BLyS in vitro at a dosage-dependent manner and demonstrated an anti-proliferative activity induced byBLyS in Daudi cells. The results have provided useful information on the evaluation of computer modeling andthe in vitro biological activity for the design of potential BLyS antagonist peptides.

7.QQ group follow up of nursing education on patients with nasopharyngeal carcinoma after radiotherapy

Objective To investigate the role of the QQ group follow up of nursing education on patients with nasopharyngeal carcinoma after radiotherapy in patients after they discharged. Methods Thirty patients with nasopharyngeal carcinoma hospitalized from January to December 2013 in our department were set as control group and another 30 patients with NPC hospitalized from January to December 2014 in our department were assined to in the experiment group. Patients of two group received health education before discharge . The controls were followed up on phone and the experimental group received QQ group follow up of education for one year, besides phone follow-up. The two groups were compared in terms of incidence of adverse reactions one year after discharge. Results One year after discharge , the incidences of side effects in the experiment group were lowered than those in the control group , and the differences were siguificant difference ( all P < 0 . 05 ) . Conclusion The QQ group follow up of nursing education can reduce the incidence of adverse reactions by radiotherapy after discharge.
8.Effects of nimodipine on reducing the complications after microvascular decompression in patients with hemifacial spasm

Objective To investigate whether nimodipine could reduce complications after microvascular decompression in patients with hemifacial spasm. Methods 220 patients with hemifacial spasm , admitted into our hospital from May 2011 to Oct. 2013 , were all treated by microvascular decompression. They were divided randomly into two groups. Patients in the treatment group were treated with nimotop 50 mg/d after the operation for the first 3 days, then was orally taken 30 mg/d for a week. Patients in the control group were treated with the same after the operation without nimodipine. The postoperative complications including hearing deficit and peripheral facial paralysis were closely observed within three months after operation. Results The total effective rate was 100% in both groups. The incidence rate of peripheral facial paralysis and hearing deficit in treatment group was significant lower than that of the control group (P < 0.05). Conclusion Nimodipine has significant effect on reducing the incidence rate of peripheral facial paralysis and hearing deficit after microvascular decompression in patients with hemifacial spasm.
9.STUDY ON THE ANTI-ANGIOGENIC ROLE OF AGARO-OLIGOSACCHARIDE

Objective: To study the inhibition effect of agaro-oligosaccharide (AOS) on neovascularization and its mechamism. Method: The anti-angiogenic effect in vivo was evaluated on chicken chorioallantoic membrane (CAM) model. Cytotoxic activity of AOS was demonstrated by inhibition of several human cell lines by MTT assay. The apoptosis of HUVECs induced by AOS was examined by Hoechst staining assay and quantified by flowcytometry. Results: In CAM, AOS caused dose-dependent decrease on the vascular density and adversely affected capillary plexus formation. Different cytotoxic sensitivities were observed for AOS towards several kinds of cell lines, and HUVECs were more sensitive. Moreover, the growth inhibitory activity was correlated with induction of apoptosis. Flowcytometric analysis also revealed that AOS arrested the cell cycle progression at S phase. Conclusion: Agaro-oligosaccharide possesses the anti-angiogenic effects, which are associated with apoptosis induction of endothelial cells.
10.Hormone Therapy on Patients with Amenorrhea Caused by Tripterygium Wilfordii Polyglycoside

Objective To discuss the clinical characteristics and treatment of amenorrhea caused by Tripterygium Wilfordii polyglycoside (TWP).Methods The sexual hormone levels of 25 outpatients of The First Affliated Hospital Of Wenzhou Medical College who suffered from secondary amenorrhea after the use of TWP were examined before and after the hormone therapy.In addition,the clinical presentations were observed.Results (1)The E2 level went down lower than 73.2pmol/L while FSH went up higher than 40U/L in all the 25 patients.Among them,24 patients were confirmed of the diagnosis of premature ovarian failure.⑵HT is an effective way for 20 patients.The total efficiency of the therapy was 80%.⑶The E2 secreted by ovary was higher (63.19?10.81)pmol/l vs. (365.45?161.60) pmol/L and FSH(110.72?21.52)U/L vs. 11.46?17.70)U/Lsecreted by pituitary was lower after therapy than before.⑷After the second period of therapy, the clinical perimenopausal symptoms of all the patients were improved to different degrees.Conclusion TWP may result in the disorders of ovarian functions, but and HT in time may be an effective therapy.
